5. Antibiotic resistant E.coli in waterborne matrices nearby intensive farming: One Health Hub project.

Abstract

The One Health Hub project aims to investigate the presence of antibiotic resistant (AR) E.coli in 5 irrigation channels and 2 private wells, in a northern Italian rural area, characterized by intensive cattle and pig farms. Water sampling occurred seasonally between 2022 and 2023. E.coli was isolated on a chromogenic agar, biochemically identified and analyzed for phenotipic resistance with the VITEK®2 card AST-N380 (Biomerieux), according to EUCAST thresholds. 17 antibiotics were tested: 3 Penicillins, 7 Cephalosporins (1st, 2nd and 3rd gen), 1 Carbapenem, 4 Aminoglycosides, 1 Fluoroquinolone, and 1 Miscellaneous agent. E.coli ranges from 170 to 10.000 CFU/100ml in 33 water samples. Fifty-nine out of 91 (64.8%) strains were sensitive to all antibiotics, while the remaining 32/91 (35.2%) were resistant at least to 1 antibiotic belonging to all classes, except Carbapenems. Major resistance was found towards Penicillins (18%), followed by miscellaneous agents (10%), Cephalosporins (6%), Fluoroquinolone and Aminoglycosides (4%). MDR (≥3antibiotics) was observed in 20/32 (62.5%) strains and ESBL in 9/91 (10%). In all detected rural channels AR E.coli were found, as well as, in the 2 wells. Notably, in these latter, E.coli was both positive for ESBL and resistant to Cephalosporins of 2nd and 3rd generation. This research clearly demonstrates the phenomenon of the spread of AR in the waterborne matrices, even for new antibiotics. The WHO considers AR as a relevant One Health issue, due to the relationship among human, animal and ecosystem domains. The role of water as a vehicle of AR bacteria is evident, assuming the hypothetical pathway of AR bacteria of both human and animal origin contaminating irrigation channels, and thus the vegetables, or when contaminated wells supply water to food-producing animals. Animal effluents are usually distributed in the nearby fields and the runoff of water during rainfall could contaminate irrigation channels also through this way. Abstract

Aeroelastic interaction in turbomachinery is of prime interest to operators, designers, and aeroelasticans. Operation at off-design conditions may promote blade stall; eventually the stall pattern will propagate around the blade annulus. The unsteady periodic nature of propagating stall will force blade vibration and blade flutter may occur if the stall propagation frequency is entrained by the blade natural frequency. In this work a computational scheme based on the vortex method is used to simulate the flow over a linear cascade of airfoils. The viscous effect is confined to a thin layer, which determines the separation points on the airfoil surfaces. The preliminary structural model is a two-dimensional characteristic section with a single degree of freedom in either bending or torsion. A study of the relationship between the stall propagation frequency and the blade natural frequency has been conducted. The study shows that entrainment, or frequency synchronization, occurs, resulting in pure torsional flutter over a certain interval of reduced frequency. A severe blade torsional amplitude (of order 20 deg) has been computed in the entrainment region, reaching its largest value in the center of the interval. However, in practice, compressor blades will not sustain this vibration and blade failure may occur before reaching such a large amplitude. Outside the entrainment interval the stall propagation is shown to be independent of the blade natural frequency. In addition, computational results show that there is no entrainment in the pure bending mode. Rather, 'de-entrainment' occurs with similar flow conditions and similar stall frequencies, resulting in blade buffeting in pure bending.

Abstract

The genome of Helicobacter pylori encodes for carbonic anhydrases (CAs, EC 4.2.1.1) belonging to the ?- and ?-CA classes, which together with urease, have a pivotal role in the acid acclimation of the microorganism within the human stomach. Recently, in the exoproteome of H. pylori, a CA with no indication of the corresponding class was identified. Here, using the protonography and the mass spectrometry, a CA belonging to the ?-class was detected in the outer membrane vesicles (OMVs) generated by planktonic and biofilm phenotypes of four H. pylori strains. The amount of this metalloenzyme was higher in the planktonic OMVs (pOMVs) than in the biofilm OMVs (bOMVs). Furthermore, the content of ?-CA increases over time in the pOMVs. The identification of the ?-CA in pOMVs and bOMVs might shed new light on the role of this enzyme in the colonization, survival, persistence, and pathogenesis of H. pylori.

