Background and Aim: Coccidiosis, caused by protozoan parasites of the genus Eimeria, is a significant concern in poultry farming, leading to substantial economic losses worldwide. In Pakistan, poultry is a major component of the agricultural sector, with both broiler and egg-laying chickens playing crucial roles in meeting the country’s protein needs. Despite the importance of the poultry industry, there is limited data on prevalence and species distribution of Eimeria in different types of chickens in District Swabi, Khyber Pakhtunkhwa, Pakistan. This study aims to estimate the prevalence and determine the distribution of Eimeria species in broiler and egg-laying chickens in this region. Materials and Methods: Nine hundred fecal samples were collected from broiler (380) and egg-laying domestic chickens (520) in District Swabi, Pakistan. Microscopic analysis was used to identify Eimeria parasites in all samples. After microscopic examination for positive identification, Eimeria species were determined using polymerase chain reaction (PCR) assays. Results: Microscopic examination identified Eimeria oocysts in 44.4% (400/900) of the samples. Eimeria parasite infection significantly varied based on chicken type, age, and gender (p < 0.05). The study found that broiler chickens (52.63%, 235/450), young chickens (4–6 weeks) (55.5%, 285/500), and females (52.2%, 200/380) were more infected with Eimeria spp. than egg-laying domestic chickens (38.5%, 200/520), adults (above 6 weeks) (28.8%), and males (36.7%, 165/450). PCR indicated a distribution rate of 42.5% (170/400) Eimeria tenella, 26.25% (105/400) Eimeria acervulina, 20% (80/400) Eimeria maxima, and 11.25% (45/400) Eimeria mitis. None of Eimeria necatrix, Eimeria brunetti, or Eimeria praecox was found in the study. Conclusion: This study underlines the essential requirement for targeted interventions due to the prevalence and predominance of E. tenella among identified Eimeria species. Future research should focus on refined sampling strategies and investigate the clinical significance of these parasites for effective disease management in the local poultry industry.

We have developed an in vitro model for the evaluation of potential anticoccidial properties of novel compounds aimed to control chicken coccidiosis, a costly disease for the poultry industry. This disease is caused by protozoan parasites of the genus Eimeria (Apicomplexa), and it is mainly controlled by chemoprophylaxis with ionophores and chemical anticoccidials; however, there is an overall agreement about the limitation of these traditional drugs and the need to improve current methods of control. Anticoccidial activities of novel compounds is currently evaluated by expensive experiments that involve large numbers of chickens. The use of our in vitro model for the pre-screening of essential oils led to a reduction of 67% of the chickens used in the in vivo trials for validation. Eimeria parasites can only complete their life cycle in their animal host, therefore chickens are required for their propagation as they cannot be propagated in vitro. In this study, we describe how further optimisation of this in vitro model by miniaturisation can have an additional impact in reduction of the number of chickens used for the generation of parasite stocks for provision of the in vitro model. We have estimated that the use of one chicken could support the evaluation of 10 compounds with a 96-well plate format versus only two compounds with a 24-well plate format, which means an 80% reduction in chicken use. In this study we have proved that the miniaturisation into a 96-well plate format perfectly mimics the invasion and replication observed before in the 24-well plate format. In addition, the 96-well plate format has allowed the simultaneous pre-screening of higher numbers of anticoccidial drugs at different concentrations following streamlined protocols in a more cost-effective way, factors that are beneficial for a wider uptake of the model by other researchers investigating anticoccidial compounds.

ABSTRACT: The black grouse (Tetrao tetrix) is an endangered species facing challenges in breeding and reintroduction programs, including parasitic infestations. This study aimed to assess natural infestations by various Eimeria species and infestation dynamics in female, male, and young black grouse kept in a stationary aviary. Faecal samples were collected from adult grouses between April and the time of chicks’ hatching and rearing (September). Faecal samples from young birds were collected from the hatching for a period of 1 year. The prevalence of Eimeria spp. was determined by a qualitative method (Fulleborn's flotation) and a quantitative method (McMaster's method with Raynaud's modification). The following Eimeria species were identified: E. lyruri, E. nadsoni and E. nonbrumpti. The average percentages of Eimeria spp. in the cock were 80.52%, 9.27%, and 10.21%, respectively; in the hen, they were 86.19%, 9.28%, and 4.53%, respectively; in the young black grouses they were 84.60%, 9.34% and 6.06%, respectively. The highest E. lyruri infestation was observed in the cock in June (144227 OPG) and July (129365 OPG). In the hen, the infestation intensity increased in May (304302 OPG) and then decreased in June (39583 OPG). Furthermore, an additional increase was observed in July (216533 OPG). Two increases in infestation intensity were also observed in young birds, with peaks in January (91387 OPG) and July (126178 OPG). A positive strong correlation was identified between Eimeria spp. in the cock and the young birds. A statistically significant positive correlation was identified in the hen between E. lyruri and E. nadsoni. No correlation was demonstrated between the infestation intensity and the age of the birds or season of the year in all the grouses under study. Despite some attempts, a comprehensive approach to the issue of coccidiosis in the black grouse as a disease that may affect the success of reintroduction has yet to be established. It seems crucial to monitor the level of Eimeria spp. invasion, and the proposed faecal sampling scheme is an important tool for achieving this goal.

