Aim:Response surface methodology (RSM) model was applied to optimize ethanol production from Calabash (Crescentia cujete) pulp juice usingCronobacter malonaticus. Study Design:The Calabash pulp was squeezed with muslin cloth, and vacuum filtered to clear solution before use. The clear juice was tested for reducing sugars using the Dinitrosalicylic acid (DNS) method. Twenty three (23) runs, including 3 controls, of the fermentation was conducted at varying temperatures, pH, and volumes of inoculum. The process parameters (input variables): volumes of inoculum, temperature, and pH were subjected to response surface model, using the Central Composite Design (CCD). Place and Duration of Study:This study was carried out in the Environmental Microbiology Laboratory, University of Port Harcourt for six months. Methodology:Fermentation was done in conical flasks covered with cotton wool and foil in a stationary incubator for four days (96 hours). Active stock culture ofCronobacter malonaticuswas used, with inoculum developed using Marcfaland’s method. Samples were collected every 24 hours, centrifuged, filtered and analyzed for measurement of the output variables: Reducing sugar, cell density and ethanol concentration. Results:The concentration of reducing sugars from Calabash pulp was 3.2 mg/ml. Results obtained also revealed that the fermentation can take place on a wide range of temperature 28-32°C. The optimal pH range for performance ofC.malonaticusfor the fermentation process was pH 5.95-6.5. The optimum volume of inoculum was 10%v/v (i.e. 10 ml in 90 ml juice). The optimized process using the RSM model gave 5.08% v/v bioethanol, being the highest achieved at pH6.08 and 28oC . Conclusion:The bioethanol yield from Calabash substrate is reasonable considering that the bacterium used is not known for ethanol production. Also the concentration of reducing sugars in the substrate and the duration of fermentation could be responsible for the yield.

Aim: This study identified and enumerated microorganisms associated with the composting of some organic wastes using the plate count method Study Design: The wastes were allowed to decompose for 70 days in greenhouse using the modified windrow method of composting. Standard methods were employed to monitor temperature changes in compost piles as well as changes in bacterial and fungal populations. Place and Duration of Study: This study was carried out at in the Agricultural Research Centre of the Federal University of Technology, Owerri, Nigeria. Methodology: The organic wastes namely Poultry Litter (PL), Pig waste (PW), Cow dung (CD) and Source-Separated Municipal Solid Waste (MSW) were composted/co-composted using the windrow method as modified .Sixty kilograms (60) each of PW, PL, CD and MSW were introduced respectively into 100-litre(L) buckets that had previously been perforated at several points. In the co-composted piles, 30 kg of both samples were introduced into the same 100 L bucket that had previously been perforated and then mixed thoroughly. The organic wastes were allowed to decompose at room temperature at a corner of the greenhouse. and initial microbial populations as well as subsequent populations in the compost bins were studied using standard microbiological methods Results: Microbial populations increased concurrently with temperature during the first 3 – 4 weeks of composting except, however, for faecal coliforms and Salmonella. The highest temperature recorded was 60°C for cow dung (CD) compost pile though at maturity the temperature in all compost piles ranged between 27°C to 30°C. The bacterial colony forming units were higher than fungal colony forming units throughout the composting period for both mesopholic and themophilic microorganisms. The population of mesophilic organisms increased in the first 14 – 15 days; for cow dung, the initial total heterotrophic bacteria count (THBC) and total coliform count (TCC) were 2.4 x107 cfu/g and 5.0 x105 cfu/g respectively and increased to 2.5x108 cfu/g and 1.7x107 cfu/g for THBC and TCC, respectively, after the 14th day. Thermophilic bacteria dominated all the composting systems after the 21st day and lasted to the 35th day except for cow dung compost where thermophilic temperatures were still observed on the 45th day with a THBC of 6.3x106 cfu/g on the 49th day. Faecal coliforms and Salmonella were completely eliminated in all the compost systems after the 28th day with temperature values between 47°C – 60°C. Conclusion: Organic wastes when managed properly through the application of knowledge of composting can be transformed into beneficial materials for human and agricultural use.

