Spodoptera littoralis is a highly polyphagous pest that attacks numerous important crops in the world and causes substantial economic losses to agricultural production. In the present study, the effects of different leafy vegetables, including Purslane, Chives, Parsley, Basil, Dill, Coriander, and Mint, were investigated on feeding responses and enzymatic activities of S. littoralis under laboratory conditions. Furthermore, the total contents of the three major secondary metabolites (phenolics, anthocyanins, and flavonoids) in the studied vegetables were determined. Our findings showed that the lowest and the highest approximate digestibility were on Basil and Purslane, respectively. The highest values of efficiency of conversion of ingested and digested food were achieved in larvae fed on Chives and Coriander, respectively, whereas the lowest values were recorded after feeding on Purslane. The highest and lowest relative growth rates were in larvae reared on Dill and Purslane, respectively. Furthermore, the highest amylolytic and proteolytic activities were in larvae fed with Coriander and Dill, respectively, while the lowest activities of these enzymes were on Purslane. In addition, correlation analysis revealed significant correlations between feeding characteristics and enzymatic activity of S. littoralis with biochemical compounds of the studied leafy vegetables. Our results suggest that Coriander is a suitable host, while Purslane displayed tolerance traits against S. littoralis, which can be used in sustainable management programs aiming to reduce chemical inputs.

Helicoverpa armigera(Hübner) is considered a serious agricultural pest worldwide. We explored the effects of artificial diets containing ten legumes, including broad beans (Shadan, Feyz, Saraziri, Barekat, and Mahta cultivars), white kidney beans (Dehghan cultivar), red kidney beans (Goli cultivar), common beans (Khomein cultivar), cowpeas (Mashhad and Arabi cultivars) on the feeding responses ofH. armigeraby quantifying specific primary and secondary metabolites in the studied legumes and determining larval nutritional indices and digestive enzyme activities. The results showed that the highest efficiency of conversion of digested food (ECD) and relative growth rate values (RGR) of whole larval instars were obtained in the Dehghan and Goli cultivars. However, the lowest values of ECD and RGR were observed in the larvae fed on the Khomein and Mahta cultivars. The highest proteolytic and amylolytic activities of larvae were found on the Dehghan and Mashhad cultivars. The highest and lowest values of standardized insect-growth index and index of plant quality were observed in larvae feeding on the Dehghan and Khomein cultivars, respectively. Additionally, significant variations in phytochemical metabolites were recorded among the studied legume cultivars. Significant negative or positive correlations were also found between feeding characteristics and enzymatic activities ofH. armigerawith the biochemical composition of the studied legumes. The cluster analysis results revealed that artificial diets containing Mahta and Khomein cultivars were unsuitable forH. armigera, and can be used as candidates for integrated pest management programs or for screening insect inhibitors to produce genetically modified pest-resistant plants.

Helicoverpa armigera (Hübner) is a key polyphagous pest with global spread that causes economic damage to agricultural crops. It is commonly managed by synthetic insecticides, which can cause negative impacts on the environment and human health, as well as toxicity to natural enemies. Hence, investigations into alternative pest management approaches such as the use of resistant host plant cultivars against H. armigera is important. In this study, the effects of various legume seeds based on meridic diets were evaluated using biology and population growth parameters of H. armigera under laboratory conditions. The results indicated that the shortest developmental time and pupal incubation periods occurred on white kidney bean Dehghan, while the longest periods occurred on broad bean Mahta. The intrinsic rate of population increase (r) was highest when H. armigera was reared on white kidney bean Dehghan and cowpea Arabi, and the lowest when reared on broad bean Mahta and common bean Khomein. Furthermore, the highest net reproductive rate (R0) occurred on white kidney bean Dehghan and the lowest on common bean Khomein. Cluster analysis revealed that white kidney bean Dehghan and cowpea Arabi were the most susceptible hosts, while common bean Khomein, and broad beans Feyz and Mahta were the poorest and perhaps least susceptible hosts for population growth of H. armigera. Our results suggest that the selective use of certain bean cultivars as potential sources of antibiosis against this pest may be useful as part of an integrated management program.

Indian meal moth, Plodia interpunctella (Hübner) (Lepidoptera: Pyralidae), is a polyphagous insect pest that causes serious damage to various food crops in storage. This study aimed to investigate the life-history and demographic parameters of P. interpunctella on 5 varieties of date palm fruits (Phoenix dactylifera L.), including Dayri, Estemaran, Fersi, Halavi, and Zahedi under laboratory conditions. Data were analyzed and compared using the age–stage, 2-sex life table. Plodia interpunctella completed its development on all date varieties. The shortest and longest pre-adult periods were recorded on Zahedi (38.47 days) and Estemaran (44.65 days) varieties, respectively. The net reproductive rates (R0) were 82.51, 59.05, 63.61, 102.27, and 114.86 offspring on Dayri, Estemaran, Fersi, Halavi, and Zahedi varieties, respectively. The intrinsic rate of increase (r) were 0.098, 0.085, 0.089, 0.109, and 0.113 day−1 on Dayri, Estemaran, Fersi, Halavi, and Zahedi varieties, respectively. The female fecundity ranged from 133.4 to 259.24 eggs on Estemaran and Zahedi varieties, respectively. The highest mean generation time (T) was obtained on Estemaran (47.984 days), and the lowest value of this parameter was obtained on Zahedi (41.722 days) variety. The results indicated that Zahedi and Halavi varieties were the susceptible hosts for P. interpunctella. In contrast, the Estemaran and Fersi were the most resistant varieties against P. interpunctella, which can be used for integrated management programs to decrease the damage of this pest.

