Abstract

As in many other small mammal groups from the Afrotropics, the number of species recognized within the ge-nus Hylomyscus has increased considerably over the past dozen years. The last comprehensive review (2005) of the genus recognized eight species. Since that time, nine additional species have been elevated from synonymy (n = 4) or described as new (n = 5). Here we describe four additional new species supported by morphological and molecular evidence, all collected by the late William Stanley. Two of the new taxa are sympatric and come from the poorly known left bank (di-rection source to mouth) of the Congo River. One of these (Hylomyscus pygmaeus sp. nov.) is easily recognized, as it is tiny and significantly smaller than any known species of the genus; the second new species (Hylomyscus thornesmithae sp. nov.) is also small, and syntopic with the first. The third new species (Hylomyscus stanleyi sp. nov.), from the SW corner of Tanzania, is quite large and had been previously included within the hypodigm of Hylomyscus anselli following its recognition from within the synonymy of Hylomyscus denniae. The fourth species (Hylomyscus mpungamachagorum sp. nov.) is from Mahale Mountains National Park, western Tanzania. Our study reveals a much higher species diversity of the genus than previously known, providing insights into additional Afrotropical and Afromontane centers of endemism that require further exploration.

Abstract

We examined den-site selection patterns for western spotted skunks (Spilogale gracilis) and striped skunks (Mephitis mephitis) co-occurring in mesquite brushland of Tom Green County, Texas. Sixteen S. gracilis and 14 M. mephitis were radiocollared to locate den sites from October 2001 through June 2003. Characteristics of dens were assessed using 23 habitat variables. S. gracilis favored den sites with dense mesquite (Prosopis glandulosa), shrubs, and cacti (Opuntia), whereas M. mephitis used these areas but also more open habitats. Den habitats of S. gracilis were more uniform than were those of M. mephitis, implying that S. gracilis is more specialized in den-site selection. Both species used multiple den sites, both above and below ground, rather than a primary den. Aboveground denning occurred year-round for both species. For S. gracilis, 56% (n = 60) of dens were above ground, but denning was more common above ground in the spring and summer. Aboveground denning was more common than belowground denning [or M. mephitis in all seasons, representing 64% (n = 50) of all dens. Den habitat analysis suggests that S. gracilis, unlike M. mephitis, may be limited to areas with sufficient cover, including prickly pear cactus, for denning. Abstract

Abstract Background All viruses in the family Bunyaviridae possess a tripartite genome, consisting of a small, a medium, and a large RNA segment. Bunyaviruses therefore possess considerable evolutionary potential, attributable to both intramolecular changes and to genome segment reassortment. Hantaviruses (family Bunyaviridae, genus Hantavirus) are known to cause human hemorrhagic fever with renal syndrome or hantavirus pulmonary syndrome. The primary reservoir host of Sin Nombre virus is the deer mouse (Peromyscus maniculatus), which is widely distributed in North America. We investigated the prevalence of intramolecular changes and of genomic reassortment among Sin Nombre viruses detected in deer mice in three western states. Methods Portions of the Sin Nombre virus small (S) and medium (M) RNA segments were amplified by RT-PCR from kidney, lung, liver and spleen of seropositive peromyscine rodents, principally deer mice, collected in Colorado, New Mexico and Montana from 1995 to 2007. Both a 142 nucleotide (nt) amplicon of the M segment, encoding a portion of the G2 transmembrane glycoprotein, and a 751 nt amplicon of the S segment, encoding part of the nucleocapsid protein, were cloned and sequenced from 19 deer mice and from one brush mouse (P. boylii), S RNA but not M RNA from one deer mouse, and M RNA but not S RNA from another deer mouse. Results Two of 20 viruses were found to be reassortants. Within virus sequences from different rodents, the average rate of synonymous substitutions among all pair-wise comparisons (πs) was 0.378 in the M segment and 0.312 in the S segment sequences. The replacement substitution rate (πa) was 7.0 × 10-4 in the M segment and 17.3 × 10-4 in the S segment sequences. The low πa relative to πs suggests strong purifying selection and this was confirmed by a Fu and Li analysis. The absolute rate of molecular evolution of the M segment was 6.76 × 10-3 substitutions/site/year. The absolute age of the M segment tree was estimated to be 37 years. In the S segment the rate of molecular evolution was 1.93 × 10-3 substitutions/site/year and the absolute age of the tree was 106 years. Assuming that mice were infected with a single Sin Nombre virus genotype, phylogenetic analyses revealed that 10% (2/20) of viruses were reassortants, similar to the 14% (6/43) found in a previous report. Conclusion Age estimates from both segments suggest that Sin Nombre virus has evolved within the past 37–106 years. The rates of evolutionary changes reported here suggest that Sin Nombre virus M and S segment reassortment occurs frequently in nature.

Abstract

During 2012, 2013 and 2015, we collected small mammals within 25 km of the town of Boende in Tshuapa Province, the Democratic Republic of the Congo. The prevalence of monkeypox virus (MPXV) in this area is unknown; however, cases of human infection were previously confirmed near these collection sites. Samples were collected from 353 mammals (rodents, shrews, pangolins, elephant shrews, a potamogale, and a hyrax). Some rodents and shrews were captured from houses where human monkeypox cases have recently been identified, but most were trapped in forests and agricultural areas near villages. Real-time PCR and ELISA were used to assess evidence of MPXV infection and other Orthopoxvirus (OPXV) infections in these small mammals. Seven (2.0%) of these animal samples were found to be anti-orthopoxvirus immunoglobulin G (IgG) antibody positive (six rodents: two Funisciurus spp.; one Graphiurus lorraineus; one Cricetomys emini; one Heliosciurus sp.; one Oenomys hypoxanthus, and one elephant shrew Petrodromus tetradactylus); no individuals were found positive in PCR-based assays. These results suggest that a variety of animals can be infected with OPXVs, and that epidemiology studies and educational campaigns should focus on animals that people are regularly contacting, including larger rodents used as protein sources.
