Tension wood (TW), characterized by increased cambium cell proliferation and few vessels, is a classical model for the mechanical analysis of wood formation. In this study, we found higher superoxide anion (O2.-) levels in the cambium zone of poplar (Populus alba × P. glandulosa clone "84K") TW than in that of opposite wood during gravistimulation. Treatment with an O2.- activator (methyl viologen) resulted in tension-wood-like xylem tissue formation, and transgenic plants with reduced cambium O2.- levels presented an attenuated gravity response. Time-course detection of O2.- and indole-3-acetic acid (IAA) levels revealed that auxin responses were enhanced following increases in O2.- levels, suggesting that IAA mediates TW induction downstream of O2.-. Rapid division but advanced programmed cell death in cambium cells was detected in both gravistimulated and O2.- activator-treated plants. The progression of gastric cancer involves a complex multi-stage process, with gastroscopy and biopsy being the standard procedures for diagnosing gastric diseases. This study introduces an innovative non-invasive approach to differentiate gastric disease stage using gastric fluid samples through machine-learning-assisted surface-enhanced Raman spectroscopy (SERS). This method effectively identifies different stages of gastric lesions. The XGBoost algorithm demonstrates the highest accuracy of 96.88% and 91.67%, respectively, in distinguishing chronic non-atrophic gastritis from intestinal metaplasia and different subtypes of gastritis (mild, moderate, and severe). Through blinded testing validation, the model can achieve more than 80% accuracy. A new marine monoraphid diatom species, Planothidiumpseudolinkei sp. nov. , is described from the coast of Guangxi, China. The detailed morphology of this epipsammic diatom is studied by using both light and scanning electron microscopy. P.pseudolinkei differs from congeners by a combination of morphological features including capitate apices, multiseriate striae, a small central area on the raphe valve and an oblong sinus on the rapheless valve. Background: Intracranial high-density areas (HDAs) have attracted considerable attention for predicting clinical outcomes; however, whether HDAs predict worse neurological function and mental health remains controversial and unclear, which requires further investigation., Aim: To investigate the predictive value of intracranial HDAs for neurological function and mental health after endovascular treatment., Methods: In this prospective study, 96 patients with acute ischemic stroke (AIS) who accepted endovascular mechanical thrombectomy (EMT) were included. The enrolled patients underwent cranial computed tomography (CT) examination within 24 hours after EMT. Clinical data in terms of National Institutes of Health Stroke Scale (NIHSS), the 3-month modified Rankin Scale (mRS), self-rating depression scale (SDS), and self-rating anxiety scale (SAS) scores were collected and compared between patients with HDAs and non-HDAs and between patients with good and poor clinical prognosis., Results: Compared to patients without HDAs, patients with HDAs presented severe neurological deficits (admission NIHSS score: 18 ± 3 vs 19 ± 4), were more likely to have post-stroke disabilities (mRS < 3: 35% vs 62%), and suffered more severe depression (SDS score: 58 ± 16 vs 64 ± 13) and anxiety disorder (SAS score: 52 ± 8 vs 59 ± 10). Compared to patients with a good prognosis, patients with a poor prognosis presented severe neurological deficits (admission NIHSS score: 17 ± 4 vs 20 ± 3), were more likely to have HDAs on CT images (64% vs 33%), and suffered more severe depression (SDS score: 55 ± 19 vs 65 ± 11) and anxiety (SAS score: 50 ± 8 vs 58 ± 12). Respiratory tract infections (RTIs) are among the most common problems in clinical settings. Rapid and accurate identification of bacterial pathogens will provide practical guidelines for managing and treating RTIs. This study describes a method for rapidly detecting bacterial pathogens that cause respiratory tract infections via multi-channel loop-mediated isothermal amplification (LAMP). LAMP is a sensitive and specific diagnostic tool that rapidly detects bacterial nucleic acids with high accuracy and reliability. The proposed method offers a significant advantage over traditional bacterial culturing methods, which are time-consuming and often require greater sensitivity for detecting low levels of bacterial nucleic acids. This article presents representative results of K. pneumoniae infection and its multiple co-infections using LAMP to detect samples (sputum, bronchial lavage fluid, and alveolar lavage fluid) from the lower respiratory tract. In summary, the multi-channel LAMP method provides a rapid and efficient means of identifying single and multiple bacterial pathogens in clinical samples, which can help prevent the spread of bacterial pathogens and aid in the appropriate treatment of RTIs.

