Abstract

In order to speed up shale gas exploration and production of Chongqing, the paper discussed construction of shale gas resource database and information retrieval platform. It determined technical methods of establishment of shale gas resource database and retrieval platform, initially identified contents of shale gas resources database and structure, functional requirements and retrieval model of retrieval platform, which provides ideas for platform construction.

47. Restoration of murine femoral segmental defect using CTGF-overexpressing MC3T3-E1 cells

Abstract

Connective tissue growth factor (CTGF) is a member of the CCN super family and is reported to widely participate in bone development and regeneration. This study aimed to restore murine femoral segmental defect using CTGF-overexpressing MC3T3-E1 cells. MC3T3-E1 cells were transinfected by lenti-CTGF (LvCTGF) and lenti-negative control (LvNC) virus to obtain stably transinfected cells. Real-time PCR, Western blot, alkaline phosphatase activity assay, and alizarin red staining demonstrated that the overexpression of CTGF enhanced osteogenesis in vitro. Cell migration assay results showed that LvCTGF cells expressed higher migration ability than LvNC cells, while CCK-8 assay revealed no significant difference in cell proliferation. The LvCTGF and LvNC cells were then seeded into a chitosan/β-TCP scaffold and were used to restore a murine femoral segmental defect. Samples were harvested by the end of 2 and 5 weeks respectively. Micro-CT analysis and Masson's trichrome staining results showed that the LvCTGF-scaffold group expressed better bone healing compared with the LvNC-scaffold and scaffold-only groups. CTGF-overexpressed cells serve as an efficient source of seeding cells for bone regeneration.

50. Inhibition of JAK2 by AG490 promotes TNF-α-induced apoptosis by inhibiting autophagy in MC3T3-E1 cells

Abstract

Tumor necrosis factor-alpha (TNF-α), one of the pro-inflammatory factors in osteoporosis, has a strong enhancement effect on osteoclastogenesis and disruption of osteoblast survival and function. JAK2 participates in a wide range of biological processes, including bone homeostasis, but its function in osteoblast survival in inflammatory environments remains unknown. In this study, flow cytometry and immunofluorescence staining of LC3B were performed under TNF-α stimulation in MC3T3-E1 cells. Apoptosis-related protein Cleaved PARP and autophagy-related protein LC3 were upregulated, meanwhile, p62 was downregulated by TNF-α. JAK2 signaling was also activated in the process. AG490 was used to inhibit JAK2 signaling, which promoted apoptosis and attenuated autophagy induced by TNF-α. Enhancement of autophagy by rapamycin reversed the promotional effect of AG490 on apoptosis, and the autophagy inhibitor chloroquine further enhanced apoptosis. Western blot analysis showed that the STAT3, Akt, and Erk signaling pathways are involved in AG490 treatment. This study demonstrated for the first time that JAK2 inhibition by AG490 may play a crucial role in TNF-α-induced apoptosis by inhibiting autophagy and inhibiting the STAT3, Akt, and Erk signaling pathways.