Abstract

This is the first study of its kind conducted with the aim to document and conserve the ethnomedicinal knowledge of plants used to cure dental diseases in Buner, Pakistan and to provide starting point for future pharmacological studies about new herbal drugs used for dental disorders. Several field trips were conducted in 2018-19 to collect indigenous knowledge of medicinal plants. A semi-structured questionnaire was used as tool for data collection in individual and group interviews and informants were selected by snowball sampling. In this study 935 men and 323 women were interviewed, yielding information on 55 plant species belonging to 34 families. Lamiaceae and Solanaceae were the dominant plant families used and the main life forms used were herbs (28 species). Leaves were the most used part (19 species). The local population was found to be sensitive and careful about oral hygiene and had rich ethnomedicinal knowledge. [ABSTRACT FROM AUTHOR]

Abstract

Background Drug toxicity currently represents the main challenge of tumour chemotherapy. Our group recently developed a new method for drug delivery inspired by the 'Trojan Horse' concept. Human mesenchymal stem cells (hMSCs) have been shown to play the role of new 'horses' in delivering anti-tumour agents, without involving any genetic manipulation. As human stromal dermal fibroblasts (hSDFs) represent an interesting alternative to hMSCs, being easy to isolate, they could be an ideal candidate for this kind of procedure. Aim To investigate whether hSDFs can take up and deliver paclitaxel (PTX) in sufficient concentrations to inhibit a very aggressive melanoma tumour (IgR39) in vitro. Methods hSDFs were primed with high doses of PTX, and then the effect of drug delivery on IgR39 melanoma proliferation in vitro was evaluated using several assays (antiproliferation, transwell cocultures, rosette assays and colony growth assays). Furthermore, the cell cycle and PTX uptake/release mechanism of hSDFs were studied both under both normal and hypoxic conditions. Results hSDFs incorporated PTX and then released it with unaffected pharmacological activity, inhibiting human IgR39 melanoma growth in vitro. The hypoxic conditions did not induce changes in cell cycle pattern and the uptake-release mechanism with PTX was not affected. Conclusions hSDFs can be used as a Trojan horse, as the released drug was functionally active. These results indicated that these cells could be used for clinical treatment as the drug was released into the cellular environment and the primed cells underwent apoptosis.

Abstract

Item does not contain fulltext BACKGROUND: Vorapaxar is a new oral protease-activated-receptor 1 (PAR-1) antagonist that inhibits thrombin-induced platelet activation. METHODS: In this multinational, double-blind, randomized trial, we compared vorapaxar with placebo in 12,944 patients who had acute coronary syndromes without ST-segment elevation. The primary end point was a composite of death from cardiovascular causes, myocardial infarction, stroke, recurrent ischemia with rehospitalization, or urgent coronary revascularization. RESULTS: Follow-up in the trial was terminated early after a safety review. After a median follow-up of 502 days (interquartile range, 349 to 667), the primary end point occurred in 1031 of 6473 patients receiving vorapaxar versus 1102 of 6471 patients receiving placebo (Kaplan-Meier 2-year rate, 18.5% vs. 19.9%; hazard ratio, 0.92; 95% confidence interval [CI], 0.85 to 1.01; P=0.07). A composite of death from cardiovascular causes, myocardial infarction, or stroke occurred in 822 patients in the vorapaxar group versus 910 in the placebo group (14.7% and 16.4%, respectively; hazard ratio, 0.89; 95% CI, 0.81 to 0.98; P=0.02). Abstract

Mesenchymal stromal cells (MSCs) have been proposed for delivering anticancer agents because of their ability to home in on tumor microenvironment. We found that MSCs can acquire strong anti-tumor activity after priming with Paclitaxel (PTX) through their capacity to uptake and then release the drug. Because MSCs secrete a high amount of membrane microvesicles (MVs), we here investigated the role of MVs in the releasing mechanism of PTX. The murine SR4987 line was used as MSC model. The release of PTX from SR4987 in the conditioned medium (CM) was checked by HPLC and the anti-tumor activity of both CM and MVs was tested on the human pancreatic cell line CFPAC-1. MVs were isolated by ultracentrifugation, analyzed by transmission (TEM) and scanning electron microscopy (SEM), and the presence of PTX by the Fourier transformed infrared (FTIR) microspectroscopy. SR4987 loaded with PTX (SR4987PTX) secreted a significant amount of PTX and their CM possessed strong anti-proliferative activity on CFPAC-1. At TEM and SEM, SR4987PTX showed an increased number of "vacuole-like" structures and shed a relevant number of MVs, but did not differ from untreated SR4987. However, SR4987PTX-derived-MVs (SR4987PTX-MVs) demonstrated a strong anti proliferative activity on CFPAC-1. FTIR analysis of SR4987PTX-MVs showed the presence of an absorption spectrum in the corresponding regions of the PTX marker, absent in MVs from SR4987. Our work is the first demonstration that MSCs are able to package and deliver active drugs through their MVs, suggesting the possibility of using MSCs as a factory to develop drugs with a higher cell-target specificity. © 2014 Elsevier B.V.