Two new diatom species belonging to the genus Gomphonemopsis are described, Gomphonemopsisnana sp. nov. and Gomphonemopsisgaoi sp. nov. These two species were compared in detail with congeners. Gomphonemopsisnana is distinguished by its high stria density and small size. This species was found so far to be epiphytic only on the eelgrass collected from Qingdao Bay (Yellow Sea). Gomphonemopsisgaoi is characterized by its isopolar valves, simple proximal raphe endings and acutely rounded apices. This taxon was separated from the exoskeleton of marine copepods sampled from the Futian Mangrove Nature Reserve (South China Sea). In addition, two new combinations, Gomphonemopsisoahuensis (Østrup) Lang Li, Yuhang Li & Junxiang Lai, comb. nov. and Gomphonemopsisplatypus (Østrup) Lang Li, Yuhang Li & Junxiang Lai, comb. nov. are proposed. Astaxanthin is widely used in food, aquaculture, cosmetics, and pharmaceuticals due to its strong antioxidant activity and coloring ability, but its production from Phaffia rhodozyma remains the main challenge due to the high fermentation cost and low content of carotenoid. In this study, the production of carotenoids from food waste (FW) by a P. rhodozyma mutant was investigated. P. rhodozyma mutant screened by UV mutagenesis and flow cytometry could stably produce high carotenoids at 25°C, with carotenoid production (32.9 mg/L) and content (6.7 mg/g), respectively, increasing by 31.6% and 32.3% compared with 25 mg/L and 5.1 mg/g of wild strain. Interestingly, the carotenoid production reached 192.6 mg/L by feeding wet FW, which was 21% higher than batch culture. The 373 g vacuum freeze-dried products were obtained from the fermentation of 1 kg FW by P. rhodozyma, which contained 784 mg carotenoids and 111 mg astaxanthin. The protein, total amino acids, and essential amino acids content of the fermentation products were 36.6%, 40.5%, and 18.2% (w/w), respectively, and lysine-added fermentation products had the potential of high-quality protein feed source. Four undescribed steroidal compounds along with twenty known compounds were isolated from n-butanol extracted fraction of the whole plants of Solanum lyratum Thunb (SLNF). Their structures were assigned based on analyses of the extensive spectroscopic data (including MS, 1D/2D NMR, and ECD) or comparisons of the NMR data with those reported. Among the knowns, three compounds were isolated from Solanum plants for the first time, while one compound was isolated from S. lyratum for the first time. In addition, the cytotoxicities of these isolates against human colon SW480 and hepatoma Hep3B cells were evaluated by a MTT assay. And, nine of them and SLNF exhibited significant activities against both SW480 and Hep3B cells, while twelve of them significantly inhibited the activities of SW480 cells. The B cell leukemia/lymphoma 2 (BCL-2) inhibitor venetoclax is effective in chronic lymphocytic leukemia (CLL); however, resistance may develop over time. Other lymphoid malignancies such as diffuse large B cell lymphoma (DLBCL) are frequently intrinsically resistant to venetoclax. Although genomic resistance mechanisms such as BCL2 mutations have been described, this probably only explains a subset of resistant cases. Using 2 complementary functional precision medicine techniques - BH3 profiling and high-throughput kinase activity mapping - we found that hyperphosphorylation of BCL-2 family proteins, including antiapoptotic myeloid leukemia 1 (MCL-1) and BCL-2 and proapoptotic BCL-2 agonist of cell death (BAD) and BCL-2 associated X, apoptosis regulator (BAX), underlies functional mechanisms of both intrinsic and acquired resistance to venetoclax in CLL and DLBCL. Additionally, we provide evidence that antiapoptotic BCL-2 family protein phosphorylation altered the apoptotic protein interactome, thereby changing the profile of functional dependence on these prosurvival proteins. Targeting BCL-2 family protein phosphorylation with phosphatase-activating drugs rewired these dependencies, thus restoring sensitivity to venetoclax in a panel of venetoclax-resistant lymphoid cell lines, a resistant mouse model, and in paired patient samples before venetoclax treatment and at the time of progression.