To treat and control parasitic infections, traditional medical remedies using plant products are utilized as antiparasitic agents rather than standard synthetic chemicals due to drug resistance. Myrrh, a resinous exudate of Commiphora myrrha (Burseraceae), is a powerful antioxidant with a variety of medicinal uses. This study aimed to investigate the effect of the myrrh methanolic extract (MyE) of three concentrations (100, 50, and 25 mg/ml) on the sporulation of oocysts and as an anthelminthic effector via in vitro study. Characterization of the plant was done by Fourier-transform infrared spectroscopy (FT-IR). The earthworm, Eisenia fetida, is used as a model worm to evaluate the anthelminthic activity of MyE. Eimeria labbeana-like oocysts are used as a model protozoan parasite in anticoccidial assays. The sporulation and inhibition (%) of E. labbeana-like were assessed by MyE compared to other chemical substances. FT-IR revealed the presence of twelve active compounds. Our results showed that paralysis and death of earthworms at MyE (100 mg/ml) were 7.88 ± 0.37 and 9.24 ± 0.60 min, respectively, which is more potency when compared to mebendazole (reference drug). In all treated worms, microscopic examinations revealed obvious surface architecture abnormality. This study shows that MyE affects oocysts sporulation in a dose-dependent manner. At 24 and 36 hr, a high concentration of MyE (100 mg/ml) inhibits sporulation by 90.95 and 87.17 %. At 36 hr, other concentrations of MyE (50 and 25 mg/ml), as well as amprolium, DettolTM, and phenol inhibits oocyst sporulation by 40.17 %, 29.34 %, 45.09 %, 85.11 %, and 61.58 %, respectively. According to our research, the MyE extract had powerful anthelmintic and anticoccidial properties.

Coccidiosis, caused by apicomplexan Eimeria species, is a protozoan disease that affects various species of wild and domestic animals. However, data available on Eimeria diversity in ruminants in Saudi Arabia is meagre. Therefore, this study was designed to investigate some eimerian parasites infecting sheep (Sawakni and Harrie breeds) using microscopy and molecular methods for the first time in Saudi Arabia. Twenty‐four fecal samples were collected from sheep farms. Based on the floatation technique, eimerian oocysts were observed in 8 of the 24 (33.33%) fecal samples. The coccidian‐positive samples were subjected to fecal culture in a shallow layer of 2.5% potassium dichromate (K2Cr2O7). Detected eimerian oocysts were described micromorphometrically as the basis for traditional oocyst identification. Morphologically, the sporulated oocysts were similar to those of sheep eimerian parasies; Eimeria faurei and Eimeria crandallis. PCR products from the two eimerian species detected from Sawakni and Harrie breeds were sequenced and were found to be distinct from each other with mutations at five positions. One of them clustered with E. crandallis with 99.8%–100% identity with sequences available in GenBank. E. crandallis was obtained from two Sawakni sheep and two Harrie sheep. The other sequences grouped with E. faurei with 99.8% identity with the only sequences available in GenBank. E. crandallis was detected from both Sawakni and Harrie breeds whereas E. faurei was detected only from Sawakni sheep. The findings of this study have implications for the importance of morphometric identification with advanced molecular tools to confirm the identities of sheep Eimeria species and to address the taxonomic study of this eimeriid parasite at the species level. Poultry coccidiosis is an intestinal infection caused by an intracellular parasitic protozoan of the genus Eimeria. Coccidia-induced gastrointestinal inflammation results in large economic losses, hence finding methods to decrease its prevalence is critical for industry participants and academic researchers. It has been demonstrated that coccidiosis can be effectively controlled and managed by employing anticoccidial chemical compounds. However, as a result of their extensive use, anticoccidial drug resistance in Eimeria species has raised concerns. Phytochemical/herbal medicines (Artemisia annua, Bidens pilosa, and garlic) seem to be a promising strategy for preventing coccidiosis, in accordance with the "anticoccidial chemical-free" standards. The impact of herbal supplements on poultry coccidiosis is based on the reduction of oocyst output by preventing the proliferation and growth of Eimeria species in chicken gastrointestinal tissues and lowering intestinal permeability via increased epithelial turnover. This review provides a thorough up-to-date assessment of the state of the art and technologies in the prevention and treatment of coccidiosis in chickens, including the most used phytochemical medications, their mode of action, and the applicable legal framework in the European Union. Enteric protozoa are major causes of economic losses in dromedary camels due to clinical and subclinical diseases. The study investigated the prevalence of enteric protozoal parasites in dromedary camels using parasitological and molecular techniques. Besides, the molecular characterization of selected enteric protozoa was performed by nucleotide sequencing and phylogenetic analysis. One hundred and twenty-one fecal samples were collected from diseased (49) and apparently healthy (72) dromedary camels during December 2020 - November 2021. Overall, 57.02% of the fecal samples were positive for enteric protozoa with the B. coli, Eimeria spp., Giardia spp., Cryptosporidium spp. were detected in 32.23%, 8.26%, 2.48%,3.30% fecal samples, respectively, while 10.74% samples had mixed infection. While the PCR technique revealed the overall prevalence of 69.56%, 13.04% and 10.14% for the B. coli, Giardia spp. and Cryptosporidium spp., respectively in the study areas. Phylogenetic analysis based on the partial nucleotide sequences of the small subunit ribosomal RNA, B beta giardin (bg) and Cryptosporidium parvum precursor of oocyst wall protein (COWP) genes of B. coli, Giardia spp. and Cryptosporidium spp., respectively revealed that the enteric protozoa from camels had high genetic similarities with strains detected from other animals and humans in Egypt and different parts of the world. Turkey coccidiosis is caused by protozoan parasites of genus Eimeria. Eimeria species are ubiquitous in intensive turkey production facilities. Seven Eimeria species have been characterised and documented in turkeys. Among the seven species, E. adenoeides, E. meleagrimitis and E. gallopavonis are considered as predominant and highly pathogenic strains of turkeys. Coccidiosis causes substantial economic losses to the turkey industry by affecting intestinal health and production performance. Background and Aim: In Indonesia, Madura cattle are native breeds that are expected to contribute to the improvement of regional meat self-sufficiency. Eimeria spp. are protozoans that are commonly found in ruminants. This study aimed to identify the occurrence and diversity of Eimeria spp. in Madura cattle. Materials and Methods: In this study, fresh fecal samples were collected from 100 cattle in Kamal Subdistrict, Bangkalan District, Madura Island, Indonesia. Morphological detection was performed using a light microscope, and molecular identification was performed using a polymerase chain reaction. DNA amplification was conducted using various species-specific primers for Eimeria bovis, Eimeria zuernii, Eimeria auburnensis, Eimeria alabamensis, Eimeria ellipsoidalis, and Eimeria cylindrica. Results: The results obtained 21% (21/100) of Eimeria spp. based on morphological detection. A total of 15 positive samples with 500–25,000/mL oocysts were selected for DNA extraction and amplification, resulting in 12 positive samples. Four Eimeria spp. were obtained based on molecular identification: E. bovis, E. zuernii, E. auburnensis, and E. cylindrica. Conclusion: Four species of Eimeria namely E. bovis, E. zuernii, E. auburnensis, and E. cylindrica were identified from fecal sample of Madura cattle using PCR method in this study. Further comprehensive studies are required to investigate the pathogenicity of Eimeria spp. in Madura cattle. Therefore, improved and integrated management practices should be strengthened by local governments to prevent pathogenic diseases and increase national livestock productivity in Indonesia.

