Abstract

Solid state fermentation (SSF) is a vital processing method used for the reduction of postharvest losses, generation of value-added products, and improvement of quality attributes of foods. In this study, the effect of inoculum size on solid state fermentation of plantain by Rhizopus oligosporus was investigated. The results revealed that the solid state fermentation of plantain by R. oligosporus at various inoculum sizes (0 - 30%) significantly (p < 0.05) reduced pH and increased the α-amylase activity, glucose and soluble proteins levels of plantain. Fermentation markedly improved the availability of glucose levels and soluble proteins of plantain. It not only improved its nutritional value but also suggested that plantain is a cheap and readily available substrate that can be used in αamylase production for various industrial applications.

34. Isolation, identification and in silicoanalysis of alpha-amylase gene of Aspergillus nigerstrain CSA35 obtained from cassava undergoing spoilage

Abstract

In this investigation, a gene (CDF_Amyl) encoding extracellular α-amylase in Aspergillus nigerstrain CSA35 associated with cassava spoilage was amplified using specific primers and characterized in silico. The gene had a partial nucleotide sequence of 968 bp and encoded a protein of 222 aa residues with a molecular weight and isoelectric point of 25.13 kDa and 4.17, respectively. Its catalytic site was located in the active site domain. BLASTp analysis showed that the protein primary sequence of the α-amylase gene had 98% and 99% homologies with the α-amylase of A. nigerand A. oryzaeRIB40,respectively. The gene is more closely related to α-amylase genes from fungi than to bacterial, plant, or animal α-amylase genes. Restriction mapping of the gene showed it can be digested with restriction enzymes like NcoI, PstI, SmaI, and BcLI among others but not with EcoRI and EcoRV. Its protein product had a hydrophobicity score of − 0.43 but no transmembrane helix. The CDF_Amyl protein was subcellularly localized in the secretory pathway, an indication of its release into extracellular space after secretion. Also, the 3D structure of the CDF-Amyl protein was barrel-shaped with domains characteristic of α-amylases. The encoded α-amylase Vmax is 6.90 U/mg protein and Km is 6.70 mg/ml. It was concluded that the unique characteristics of the CDF_Amyl gene and its deduced protein could find applications in biotechnological, food and pharmaceutical industries where cloning and further modification of this gene would be required for product development and improvement.

Abstract

α-Amylase is a foremost hydrolytic enzyme used for various industrial processes. In the present study, Rhizopus oligosporus α-amylase (ROAA) produced during solid state fermentation of cassava (Manihot esculenta) tubers by the fungus was partially purified and biochemically characterized. The kinetic and thermal inactivation thermodynamics properties of the partially purified enzyme were also evaluated as well as the potential application in cloth washing. The ROAA was purified by 4.11-fold and had a molecular weight of 60 kDa. ROAA was optimally active at 60 °C, pH 5.0 and had a salt tolerance of 1–100 mM NaCl for 120 min. The activity of the α-amylase was significantly (p < 0.05) increased in the presence of various inorganic salts especially at 10 mM. Ethylenediaminetetraacetic acid (EDTA), urea and thiourea enhanced ROAA activity. Triton X-100 enhanced ROAA activity by 141.97%, whereas sodium dodecyl sulphate (SDS) decreased it by 28.58%. Tween 20 and Tween 80 did not significantly affect ROAA activity at 1 mM. V max , K m and K cat of ROAA were 37.59 U/mg protein, 16.39 mg/mL and 0.016 s−1, respectively. Thermal stability indicators such as half-life, D-value at 70 °C and Z-value were 101.91 min, 338.70 min, and 48.78 °C, respectively. Cloth washing efficiency of a mixture of the isolated enzyme and commercial detergents was high relative to detergent-only trial. The enzyme had high thermal stability, optimized activity in the presence of some inorganic salts, and resistance to several surfactants and a chelating agent. The fungal α-amylase reported in this study would find relevance not only in starch-based industries but also in laundry. Abstract

The CREA gene has been implicated in glucose repression in several fungi. The product of this gene, CreA, binds to the promoter region of several enzymes and down-regulates gene expression. An ortholog of CREA was isolated and characterized from the maize pathogenic fungus, Cochliobolus carbonum . The deduced amino acid sequence of the C. carbonum CREA gene is very similar to the CreA proteins of Aspergillus niger , Gibberella fujikuroi , Sclerotinia sclerotiorum and Trichoderma reesei , as well as the Mig1 protein of Saccharomyces cerevisiae . And like the other fungal proteins, C. carbonum CreA has two zinc finger regions and a nuclear localization signal. Putative CreA binding sequences were also identified in the 5′ region of three C. carbonum cell wall degrading enzyme genes suggesting that the protein may play a role in the regulatory process that controls these enzymes expression. 40. Role of Anthocleista vogeliiin serum antioxidant defence system in cadmium-induced oxidative stress in Wistar rats

Abstract

Background: Cadmium (Cd) toxicity, which runs across the food chain, is chiefly regulated by in vivo antioxidant defence system or through antioxidant supplementation of biological systems predisposed to this environmental stressor. The present study was designed to examine the role of Anthocleista vogeliileaves in Cd-induced oxidative stress in the serum of Wistar rats through the application of response surface methodology (RSM) and biomonitoring of selective responses: malondialdehyde (MDA), catalase (CAT), superoxide dismutase (SOD), glutathione-s-transferase (GST) and peroxidase (POD) activities, respectively. The cold macerated plant leaves were subjected to fractionation process using methanol-hexane-chloroform (3:2:1?v/v) solvent system such that n-hexane fraction with ample antioxidant levels in terms of total phenolic content (TPC) and total flavonoid content (TFC) among others at p?&lt;? 0.05 was selected for the study. The study employed central composite design (CCD) with twenty experimental “runs” of male Wistar rats for twenty-eight days, following a week of acclimatization, where n-hexane fraction of A. vogelii(NFAV), cadmium chloride (CdCl2) and body weights of rats were considered input factors in the study. Results: The study generated five quadratic models, which differed significantly at p?&lt;? 0.05 for MDA levels as well as CAT, SOD, GST and POD activities in the sera of Wistar rats. The study revealed that exposure to Cd toxicity caused a marked increase (p?&lt;? 0.05) in serum MDA levels, but a significant inhibition (p?&lt;? 0.05) of serum SOD, CAT, GST and POD activities. However, Cd interaction with NFAV showed marked amelioration of Cd-induced oxidative stress, which was confirmed by significant decrease in serum MDA levels, but significant increase in serum SOD, CAT, GST and POD activities at p?&lt;? 0.05 via the response surface plots. The study also confirmed the reliability and adequacy of the models for accurate prediction of the responses since R-squared (R2) values obtained were greater than 90%. Conclusion: It was inferred from the present study that the adequacy of the models validated the potency of A. vogeliileaves graphically in the amelioration of Cd-induced oxidative stress in the serum of Wistar rats. Hence, the plant was considered a rich source of bioactive compounds with significant antioxidant properties.
