Las enfermedades neurodegenerativas constituyen un grupo heterogéneo de trastornos crónicos que se caracterizan principalmente por la pérdida progresiva de neuronas y la aparición de alteraciones sinápticas1 . Se trata de un conjunto de dolencias cuyas manifestaciones clínicas pueden ser muy variadas, afectan a millones de personas y suponen una de las principales causas de discapacidad a nivel mundial. La enfermedad de Alzheimer (EA) junto con otros tipos de demencia, la enfermedad de Parkinson (EP), la esclerosis lateral amiotrófica, las patologías causadas por priones o la enfermedad de Huntington, son algunas de las más destacadas. A pesar de que existen diferencias considerables entre todas ellas, cada vez tenemos más evidencias de que los mecanismos fisiopatológicos subyacentes pueden ser compartidos. El mal plegamiento y la agregación de distintas proteínas amiloidogénicas, es uno de los principales eventos involucrados en estas patologías. De hecho, estas enfermedades se clasifican desde el punto de vista patológico por la presencia de depósitos anormales de diversas proteínas2 . Así, por ejemplo, sabemos que la -sinucleína es la principal proteína implicada en la EP la demencia por cuerpos de Lewy (DCL) o la atrofia multisistema (AMS) y que la proteína tau está involucrada en la enfermedad de Alzheimer (EA), la parálisis supranuclear progresiva (PSP) o la degeneración corticobasal, entre otras. La agregación de la proteína priónica (PrP), desempeña un papel fundamental en el desarrollo de las patologías priónicas como la enfermedad de Creutzfeldt-Jakob. Sin embargo, estos depósitos no son exclusivos de cada enfermedad y, de hecho, se pueden encontrar de manera simultánea agregados de diferentes proteínas en un mismo paciente3 . Estos mecanismos tampoco son exclusivos de las enfermedades neurodegenerativas ya que también se pueden hallar depósitos anormales de proteínas amiloidogénicas en otras patologías como es el caso de la amilina en el páncreas de pacientes con diabetes mellitus tipo 2 (DM2) o la transtiretina en el miocardio de los que sufren amilodosis sistémica.

The co-administration of glial cell line-derived neurotrophic factor (GDNF) and mesenchymal stem cells (MSCs) in hydrogels (HGs) has emerged as a powerful strategy to enhance the efficient integration of transplanted cells in Parkinson's disease (PD). This strategy could be improved by controlling the cellular microenvironment and biomolecule release and better mimicking the complex properties of the brain tissue. Here, we develop and characterize a drug delivery system for brain repair where MSCs and GDNF are included in a nanoparticle-modified supramolecular guest-host HA HG. In this system, the nanoparticles act as both carriers for the GDNF and active physical crosslinkers of the HG. The multifunctional HG is mechanically compatible with brain tissue and easily injectable. It also protects GDNF from degradation and achieves its controlled release over time. The cytocompatibility studies show that the developed biomaterial provides a friendly environment for MSCs and presents good compatibility with PC12 cells. Finally, using RNA-sequencing (RNA-seq), we investigated how the three-dimensional (3D) environment, provided by the nanostructured HG, impacted the encapsulated cells. The transcriptome analysis supports the beneficial effect of including MSCs in the nanoreinforced HG. An enhancement in the anti-inflammatory effect of MSCs was observed, as well as a differentiation of the MSCs toward a neuron-like cell type. In summary, the suitable strength, excellent self healing properties, good biocompatibility, and ability to boost MSC regenerative potential make this nanoreinforced HG a good candidate for drug and cell administration to the brain.

Parkinson’s disease is characterized by motor and nonmotor symptoms that gradually appear as a consequence of the selective loss of dopaminergic neurons in the substantia nigra pars compacta. Currently, no treatment can slow Parkinson’s disease progression. Inasmuch, there is a need to develop animal models that can be used to understand the pathophysiological mechanisms underlying dopaminergic neuron death. The initial goal of this study was to determine if canine adenovirus type 2 (CAV-2) vectors are effective gene transfer tools in the monkey brain. A second objective was to explore the possibility of developing a large nonhuman primate that expresses one of the most common genetic mutations causing Parkinson’s disease. Our studies demonstrate the neuronal tropism, retrograde transport, biodistribution, and efficacy of CAV-2 vectors expressing GFP and leucine-rich repeat kinase 2 (LRRK2G2019S) in the Macaca fascicularis brain. Our data also suggest that following optimization CAV-2-mediated LRRK2G2019S expression could help us model the neurodegenerative processes of this genetic subtype of Parkinson’s disease in monkeys.

Parkinson's disease (PD) is a progressive CNS disorder that is primarily associated with impaired movement. PD develops over decades and is linked to the gradual loss of dopamine delivery to the striatum, via the loss of dopaminergic (DA) neurons in the substantia nigra pars compacta (SNpc). While the administration of L-dopa and deep brain stimulation are potent therapies, their costs, side effects and gradual loss of efficacy underlines the need to develop other approaches. Unfortunately, the lack of pertinent animal models that reproduce DA neuron loss and behavior deficits-in a timeline that mimics PD progression-has hindered the identification of alternative therapies. A complementary approach to transgenic animals is the use of nonhuman primates (NHPs) combined with the overexpression of disease-related genes using viral vectors. This approach may induce phenotypes that are not influenced by developmental compensation mechanisms, and that take into account the personality of animals. In this review article, we discuss the combination of gene transfer and NHPs to develop "genetic" models of PD that are suitable for testing therapeutic approaches.