Abstract With a worldwide distribution, Eimeria spp. could result in serious economic losses to the poultry industry. Due to drug resistance and residues, there are no ideal drugs and vaccines against Eimeria spp. in food animals. In the current study, a bioinformatics approach was employed to design a multiepitope antigen, named NSLC protein, encoding antigenic epitopes of E. necatrix NA4, E. tenella SAG1, E. acervulina LDH, and E. maxima CDPK. Thereafter, the protective immunity of NSLC protein along with five adjuvants and two nanospheres in laying chickens was evaluated. Based on the humoral immunity, cellular immunity, oocyst burden, and the coefficient of growth, the optimum adjuvant was evaluated. Furthermore, the optimum immune route and dosage were also investigated according to the oocyst burden and coefficient of growth. Accompanied by promoted secretion of antibodies and enhanced CD4+ and CD8+ T lymphocyte proportions, NSLC proteins entrapped in PLGA nanospheres were more effective in stimulating protective immunity than other adjuvants or nanospheres, indicating that PLGA nanospheres were the optimum adjuvant for NSLC protein. In addition, a significantly inhibited oocyst burden and growth coefficient promotion were also observed in animals vaccinated with NSLC proteins entrapped in PLGA nanospheres, indicating that the optimum adjuvant for NSLC proteins was PLGA nanospheres. The results also suggested that the intramucosal route with PLGA nanospheres containing 300 μg of NSLC protein was the most efficient approach to induce protective immunity against the four Eimeria species. Collectively, PLGA nanospheres loaded with NSLC antigens are potential vaccine candidates against avian coccidiosis.

SUMMARY: Avian coccidiosis is an intestinal infection caused by a parasitic intracellular protozoan of Eimeria species. The effect of coccidian intestinal infection in birds renders significant economic losses in production, and therefore, it is crucial for industry players and researchers to find solutions to its occurrence. The use of synthetic additives and anticoccidial supplements has been established to be effective in preventing and controlling coccidiosis. However, concerns about anticoccidial drug resistance in Eimeria species due to frequent use have been reported. Alternatively, the use of attenuated live vaccines has been successful but it comes with challenges because of poor farm management. Garlic and its bioactive compounds have been found to exhibit anti-inflammatory and antioxidant properties. The anticoccidial potential of garlic is linked to its immunostimulatory activity. Garlic and its derivatives inhibit the sporulation of oocysts in vitro. Supplementation of garlic in coccidiosis-infected broilers improves weight gain and feed efficiency, and reduces fecal oocysts output, lesion score, and clinical signs postinfection. Thus, this review aims to highlight the clinical signs of coccidiosis, the techniques involved in oocysts determination, and the anticoccidial potential of garlic in improving the health of broiler chickens. Future studies can bridge the knowledge gap on the mode of action of the bioactive compounds of garlic against coccidiosis and the Eimeria species-specific gene and immune-protective antigens that they inhibit and improve in infected birds.

Coccidiosis is an intestinal infection caused by Eimeria spp. that results in economic losses owing to morbidity and mortality in young buffalo calves. This study aimed for molecular diagnosis and phylogenetic analysis of Eimeria spp. in buffaloes of Meghalaya's sub-tropical mountainous terrain. Fresh buffaloes' fecal samples were collected from buffalo farms of Umling, Umsning and Bhoirymbong blocks, Ri Bhoi, Meghalaya and screened for Eimeria oocysts using flotation and modified McMaster methods. Fecal sample examination revealed 27.44 % (87/317) infection in buffaloes. Age wise, 64.44 % (29/45), 25.35 % (36/142) and 16.92 % (22/130) infections were recorded in <6 months, 6 months to 1 year and 1-2 year old buffaloes, respectively. Morphological characterization of Eimeria spp. revealed E. bovis (21.83 %), E. bareillyi (18.39 %), E. zuernii (11.49 %), E. ellipsoidalis (3.44 %) and mixed infection (44.82 %). Amplification of ITS-1 gene confirmed Eimeria spp. (410 bp), E. bovis (238 bp) and E. zuernii (344 bp). Phylogenetic analysis of E. bovis Umiam isolate revealed that these were closely related to the E. bovis isolate from South Korea (MH245198.1), and Turkey (KU351711.1) and distantly related to the isolates from Jammu and Kashmir (OQ103422.1) and Uttar Pradesh, Mathura (OK486542.1). E. zuernii isolate from Umiam, Meghalaya was observed to be phylogenetically close to the isolates from South Korea (MH245202.1), Japan (LC171339.1) and Turkey (KU351715.1), whereas phylogenetic divergence was observed between, E. zuernii isolate from Umiam, Meghalaya with isolates of Andhra Pradesh, Tirupati (MN601278.1) and Jammu and Kashmir (OQ103424.1). The current in vitro study aimed to investigate the effects of a processed sugarcane extract on the viability of avian Eimeria sporozoites. Treatments were applied to hatched sporozoites: 1) without additives (no-treatment control); 2) with ethanol; 3) with salinomycin; 4) with Polygain™. All treatments were incubated in RPMI media containing live sporozoites at 37 °C for 14 h and then the number of viable sporozoites were counted. Compared to the no-treatment control, Polygain™ decreased (P 0.05). In conclusion, Polygain™ could be a potential candidate as an anticoccidial agent.

