Proteoglycans (PGs), which have glycosaminoglycan chains attached to their protein cores, are essential for maintaining the morphology and function of healthy body tissues. Extracellular PGs perform various functions, classified into the following four categories: i) the modulation of tissue mechanical properties; ii) the regulation and protection of the extracellular matrix; iii) protein sequestration; and iv) the regulation of cell signaling. The depletion of PGs may significantly impair tissue function, encompassing compromised mechanical characteristics and unregulated inflammatory responses. Since PGs play critical roles in the function of healthy tissues and their synthesis is complex, the development of PG mimetic molecules that recapitulate PG functions for tissue engineering and therapeutic applications has attracted the interest of researchers for more than 20 years. These approaches have ranged from semisynthetic graft copolymers to recombinant PG domains produced by cells that have undergone genetic modifications. This study objective was to evaluate the hygienic condition of surfaces by microbiological swabs and the effectiveness of disinfectant Virkon S. Level of hygiene was evaluated in a small-capacity slaughterhouse located in the Košice region with a maximum weekly capacity of 5 large livestock units. Microbiological swabs were taken from an area of 10 cm² before the process of slaughtering, during the process, and after disinfection. Disinfectant Virkon S was used in a 1% concentration during an exposure time of 30 minutes for disinfection of monitored surfaces. Disinfectant was effective on surfaces of cage, wall, floor, and (p<0.0001), where was determined significant decrease of microorganisms, but we recorded 1.0 x 104 colony forming units (CFU)/10 cm² of the total count of bacteria (TCB) on the cage, 7.5 x 10³ CFU/10 cm² of TCB on the wall and 1.5 x 104 CFU/10 cm² of TCB on the floor after disinfection, which indicates an insufficient level of disinfection. From the achieved results, it is clear, that the disinfection provided by the disinfectant Virkon S was not effective, as the evaluated disinfectant did not achieve a decrease in the number of microorganisms and thus did not ensure a sufficient level of hygiene. We concluded that it is crucial to effectively disinfect products using the right disinfectant at the right exposure period since it helps to stop the spread of several germs that could contaminate products and have a negative impact on consumers' health. Mastitis is an inflammation of the mammary gland which has a particular importance in ruminants. The purpose of this study was to evaluate and compare two methods for cultivation of udder pathogens: classical laboratory cultivation on Columbia agar with 5% sheep blood versus MicroMast rapid plates. The results allowed assessing the incidence of mastitis and the prevalence of the pathogens. During the study have been investigated 227 cows in a dairy farm localized in the east of Slovakia. Subsequently, 141 quarter milk samples from the positive cows with California mastitis test score 1-4 have been undergone the laboratory culture on Columbia agar and MicroMast test in accordance with their respective steps. The values obtained from these tests showing sensitivity of positive samples using the MicroMast test at the level of 64.5%, and sensitivity of cultures on Columbia blood agar at the level of 61.7%. After biochemical identification of cultured isolates, the Columbia blood agar and MicroMast test identified both as the main pathogen present Staphylococcus aureus (S. aureus) with results of 13.4% and 14.9%, respectively. On the base of results, both tests are comparable and therefore a test that speeds up testing is more useful in practice. Nitrate poisoning due to the consumption of cabbages was diagnosed in a small goat flock, in Qena city, Egypt. The methylene blue 1% antidote (1 mg/kg BW) was given intravenously to the poisoned goats. Nitrate poisoning was confirmed in blood plasma, saliva, urine, and cabbage dry matter samples. The dark brown blood and methemoglobin fraction were 47.25 ± 0.58%. Green cabbage containing 7.1% nitrate on dried materials was fed to the goats. The nitrate concentration in the saliva was (59.50 ± 4.67), compared to the healthy goats reared in the same area and tested positive in the plasma and urine of intoxicated goats. Intoxication induced a significant decline in the Red Blood Cells count, hemoglobin concentration, and HCT % (P < 0.001), while the WBC count (P < 0.001), neutrophils (P < 0.001), and monocyte percentage were elevated (P < 0.002) in intoxicated goats, while the lymphocyte % (P < 0.01) significantly decreased when compared to the control. For the oxidative stress in the red blood cells, nitrate toxicity caused a significant elevation in the concentration of Malondialdehyde, (P < 0.001) while Superoxide Dismutase, Glutathione, and Glutathione S-Transferase levels were significantly reduced (P < 0.001) compared to the control. Simple Summary: The ability of S. aureus to attach to different surfaces and assist with biofilm formation is described in many types of environments, but mainly in the milk processing environment. The development of biofilm is one of the most significant virulence mechanisms involved in the adherence of staphylococcal strains to living or non-living surfaces. Recently, many studies have revealed that biofilms formed in milking equipment may be a source of ongoing S. aureus contamination. The