Objective: The aim of this study was to compare perioperative outcomes of patients with low rectal cancer after stoma-site approach single-port laparoscopic Miles procedure or conventional multi-port laparoscopic Miles procedure, as well as to evaluate the safety and efficacy of stoma-site approach single-port laparoscopic surgery in low rectal cancer., Methods: Between September 2020 and September 2021, 51 low rectal cancer patients scheduled for Miles procedure at the Department of Gastrointestinal Surgery of Affiliated Hospital of North Sichuan Medical College were randomly assigned to the single-port laparoscopic surgery group (SPLS) and the multi-port laparoscopic surgery (MPLS) group. The perioperative outcomes were compared between the two groups., Results: In this study, 25 patients underwent SPLS and 26 underwent MPLS. All patients completed the study, and there were no perioperative deaths in either group. Observation indicators such as intraoperative bleeding (39 mL vs. 41 mL), number of lymph nodes (20.12 ± 3.29 vs. 21.84 ± 3.74), average hospital stay (7.15 ± 1.52 vs. 7.64 ± 1.66), and time to flatulence (2.5d vs. 2.5d) showed no significant differences between the SPLS and MPLS groups (p > 0.05). However, the operation duration (180 min vs. 118 min) and perioperative complications showed statistically significant differences between the two groups (p < 0.05). In addition, patients in the SPLS group had significantly higher satisfaction scores than those in the MPLS group (p < 0.05)., Conclusion: For patients with low rectal cancer requiring Miles surgery, stoma-site approach single-port laparoscopic surgery has comparable safety and efficacy to multi-port laparoscopic surgery., (Copyright © 2023 Asian Surgical Association and Taiwan Robotic Surgery Association. Chemical investigation on the water-soluble constituents of Stemona tuberosa Lour. resulted in the isolation of a previously undescribed furfural derivative namely (S)-5-((R)-hydroxy(5-(hydroxymethyl)furan-2-yl)methyl)-5-methylfuran-2(5H)-one and twenty-five known compounds from the water decoction of the dried root tubers. Their structures were determined by analysis of the extensive spectroscopic data, including 1D/2D NMR, HR-ESI-MS, and ORD, as well as the ECD simulation and comparison. Most of them were phenolic and among them, four compounds were isolated from Stemona plants for the first time. Background: The rapid and reliable detection of pathogenic bacteria at an early stage is a highly significant research field for public health. However, most traditional approaches for pathogen identification are time-consuming and labour-intensive, which may cause physicians making inappropriate treatment decisions based on an incomplete diagnosis of patients with unknown infections, leading to increased morbidity and mortality. Therefore, novel methods are constantly required to face the emerging challenges of bacterial detection and identification. In particular, Raman spectroscopy (RS) is becoming an attractive method for rapid and accurate detection of bacterial pathogens in recent years, among which the newly developed surface-enhanced Raman spectroscopy (SERS) shows the most promising potential., Aim of Review: Recent advances in pathogen detection and diagnosis of bacterial infections were discussed with focuses on the development of the SERS approaches and its applications in complex clinical settings., Key Scientific Concepts of Review: The current review describes bacterial classification using surface enhanced Raman spectroscopy (SERS) for developing a rapid and more accurate method for the identification of bacterial pathogens in clinical diagnosis. The initial part of this review gives a brief overview of the mechanism of SERS technology and development of the SERS approach to detect bacterial pathogens in complex samples. The development of the label-based and label-free SERS strategies and several novel SERS-compatible technologies in clinical applications, as well as the analytical procedures and examples of chemometric methods for SERS, are introduced. The purple soil slope farmland is an important agricultural land in southwest China but is also one of the main sources of agricultural non-point source pollution in the Three Gorges Reservoir area. Taking reasonable measures to control the loss of soil nutrients is of great significance to the treatment of non-point source pollution in the region. Here, a three-year (2018-2020) field runoff experiment was conducted to monitor and evaluate the phosphorus (P) loss in sloping farmland via surface runoff (i.e., surface flow, 0-20 cm) and subsurface runoff (i.e., subsurface flow, 20-60 cm), with five treatments including no fertilization (CK), conventional fertilization (CF), optimal fertilization (OF), biochar combined with 85% of OF (BF), and straw combined with 85% of OF (SF). The results showed that fertilization application reduced the sediment yields and surface runoff flux but increased the subsurface runoff flux. The total loss flux of phosphate (PO 3- 4 -P), total phosphorus (TP), and particulate phosphorus (PP) in surface flow were the highest in the BF treatment and the lowest in the SF treatment. All fertilization applications increased the P loss fluxes in subsurface flow relative to that in CK. The highest PO 3- 4 -P and TP loss flux in subsurface flow was found in the BF (213.88 g·hm -2 and 694.54 g·hm -2 , respectively) treatment, followed by that in the OF and SF treatments. Redundancy analysis (RDA) results showed that surface runoff flux and biochar application were the main factors contributing to increased P loss in surface flow, and subsurface runoff flux was the main factor contributing to increased P loss in subsurface flow. In summary, the SF treatment reduced the amount of sediment yield and surface runoff flux in sloping farmland of purple soil and was the most effective for controlling P loss, whereas the risk of subsurface runoff flux requires further attention.

