Generally failures of different aircraft components and parts are revealed and examined by the use of non-destructive examination methods. In further detailed explanation and interpretation of failures optical and scanning electron microscopy are used. This paper deals with a problem of a crack on aircraft wheel rim made from aluminium alloy 2014-T6. The crack was observed during regular control by the maintenance unit for non-destructive examination of the Slovenian air carrier Adria Airways. The crack on the rim of an aircraft wheel investigated was a typical fatigue crack. At same time a numerous pits were found which served as stress concentrations on the rim surface. The reduction of area in the through thickness direction is an essential mechanical property of thick steel heavy plates. By a routine control, a very small through thickness reduction of area was found for tensile specimen of a 90 mm plate. Careful investigations of the fracture and section of specimens cut from the as solidified continuously cast 250 mm thick slab showed that the cause was the presence of coarse particles of niobium carbonitride as constituent of the quasi eutectic Fe-Nb(C, N) that form because of the centerline segregation of niobium. Presented work discusses a complex process of inductive heating and quenching of carbon steel planetary shafts for diesel engine starters. On the measurements base of temperature fields on the surface of the planetary shafts by thermographic camera and theoretical knowledge, a mathematical model for temperature conditions determination in the shaft during the entire process of heating and quenching was carried out. On the basis of developed mathematical model a computer program was developed, and used for analyses and induction hardening process optimization of planetary shafts. [ABSTRACT FROM AUTHOR]

Zeaxanthin, a vital dietary carotenoid, is naturally synthesized by plants, microalgae, and certain microorganisms. Large-scale zeaxanthin production can be achieved through plant extraction, chemical synthesis, or microbial fermentation. The environmental and health implications of the first two methods have made microbial fermentation an appealing alternative for natural zeaxanthin production despite the challenges in scaling up the bioprocess. An intermediate between β-carotene and zeaxanthin, β-cryptoxanthin, is found only in specific fruits and vegetables and has several important functions for human health. The low concentration of β-cryptoxanthin in these sources results in low extraction yields, making biotechnological production a promising alternative for achieving higher yields. Currently, there is no industrially relevant microbial fermentation process for β-cryptoxanthin production, primarily due to the lack of identified enzymes that specifically convert β-carotene to β-cryptoxanthin without further conversion to zeaxanthin. In this study, we used genetic engineering to leverage the oleaginous yeast Yarrowia lipolytica as a bio-factory for zeaxanthin and β-cryptoxanthin production. We screened 22 β-carotene hydroxylases and identified eight novel enzymes with β-carotene hydroxylating activity: six producing zeaxanthin and two producing only β-cryptoxanthin. By introducing the β-carotene hydroxylase from the bacterium Chondromyces crocatus (CcBCH), a β-cryptoxanthin titer of 24 ± 6 mg/L was achieved, representing the highest reported titer of sole β-cryptoxanthin in Y. lipolytica to date. By targeting zeaxanthin-producing β-carotene hydroxylase to the endoplasmic reticulum and peroxisomes, we increased the production of zeaxanthin by 54% and 66%, respectively, compared to untargeted enzyme. The highest zeaxanthin titer of 412 ± 34 mg/L was achieved by targeting β-carotene hydroxylases to peroxisomes. In addition, by constructing multienzyme scaffold-free complexes with short peptide tags RIDD and RIAD, we observed a 39% increase in the zeaxanthin titer and a 28% increase in the conversion rate compared to the strain expressing unmodified enzyme. The zeaxanthin titers obtained in this study are not the highest reported; however, our goal was to demonstrate that specific approaches can enhance both titer and conversion rate, rather than to achieve the maximum titer. We present an algorithm for fast generation of quasi-uniform and variable-spacing nodes on domains whose boundaries are represented as computer-aided design (CAD) models, more specifically non-uniform rational B-splines (NURBS). This new algorithm enables the solution of partial differential equations within the volumes enclosed by these CAD models using (collocation-based) meshless numerical discretizations. Our hierarchical algorithm first generates quasi-uniform node sets directly on the NURBS surfaces representing the domain boundary, then uses the NURBS representation in conjunction with the surface nodes to generate nodes within the volume enclosed by the NURBS surface. We provide evidence for the quality of these node sets by