Abstract

The prevalence of the internalization-associated prtF1 gene was studied in 837 isolates of Streptococcus pyogenes obtained from 713 pediatric patients presenting with acute pharyngotonsillitis before and after antibiotic therapy. Its association with macrolide resistance and with bacteriological treatment failure was determined. The bacterial population isolated from baseline pharyngeal swabs showed an overall prtF1 positivity rate of 33%. A higher prtF1 positivity was found among erythromycin-resistant strains (45%) showing, however, marked differences between the inducible (iMLS), constitutive (cMLS), and efflux pump (M) resistance phenotypes. The prevalence was statistically higher (p < 0.001) in strains belonging to iMLS (84%) and cMLS (67%) phenotypes as compared to the M phenotype (15%). Interestingly, the prevalence of the prtF1 gene was significantly lower (p = 0.04) in strains belonging to M resistance phenotype as compared to erythromycin-susceptible strains (28%). Failed bacterial eradication was demonstrated in 124 patients. The prtF1 positivity rate remained unchanged in strains isolated before and after therapy in patients treated with macrolides (9/54). On the other hand, the positivity rate for the prtF1 gene was significantly higher (p = 0.015) in strains isolated after therapy with beta-lactams (21/70) as compared to baseline isolates (6/70), indicating a differential selection imposed on the organism by these agents. Finally, a high overall eradication rate (88%) of prtF1-positive isolates, belonging to both the erythromycin-susceptible and -resistant phenotypes, was demonstrated following macrolide treatment

Abstract

The fundamental and lowest frequency natural modes in a cantilever fan blade exhibit significant amounts of flexure and torsion coupled by pretwist and operation in a rotational force field. Consequently the flutter estimation of such blades requires an accurate structural description that incorporates these two effects, amongst others. A beam-type finite element model is used in this study with up to six spanwise elements, each element being pretwisted. Coalescence-type flutter is found with subsonic aerodynamics. Evidence of the aerodynamic resonance phenomenon is exhibited and the importance of including radially varying aerodynamic forces is brought out.

The structural dynamics of a cantilever turbomachine blade mounted on a spinning and precessing rotor are investigated. Both stability and forced vibration are considered with a blade model that increases in complexity (and verisimilitude) from a spring-restrained point mass, to a uniform cantilever, to a twisted uniform cantilever turbomachine blade mounted on a spinning and precessing rotor are investigated. In every instance the formulation is from first principles using a finite element based on beam theory. Both ramp-type and periodic-type precessional angular displacements are considered. In concluding, forced vibrating and flutter are studied using the final and most sophisticated structural model. The analysis of stability is presented and a number of numerical examples are worked out.

Abstract

A finite element method of discretizing beam segments of pretwisted rotating blades is presented. Employing the matrix displacement method, stiffness and mass properties are developed from basic mechanics of a pretwisted beam theory. By introducing the proper displacement functions, the effect of rotor blade rotational motion on the stiffness matrix is obtained systematically from the kinetic energy expression. Comparing with other beam elements the derivation of this element is more fundamental. This allows one to apply the same approach to more complicated problems including nonlinear effects or complex dynamic motions. Illustrative examples are given comparing numerical results with available data and other numerical solutions from rotating and nonrotating force fields. These examples show that accurate prediction of vibration frequencies for pretwisted blades can be obtained by employing a quite modest number of degrees of freedom.

The question of divergent oscillatory stability of an axial turbomachine blade when the bladed rotor system is subjected to a forced precessional motion is studied by using two simplified models of the blade. The simplest (lumped) model is a point mass restrained by three orthogonal linear springs attached to the rotor. An improved distributed parameter model represents the blade by a uniform cantilever beam with bending in one plane. In both instances instability is shown to be strongly dependent upon the ratio of blade root radius to blade length and the magnitude of the rotor spin relative to the nonrotating blade natural frequency for realistic values of precession rate. Results are presented on augmented Campbell diagrams. Implications for design and operation are drawn as are the characteristics of needed future studies.

40. Adipose Tissue-Derived Stromal Cells Primed in Vitrowith Paclitaxel Acquire Anti-Tumor Activity

Abstract

Many strategies, including those based on genetically modified Mesenchymal Stromal Cells (MSCs), have been developed in recent years in order to obtain high concentrations of anticancer drugs effective on tumor mass. In previous studies, we showed that human and murine bone marrow-derived MSCs (BM-MSCs) and human skin-derived stromal fibroblasts (hSDFs) acquired strong anti-tumor capacity, both in vitroand in vivo, once primed with Paclitaxel (PTX). In this report we investigate whether adipose tissue-derived MSCs (AT-MSCs) behave similarly to BM-MSCs in their uptake and release of PTX in sufficient amounts to inhibit tumor proliferation in vitro. According to a standardized procedure, PTX primed AT-MSCs (AT-MSCsPTX) were washed and then subcultured to harvest their conditioned medium, which was then tested to evaluate its in vitroanti-tumor potential. We observed that AT-MSCsPTX were able to uptake PTX and release it in a time-dependent manner and that the released drug was active in vitroagainst proliferation of leukemia, anaplastic osteosarcoma, prostatic carcinoma and neuroblastoma cell lines. These data confirm that AT-MSCs, as well as BM-MSCs, can be loaded in vitrowith anti-cancer drugs. While the harvesting of BM-MSCs requires invasive procedures, AT-MSCs can be prepared from fat samples taken with little patient discomfort. For this reason, this source of stromal cells represents an important alternative to BM-MSCs in developing new tools for carrying and delivering anti-cancer drugs into tumor microenvironments.