results of this study indicate that S. aureus was the most represented pathogen in milk and on surfaces. In the current study, the biofilm-producing ability of the reference strain and isolates of S. aureus obtained from surfaces and milk was determined. In all strains, the counts necessary for biofilm formation (>5 Log10 CFU/cm2) were detected, except for the reference strain. The ability to produce biofilm from isolates of S. aureus was higher in comparison with the reference strain during the first 3 h. In addition, significant differences between types of environmental contamination—occurrence of S. aureus on the floor, teat cup, and cow restraints, and the frequency of mastitis caused by S. aureus (p < 0.05)—were confirmed, which potentially poses a serious risk of this pathogen persisting in the milking environment and during biofilm formation. This study was conducted to evaluate the incidence of mastitis in 153 dairy cows and to evaluate the kinetics of adhesion of isolates obtained from surfaces and milk in comparison with the reference strain (RS), CCM 4223. The surfaces of the floor, teat cup, and cow restraints were aseptically swabbed in three replicates (n = 27). Of the total number of infected cows (n = 43), 11 samples were found to be positive for Staphylococcus aureus, 12 samples tested positive for non-aureus staphylococci, 6 samples tested positive for Streptococcus spp., and 11 samples tested positive for other bacteria (Escherichia coli, Pseudomonas spp.) or a mixed infection. The most represented pathogen in milk (11/43) and on surfaces (14/27) was S. aureus. The kinetics of adhesion of the reference strain and isolates of S. aureus on stainless steel surfaces were determined after 3, 6, 9, 12, 24, and 48 h, and 3, 6, 9, 12, and 15 days of incubation. All strains reached counts higher than 5 Log10 CFU/cm2 needed for biofilm formation, except RS (4.40 Log10 CFU/cm2). The isolates of S. aureus revealed a higher capability to form biofilm in comparison with RS during the first 3 h (p < 0.001). Thus, there is a significant difference between the occurrence of S. aureus on monitored surfaces—floor, teat cup, and cow restraints—and the frequency with which mastitis is caused by S. aureus (p < 0.05). This finding raises the possibility that if various surfaces are contaminated by S. aureus, it can result in the formation of biofilm, which is a significant virulence factor. [ABSTRACT FROM AUTHOR]

Objective: The purpose of this study was to measure the mean concentrations of heavy metals including aluminum (Al), arsenic, nickel (Ni), mercury, lead (Pb), and cadmium (Cd) and to assess the health hazards due to the exposure of cattle/human population to a distinct or the mixture of heavy metals through various sources. Materials and methods: A total of 180 samples including water sources, animal feed, and raw cows' milk from rural regions in Qena, Egypt, were examined using the inductively coupled plasma emission spectrometer (ICP; iCAP 6200). Results: The data highlighted heavy metal pollution with variable concentrations among most of the investigated regions. All concentrations of Al, Ni, and Cd detected in the feeding stuff showed a strong correlation to their respective levels in milk rather than those detected in water (R2= 0.072 vs. 0.039, 0.13 vs. 0.10, and 0.46 vs. 0.014, respectively) (p < 0.05). Anisocytosis and poikilocytosis with a tendency to rouleaux formation were evident, and basophilic stippling was a pathognomic indicator for heavy metal toxicity, especially Pb. Leukopenia and macrocytic anemia were shown in 50% and 65% of examined cattle, respectively. The target hazard quotients values were more than one (>1) for all heavy metals from water intake for both children and adults and Al and Cd in milk for children, and the hazard index values were indicated higher for noncarcinogenic health hazards. The target cancer risk values predispose people in the surveyed villages to higher cancerous risks due to exposures to the mixture of heavy metal through the consumption of water and milk. Conclusion: The bioaccumulation and transmission of heavy metal mixtures from water sources and feeding material have detrimental influences on milk pollution and cattle health which seem to be a serious issue affecting public health in those rural communities. This study was aimed to explore differences between day and night feeding of farmed European eels (Anguilla anguilla) on: (1) the growth and feeding behaviour of eels; and (2) the subsequent impact on the quality of the culture water. A 30 day field experiment was conducted in which 40 European eels (186.4 ± 9.26 g) were arbitrarily allocated into two replicate concrete ponds (200 x 100 x 125cm) that were divided into two treatment groups, one receiving feed by day (DF) and the other receiving feed at night (NF). Results revealed a significant improvement in eel's growth performance (larger final weight and faster growth) of the fish that were fed by night (NF). Moreover, feeding behaviour was also improved in the NF group with a significantly shorter latency to start feeding and shorter feeding duration. As a consequence of improved feeding behaviour, more favourable water quality conditions (higher dissolved oxygen concentrations and lower ammonia concentrations) were observed in the NF group. In conclusion, feeding eels by night could considerably enhance the culturing of eels by promoting enhanced growth and feeding behavior, and simultaneously improving water quality.