Marine sediments represent an important sink of harmful petroleum hydrocarbons after an accidental oil spill. Electrobioremediation techniques, which combine electrokinetic transport and biodegradation processes, represent an emerging technological platform for a sustainable remediation of contaminated sediments. Here, we describe the results of a long-term mesocosm-scale electrobioremediation experi- ment for the treatment of marine sediments contaminated by crude oil. A dimensionally stable anode and a stainless-steel mesh cathode were employed to drive seawater electrolysis at a fixed current density of 11 A/m2. This approach allowed establishing conditions conducive to contaminants biodeg- radation, as confirmed by the enrichment of Alcanivorax borkumensis cells harboring the alkB-gene and other aerobic hydrocarbonoclastic bacteria. Moderately thermophilic (T-max, similar to 55 degrees C) methanogens are identified after extended enrichments from temperate, tropical and low-temperature environments. However, thermophilic methanogens with higher growth temperatures (T-opt >= 60 degrees C) are only reported from high-temperature environments. A microcosm-based approach was used to measure the rate of methane production and methanogen community structure over a range of temperatures and salinities in sediment from a temperate estuary. Interpretation of bacteriohopanepolyol (BHP) biomarkers tracing microbiological processes in modern and ancient sediments relies on understanding environmental controls of production and preservation. BHPs from methanotrophs (35-aminoBHPs) were studied in methane-amended aerobic river-sediment incubations at different temperatures. It was found that: 1. With increasing temperature (4-40°C) a tenfold increase in aminopentol (associated with Crenothrix and Methylobacter spp. growth) occurred with only marginal increases in aminotriol and aminotetrol; 2. A further increase in temperature (50°C) saw selection for the thermophile Methylocaldum and mixtures of aminopentol and C-3 methylated aminopentol, again, with no increase in aminotriol and aminotetrol. 3. At 30°C more aminopentol and an aminopentol isomer and unsaturated aminopentol were produced after methanotroph growth and the onset of substrate starvation/oxygen depletion; 4. At 50°C, aminopentol and C-3 methylated aminopentol, only accumulated during growth but were clearly resistant to remineralisation despite cell death. These results have profound implications for the interpretation of aminoBHP distributions and abundances in modern and past environments. For instance, a temperature regulation of aminopentol production but not aminotetrol or aminotriol is consistent with and, corroborative of, observed aminopentol sensitivity to climate warming recorded in a stratigraphic sequence deposited during the Paleocene-Eocene thermal maximum (PETM). N and P are the key limiting nutrients considered most important for the stimulation of crude oil degradation but other trace nutrients may also be important. Experimental soil microcosms were setup to investigate crude oil degradation in the context of Ni amendments. Amended Nickel as NiO, NiCl2, or, a porphyrin complex either inhibited, had no effect, or, enhanced aerobic hydrocarbon degradation in an oil-contaminated soil. Biodegradation was significantly (95% confidence) enhanced (70%) with low levels of Ni-Porph (12 mg/kg) relative to an oil-only control; whereas, NiO (200 and 350 mg/kg) significantly inhibited (36 and 87%) biodegradation consistent with oxide particle induced reactive oxygen stress. Microbial community compositions were also significantly affected by Ni. In 16S rRNA sequence libraries, the enriched hydrocarbon degrading genus, Rhodococcus, was partially replaced by a Nocardia sp. in the presence of low levels of NiO (12 and 50 mg/kg). In contrast, the highest relative and absolute Rhodococcus abundances were coincident with the maximal rates of oil degradation observed in the Ni-Porph-amended soils. Growth dependent constitutive requirements for Ni-dependent urease or perhaps Ni-dependent superoxide dismutase enzymes (found in Rhodococcus genomes) provided a mechanistic explanation for stimulation. These results suggest biostimulation technologies, in addition to N and P, should also consider trace nutrients such as Ni tacitly considered adequately supplied and available in a typical soil.

Marine sediments represent an important sink for a number of anthropogenic organic contaminants, including petroleum hydrocarbons following an accidental oil spill. Degradation of these compounds largely depends on the activity of sedimentary microbial communities linked to biogeochemical cycles, in which abundant elements such as iron and sulfur are shuttled between their oxidized and reduced forms. Here we show that introduction of a small electrically conductive graphite rod ("the electrochemical snorkel") into an oil-contaminated River Tyne (UK) sediment, so as to create an electrochemical connection between the anoxic contaminated sediment and the oxygenated overlying water, has a large impact on the rate of metabolic reactions taking place in the bulk sediment. The electrochemical snorkel accelerated sulfate reduction processes driven by organic contaminant oxidation and suppressed competitive methane-producing reactions. The application of a comprehensive suite of chemical, spectroscopic, biomolecular and thermodynamic analyses suggested that the snorkel served as a scavenger of toxic sulfide via a redox interaction with the iron cycle. Taken as a whole, the results of this work highlight a new strategy for controlling biological processes, such as bioremediation, through the manipulation of the electron flows in contaminated sediments.
