Digital photogrammetry has attracted widespread attention in the field of geotechnical and geological surveys due to its low-cost, ease of use, and contactless mode. In this work, with the purpose of studying the progressive block surficial detachments of a landslide, we developed a monitoring system based on fixed multi-view time-lapse cameras. Thanks to a newly developed photogrammetric algorithm based on the comparison of photo sequences through a structural similarity metric and the computation of the disparity map of two convergent views, we can quickly detect the occurrence of collapse events, determine their location, and calculate the collapse volume. With the field data obtained at the Perarolo landslide site (Belluno Province, Italy), we conducted preliminary tests of the effectiveness of the algorithm and its accuracy in the volume calculation. The method of quickly and automatically obtaining the collapse information proposed in this paper can extend the potential of landslide monitoring systems based on videos or photo sequence and it will be of great significance for further research on the link between the frequency of collapse events and the driving factors. Monitoring surface displacements of landslides is essential for evaluating their evolution and the effectiveness of mitigation works. Traditional methods like robotic total stations (RTSs) and GNSS provide high-accuracy measurements but are limited to discrete points, potentially missing the broader landslide's behavior. On the contrary, laser scanner surveys offer accurate 3D representations of slopes and the possibility of inferring their movements, but they are often limited to infrequent, high-cost surveys. Monitoring techniques based on ground-based digital photogrammetry may represent a new, robust, and cost-effective alternative. This study demonstrates the use of multi-temporal images from fixed and calibrated cameras to achieve the 3D reconstruction of landslide displacements. The method presented offers the important benefit of obtaining spatially dense displacement data across the entire camera view and quasi-continuous temporal measurement. This paper outlines the framework for this prototyping technique, along with a description of the necessary hardware and procedural steps. Furthermore, strengths and weaknesses are discussed based on the activities carried out in a landslide case study in northeastern Italy. The results from the photo-monitoring are reported, discussed, and compared with traditional topographical data, validating the reliability of this new approach in monitoring the time evolution of surface displacements across the entire landslide area. In this work, we present a numerical study of a granular collapse in a viscous fluid adopting a DEM-PFV (Pore Finite Volume) coupled model. We analyze the time evolution of the collapse and the final morphology of the deposit (triangular or trapezoidal) as a function of the initial solid fraction and column aspect ratio. Finally, we discuss the role of the initial packing fraction of the granular material on the time evolution of the pore pressure. The numerical results are consistent with experimental observation [3] showing the capability of the DEM-PFV coupled model to simulate saturated granular media from triggering to flowing and stopping conditions.

- Abstract
Debris flows are one of the most important hazards in mountainous areas because of their paroxysmal nature, the high velocities, and energy carried by the transported material. The monitoring of these phenomena plays a relevant role in the prevention of the effects of these events. Among different possibilities, fiber optical sensors appear well-suited for this purpose thanks to their fair cheapness (with the exception of the interrogator), the robustness to electromagnetic interferences, the adaptability in extreme harsh conditions (no power supply is required), the possibility of locating the interrogator many kilometers far away from the monitored site, and the unique feature to provide very-dense multipoint distributed measurements along long distances. In this work, the vibro-acoustics signal produced by these phenomena has been focused as a possible source of information for the prediction of incipient movement, and the tracking of their path, velocity and thickness. Few literature works investigate these aspects, and for this reason, a preliminary laboratory and numerical campaign have been carried out with dry granular flume tests on an inclined chute. The discrete element method has been used to simulate the tests and to synthesize the signal measured on an instrumented mat along the channel. The grain shapes of the granular material used in simulations have been obtained by a photogrammetric tridimensional reconstruction. The force-time signal has been also analyzed in time-frequency domain in order to infer the features of the flow. The numerical activity has been preparatory for the experiments carried out by instrumenting the flume with an optical fiber distributed vibration sensing system.