Coccidiosis is an intestinal disease caused by a parasite of the genus Eimeria. This parasite mainly affects poultry species and causes great economic losses in the poultry industry. This study was designed to estimate the prevalence of coccidiosis in the local breed of domestic chicken in Nineveh Governorate, Iraq. 450 faecal swabs and intestinal samples (intestinal scraping) were collected from different local breeds of home-bred chickens from October 2020 to the end of March 2021. All fecal samples were examined using the flotation method by using sugar solution, and Eimeria was confirmed by the polymerase chain reaction method. Fecal examination results showed that 32.6% of the total samples were positive for Eimeria oocysts, classified into six species including E. brunetti, E. mitis.E. maxima E. acervulina E. necatrix, E. tenella with infection rates are 57.5, 44.2, 42.1, 26.5, 20.4, 16.3%, respectively. The phenotypic results were genetically confirmed by the result of the reaction of 455 base pairs. The prevalence of coccidiosis was highest in chickens less than three months of age 49.2% and lowest in chickens older than 6 months 23.9%

Background and Aim: In Indonesia, Madura cattle are native breeds that are expected to contribute to the improvement of regional meat self-sufficiency. Eimeria spp. are protozoans that are commonly found in ruminants. This study aimed to identify the occurrence and diversity of Eimeria spp. in Madura cattle. Materials and Methods: In this study, fresh fecal samples were collected from 100 cattle in Kamal Subdistrict, Bangkalan District, Madura Island, Indonesia. Morphological detection was performed using a light microscope, and molecular identification was performed using a polymerase chain reaction. DNA amplification was conducted using various species-specific primers for Eimeria bovis, Eimeria zuernii, Eimeria auburnensis, Eimeria alabamensis, Eimeria ellipsoidalis, and Eimeria cylindrica. Results: The results obtained 21% (21/100) of Eimeria spp. based on morphological detection. A total of 15 positive samples with 500-25,000/mL oocysts were selected for DNA extraction and amplification, resulting in 12 positive samples. Four Eimeria spp. were obtained based on molecular identification: E. bovis, E. zuernii, E. auburnensis, and E. cylindrica. Conclusion: Four species of Eimeria namely E. bovis, E. zuernii, E. auburnensis, and E. cylindrica were identified from fecal sample of Madura cattle using PCR method in this study. Further comprehensive studies are required to investigate the pathogenicity of Eimeria spp. in Madura cattle. Therefore, improved and integrated management practices should be strengthened by local governments to prevent pathogenic diseases and increase national livestock productivity in Indonesia. Objectives: The purpose of this study was to determine the species composition of Eimeria circu¬lating in Mymensingh district, Bangladesh, using Internal Transcribed Spacer 1 (ITS1) sequences in polymerase chain reaction (PCR) assay. Materials and Methods: Coccidian oocysts were isolated and sporulated in a solution containing 2% potassium dichromate from litter slurry collected from 13 commercially active broiler farms in the research region. Genomic DNA was isolated from sporulated oocysts and used to amplify the Eimeria species-specific ITS1 gene by PCR amplification. Electrophoresis of 1.5% agarose gel was used to visualize the amplified PCR products. Results: In the study samples from Mymensingh district, Bangladesh, the presence of Eimeria brunetti, Eimeria acervulina, Eimeria necatrix, Eimeria mitis, and Eimeria tenella was identified. Conclusions: The findings of this study may shed light on the zonal approach to chicken coccidio¬sis control. BACKGROUND: Prebiotics are non-digestible feed ingredients that improve the immune system. OBJECTIVES: The present study was designed to assess the changes caused by the addition of prebiotics to the feed on carcass characteristics and also chemical composition, physical characteristics, color, texture, and fatty acid profile of chicken pectoral muscles containing Eimeria species. METHODS: Forty-one-day-old male Ross 308 broiler chickens were assigned to four treatments, including nega-tive control (NC), positive control (PC), positive medicated with coxidine (COX), and positive medicated with prebiotics (PRE). After 42 days, carcass characteristics of the chickens were recorded, and also physical character-istics, chemical composition, color, texture, and fatty acid analysis of breast meat were determined. RESULTS: Infection with Eimeria species diminished carcass characteristics. PRE had higher final body weight, hot carcass weight, and breast and thigh muscle weights. Drip loss, pH, cooking loss, fat, ash, dry matter, and texture properties of broilers’ breast meat did not show any significant differences among the experimental groups. Dietary supplementation with prebiotics increased the crude protein content of breast meat. Infection with Eimeria species decreased the a-value of breast meat. Dietary supplementation with prebiotics decreased the amount of fatty acids 16:1 and 18:1 and monounsaturated fatty acids (MUFAs) compared to NC. CONCLUSIONS: Dietary supplementation with prebiotics is a promising strategy with the potential to compensate for the negative effects of infection with Eimeria spp. on carcass characteristics, protein content, and color of breast meat of broiler chickens.

