- Abstract
Objective To observe change of proliferation, migration and invasion abilities of pancreatic cancer cells with over-/down-expression of circSTON2, and to preliminarily investigate the biological function of circSTON2 in pancreatic cancer. Methods Human pancreatic cancer cell line PANC-1 was in vitro cultured. CircSTON2 over-expression group, circSTON2 down-expression group, and control group were established. Plasmid transfection and siRNA interference were used to transfect the circSTON2 overexpression plasmid and circSTON2-siRNA interference plasmid and blank control plasmid into the above groups. At 24, 36 and 48 hours of culture, CCK-8 assay was used to detect cell proliferation and Transwell assay was used to detect cell migration and invasion. Results At 24, 36 and 48 hours of culture, cell proliferation ability was as follows: circSTON2 over-expression group > control group > circSTON2 down-expression group (all P<0. 05); the number of migration cells and the number of invasion cells were as follows: circSTON2 over-expression group > control group > circSTON2 down-expression group (all P<0. 05). Conclusions The over-expression of circSTON2 promotes proliferation, migration, and invasion of pancreatic cancer cells. However, the effect of down-expression of circSTON2 is the opposite.