Abstract

The electron temperature of a large diameter (8 cm)Qmachine has been increased by a factor of 3 by electron cyclotron resonance heating. Using this technique we have eperimentally studied the effect of electron temperature on coherent flute modes driven by a strong radial electric field. We have found a strong stabilizing effect, as shown by the relationship between the threshold voltage required to excite the instability and the electron temperature. In fact the threshold seems to be proportional to a positive power of temperature. La temperatura degli elettroni di una macchinaQdi grande diametro è stata aumentata di un fattore 3 sfruttando la risonanza ciclotronica degli elettroni. Mediante questa tecnica si è studiato sperimentalmente l’effetto della temperatura elettronica sulla stabilità dei modi di oscillazione generati da un intenso campo elettrico radiale. Si è trovato un forte effetto stabilizzante, come si vede dalla relazione fra la tensione di soglia necessaria per eccitare tali instabilità e la temperatura elettronica. Infatti la soglia risulta proporzionale ad una potenza positiva della temperatura. 42. The Reversed Brayton Cycle Heat Pump—A Natural Open Cycle for HVAC Applications

Using reasonable assumptions for turbomachinery efficiencies and heat exchanger effectivenesses, the Coefficient of Performance (COP) and Specific Heating Effect (SHE) of the regenerated and unregenerated cycles are computed and optimized. Only heating data are presented and both open and closed cycles are considered. An effort is made to indicate those cases where (the working fluid being air) the cycle may be appropriately open to combine the heating function with ventilation. When driven by an appropriately small direct Brayton cycle prime mover, the total potential for a new and widespread gas turbine application of literally millions of units becomes defined.

44. Vortex Simulation of Propagating Stall in a Linear Cascade of Airfoils

Abstract

A numerical simulation of propagating stall in a linear cascade of airfoils at high Reynolds numbers is conducted using a vortex method which was first developed by Spalart [7] for this problem. In this approach, the vorticity is discretized into a large collection of vortex blobs whose motion is tracked in time by the use of a well-known vortex tracing algorithm based on the Euler equation. The near-wall effects of viscosity are accounted for by the creation of discrete vortex sheets at the boundaries of the airfoils consistent with the no-slip condition. These boundary vortices are then released into the flow field downstream of the separation points which are obtained from a boundary-layer routine. Calculations are presented for a variety of flow geometries. It is demonstrated that (for a given cascade of airfoils, disturbance wavelength, and stagger angle) several different flow regimes are obtained: Attached flow at lower angles of attack and a chaotic deep stall configuration at larger angles of attack with a narrow intermediate range of such angles where propagating stall occurs. The physical characteristics of this propagating stall are parameterized and a quantitative study of the effects of camber and imposed wavelength is conducted. Comparisons are made with previous theoretical and experimental studies.

Abstract

The “time marching” technique is successfully applied to the numerical computation of the nonstationary aerodynamics of a flat plate cascade for compressible flow of either subsonic or supersonic nature. The unsteady perturbation amplitudes of fluid properties are used as the dependent variables so that the computational domain can be reduced to a two-dimensional channel guided by two adjacent blades for any interblade phase angle. A new method of handling the boundary condition is developed in which the order of accuracy for the boundary points will be the same as the interior points. The wake region behind the trailing edge of each blade is treated as a “slip plane” as done in two-dimensional steady state supersonic flow. Results are in good agreement with existing analytical solutions.

Abstract

The objectives of this research program were to measure the unsteady flow field in the rotor passage of an axial compressor and map this velocity field when operating near rotor blade stall. A laser doppler velocimeter system was developed for this purpose and used in conjunction with a single stage compressor designed expressly to facilitate these measurements. An extensive data acquisition system utilizing microcomputer components was designed and partially assembled an data processing algorithms were developed to yield the velocity mapping from the data. Various program sub-elements were completed, but a successful on-rotor velocity realization was not achieved at contract termination. Propagating stall was observed on the compressor and modelling studies using simulated inputs to the data processing system indicated a potential for successful unsteady velocity field mapping.