CoFe-based alloys and rare earth (RE) elements are among the most studied materials in applying magnetic devices to improve soft magnetic characteristics. A series of Co 40 Fe 40 Sm 20 films are deposited on a glass substrate via the sputtering technique, followed by an annealing process to investigate their effect on microstructural and optical properties of Co 40 Fe 40 Sm 20 films. In this study, the increase in the thickness of Co 40 Fe 40 Sm 20 films and annealing temperatures resulted in a smoother surface morphology. The 40 nm Co 40 Fe 40 Sm 20 films annealed 300 °C are expected to have good wear resistance and adhesive properties due to their high values of H/E ratio and surface energy. Optical transparency also increased due to the smoother surface of the Co 40 Fe 40 Sm 20 films.

Helicobacter pylori is a major human pathogen that infects approximately half of the global population and is becoming a serious health threat due to its increasing antibiotic resistance. It is the causative agent of chronic active gastritis, peptic ulcer disease, and gastric cancer and has been classified as a Group I Carcinogen by the International Agency for Research on Cancer. Therefore, the rapid and accurate diagnosis of H. pylori and the determination of its antibiotic resistance are important for the efficient eradication of this bacterial pathogen. Currently, H. pylori diagnosis methods mainly include the urea breath test (UBT), the antigen test, the serum antibody test, gastroscopy, the rapid urease test (RUT), and bacterial culture. Among them, the first three detection methods are noninvasive, meaning they are easy tests to conduct. However, bacteria cannot be retrieved through these techniques; thus, drug resistance testing cannot be performed. The last three are invasive examinations, but they are costly, require high skills, and have the potential to cause damage to patients. Therefore, a noninvasive, rapid, and simultaneous method for H. pylori detection and drug resistance testing is very important for efficiently eradicating H. pylori in clinical practice. This protocol aims to present a specific procedure involving the string test in combination with quantitative polymerase chain reaction (qPCR) for the rapid detection of H. pylori infection and antibiotic resistance. Unlike bacterial cultures, this method allows for easy, rapid, noninvasive diagnosis of H. pylori infection status and drug resistance. Specifically, we used qPCR to detect rea for H. pylori infection and mutations in the 23S rRNA and gyrA genes, which encode resistance against clarithromycin and levofloxacin, respectively. Compared to routinely used culturing techniques, this protocol provides a noninvasive, low-cost, and time-saving technique to detect H. pylori infection and determine its antibiotic resistance using qPCR.

Nardosinone, a predominant bioactive product from Nardostachys jatamansi DC, is well-known for its promising therapeutic applications, such as being used as a drug on anti-inflammatory, antidepressant, cardioprotective, anti-neuroinflammatory, anti-arrhythmic, anti-periodontitis, etc. However, its stability under varying environmental conditions and its degradation products remain unclear. In this study, four main degradation products, including two previously undescribed compounds [2-deoxokanshone M (64.23%) and 2-deoxokanshone L (1.10%)] and two known compounds [desoxo-narchinol A (2.17%) and isonardosinone (3.44%)], were firstly afforded from the refluxed products of nardosinone in boiling water; their structures were identified using an analysis of the extensive NMR and X-ray diffraction data and the simulation and comparison of electronic circular dichroism spectra. Compared with nardosinone, 2-deoxokanshone M exhibited potent vasodilatory activity without any of the significant anti-neuroinflammatory activity that nardosinone contains. Secondly, UPLC-PDA and UHPLC-DAD/Q-TOF MS analyses on the degradation patterns of nardosinone revealed that nardosinone degraded more easily under high temperatures and in simulated gastric fluid compared with the simulated intestinal fluid. A plausible degradation pathway of nardosinone was finally proposed using nardosinonediol as the initial intermediate and involved multiple chemical reactions, including peroxy ring-opening, keto-enol tautomerization, oxidation, isopropyl cleavage, and pinacol rearrangement. Our findings may supply certain guidance and scientific evidence for the quality control and reasonable application of nardosinone-related products.

