BackgroundThe rise of antibiotic-resistant pathogens has intensified the search for novel antimicrobial agents. This study aimed to isolate Streptomyces kanamyceticus from local soil samples and evaluate its antimicrobial properties, along with optimizing the production of bioactive compounds.MethodsSoil samples were collected from local regions, processed, and analysed for Streptomyces strains isolation using morphological characteristics and molecular identification through 16S rRNA gene PCR assay. Antimicrobial activity was assessed against Escherichia coli, Staphylococcus aureus, Bacillus subtilis, and Candida albicans using the double-layer method, while Minimum Inhibitory Concentration (MIC) values were determined. The extracted compounds underwent Fourier Transform Infrared Spectroscopy (FTIR) analysis for functional group identification. Optimization of bioactive compound production was performed using a Central Composite Design (CCD) coupled with Partial Least Squares Regression (PLSR).ResultsA total of 25 distinct Streptomyces strains were isolated, with seven confirmed as S. kanamyceticus. These strains exhibited antimicrobial activity, with inhibition zones reaching 30 mm and MIC values between 20 and 70 µg/mL. The extraction yielded 150-200 mL of bioactive compounds. Optimization studies revealed that a medium containing 10 g/L glucose and 10 g/L glycine max meal maximized antibiotic production.ConclusionThis study confirmed that S. kanamyceticus is a promising source of novel antibiotics. The combination of microbial isolation, antimicrobial testing, and statistical optimization successfully enhanced the production of bioactive compounds, contributing to the search for effective antimicrobial agents against resistant pathogens.

BackgroundThe emergence of ESBLs producing cephalosporin-resistant Escherichia coli isolates poses a threat to public health. This study aims to decipher the genetic landscape and gain insights into ESBL-producing E. coli strains belonging to the high-risk clone ST410 from pediatric patients.Methods29 E. coli ST410 isolates were collected from young children and subjected to antimicrobial susceptibility testing, Whole-genome sequencing (WGS), serotype analysis, MLST, ESBL genes, virulence genes, and plasmid profiling.ResultsAntimicrobial susceptibility testing demonstrated a high level of resistance to cephalosporins followed by aminoglycoside, sulfonamide, carbapenem and penicillin group of antibiotics. However, n=20/29 shows MDR phenotype. Phylogenetic group B2 (n=15) dominated, followed by group D (n=7), group A (n=4), and group B1 (n=3). Serotyping analysis identified O1:H7 (n=8), O2:H1 (n=6), O8:H4 (n=5), O16:H5 (n=4), and O25:H4 (n=3). Other serotypes identified included O6:H1, O15:H5, and O18:H7 (n=1 each). The most commonly detected ESBL genes were blaCTX-M, (n=26), followed by blaTEM (n=23), and blaSHV (n=18). Additionally, blaOXA-1 (n=10), blaOXA-48 (n=5), blaKPC-2 (n=3), blaKPC-3 (n=2), blaNDM-1 (n=4), blaNDM-5 (n=1), blaGES-1 (n=2), blaGES-5 (n=1), and blaCYM-1 (n=3). Notable virulence genes identified within the ST410 isolates included fimH (n=29), papC (n=24), hlyA (n=22), and cnf1 (n=18), among others. Diverse plasmids were observed including IncFIS, IncX4, IncFIA, IncCol, IncI2 and IncFIC with transmission frequency ranges from 1.3X10-2 to 2.7X10-3.ConclusionThe ST410 clone exhibited a complex resistance profile, diverse serotypes, the presence of specific resistance genes (ESBL genes), virulence gene repertoire, and diverse plasmids. The blaCTX-M was the most prevalent ESBL gene detected.

Strategies for reconciling gut microbes through diets, precision nutrition, and clinical indications showcase promising avenues for managing gastrointestinal distress and revolutionizing healthcare. From the Low-FODMAP diet to neuro-gut interventions, these strategies provide a holistic understanding of the gut’s dynamic world. Precision nutrition, as a groundbreaking discipline, holds transformative potential by tailoring dietary recommendations to individual gut microbiota compositions, reshaping the landscape of gastrointestinal health. Recent advancements in clinical indications, including exact probiotics, fecal microbiota transplantation, and neuro-gut interventions, signify a new era where the gut microbiome actively participates in therapeutic strategies. As the microbiome takes center stage in healthcare, a paradigm shift toward personalized and effective treatments for gastrointestinal disorders emerges, reflecting the symbiotic relationship between the human body and its microbial companions.