analyzing them in terms of local regularity and separation distances. Finally, we demonstrate that these node sets are well-suited (both in terms of accuracy and numerical stability) for meshless radial basis function generated finite differences discretizations of the Poisson, Navier-Cauchy, and heat equations. Addressing the elusive specificity of cysteine cathepsins, which in contrast to caspases and trypsin-like proteases lack strict specificity determining P1 pocket, calls for innovative approaches. Proteomic analysis of cell lysates with human cathepsins K, V, B, L, S, and F identified 30,000 cleavage sites, which we analyzed by software platform SAPS-ESI (Statistical Approach to Peptidyl Substrate-Enzyme Specific Interactions). SAPS-ESI is used to generate clusters and training sets for support vector machine learning. Cleavage site predictions on the SARS-CoV-2 S protein, confirmed experimentally, expose the most probable first cut under physiological conditions and suggested furin-like behavior of cathepsins. Crystal structure analysis of representative peptides in complex with cathepsin V reveals rigid and flexible sites consistent with analysis of proteomics data by SAPS-ESI that correspond to positions with heterogeneous and homogeneous distribution of residues. Streptomyces rimosus ATCC 10970 is the parental strain of industrial strains used for the commercial production of the important antibiotic oxytetracycline. As an actinobacterium with a large linear chromosome containing numerous long repeat regions, high GC content, and a single giant linear plasmid (GLP), these genomes are challenging to assemble. Here, we apply a hybrid sequencing approach relying on the combination of short- and long-read next-generation sequencing platforms and whole-genome restriction analysis by using pulsed-field gel electrophoresis (PFGE) to produce a high-quality reference genome for this biotechnologically important bacterium. By using PFGE to separate and isolate plasmid DNA from chromosomal DNA, we successfully sequenced the GLP using Nanopore data alone. Using this approach, we compared the sequence of GLP in the parent strain ATCC 10970 with those found in two semi-industrial progenitor strains, R6-500 and M4018. Sequencing of the GLP of these three S. rimosus strains shed light on several rearrangements accompanied by transposase genes, suggesting that transposases play an important role in plasmid and genome plasticity in S. rimosus . The polished annotation of secondary metabolite biosynthetic pathways compared to metabolite analysis in the ATCC 10970 strain also refined our knowledge of the secondary metabolite arsenal of these strains. The proposed methodology is highly applicable to a variety of sequencing projects, as evidenced by the reliable assemblies obtained. IMPORTANCE The genomes of Streptomyces species are difficult to assemble due to long repeats, extrachromosomal elements (giant linear plasmids [GLPs]), rearrangements, and high GC content. To improve the quality of the S. rimosus ATCC 10970 genome, producer of oxytetracycline, we validated the assembly of GLPs by applying a new approach to combine pulsed-field gel electrophoresis separation and GLP isolation and sequenced the isolated GLP with Oxford Nanopore technology. By examining the sequenced plasmids of ATCC 10970 and two industrial progenitor strains, R6-500 and M4018, we identified large GLP rearrangements. Analysis of the assembled plasmid sequences shed light on the role of transposases in genome plasticity of this species. The new methodological approach developed for Nanopore sequencing is highly applicable to a variety of sequencing projects. In addition, we present the annotated reference genome sequence of ATCC 10970 with a detailed analysis of the biosynthetic gene clusters.

Three charges of scrap-based, Ti-stabilized, Cr-Ni-Mo austenitic stainless steel in the form of hot-rolled steel plates were characterized. Based on automated metallographic analyses of representative microstructures, a quality characterization in terms of cleanliness of the hot-rolled steel plates was performed. Elevated contents of impurities, especially Pb, Bi, and oxygen, which affect the hot workability of stainless steels, were detected. The recycled FeTi-cored wire was the main source of the elevated levels of impurities detected in the hot-rolled, Ti-stabilized, stainless-steel plates. Related to this, elevated levels of nonmetallic inclusions (NMIs) and segregations were formed. The three charges were classified based on calculations of the Mahalanobis distance (MD) between the inclusions. The charge with the smallest number of nonmetallic inclusions was set as the reference class. The selection of outlier inclusions based on their MDs and their back-representation into ternary diagrams gave relevant metallurgical information about the abnormalities. The advantage of this technique is that the calculations of the MD and the threshold can be fully automated.