Coccidiosis is one of the most prevalent and economically important parasitic diseases caused by the infection with Eimeria species contributing to major economic losses of poultry industry worldwide. In this study, occurrence of Eimeria spp. in chicken reared under different management systems was studied. A total of 300 faecal samples from chicken were collected from six organised poultry farms and six backyard poultry units in and around Thrissur, Kerala. Out of this, 167 faecal samples were from organised farms and 133 from backyard poultry units. All the samples were artificially sporulated and examined for studying the oocysts morphology and morphometry. Out of 167 samples from organised farms 52 were found to be positive for Eimeria spp. while 61 out of 133 samples from backyard poultry were positive. The overall occurrence of Eimeria spp. in chicken from 12 different areas in and around Thrissur was 37.66 per cent (113/300). The species of Eimeria identified on morphological examination were E. tenella, E. necatrix and E. maxima. The occurrence rate of E. tenella was found to be significantly higher (46.01 %) compared to E. necatrix (39.82 %) and E. maxima (14.15 %). The rate of occurrence of Eimeria spp. infection was significantly higher in backyard poultry (45.86 %) compared to that in organised farms (31.13 %).

With a worldwide distribution, Eimeria spp. could result in serious economic losses to the poultry industry. Due to drug resistance and residues, there are no ideal drugs and vaccines against Eimeria spp. in food animals. In the current study, a bioinformatics approach was employed to design a multiepitope antigen, named NSLC protein, encoding antigenic epitopes of E. necatrix NA4, E. tenella SAG1, E. acervulina LDH, and E. maxima CDPK. Thereafter, the protective immunity of NSLC protein along with five adjuvants and two nanospheres in laying chickens was evaluated. Based on the humoral immunity, cellular immunity, oocyst burden, and the coefficient of growth, the optimum adjuvant was evaluated. Furthermore, the optimum immune route and dosage were also investigated according to the oocyst burden and coefficient of growth. Accompanied by promoted secretion of antibodies and enhanced CD4+ and CD8+ T lymphocyte proportions, NSLC proteins entrapped in PLGA nanospheres were more effective in stimulating protective immunity than other adjuvants or nanospheres, indicating that PLGA nanospheres were the optimum adjuvant for NSLC protein. In addition, a significantly inhibited oocyst burden and growth coefficient promotion were also observed in animals vaccinated with NSLC proteins entrapped in PLGA nanospheres, indicating that the optimum adjuvant for NSLC proteins was PLGA nanospheres. The results also suggested that the intramucosal route with PLGA nanospheres containing 300 μg of NSLC protein was the most efficient approach to induce protective immunity against the four Eimeria species. Quails have many advantages over other poultry species. Its meat has achieved great popularity as an excellent source of protein and other important nutrients. However, there are some limitations to quails production. One of them is the susceptibility to parasitic diseases that cause severe economic losses. Therefore, this work aimed to determine the infection rate and morphology of parasites infecting quails in El-Behera governorate, Egypt. 100 quails (50 migratory, Coturnix coturnix japonica and 50 domesticated quails) were collected. The gastrointestinal tracts of each bird were examined to collect helminths. Fecal materials were examined by direct and flotation methods to detect any coccidian species. The results showed that the total percentage of infection with parasites was 55%. The prevalence of parasitic infection in migratory and domesticated quails was 40% and 70%, respectively. Two species of helminths were recorded, Raillitina tetragona and Heterakis gallinarum with a prevalence of 87.5% and 22.22%, respectively. The morphology of helminths was described using a scanning electron microscope. Eimeria bateri, Eimeria tsunodai, and Eimeria uzura were among the protozoa identified. The histopathological changes in infected tissue with Eimeria species were recorded. In conclusion, this study presented the parasites' prevalence, morphology, and histopathological changes in infected tissue with Eimeria species in examined domesticated and migratory quails. Abstract Dendritic cells (DCs) play a pivotal role to amplify antigen-specific immune responses. Antigens that sensitize T cells via antigen-presentation by DCs could enhance the capacity of host immunity to fight infections. In this study, we tested the immunogenic profiles of chicken DCs towards Glyceraldehyde-3-phosphate dehydrogenase from Eimeria acervulina (EaGAPDH). Immunoblot analysis showed that recombinant EaGAPDH (rEaGAPDH) protein was successfully recognized by rat sera generated against rEaGAPDH. Interaction and internalisation of rEaGAPDH by chicken splenic-derived DCs (chSPDCs) was confirmed by immunofluorescence analysis. Flow cytometry revealed that chSPDCs upregulated MHCII, CD1.1, CD11c, CD80, and CD86 cell-surface markers. Moreover, mRNA expressions of DC maturation biomarkers (CCL5, CCR7, and CD83) and TLR signalling genes (TLR15 and MyD88) were also upregulated whereas those of Wnt signalling were non-significant compared to negative controls. rEaGAPDH treatment induced IL-12 and IFN-γ secretion in chSPDCs but had no effect on IL-10 and TGF-β. Likewise, DC-T cell co-culture promoted IFN-γ secretion and the level of IL-4 was unaffected. Proliferation of T cells and their differentiation into CD3+/CD4+ T cells were triggered in chSPDCs-T cells co-culture system. Taken together, rEaGAPDH could promote Th1 polarization by activating both host DCs and T cells and sheds new light on the role of this important molecule which might contribute to the development of new DCs-based immunotherapeutic strategies against coccidiosis.