Introduction: It has been established that UBR4 encodes E3 ubiquitin ligase, which determines the specificity of substrate binding during protein ubiquitination and has been associated with various functions of the nervous system but not the reproductive system. Herein, we explored the role of UBR4 on fertility with a Drosophila model ., Methods: Different Ubr4 knockdown flies were established using the UAS/GAL4 activating sequence system. Fertility, hatchability, and testis morphology were studied, and bioinformatics analyses were conducted. Our results indicated that UBR4 deficiency could induce male sterility and influent egg hatchability in Drosophila ., Results: We found that Ubr4 deficiency affected the testis during morphological analysis. Proteomics analysis indicated 188 upregulated proteins and 175 downregulated proteins in the testis of Ubr4 knockdown flies. Gene Ontology analysis revealed significant upregulation of CG11598 and Sfp65A , and downregulation of Pelota in Ubr4 knockdown flies. These proteins were involved in the biometabolic or reproductive process in Drosophila . These regulated proteins are important in testis generation and sperm storage promotion. Cyperus rotundus L. has been widely used in the treatment and prevention of numerous diseases in traditional systems of medicine around the world, such as nervous, gastrointestinal systems diseases and inflammation. In traditional Chinese medicine (TCM), its rhizomes are frequently used to treat liver disease, stomach pain, breast tenderness, dysmenorrheal and menstrual irregularities. The review is conducted to summarize comprehensively the plant's vernacular names, distribution, phytochemistry, pharmacology, toxicology and analytical methods, along with the data mining for TCM prescriptions containing C. rotundus . Herein, 552 compounds isolated or identified from C. rotundus were systematically collated and classified, concerning monoterpenoids, sesquiterpenoids, flavonoids, phenylpropanoids, phenolics and phenolic glycosides, triterpenoids and steroids, diterpenoids, quinonoids, alkaloids, saccharides and others. Their pharmacological effects on the digestive system, nervous system, gynecological diseases, and other bioactivities like antioxidant, anti-inflammatory, anti-cancer, insect repellent, anti-microbial activity, etc. were summarized accordingly. Moreover, except for the data mining on the compatibility of C. rotundus in TCM, the separation, identification and analytical methods of C. rotundus compositions were also systematically summarized, and constituents of the essential oils from different regions were re-analyzed using multivariate statistical analysis. In addition, the toxicological study progresses on C. rotundus revealed the safety property of this herb. Although the incidence and mortality of gastric cancer (GC) have been decreasing steadily worldwide, especially in East Asia, the disease burden of this malignancy is still very heavy. Except for tremendous progress in the management of GC by multidisciplinary treatment, surgical excision of the primary tumor is still the cornerstone intervention in the curative-intent treatment of GC. During the relatively short perioperative period, patients undergoing radical gastrectomy will suffer from at least part of the following perioperative events: Surgery, anesthesia, pain, intraoperative blood loss, allogeneic blood transfusion, postoperative complications, and their related anxiety, depression and stress response, which have been shown to affect long-term outcomes. A taxonomic study was carried out on strain BGMRC 0090 T , which was isolated from seawater. The isolate was a Gram-negative, aerobic, flagellated, rod-shaped bacterium with algicidal activity. Optimal growth was observed at 30 °C, pH 6.0 and with 2 % (w/v) NaCl. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain BGMRC 0090 T belonged to the genus Parvularcula , with highest sequence similarity to Parvularcula lutaonensis CC-MMS-1 T (98.4 %). Average nucleotide identity, amino acid identity and digital DNA-DNA hybridization values between strain BGMRC 0090 T and five strains of the genus Parvularcula with publicly available genomes were below 84.0, 69.2 and 21.4 %, respectively. The genome of strain BGMRC 0090 T was 3.2 Mb with 64.8 mol% DNA G+C content and encoded 2905 predicted proteins, three rRNA, 42 tRNA and four ncRNA genes. Some algicidal biosynthesis-associated genes were detected in the genome. Strain BGMRC 0090 T contained Q-10 as the major quinone. The predominant fatty acids were identified as summed feature 8 (C 18 : 1 ω7 c /ω6 c ) and C 16 : 0 . Based on the polyphasic evidence presented in this paper, strain BGMRC 0090 T is concluded to represent a novel species of the genus Parvularcula , for which the name Parvularcula maris sp. nov. is proposed. The type strain is BGMRC 0090 T (= KCTC 92591 T =MCCC 1K08100 T ).

