13. Dynamic Adhesive Behavior and Biofilm Formation of Staphylococcus aureus on Polylactic Acid Surfaces in Diabetic Environments.

Abstract

Interest in biodegradable implants has focused attention on the resorbable polymer polylactic acid. However, the risk of these materials promoting infection, especially in patients with existing pathologies, needs to be monitored. The enrichment of a bacterial adhesion medium with compounds that are associated with human pathologies can help in understanding how these components affect the development of infectious processes. Specifically, this work evaluates the influence of glucose and ketone bodies (in a diabetic context) on the adhesion dynamics of S. aureus to the biomaterial polylactic acid, employing different approaches and discussing the results based on the physical properties of the bacterial surface and its metabolic activity. The combination of ketoacidosis and hyperglycemia (GK2) appears to be the worst scenario: this system promotes a state of continuous bacterial colonization over time, suppressing the stationary phase of adhesion and strengthening the attachment of bacteria to the surface. In addition, these supplements cause a significant increase in the metabolic activity of the bacteria. Compared to non-enriched media, biofilm formation doubles under ketoacidosis conditions, while in the planktonic state, it is glucose that triggers metabolic activity, which is practically suppressed when only ketone components are present. Both information must be complementary to understand what can happen in a real system, where planktonic bacteria are the ones that initially colonize a surface, and, subsequently, these attached bacteria end up forming a biofilm. Abstract

Deficiencies in the BRCA1 tumor suppressor gene are the main cause of hereditary breast and ovarian cancer. BRCA1 is involved in the Homologous Recombination DNA repair pathway and, together with BARD1, forms a heterodimer with ubiquitin E3 activity. The relevance of the BRCA1/BARD1 ubiquitin E3 activity for tumor suppression and DNA repair remains controversial. Here, we observe that the BRCA1/BARD1 ubiquitin E3 activity is not required for Homologous Recombination or resistance to Olaparib. Using TULIP2 methodology, which enables the direct identification of E3-specific ubiquitination substrates, we identify substrates for BRCA1/BARD1. We find that PCNA is ubiquitinated by BRCA1/BARD1 in unperturbed conditions independently of RAD18. PCNA ubiquitination by BRCA1/BARD1 avoids the formation of ssDNA gaps during DNA replication and promotes continuous DNA synthesis. Abstract

OBJECTIVE: To identify the scientific journals asked by users, not available in Spanish libraries. MATERIAL AND METHODS: We analyzed article applications processed during the years 2012 and 2013 (a period of important budget cuts in many libraries) by the interlending services of four hospitals of the Spanish region Castilla-La Mancha, two of them university hospitals. We identified those article that could not be served through national interlibrary loan, excluding from the sample those that were not served for the following reasons: Erroneous citation or not enough data, user cancellations because of cost or journal being binded at the time of request. RESULTS: Out of a total of 19.159 requests received from our users, we identified 1.033 requests that could not be served, corresponding to 665 journal titles. Problems encountered include: • Old references not covered by current collections • References in rare languages ​​(Chinese, Japanese ...), Only a small core of journals had a certain demand level (there were only 2 with with more that 10 requests). Among them, some titles have been identified that have been rejected by some library and obtained otherwise, usually using the lists of searches. CONCLUSIONS • We believe that the method could be extended to other libraries and would serve to identify certain gaps in our collections that could be solved cooperatively. • Given the dispersion of the titles requested, it could be more advantageous to purchase articles on demand than to subscribe to journals, but we do not have administrative mechanisms that assure us a speedy purchase. • Further updating of collective catalogs is required. • We detected some degree of "variation in practice" that influences the results. • Some requests indicated that users need some training.

34. Optimization of iron oxide nanoparticles for MRI-guided magnetic hyperthermia tumor therapy: reassessing the role of shape in their magnetocaloric effect.