The etiology and pathogenesis of Parkinson’s disease (PD) is unknown, aging being the strongest risk factor for brain degeneration. Understanding PD pathogenesis and how aging increases the risk of disease would aid the development of therapies able to slow or prevent the progression of this neurodegenerative disorder. In this review we provide an overview of the most promising therapeutic targets and strategies to delay the loss of dopaminergic neurons observed both in PD and aging. Among them, handling alphasynuclein toxicity, enhancing proteasome and lysosome clearance, ameliorating mitochondrial disruptions and modifying the glial environment are so far the most promising candidates. These new and conventional drugs may present problems related to their labile nature and to the difficulties in reaching the brain. Thus, we highlight the latest types of drug delivery system (DDS)-based strategies for PD treatment, including DDS for local and systemic drug delivery. Finally, the ongoing challenges for the discovery of new targets and the opportunities for DDS-based therapies to improve and efficacious PD therapy will be discussed.

La degeneración lobar frontotemporal engloba tres síndromes diferentes, que comparten características clínicas y patológicas comunes, dificultando así su diagnóstico en estadios iniciales. Se incluyen en este grupo las tres variantes de la demencia frontotemporal, el síndrome corticobasal y el síndrome de parálisis supranuclear progresiva. Se ha llevado a cabo una revisión del perfil neuropsicológico de cada uno de los síndromes, que permita clarificar las características fundamentales que los definen y ayudar a diferenciarlos de otras demencias. Se ha hecho una revisión de los diferentes trabajos publicados en la literatura al respecto, describiendo las características clínicas, patológicas y los hallazgos de imagen fundamentales de cada entidad para describir de manera exhaustiva los hallazgos en los diferentes dominios neuropsicológicos y su progresión. Aunque existe un solapamiento entre los síndromes que conforman la degeneración lobar frontotemporal, la comparación del perfil neuropsicológico de las mismas entre sí y frente a otras demencias permite establecer características propias de su perfil neuropsicológico para llevar a cabo un diagnóstico diferencial. Frontotemporal lobar degeneration encompasses three different syndromes, with clinical and pathologic commonalities, making diagnosis difficult in early stages. Three subtypes are recognized: frontotemporal dementia and its three variants, corticobasal syndrome and supranuclear palsy syndrome. The objective of this study is to review the neuropsychological features of each syndrome in order to differentiate amongst subtypes as well as from other forms of dementia. We review multiple studies from the literature, highlighting the main clinical features, neuropathology and changes in brain imaging of each syndrome. Subsequently, we describe the neuropsychological profile compared to other dementias, and how it progresses over time. Although there is an overlap amongst the different subtypes of frontotemporal lobar degeneration, neuropsychological profiles can help identify subtypes and discriminate frontotemporal lobar degeneration from other forms of dementia.