The aim of the study was to understand the impact of biochar and straw return on soil aggregates and organic carbon for soil improvement of the newly cultivated purple soil dry slope land in the Three Gorges Reservoir area. In this study, a field test was used to set five treatment pairs with regards to soil aggregate composition and organic carbon distribution:no fertilization(CK), conventional fertilization(NPK), optimized fertilization(GNPK), chemical fertilizer reduction combined with straw(RSD), and chemical fertilizer reduction combined with biochar(BC). The results showed that fertilization can improve the level of soil fertility, especially with the RSD and BC treatments. The soil aggregates of each fertilization treatment were<0.25 mm in size. Compared with the CK, each treatment significantly increased the aggregate content of 0.5-5 mm particles, and the values of MWD, GMD, and R 0.25 . Further, the treatments significantly reduced the value of D and PAD 0.25 ( P <0.05), and each fertilization treatment significantly increased the soil organic carbon content, of which BC(6.73 g·kg -1 ) and RSD(5.45 g·kg -1 ) were significantly better than NPK(5.05 g·kg -1 ) and GNPK(3.63 g·kg -1 ). The<0.25 mm aggregates had the highest contribution rate of organic carbon(34.92%-59.49%), while the>5 mm aggregates had the lowest contribution rate of organic carbon(1.55%-6.01%). The BC treatment significantly increased the organic carbon contribution rate of 5-2 mm and 2-1 mm agglomerates( P <0.05), while the contribution rate of NPK, RSD, and GNPK was the most significant for 0.5-0.25 mm( P <0.05). Each fertilization treatment increased the yield of rapeseed and corn, with large inter-annual differences, but the overall difference between treatments was not significant. The stability of soil aggregates and crop yields showed an upward trend with the increase of soil organic carbon. Biochar and straw returning to the field may promote the formation of large and medium aggregates in soil, effectively improve the stability of aggregates, increase organic carbon content, and promote crop yields. It is therefore an effective measure to improve the soil structure of purple soil and improve soil quality.

This research explores the behavior of Co 40 Fe 40 W 10 B 10 when it is sputtered onto Si(100) substrates with a thickness (t f ) ranging from 10 nm to 100 nm, and then altered by an annealing process at temperatures of 200 °C, 250 °C, 300 °C, and 350 °C, respectively. The crystal structure and grain size of Co 40 Fe 40 W 10 B 10 films with different thicknesses and annealing temperatures are observed and estimated by an X-ray diffractometer pattern (XRD) and full-width at half maximum (FWHM). The XRD of annealing Co 40 Fe 40 W 10 B 10 films at 200 °C exhibited an amorphous status due to insufficient heating drive force. Moreover, the thicknesses and annealing temperatures of body-centered cubic (BCC) CoFe (110) peaks were detected when annealing at 250 °C with thicknesses ranging from 80 nm to 100 nm, annealing at 300 °C with thicknesses ranging from 50 nm to 100 nm, and annealing at 350 °C with thicknesses ranging from 10 nm to 100 nm. The FWHM of CoFe (110) decreased and the grain size increased when the thickness and annealing temperature increased. The CoFe (110) peak revealed magnetocrystalline anisotropy, which was related to strong low-frequency alternative-current magnetic susceptibility (χ ac ) and induced an increasing trend in saturation magnetization (Ms) as the thickness and annealing temperature increased. The contact angles of all Co 40 Fe 40 W 10 B 10 films were less than 90°, indicating the hydrophilic nature of Co 40 Fe 40 W 10 B 10 films. Furthermore, the surface energy of Co 40 Fe 40 W 10 B 10 presented an increased trend as the thickness and annealing temperature increased. According to the results, the optimal conditions are a thickness of 100 nm and an annealing temperature of 350 °C, owing to high χ ac , large Ms, and strong adhesion; this indicates that annealing Co 40 Fe 40 W 10 B 10 at 350 °C and with a thickness of 100 nm exhibits good thermal stability and can become a free or pinned layer in a magnetic tunneling junction (MTJ) application.