Spodoptera littoralis (Boisd) is globally recognized as a destructive polyphagous insect pest of various crops in the world. It is commonly managed by chemical pesticides, which can cause deleterious effects such as environmental pollution, toxicity to non-target organisms and the emergence of secondary pests. Hence, investigations into alternative pest control strategies such as the use of resistant host plant cultivar against S. littoralis is important. This study aimed to explore the nutritional performance of S. littoralis larvae in dependence on total anthocyanin, flavonoid, and phenol levels across 11 bean cultivars (Phaseolus and Vigna spp.) under laboratory conditions. The results revealed that the Mashhad cultivar accumulated the highest amount of total phenols (13.59 mg ml−1), whereas Yaghout and Arabi cultivars posed the lowest total phenols contents (1.80 and 1.90 mg ml−1, respectively). Across larval instars (third to sixth), the highest consumption index and relative consumption rate were recorded on the Mashhad cultivar. The lowest values of efficiency of conversion of ingested food and the efficiency of conversion of digested food of total larval instars were detected in the larvae which were reared on the Mashhad cultivar. Likewise, the lowest value of the index of plant quality (IPQ) was obtained in the Mashhad cultivar; however, IPQ was figured out at the highest level in the Arabi cultivar. Our findings show that the differential accumulation of secondary metabolites would change the nutritional quality of plants for S. littoralis. Based on the findings, the Mashhad cultivar may serve as a candidate for either integrated pest management or breeding programs aiming at controlling this pest.

Spodoptera frugiperda (J.E. Smith) is an invasive pest in agriculture. It can potentially damage yield resulting severe crop losses and subsequently significant economic damage each year. S. frugiperda is predominantly managed using traditional chemical pesticides. Accordingly, sustainable alternatives such as digestive enzymes inhibitors can be used as an efficient pest management that protects the environment. This contribution aims to examine the pro-region of S. frugiperda trypsin as specific inhibitor of the pest protease enzyme. Structural modeling in conjunction with molecular docking simulations were conducted to design a peptide sequence with the best docking scores and strong binding energy to the target enzyme. The structural models of six pro-peptides were produced based on modification of 7-amino acids of the pro-region of S. frugiperda trypsin. VERIFY_3D, ERRAT, PROCHECK, PROSA and WHAT-IF scores validated the reliability of the predicted model of S. frugiperda trypsin. Molecular docking studies between the six designed inhibitor peptides and the predicted model structure at three different pH conditions were carried out. Data revealed that VPSNPQR at pH 11.0 with the best docking score, the lowest binding energy (ΔG) and dissociation constant (Kd) indicated a potent binding affinity towards S. frugiperda trypsin’s active site. Moreover, the peptide showed a weak potential for interaction with the human trypsin. The results indicated the importance of computational studies in design and selection of inhibitor peptides against target enzymes. Such inhibitors can be used for S. frugiperda control, which can be further applied in other pest management programs. Docking simulations between the pro-peptide inhibitor and Spodoptera frugiperda midgut trypsin confirmed the capacity of the designed pro-region in inhibiting the insect trypsin.

Stethorus&nbsp, gilvifrons is an acarophagous coccinellid distributed in the Mediterranean region and could potentially be mass-reared for the augmentative biological control of Tetranychus&nbsp, turkestani and related species on crop plants. The hypothesis that brine shrimp Artemia&nbsp, franciscana cysts can improve diets for rearing of S. gilvifrons was tested in laboratory experiments. The diet treatments included A. franciscana cysts (D1), A. franciscana cysts plus a vitamin B complex (D2), A. franciscana cysts plus date palm pollen (D3), and A. franciscana cysts plus date palm pollen and Ephestia&nbsp, kuehniella eggs (D4). The results indicated that D1 did not support immature development. D2 supported egg–larval development but not pupal–adult development. Both D3 and D4 supported development to the adult stage and reproduction. However, D4 was the most effective diet, determined by observations of S. gilvifrons oviposition behavior and fecundity. A life table analysis corroborated these results, an intrinsic rate of increase, net and gross reproductive rates, and mean generation time were best for S. gilvifrons fed D4 rather than D3. A mixed diet composed of A. franciscana cysts, date palm pollen, and E. kuehniella eggs can be used to mass rear S. gilvifrons.