La enfermedad de Parkinson (EP) es una entidad frecuentre que afecta fundamentalmente a personas de edad avanzada. Los fármacos más utilizados para el control de los síntomas de esta entidad son la levodopa y los agonistas dopaminérgicos. Diversos estudios de tomografía de emisión de positrones (PET) con 18F-fluorodopa y DAT-scan realizados en pacientes tratados han mostrado, que tras 5 años dde tratamiento, la actividad dopa-descarboxilasa y del transportador de dopamina, fue mayor en los pacientes tratados con fármacos agonistas que en los tratados con levodopa. Estos hallazgos podrían indicar que los agonistas dopaminérgicos podrían evitar la pérdida de neuronas nigroestriatales. En contraposición, otros autores no consideran que estos hechos indiquen realmente un efecto neuroprotector, y se inclinan a pensar que se podría tratar de un efecto modulador enzimático inducido por el tratamiento. El objetivo de esta tesis es estudiar las modificaciones funcionales de la vía nigroestriada inducida por la administración crónica de levodopa a monos parkinsonianos. Se incluyeron once monos (Macaca fascicularis) en el estudio. Ocho de ellos recibieron inyecciones semanales de 1-metil-4-feni-1,2,3,6-tetrahidropiridina (MPTP) durante 7 semanas consecutivas para inducirles un parkinsonismo leve. Tras un mes de estabilización, cuatro de ellos fueron tratados con levodopa y otros cuatro con placebo (grupo placebo). El tratamiento con levodopa y placebo se administró durante 11 meses consecutivos. Además, se incluyeron tres animales control, intactos para el estudio histológico (grupo control). Se realizaron estudios de PET de 18F-Fluorodopa a los dos grupos de monos parkinsonianos según el siguiente esquema: 1. Basal, antes del tratamiento con MPTP; 2. Al mes de finalizar el tratamiento con MPTP y antes de iniciar la administración de levodopa; 3. Al día siguiente de terminar el tratamiento con levodopa; 4. Al mes de haber finalizado el tratamiento con levodopa; 5. A los 3 meses de haber finalizado el tratamiento con levodopa; 6. A los 6 meses de haber finalizado el tratamiento con levodopa. Tras la realización del último PET los animales fueron sacrificados y los cerrbros fijados para su posterior análisis histológico. Este análisis incluyó la cuantificación de neuronas tiroina hidroxilasa (TH) positivas en la sustancia negra, neuronas dopaminérgicas estriatales y de la expresión de las enzimas dopadescarboxiladsa (DDC), transportador de dopamina (DAT) y TH mediante Western blot. En el grupo de animales parkinsonianos tratados con levodopa se observó un incremento de la captación de 18F-Fluorodopa en el putamen anterior a los 3 y 6 meses de haber suspendido el tratamiento farmacológico, comparado con el grupo placebo. Ambos grupos de animales mostraron una reducción similar del número de neuronas dopaminérgicas en la sustancia negra, comparado con el grupo control. En cuanto al número de neuronas dopaminérgicas estriatales, se observó un aumento en los monos del grupo placebo con respecto al grupo control. En el grupo de animales tratados con levodopa, el número de estas células fue signigicativamente mayor que en el grupo placebo. Los niveles de DAT en el estriado se redujeron de igual manera en los dos grupos de monos parkinsonianos con respecto al grupo control. En los animales del grupo placebo se observó una disminución significativa del contenido de TH en todas las regiones del estriado con respecto al grupo control. En el grupo tratado con levodopa se observó una reducción significativa de TH en el putamen anterior y posterior. En este grupo de animales se objetivó una reducción mayor de los niveles de TH en el putamen posterior con respecto al grupo placebo. Los niveles de DDC fueron similares en los animales tratados con respecto al grupo control. Este estudio muestra que la administración crónica de levodopa a monos con una denervación parcial de la vía nigroestriada produce modificaciones en la actividad funcional de la misma.

Normalization of neuroimaging studies to a stereotaxic space allows the utilization of standard volumes of interest (VOIs) and voxel-based analysis (SPM). Such spatial normalization of PET and MRI studies requires a high quality template image. The aim of this study was to create new MRI and PET templates of 18F-DOPA and 11C-(+)-α-dihydrotetrabenazine (11C-DTBZ) of the Macaca fascicularis brain, an important animal model of Parkinson's disease. MRI template was constructed as a smoothed average of the scans of 15 healthy animals, previously transformed into the space of one representative MRI. In order to create the PET templates, 18F-DOPA and 11C-DTBZ PET of the same subjects were acquired in a dedicated small animal PET scanner and transformed to the created MRI template space. To validate these templates for PET quantification, parametric values obtained with a standard VOI-map applied after spatial normalization to each template were statistically compared to results computed using individual VOIs drawn for each animal. The high correlation between both procedures validated the utilization of all the templates, improving the reproducibility of PET analysis. To prove the utility of the templates for voxel-based quantification, dopamine striatal depletion in a representative monkey treated with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) was assessed by SPM analysis of 11C-DTBZ PET. A symmetric reduction in striatal 11C-DTBZ uptake was detected in accordance with the induced lesion. In conclusion, templates of M. fascicularis brain have been constructed and validated for reproducible and automated PET quantification. All templates are electronically available via the internet.

Thymidine analogs, including bromodeoxyuridine, chlorodeoxyuridine, iododeoxyuridine, and tritiated thymidine, label dividing cells by incorporating into DNA during S phase of cell division and are widely employed to identify cells transplanted into the central nervous system. However, the potential for transfer of thymidine analogs from grafted cells to dividing host cells has not been thoroughly tested. We here demonstrate that graft-derived thymidine analogs can become incorporated into host neural precursors and glia. Large numbers of labeled neurons and glia were found 3–12 weeks after transplantation of thymidine analog-labeled live stem cells, suggesting differentiation of grafted cells. Remarkably, however, similar results were obtained after transplantation of dead cells or labeled fibroblasts. Our findings reveal for the first time that thymidine analog labeling may not be a reliable means of identifying transplanted cells, particularly in highly proliferative environments such as the developing, neurogenic, or injured brain.

This article reviews the most useful animal models of Parkinson's disease available. Basically they include the model of unilateral nigrostriatal lesion induced by 6-hydroxydopamine (6-OHDA) in rodents and the 1-methy-4-phenyl-1,2,3,6, tetrahydropyridine (MPTP) model of parkinsonism in non-human primates. The major neurochemical and histological alterations found in these two models are also analyzed. Finally, possible applications of these models of parkinsonism are also discussed.

The mechanism involved in dopaminergic neuronal death remains unknown. Increased oxidative stress, inhibition of mitochondrial respiratory chain and apoptosis have been suggested as possible factors mediating cellular death. This article reviews the most important findings reported in parkinsonian brains related to nigral neuronal death.