ABSTRACT: Objectives: Emergence of the plasmid-born mobile colistin resistance (mcr) gene is a growing concern in healthcare. Therefore, this study aimed to genomically characterise multidrug-resistant Escherichia coli and Klebsiella pneumoniae co-harbouring the mcr-1 and mcr-3 genes in young children. Methods: E. coli (n = 3) and K. pneumoniae (n = 2) were collected from abdominal secretions and blood, respectively. The isolates were screened using tryptone soy broth with 4 µL/mL polymyxin-B. Growing bacteria were identified using the VITEK-2 system, matrix-assisted laser desorption/ionisation time-of-flight, and 16s RNA sequencing, followed by antibiotic susceptibility testing. Metallo-β-lactamase (MBL) and extended-spectrum β-lactamase (ESBL) production was also detected. Afterwards, strains were subjected to molecular screening targeting mcr variants and ESBL/MBL-encoding genes. Conjugation, pulsed-field gel electrophoresis, Southern hybridisation, multilocus sequence typing, and phylogenic group detection were performed, along with plasmid-genome sequencing and bioinformatics analysis. Results: E. coli isolates (EC-19–322, 323, and 331) and K. pneumoniae isolates (KP-19–225 and 226) harboured both mcr-1 and mcr-3 genes. These strains were also found to be resistant to more than three classes of antibiotics. The conjugation experiment revealed the presence of mcr-1 and mcr-3 on a single plasmid, and the transmission frequency was 10–2 to 10–3. Both strains were found to be able to produce ESBLs and MBL. E. coli EC-19–322 and 323 were identified as ST131(O25a:H41); SP-19–331, as ST1577 (O16:H30); and K. pneumoniae, as ST231 (K2). All E. coli strains belonged to phylogenetic group B2, and the results of pulsed-field gel electrophoresis supported the multilocus sequence typing findings. Conclusion: This study reported the co-occurrence of mcr-1 and mcr-3 genes on a single plasmid in pathogenic ESBL/MBL-producing E. coli and K. pneumoniae isolated from young children.

With the improvement of coal mining mechanization level, dust has become a serious problem that restricts coal mine safety production and endangers occupational health. Wet dust collectors are widely used because of their low investment and simple structure. In order to further improve the performance of wet dust collector, the overall structure and principle are analyzed in detail, the components, working principle, influencing factors, advantages and disadvantages of the structure of power, dust removal and dehydration are analyzed systematically. On this basis, in view of the existing shortcomings of the existing wet dust collectors, it is proposed that the future should be considered from the perspective of integrity, locality, economy, environmental protection to develop a new type of wet dust collector with integrated structure, suitable for the narrow underground space, diversified functions, low maintenance cost, high dust removal efficiency, sewage and sludge recycling and intelligent management to provide technical support for the realization of safe production and the guarantee of occupational health of operators.

This study presents an innovative home energy management system (HEMS) that incorporates PV, WTs, and hybrid backup storage systems, including a hydrogen storage system (HSS), a battery energy storage system (BESS), and electric vehicles (EVs) with vehicle-to-home (V2H) technology. The research, conducted in Liaoning Province, China, evaluates the performance of the HEMS under various demand response (DR) scenarios, aiming to enhance resilience, efficiency, and energy independence in green buildings. Four DR scenarios were analyzed: No DR, 20% DR, 30% DR, and 40% DR. The findings indicate that implementing DR programs significantly reduces peak load and operating costs. The 40% DR scenario achieved the lowest cumulative operating cost of $749.09, reflecting a 2.34% reduction compared with the $767.07 cost in the No DR scenario. The integration of backup systems, particularly batteries and fuel cells (FCs), effectively managed energy supply, ensuring continuous power availability. The system maintained a low loss of power supply probability (LPSP), indicating high reliability. Advanced optimization techniques, particularly the reptile search algorithm (RSA), are crucial in enhancing system performance and efficiency. These results underscore the potential of hybrid backup storage systems with V2H technology to enhance energy independence and sustainability in residential energy management.

Recently,knowledge base(KB) has been widely adopted to the task of question answering(QA) to provide a proper answer for a given question,known as the KBQA problem.However,knowledge base itself may be incomplete(e.g.KB does not contain the answer to the question,or some of the entities and relationships in the question),limiting the overall performance of existing KBQA models.To resolve this issue,this paper proposes a new model to leverage textual documents for KBQA task by providing additional answers to enhance knowledge base coverage and background information to enhance the representation of questions.Specifically,the proposed model consists of three modules,namely entity and question representation module,document and enhanced-question representation module and answer prediction module.The first module aims to learn the representations of entities from the retrieved subgraph of knowledge base.Then,the question representation can be updated with the fusion of seed entities.The second module attempts to learn a proper representation of the document that is relevant to the given question.Then,the question representation can be further improved by fusing the document information.Finally,the last module makes an answer prediction based on the information of knowledge base,updated question and documents.Extensive experiments are conducted on the WebQuestionsSP dataset,and the results show that better accuracy can be obtained in comparison with other counterparts.

Biofilms are a common survival strategy employed by bacteria in healthcare settings, which enhances their resistance to antimicrobial and biocidal agents making infections difficult to treat. Mechanisms of biofilm-induced antimicrobial resistance involve reduced penetration of antimicrobial agents, increased expression of efflux pumps, altered microbial physiology, and genetic changes in the bacterial population. Factors contributing to the formation of biofilms include nutrient availability, temperature, pH, surface properties, and microbial interactions. Biofilm-associated infections can have serious consequences for patient outcomes, and standard antimicrobial therapies are often ineffective against biofilm-associated bacteria, making diagnosis and treatment challenging. Novel strategies, including antibiotics combination therapies (such as daptomycin and vancomycin, colistin and azithromycin), biofilm-targeted agents (such as small molecules (LP3134, LP3145, LP4010, LP1062) target c-di-GMP), and immunomodulatory therapies (such as the anti-PcrV IgY antibodies which target Type IIIsecretion system), are being developed to combat biofilm-induced antimicrobial resistance. A multifaceted approach to diagnosis, treatment, and prevention is necessary to address this emerging problem in healthcare settings.