In this work, a fluorescent nanoparticles labeling-free fluorescence enzyme-linked immunoassay (FELISA) has been established for the ultrasensitive detection of microcystin-LR (MC-LR) in water and fish samples. Polyclonal antibody against MC-LR was labeled with horseradish peroxidase (HRP) and used as signal probe for binding with analyte in sample or for coating antigen. After washing of the unbound antibody, the substrate system (2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate) (ABTS)/H 2 O 2 ) was added. The oxidation product of ABTS (ox-ABTS) catalyzed by HRP effectively caused the fluorescence quenching of subsequently added silane-doped carbon dots (Si-CDs), and the change in fluorescence intensity of Si-CDs was used to realize the quantitative detection of MC-LR. Under the optimum conditions, the Si-CDs based FELISA method showed a good linear relationship from 0.001 to 3.20 μg L -1 (R 2  = 0.994) and provided a low detection limit of 0.6 ng L -1 , which was approximately 30-fold lower than that of traditional indirect competitive ELISA. Average recovery values from 79.9% to 109.2% was obtained from spiked water and crucian samples, suggesting its potential application on the monitoring of MR-LR at a trace level., (Copyright © 2019 Elsevier B.V. Objective: To investigate and analyze the clinical efficacy of different side branch protection techniques on patients receiving coronary intervention and the patient's prognosis., Patients and Methods: A total of 80 patients with coronary heart disease treated in Jiangmen Central Hospital from January 2014 to January 2017 were collected. According to different side branch protection strategies selected during operation, they were divided into jailed wire technique (JWT) group (n=20), jailed balloon technique (JBT) group (n=20), balloon-stent kissing technique (BSKT) group (n=20), and BSKT+RW group (n=20). The relevant operation parameters and the prevalence of adverse reactions at 1 month and 6 months after operation were compared among the four groups., Results: The success rate of operation and relevant operation parameters in BSKT+RW group were slightly superior to those in the other three groups, but there were no significant differences among the four groups (p>0.05). Besides, the prevalence rates of adverse reactions at 1 month and 6 months after the operation had no significant differences among the four groups, but they were slightly lower in BSKT+RW group than those in the other three groups., Conclusions: There are no significant differences in the clinical efficacy and postoperative recovery of patients receiving coronary intervention among the four kinds of different side branch protection techniques. However, BSKT+RW is slightly superior to the other three treatment methods, which, therefore, is a preferred choice if the patient's economic conditions permit.

N-methyl-D-aspartate receptors (NMDARs), a subtype of glutamate-gated ion channels, play a central role in epileptogenesis. Recent studies have identified an increasing number of GRIN2A (a gene encoding the NMDAR GluN2A subunit) mutations in patients with epilepsy. Phenotypes of GRIN2A mutations include epilepsy-aphasia disorders and other epileptic encephalopathies, which pose challenges in clinical treatment. Here we identified a heterozygous GRIN2A mutation (c.1341T>A, p.N447K) from a boy with Rolandic epilepsy by whole-exome sequencing. The patient became seizure-free with a combination of valproate and lamotrigine. Functional investigation was carried out using recombinant NMDARs containing a GluN2A-N447K mutant that is located in the ligand-binding domain of the GluN2A subunit. Whole-cell current recordings in HEK 293T cells revealed that the N447K mutation increased the NMDAR current density by ~1.2-fold, enhanced the glutamate potency by 2-fold, and reduced the sensitivity to Mg 2+ inhibition. These results indicated that N447K is a gain-of-function mutation. Interestingly, alternative substitutions by alanine and glutamic acid at the same residue (N447A and N447E) did not change NMDAR function, suggesting a residual dependence of this mutation in altering NMDAR function. Taken together, this study identified human GluN2A N447K as a novel mutation associated with epilepsy and validated its functional consequences in vitro. Identification of this mutation is also helpful for advancing our understanding of the role of NMDARs in epilepsy and provides new insights for precision therapeutics in epilepsy.