The black grouse (Tetrao tetrix) is an endangered species facing challenges in breeding and reintroduction programs, including parasitic infestations. This study aimed to assess natural infestations by various Eimeria species and infestation dynamics in female, male, and young black grouse kept in a stationary aviary. Faecal samples were collected from adult grouses between April and the time of chicks' hatching and rearing (September). Faecal samples from young birds were collected from the hatching for a period of 1 year. The prevalence of Eimeria spp. was determined by a qualitative method (Fulleborn's flotation) and a quantitative method (McMaster's method with Raynaud's modification). The following Eimeria species were identified: E. lyruri, E. nadsoni and E. nonbrumpti. The average percentages of Eimeria spp. in the cock were 80.52%, 9.27%, and 10.21%, respectively; in the hen, they were 86.19%, 9.28%, and 4.53%, respectively; in the young black grouses they were 84.60%, 9.34% and 6.06%, respectively. The highest E. lyruri infestation was observed in the cock in June (144227 OPG) and July (129365 OPG). In the hen, the infestation intensity increased in May (304302 OPG) and then decreased in June (39583 OPG). Furthermore, an additional increase was observed in July (216533 OPG). Two increases in infestation intensity were also observed in young birds, with peaks in January (91387 OPG) and July (126178 OPG). A positive strong correlation was identified between Eimeria spp. in the cock and the young birds. A statistically significant positive correlation was identified in the hen between E. lyruri and E. nadsoni. No correlation was demonstrated between the infestation intensity and the age of the birds or season of the year in all the grouses under study. Despite some attempts, a comprehensive approach to the issue of coccidiosis in the black grouse as a disease that may affect the success of reintroduction has yet to be established. The current in vitro study aimed to investigate the effects of a processed sugarcane extract on the viability of avian Eimeria sporozoites. Treatments were applied to hatched sporozoites: 1) without additives (no-treatment control); 2) with ethanol; 3) with salinomycin; 4) with Polygain™. All treatments were incubated in RPMI media containing live sporozoites at 37 °C for 14 h and then the number of viable sporozoites were counted. Compared to the no-treatment control, Polygain™ decreased (P < 0.001) the counts of E. maxima , E. acervulina , E. bruneti , and E. mitis sporozoites to a level similar to salinomycin (P > 0.05). Avian coccidiosis has a major economic impact on the poultry industry, it is caused by 7 species of Eimeria, and has been primarily controlled using chemotherapeutic agents. Due to the emergence of drug-resistant strains, alternative control strategies are needed. We assessed anticoccidial effects of berberine-based diets in broiler chickens following oral infection with 5 Eimeria species (E. acervulina, E. maxima, E. tenella, E. mitis, and E. praecox). When 0.2% berberine, a concentration that does not affect weight gain, was added to the diet, the 4 groups infected with E. acervulina, E. tenella, E. mitis, or E. praecox showed significant reductions in fecal oocyst shedding (P<0.05) compared to their respective infected and untreated controls. In chickens treated 0.5% berberine instead of 0.2% and infected with E. maxima, fecal oocyst production was significantly reduced, but body weight deceased, indicating that berberine treatment was not useful for E. maxima infection. Taken together, these results illustrate the applicability of berberine for prophylactic use to control most Eimeria infections except E. maxima. Objective: The current study aimed to determine the prevalence, infection burden, and risk factors associated with the occurrence of gastrointestinal (GI) parasites in different avian species in Ilorin, Nigeria. Materials and Methods: This study was conducted in Ilorin, involving 597 fecal samples and GI tracts from a variety of sold and slaughtered avian species. The study was conducted between September 2017 and February 2018. Fecal samples were examined using floatation technique, while the GI tracts were examined for gross helminths and its content were subjected to the direct wet mount examination. Data were analyzed using percentages (descriptive) and the Chi-square (χ2 ) test (inferential). p < 0.05 was considered significant for all analysis. Results: Ten GI parasites were detected with Eimeria species (32.83%), Ascaridia galli (30.15%) and Heterakis gallinarum (24.79%) as the most prevalent ones. Multiple parasites co-infection was recorded in all the avian species: broilers (77.78%), layers (33.33%), cockerels (45.16%), indigenous chickens (17.91%), ducks (69.70%), pigeons (94.12%), turkeys (47.83%), and guinea fowls (77.36%). Pigeons (100.00%) and turkeys (95.65%) were the most infected avian species. Age, sex, and avian types were significantly (p < 0.05) associated with the occurrence of GI parasites infection. Conclusion: This study gives a reflection of the GI parasites fauna of avian species in Nigeria. The GI parasites are endemic among different avian speciesin Ilorin, North Central Nigeria. Knowledge on the epidemiology of these parasites is important in instituting a good preventive and control measures against GI parasites, so as to have maximum production and reproduction effects in the poultry industry. In this report, the site‐specific co‐delivery of green tea/aluminium magnesium silicate (AMS) was reported and the specific target delivery was achieved orally. The new co‐precipitation process was developed to synthesis the green tea/AMS hybrid complex and using energy‐dispersive X‐ray spectroscopy, Fourier‐transform infrared spectroscopy and Raman confirmed its successful synthesis. The blood biocompatibility of the green tea/AMS was tested using chicken blood, and the compound is safe up to 500 mg/ml. After mixed with hydroxypropyl methylcellulose phthalate, the oral beads were synthesised using a linking agent. The oral beads underwent different pH‐based dissolution studies and the results indicated that the beads specifically dissolved in gastric pH (6.5). The pharmaco kinetic studies were performed to estimate the delivery kinetics. The results revealed that the beads underwent as per the Higuchi model. The anticoccidial effects of the beads were tested using chicken. The animal studies were performed in two different modes such as prophylactic treatment and active treatment after Eimeria species challenge. The results indicated that the prophylactic treatment with beads 100% protected the chicken and the active treatment with beads after the Eimeria challenge significantly protected against the intestinal damage and it also enhanced the anticoccidial effect. Aims: The study was conducted to investigate the combination of a probiotic strain of Enterococcus faecium and diclazuril to control coccidiosis in broilers. Methods and Results: A total of 240 one‐day‐old female broiler chicks were divided into eight groups (30 chicks per group): prophylactic groups (G1, G2 and G3) and therapeutic groups (G4, G5 and G6) and two control groups (untreated infected, G7 and untreated uninfected, G8 controls). In the prophylactic approach, diclazuril alone (G1), probiotic alone (G2) or a mixture of both probiotic and diclazuril (G3) was orally administered to the chicks via drinking water 10 days prior to the infection. However, in the therapeutic approach, G4, G5 and G6 birds were administered diclazuril alone, probiotic alone and diclazuril+probiotic mix, respectively, in drinking water for five consecutive days after the appearance of clinical signs of coccidiosis. Birds of both approaches and G7 were experimentally infected with 25 × 103Eimeria‐sporulated oocysts. Chicks in G3 showed the highest weight gain, the lowest lesion score, a low oocyst count and mortality rate among the challenged groups. Moderate lesion scores and oocyst counts were observed in chickens administered probiotics prophylactically. In the therapeutic approach, broilers in G6 but not G5 displayed a decreased mortality rate and lesion score in comparison to those in G7 and G8. However, the result of the probiotic‐treated group was not significantly different from that in the untreated infected control group. Conclusion: The probiotic supplementation as a prophylactic approach can decrease the adverse effects of eimerian infection. In addition, the probiotic and diclazuril mix achieved a considerable improvement in the growth performance. Therefore, probiotic plus diclazuril combination achieved a synergistic effect. The Coccidia are a subclass of the Apicomplexa and include several genera of protozoan parasites that cause important diseases in humans and animals, with Toxoplasma gondii becoming the ‘model organism’ for research into the coccidian molecular and cellular processes. The amenability to the cultivation of T. gondii tachyzoites and the wide availability of molecular tools for this parasite have revealed many mechanisms related to their cellular trafficking and roles of parasite secretory organelles, which are critical in parasite-host interaction. Nevertheless, the extrapolation of the T. gondii mechanisms described in tachyzoites to other coccidian parasites should be done carefully. In this review, we considered published data from Eimeria parasites, a coccidian genus comprising thousands of species whose infections have important consequences in livestock and poultry. These studies suggest that the Coccidia possess both shared and diversified mechanisms of protein trafficking and secretion potentially linked to their lifecycles. Whereas trafficking and secretion appear to be well conversed prior to and during host-cell invasion, important differences emerge once endogenous development commences. Therefore, further studies to validate the mechanisms described in T. gondii tachyzoites should be performed across a broader range of coccidians (including T. gondii sporozoites). In addition, further genus-specific research regarding important disease-causing Coccidia is needed to unveil the individual molecular mechanisms of pathogenesis related to their specific lifecycles and hosts.