We report that at least 101 of 987 patients with life-threatening COVID-19 pneumonia had neutralizing IgG auto-Abs against IFN-ω (13 patients), the 13 types of IFN-α (36), or both (52), at the onset of critical disease; a few also had auto-Abs against the other three type I IFNs. The auto-Abs neutralize the ability of the corresponding type I IFNs to block SARS-CoV-2 infection in vitro. These auto-Abs were not found in 663 individuals with asymptomatic or mild SARS-CoV-2 infection and were present in only 4 of 1,227 healthy individuals. Patients with auto-Abs were aged 25 to 87 years and 95 were men. Objective: Primary Sjögren's syndrome (SS) is the second most frequent systemic autoimmune disease, affecting 0.1% of the general population. To characterize the molecular and clinical variabilities among patients with primary SS, we integrated transcriptomic, proteomic, cellular, and genetic data with clinical phenotypes in a cohort of 351 patients with primary SS. Methods: We analyzed blood transcriptomes and genotypes of 351 patients with primary SS who were participants in a multicenter prospective clinical cohort. We replicated the transcriptome analysis in 3 independent cohorts (n = 462 patients). We determined circulating interferon-α (IFNα) and IFNγ protein concentrations using digital single molecular arrays (Simoa). Results: Transcriptome analysis of the prospective cohort showed a strong IFN gene signature in more than half of the patients; this finding was replicated in the 3 independent cohorts. Because gene expression analysis did not discriminate between type I IFN and type II IFN, we used Simoa to demonstrate that the IFN transcriptomic signature was driven by circulating IFNα and not by IFNγ protein levels. IFNα protein levels, detectable in 75% of patients, were significantly associated with clinical and immunologic features of primary SS disease activity at enrollment and with increased frequency of systemic complications over the 5-year follow-up. Genetic analysis revealed a significant association between IFNα protein levels, a major histocompatibility (MHC) class II haplotype, and anti-SSA antibody. Additional cellular analysis revealed that an MHC class II HLA–DQ locus acts through up-regulation of HLA class II molecules on conventional dendritic cells. International audience; The interleukin-12 (IL-12) family comprises the only heterodimeric cytokines mediating diverse functional effects. We previously reported a striking bimodal IL-12p70 response to lipopolysaccharide (LPS) stimulation in healthy donors. Herein, we demonstrate that interferon β (IFNβ) is a major upstream determinant of IL-12p70 production, which is also associated with numbers and activation of circulating monocytes. Integrative modeling of proteomic, genetic, epigenomic, and cellular data confirms IFNβ as key for LPS-induced IL-12p70 and allowed us to compare the relative effects of each of these parameters on variable cytokine responses. Clinical relevance of our findings is supported by reduced IFNβ-IL-12p70 responses in patients hospitalized with acute severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection or chronically infected with hepatitis C (HCV). Importantly, these responses are resolved after viral clearance. Our systems immunology approach defines a better understanding of IL-12p70 and IFNβ in healthy and infected persons, providing insights into how common genetic and epigenetic variation may impact immune responses to bacterial infection.

By characterizing gut microbiome composition across 946 healthy donors, Byrd et al. demonstrate strong influences of age and biological sex. Additionally, they define global shifts in the gut microbiome composition of patients with non-gastrointestinal tumors compared with healthy donors., As microbial therapeutics are increasingly being tested in diverse patient populations, it is essential to understand the host and environmental factors influencing the microbiome. Through analysis of 1,359 gut microbiome samples from 946 healthy donors of the Milieu Intérieur cohort, we detail how microbiome composition is associated with host factors, lifestyle parameters, and disease states. Using a genome-based taxonomy, we found biological sex was the strongest driver of community composition. Additionally, bacterial populations shift across decades of life (age 20–69), with Bacteroidota species consistently increased with age while Actinobacteriota species, including Bifidobacterium, decreased. Longitudinal sampling revealed that short-term stability exceeds interindividual differences. By accounting for these factors, we defined global shifts in the microbiomes of patients with non-gastrointestinal tumors compared with healthy donors. Together, these results demonstrated that the microbiome displays predictable variations as a function of sex, age, and disease state. These variations must be considered when designing microbiome-targeted therapies or interpreting differences thought to be linked to pathophysiology or therapeutic response., Graphical Abstract