Objective: To explore the potential causative genes of paroxysmal hypnogenic dyskinesia (PHD), which was initially considered a subtype of paroxysmal dyskinesia and has been recently considered a form of nocturnal frontal lobe epilepsy (NFLE)., Methods: Eleven patients with PHD were recruited. Mutations in proline-rich region transmembrane protein-2 (PRRT2), myofibrillogenesis regulator 1 (MR-1), solute carrier family 2, member 1 (SLC2A1), calcium-activated potassium channel alpha subunit (KCNMA1), cholinergic receptor, nicotinic, alpha 4 (CHRNA4), cholinergic receptor, nicotinic, beta 2 (CHRNB2), cholinergic receptor, nicotinic, alpha 2 (CHRNA2), and potassium channel subfamily T member 1 (KCNT1) were screened by direct sequencing., Results: Two PRRT2 mutations were identified in patients with typical PHD. A mutation of c.649dupC (p.Arg217ProfsX8) was identified in a patient with PHD and his father who was diagnosed with paroxysmal kinesigenic dyskinesia. An additional mutation of c.640G>C (p.Ala214Pro) was identified in a sporadic patient and his asymptomatic mother. No mutations were found in the other screened genes., Conclusions: The present study identified PRRT2 mutations in PHD, extending the phenotypic spectrum of PRRT2 and supporting the classification of PHD as a subtype of paroxysmal dyskinesia but not NFLE. Based on the results of this study, screening for the PRRT2 mutation is recommended in patients with PHD.

Three cruises were carried out in the Yangtze Estuary and its adjacent areas in May, November, June during 2009-2010. The spatial variations of phytoplankton community structure were investigated based on RP-HPLC analysis of pigments and CHEMTAX processing of the pigment data. 21 kinds of pigments were detected, among which chlorophyll a, peridinin, fucoxanthin, 19'-butanoyloxyfucoxanthin, 19'-hexanoyloxyfucoxanthin, chlorophyll b, diadinoxanthin, alloxanthin and zeaxanthin were the major pigments in the Yangtze Estuary and its adjacent areas. Chlorophyll a was the most abundant in all pigments, followed by fuxoxanthin. Other pigments generally contributed a minor proportion to the total pigments. High concentrations of fucoxanthin and peridinin were observed in May 2009 and June 2010, indicating blooms of diatoms and dinoflagellates. The results showed that the composition and distribution of phytoplankton pigments were influenced by environmental factors. The phytoplankton community, as determined by biomarker pigment concentration using HPLC and CHEMTAX, was composed mainly of diatoms, dinoflagellates, cryptophytes, chlorophytes, cyanobacteria, prymnesiophytes, chrysophytes and prasinophytes. The dominant algal groups were diatoms, dinoflagellates and chlorophytes in May 2009. The phytoplankton community was characterized by high contribution of diatoms in November 2009. Diatoms, dinoflagellates and cryptophytes accounted for 62.5% of chlorophyll a in June 2010, and the relative abundance of cyanobacteria was higher in this cruise. The spatial variations of phytoplankton community structure featured distinct regionality. Diatoms, chlorophytes and cryptophytes were the main groups in the inshore waters, and the abundances of prymnesiophytes, chrysophytes and cyanobacteria were increasing from inshore to the open sea.

The asymmetric unit of the title compound, (C(15)H(25)N(2)O(2))[FeCl(4)], contains a tetra-chloridoferrate(III) anion and a oxymatrinium cation [oxymatrine is (4R,7aS,13aR,13bR,13cS)-dodeca-hydro-1H,5H,10H-dipyrido[2,1-f:3',2',1'-ij][1,6]naphthyridin-10-one 4-oxide]. The conformation of oxymatrine is similar to that of matrine with one ring having a half-chair conformation, while the others have chair conformations. Chiral chains of cations along the c axis are formed by O-H⋯O hydrogen bonds.