Prevention of coccidiosis is one of the best ways of controlling disease. Therefore, the present study was carried out to evaluate the protective effect of ultraviolet (UV)-irradiated sporulated oocysts of Eimeria species against coccidiosis in layer chickens. One hundred forty-four one-day-old layer chicks were randomly divided into 4 groups (n = 36), including non-immunized/non-challenged negative control group (NC group), non-immunized/challenged control group (NIC group), non-irradiated sporulated oocyst/challenged group (CA group), and UV-irradiated sporulated oocyst/challenged (UV group). At the age of 4 days, chickens in groups UV and CA were both orally inoculated with 1.0 × 104 UV-irradiated and non-irradiated sporulated oocysts of Eimeria species, respectively. Chickens in groups NIC and NC were served as positive and negative controls, respectively. Chickens in all groups were orally challenged with 7.5 × 104 sporulated oocysts of Eimeria species except the NC group at the age of 21 days. The results revealed that chicks receiving UV-irradiated sporulated oocysts had no signs of illness with minimal or no changes in the cecal integrity and a significantly lower oocyst shedding (OPG) than in the NIC group. Additionally, the cytokine gene expression profiles were evaluated. Expression levels of IL-2, IL-12, and IFN-γ were significantly higher in the spleen of chicks in the UV and CA groups than in the NC group post-challenge. As expected, treatment with irradiated oocysts resulted in a significant reduction in oocyst shedding and maintenance of cecal mucosal integrity. Furthermore, the body weight was higher in chickens inoculated with UV-irradiated oocysts than their non-irradiated counterparts. In conclusion, our results demonstrate that inoculation with UV-irradiated sporulated oocysts of Eimeria species can produce a substantial reduction in infection symptoms. [ABSTRACT FROM AUTHOR]