Abstract

Superparamagnetic iron oxide nanoparticles have hogged the limelight in different fields of nanotechnology. Surprisingly, notwithstanding the prominent role played as agents in magnetic hyperthermia treatments, the effects of nanoparticle size and shape on the magnetic hyperthermia performance have not been entirely elucidated yet. Here, spherical or cubical magnetic nanoparticles synthesized by a thermal decomposition method with the same magnetic and hyperthermia properties are evaluated. Interestingly, spherical nanoparticles displayed significantly higher magnetic relaxivity than cubic nanoparticles; however, comparable differences were not observed in specific absorption rate (SAR), pointing out the need for additional research to better understand the connection between these two parameters. Additionally, the as-synthetized spherical nanoparticles showed negligible cytotoxicity and, therefore, were tested in vivo in tumor-bearing mice. Following intratumoral administration of these spherical nanoparticles and a single exposure to alternating magnetic fields (AMF) closely mimicking clinical conditions, a significant delay in tumor growth was observed. Although further in vivo experiments are warranted to optimize the magnetic hyperthermia conditions, our findings support the great potential of these nanoparticles as magnetic hyperthermia mediators for tumor therapy. Abstract

Objective: To identify and quantify scientific information resources, and to describe the present situation of libraries in the Spanish public health system. Methods and Materials: A survey sent to the heads of the health science libraries in the public health system and the virtual libraries in each Health Council. Variables: number of centres, number of users and their distribution by type, number of libraries and their staff, and shared scientific information resources. Results: 14 of the 17 Autonomous Communities have responded. All except Canarias, La Rioja and Comunidad Valenciana. In 14 communities are 201 specialized libraries, with 295 librarians, that serve more than 130.000 professionals in the public health system. The Health Councils have started the development of virtual libraries, which are in different development phases. On 2006 the libraries have subscribed, via shared access through virtual libraries, more than 17 000 journals in electronic format and 36 bibliographic databases. The presence of bibliographic management programs, meta-search engines and linksolvers is still low. Conclusions: The libraries in the public health system are subscribed to a significant volume of scientific information resources, based on the number of centers and users they give service to. A first approximation reveals that most resources overlap between the different communities. A more detailed study could corroborate those results and suggest cooperation systems between communities. The recent development of virtual libraries in the counties is promoting the access for all the health professionals to electronic resources, but would be necessary to evaluate the use of those resources. In spite of these advances, there is still reluctance to subscribe to "next generation" resources, such as meta-search engines or linksolvers.

Abstract

Four tropical transition (TT) events in the North Atlantic basin are simulated with the Weather Research and Forecasting (WRF) and the HARMONIE‐AROME (HAR) models to study the main features of the horizontal kinetic energy (HKE) spectra of these kinds of high‐energetic atmospheric system. Though most of the times similar results are obtained with both models, HAR shows a more intense filtering and numerical dissipation, whereas WRF tends to represent overenergized spectra in the synoptic scale and especially at smaller wavelengths. Predictability is dissimilar for the four TTs studied due to the different spectral curve slope obtained for each case, ranging from unlimited to very poor predictability at synoptic scale. Additionally, an increased HKE is presented in the middle–upper troposphere spectra. A deep analysis of the different terms involved in the equation of the spectral energy budget is presented through a detailed study of one of these TTs. The role of all of them is studied, connecting the energy spectra and the meteorological processes involved. The energy budget terms related to the nonlinear spectral transfer, the three‐dimensional divergence, and diabatic process tendencies are identified as the key ones, whereas the potential and kinetic conversion terms and the vertical flux HKE and pressure divergence terms play a secondary role on modulating the spectrum behaviour. The major energetic contributions are found at the synoptic scale, but results show that a two‐dimensional energy cascade does not fully capture the whole spectrum of a TT. The role of convection, latent heat release, and moist convection outbursts is sketched and a link within different vertical levels is found. Results show that a high‐energetic system, such as a TT, can effectively alter the atmospheric energy behaviour. [ABSTRACT FROM AUTHOR]