Research is evaluating the capacity of Trichogramma parasitoids to control H. armigera populations on field crops. The objective of this research was to determine if young rather than old H. armigera eggs were optimal for Trichogramma&nbsp, euproctidis development, reproduction, and life table parameters. Bioassays involved exposing T. euproctidis mated females to H. armigera 14, 38, or 62 h old eggs within 24 h in laboratory arenas. Results indicated that the number of host eggs parasitized successfully by T. euproctidis decreased as host egg age increased. Host egg age had no significant effect on T. euproctidis adult emergence. Adults that developed in 14 h old eggs had greater longevity and fecundity than those that developed in 38 h or 62 h old eggs. The intrinsic rate of increase (r) was greatest, and the mean generation time (T) was lowest for T. euproctidis reared in 14 h old eggs. This study indicates that young H. armigera eggs are more suitable than old ones for T. euproctidis development and reproduction. This study is important because it provides evidence, for the first time, that T. euproctidis can utilize H. armigera as a rearing host. Using young rather than old host eggs could ensure the persistence of a T. euproctidis mass production system to support augmentative releases.

Background: The ladybird beetle Stethorus gilvifrons (Mulsant) is an important natural enemy of tetranychid mites and functions as a biological control of these plant pests. The development, survival and reproduction of S. gilvifrons were studied when fed on three artificial diets. The components of the artificial diet that S. gilvifrons could be reared successfully on for one generation with no use of tetranychid mites were examined. Methods: Artificial diets consisted of sucrose, honey, royal jelly, agar, yeast, date palm pollen supplemented in different diets with hen&rsquo, s egg yolk (AD1, as basic diet), Ephestia kuehniella Zeller eggs (AD2), or E. kuehniella eggs and 2,4-dihydroxybenzoic acid (AD3). Results: Adults and larvae of Stethorusgilvifrons fed on AD1 had a shorter immature development and preoviposition periods than those fed on AD2 and AD3. The total number of deposited eggs was significantly higher for the females fed on AD3 than on the other diets. The intrinsic rate of increase (r) of S. gilvifrons was highest on AD3, followed by AD2, and AD1. Conclusion: Stethorus gilvifrons performed best on AD3, indicating the potential of this artificial diet for the mass rearing of this important predatory ladybird beetle.

In the present study, trypsin from Plodia interpunctella (Hubner) is characterized to discover sequence, biochemical and structural features. This enzyme is purified by ion exchange chromatography using fast protein liquid chromatography on proteins from fifth-instar larvae. The enzyme is optimally active at 50 °C and pH 11.0. The kinetic parameters (Km and Vmax) of the enzyme are 5.3 ± 0.6 µm and 31 ± 1.3 nmol min−1 mg−1, respectively (using Nα-benzoyl-l-arginine ρ-nitroanilide hydrochloride as substrate). The enzyme is inhibited by the addition of Cu2+ and Mn2+, whereas it is activated by Li+ at high concentrations. Moreover, the enzyme is almost completely inhibited in the presence of Nα-tosyl-l-lysine chloromethyl ketone hydrochloride and phenylmethanesulphonyl fluoride. To understand some characteristics of P. interpunctella trypsin, including active site structure and alkaline pH profile, a reliable structural model of P. interpunctella trypsin is built based on the Fusarium oxisporum (Schlecht) trypsin cystal structure (Protein Data Bank code: 1GDU). The secondary structure content of the purified trypsin from near-ultraviolet circular dichroism data shows considerable similarities with that of P. interpunctella trypsin predicted structure. Analysis of pKa values of active site residues, a type of amino acid residue in the active site cleft and the surface charges of the model and Tribolium castaneum (Herbst) trypsin structure as an insect species from different orders reveals some differences between them. These differences might effect on the microenvironment of the active site cleft and consequently shift its pH profile. The application of multiple theoretical and experimental techniques is well adapted to predict the enzyme structure with high accuracy and this could help in the design of a powerful inhibitor for trypsin with ideal properties.

The inhibitory potential of an inhibitor peptide based on the pro-region of trypsin zymogen was investigated in Indianmeal moth, P. interpunctella, which is a world-wide insect pest of stored food. Five peptides were designed based on molecular docking simulations. The designed peptide with the best score was selected and synthesized for further screening in vitro and in vivo. The peptide was characterized and its inhibitory effects towards the insect trypsin were evaluated and the kinetic analysis revealed a competitive type of inhibition against the target enzyme. The results showed that the peptide could successfully suppress the pest midgut trypsin, and more interestingly, it did not show considerable inhibitory effects on a mammalian trypsin. We also aimed to assess the effect of dietary insect meal treated with different concentrations of the peptide and observed a significant growth and development retardation in pupa and adult insects fed with the inhibitor peptide. The outcomes of the present study suggest an efficient inhibitor peptide that could specifically bind the P. interpunctella trypsin and inhibit its activity, which would be safe against human being health and environment. Notably, this is the first report on in vivo assessment of the direct effect of a pro-region as the specific inhibitor in development as well as survival of the pest insect. Furthermore, our findings could be a promising for future designed pesticides used in pest management.