Coccidiosis is one of the biggest challenges faced by the global poultry industry as the protozoan is found anywhere poultry are reared. Before now, there have been no documents on the intensity and diversity of Eimeria species of poultry in Kwara State. This study aimed to determine the prevalence and intensity of Eimeria species and its species diversity among poultry in Kwara Central, North-Central Nigeria. Five hundred and two fecal samples (from four hundred and seventy two chickens and thirty turkeys) were collected from chickens and turkeys from 15 farms from December 2017 to May 2018. The samples were subjected to floatation technique. Positive samples were further subjected to the McMaster counting technique for determining the intensity of infection. Sporulation of unsporulated oocysts was carried out using 2.5% potassium dichromate (K2Cr2O7), for the purpose of identifying the different Eimeria species. Ninety five (18.9%) of the sampled population, 10 (66.7%) of the 15 sampled farms were positive for Eimeria species. The total mean oocyst per gram was 6325.0. Eight Eimeria species were detected in the study: 7 in chickens and 1 in turkeys. Eimeria tenella was the most predominant among chickens, E. meleagrimitis was the only species detected in turkeys. Age, sex, bird type, physiological status, farm age, farm size (acres), management system, frequency of anthelmintic use, frequency of anticoccidial use, distance to dumping site (meters), level of biosecurity and frequency of cleaning the pen were significantly associated (P < 0.05) with the occurrence of Eimeria infection. It is envisaged that the information obtained in this study will contribute to a clearer understanding of the epidemiology of poultry coccidiosis, for better management protocol which will improve the productivity of the sector in Kwara State and Nigeria. Objective: The current study aimed to determine the prevalence, infection burden, and risk factors associated with the occurrence of gastrointestinal (GI) parasites in different avian species in Ilorin, Nigeria. Materials and Methods: This study was conducted in Ilorin, involving 597 fecal samples and GI tracts from a variety of sold and slaughtered avian species. The study was conducted between September 2017 and February 2018. Fecal samples were examined using floatation technique, while the GI tracts were examined for gross helminths and its content were subjected to the direct wet mount examination. Data were analyzed using percentages (descriptive) and the Chi-square (X2) test (inferential). p < 0.05 was considered significant for all analysis. Results: Ten GI parasites were detected with Eimeria species (32.83%), Ascaridia galli (30.15%) and Heterakis gallinarum (24.79%) as the most prevalent ones. Multiple parasites co-infection was recorded in all the avian species: broilers (77.78%), layers (33.33%), cockerels (45.16%), indigenous chickens (17.91%), ducks (69.70%), pigeons (94.12%), turkeys (47.83%), and guinea fowls (77.36%). Pigeons (100.00%) and turkeys (95.65%) were the most infected avian species. Age, sex, and avian types were significantly (p < 0.05) associated with the occurrence of GI parasites infection. Conclusion: This study gives a reflection of the GI parasites fauna of avian species in Nigeria. The GI parasites are endemic among different avian species in Ilorin, North Central Nigeria. Knowledge on the epidemiology of these parasites is important in instituting a good preventive and control measures against GI parasites, so as to have maximum production and reproduction effects in the poultry industry. Intestinal coccidiosis, caused by Eimeria species, is an economically-important disease of poultry production industry worldwide. This study was designed to investigate the prevalence of different Eimeria species in the farmed broilers of Khoy city, West Azarbaijan, North West Iran. A total of 26 broiler farms of different production capacities were arbitrarily selected and examined in 2013. In each of the farms, Litters of two broilers farms were randomly sampled twice a week and examined. The intensity of infection with each of the Eimeria species was assessed on the basis of number of oocysts per gram of litter using Clayton-Lane and McMaster methods. Eimeria species diversity was determined by using oocyst sporulation technique in 2% potassium dichromate solution. Results indicated that 23.08% (6/26) of the broiler farms were infected with Eimeria oocysts. The maximum litter infection rate (7.5×103) was observed in fifth week of the rearing period. The litter infection rate was significantly correlated with kinds of water dispenser, feeder, ventilation, and density. The litters were infected with five Eimeria species; E. maxima (32.67%) in 6 farms (23.07%), E. mitis (24%) in 6 farms (23.07%), E. acervulina (18%) in 5 farms (19.23%), E. tenella (14.67%) in 4 farms (15.38%), and E. necatrix (10.67%) in 3 farms (11.58%). Results of this study uncovered high rates of litter infection with various Eimeria species in the studied farms, suggesting the establishment of firm health management strategies in the region.

Camelids (llamas, alpacas, vicuñas, guanacos) are important for the economy of South America and Eimeria infections are important as cause of mortality in camelids. Of the five most prevalent species of Eimeria in South American camelids, Eimeria macusaniensis, Eimeria lamae, Eimeria alpacae, Eimeria punoensis, and Eimeria ivitaensis, E. macusaniensis is considered the most pathogenic. There is considerable confusion concerning the endogenous developmental stages of Eimeria spp. in camelids. Many papers on camelid coccidiosis were published in local Peruvian journals, not easily accessible to wider audience. The water buffalo ( Bubalus bubalis ) is important to the economy of several countries in Asia, and South America and there are also isolated herds in Europe. In India, buffalo is the main dairy animal. Coccidiosis due to Eimeria is an important cause of diarrhea in livestock worldwide. Eimeria species are generally considered host specific. More than 11 Eimeria species are considered common in buffaloes and cattle ( Bos taurus , Bos indicus ). Of the numerous species of Eimeria in cattle, Eimeria bovis , Eimeria zuernii , Eimeria auburnensis and Eimeria alabamensis are considered pathogenic in cattle, but there is no report of their pathogenicity in naturally infected water buffaloes. Instead, Eimeria bareillyi has been documented to be the main pathogenic species in buffalo; it is not transmissible to cattle. There are many reports of Eimeria infections in water buffaloes, scattered in many local journals, and there are conflicting life cycle data on Eimeria species in buffalo. The prevalence of Eimeria in 50 examined rabbits was 84% (42/50). The prevalence was 92.6% in male rabbits while it was73.9% in female rabbits. According to age, the prevalence was determined 88% in rabbits less than 4 months of age while it was 80% in rabbits 4 months of age or more. No significant difference was recorded between rabbits in correlation to age and sex. Ten species of Eimeria infecting rabbits were isolated in Assiut by parasitological examination. The prevalence of Eimeria species were E. perforans (66.7%) followed by E. exigua (26.2%), E. media (26.2%), E. magna (21.4%), E. intestinalis (19%), E. coecicola (19%), E. irresidua (19%), E. piriformis (14.3%), E. flavescens (7.1%) and E. stiedae (7.1%). Single infection of Eimeria spp. was found in 23.8% of the infected rabbits, where as mixed infection involved two, three or four Eimeria spp. was observed in 76.2% of the infected rabbits. Clinical signs were depression, anorexia, diarrhea while postmortem examination revealed hepatomegaly with presence of separate yellowish-white nodules of varying sizes spread over the surface with distended gall bladder. The intestinal lesions revealed varying degree of congestion, thickening of intestinal wall. Histopathological examination of the liver revealed dilated bile ducts and formation of papilliform projections of epithelium containing different developmental stages of Eimeria, associated with degeneration and pathological changes in hepatic parenchyma. The intestinal coccidiosis revealed hyperplasia of the epithelial cells and presence of Eimeria oocytes and gametocytes within the epithelial cells of the villi associated with lymphocytic infiltration in the lamina propria of the villi.
