Understanding the genetic differentiation among populations of most marine fish requires investigating the differences among spawning grounds. However, this can be challenging as spawning grounds for some species are not well known, or spawning fish are difficult to collect. An alternative is to collect juvenile fish in nursery habitats closely associated with potential spawning grounds. Greenland halibut is a deep-dwelling, commercially important species with at least two identified major offshore spawning grounds in the North Atlantic and weak genetic differentiation across the Atlantic. In this study, we sampled juveniles from three sites representing the Davis Strait spawning area in the northwest Atlantic and one site in the northeast Atlantic representing the primary spawning area along the western slope of the Barents Sea. We applied genotype by sequencing and discovered 90 genetic markers that could be used to assess genetic differentiation among the four sites. The northeast and northwest Atlantic showed major genetic differentiation, supporting the existence of the two primary spawning clusters. Additionally, we found genetic differentiation between the three northwest Atlantic samples implying the existence of more than one spawning area in the northwest.

The Arctic is experiencing increasing water temperatures, leading to a northward shift of Atlantic species into Arctic waters. Arctic marine ecosystems are therefore subject to sub- stantial changes in species distributions and occurrence due to anthropogenic climate change. Atlantic cod is one of the most important commercial fish species in the northern seas. The largest known stock is the migrating Northeast Arctic cod (NEAC) that is distributed along the Norwe- gian coast, the Barents Sea and off Svalbard. Atlantic cod in Svalbard waters are generally reported in the literature as belonging to the NEAC ecotype. The more stationary coastal cod (CC) spawn together with NEAC in the Lofoten region and several other areas along the Norwegian coast. The aim of this study was to investigate the population structure of Atlantic cod in Svalbard waters. We used single nucleotide polymorphic (SNP) markers, the pantophysin locus (Pan I) and otolith structure to categorize the 2 cod ecotypes collected in Svalbard fjords between 2017 and 2019. Our results show that both NEAC and CC appear in Svalbard fjords and revealed that 0- group and adult CC individuals caught in Svalbard fjords differ genetically from those along the Norwegian coast, indicating a separation into a local Svalbard CC population. The establishment of CC in Svalbard fjords may be another keystone of the ongoing borealization of the Arctic, with consequences for the local Arctic fjord ecosystem.

Marine fisheries are often allocated to stocks that reflect pragmatic considerations and may not represent the species’ spatial population structure, increasing the risk of mismanagement and unsustainable harvesting. Here we compile mark–recapture data collected across the North Atlantic to gain insight into the spatial population structure of Greenland halibut (Reinhardtius hippoglossoides), an issue that has been unresolved for decades. The dataset contains 168130 fish tagged from 1952 to 2021, with 5466 (3.3%) recaptured individuals. Our results indicate that fish tagged at <50 cm body length migrate at higher rates, suggesting that mark–recapture studies on adult individuals underestimate population-level migration rates. We find evidence for migrations across management units in the North Atlantic indicating two regional offshore populations: one in the Northeast Atlantic, where the West Nordic and Northeast Arctic stocks, currently managed separately, likely belong to a single population that spans from the Kara Sea to Southeast Greenland; and one in the Northwest Atlantic where migration was observed between the Newfoundland and Labrador stock and the Northwest Arctic stock in Davis Strait and Baffin Bay. Our findings indicate complex population structure with implications for international and domestic fisheries management of this long-lived species.

Science-based management of marine fisheries and effective ecosystem monitoring both require the analysis of large amounts of often complex and difficult to collect information. Legislation also increasingly requires the attainment of good environmental status, which again demands collection of data to enable efficient monitoring and management of biodiversity. Such data is traditionally obtained as a result of research surveys through the capture and/or visual identification of organisms. Recent years have seen significant advances in the utilisation of environmental DNA (eDNA) in the marine environment in order to develop alternative cost-effective ways to gather relevant data. Such approaches attempt to identify and/or quantify the species present at a location through the detection of extra-organismal DNA in the environment. These new eDNA based approaches have the potential to revolutionise data collection in the marine environment using non-invasive sampling methods and providing snapshots of biodiversity beyond the capacity of traditional sampling. Here we present a non-technical summary of different approaches in the field of eDNA, and emphasise the broad application of this approach, with value for the governance and management of marine aquatic ecosystems. The review focuses on identifying those tools which are now readily applicable and those which show promise but are currently in development and require further validations. The aim is to provide an understanding of techniques and concepts that can be used by managers without genetic or genomic expertise when consulting with specialists to perform joint evaluations of the utility of the approaches.

The diverse biology and ecology of marine organisms may lead to complex patterns of intraspecific diversity for both neutral and adaptive genetic variation. Sebastes mentella displays a particular life-history as livebearers, for which existence of multiple ecotypes has been suspected to complicate the genetic population structure of the species. Double digest restriction-site associated DNA was used to investigate genetic population structure in S. mentella and to scan for evidence of selection. In total, 42,288 SNPs were detected in 277 fish, and 1,943 neutral and 97 tentatively adaptive loci were selected following stringent filtration. Unprecedented levels of genetic differentiation were found among the previously defined ‘shallow pelagic’, ‘deep pelagic’ and ‘demersal slope’ ecotypes, with overall mean FST = 0.05 and 0.24 in neutral and outlier SNPs, respectively. Bayesian computation estimated a concurrent and historical divergence among these three ecotypes and evidence of local adaptation was found in the S. mentella genome. Overall, these findings imply that the depth-defined habitat divergence of S. mentella has led to reproductive isolation and possibly adaptive radiation among these ecotypes. Additional sub-structuring was detected within the ‘shallow’ and ‘deep’ pelagic ecotypes. Population assignment of individual fish showed more than 94% agreement between results based on SNP and previously generated microsatellite data, but the SNP data provided a lower estimate of hybridization among the ecotypes than that by microsatellite data. We identified a SNP panel with only 21 loci to discriminate populations in mixed samples based on a machine-learning algorithm. This first SNP based investigation clarifies the population structure of S. mentella, and provides novel and high-resolution genomic tools for future investigations. Population connectivity is an increasingly important focal area for the understanding of how marine fish populations respond to anthropogenic pressures like climate change and fisheries. Our model species, the saithe Pollachius virens (Linnaeus, 1758), was chosen because genetic analyses have documented a mismatch between the assessed stocks and the biological populations. We combined laboratory experiments of saithe egg buoyancy and temperature-modulated development time, genetic field data, and high-resolution oceanographic models to disentangle the mechanisms causing isolation and mixing between the management units and the biological populations. Saithe egg buoyancy and development data were included in an individual-based model to simulate transport from all known spawning grounds in the Northeast Atlantic. The results show that interannual variability in the transport of early life stages is strongly influenced by wider climate systems (e.g. the North Atlantic Oscillation). One sample (Rockall) showed genetic differences from the other samples, and this finding was supported by the model showing low mixing with other populations and strong local retention. Strong retention of early life stages around Iceland could indicate an isolated population; however, this possible isolation is counteracted by active migration of adults westward from the Norwegian coast, and no genetic differentiation from other populations was found. Overall, the dispersal modeling supports the genetic analysis, showing a large and well-connected Central Northeast Atlantic population distributed across several management units. This mismatch between population structure and management units can potentially increase the risk for overexploitation of saithe.
