WOS: 000435647500033, Aim: The aim of this study is to examine the effects of orally administered palmatine on periodontium in an experimental periodontitis (EP) rat model. Material and Method: A total of 40 rats were divided into 4 groups as group 1, no EP and no palmatine (CTRL); group 2, EP and no palmatine (EP); group 3, EP and 5mg/kg of palmatine (EP-5P); group 4, EP and 10mg/kg palmatine (EP-10P). Experimental periodontitis was achieved by plating 3.0 silk sutures on the cervical margins of the right mandibular first molar teeth of the rats. The alveolar bone loss, attachment loss and alveolar bone area were evaluated by histomorphometric evaluation. Immuno-histochemical evaluation was used to assess the Receptor activator of nuclear factor kappa-B ligand (RANKL) ve osteoprotegerin (OPG) immunoreactivity and RANKL/OPG ratio. Results: Alveolar bone area (ABA) values in palmatine groups were statistically higher than EP group (p0.05). According to the RANKL / OPG ratio, a significant decrease was observed in the palmatine-treated groups during the experimental period (p0.05). Discussion: Palmatine might have protective effects on alveolar bone loss and connective tissue destruction in periodontal disease.

Material and Method: A critical-size defect of 5-mm diameter was created in the calvarium of twenty-four rats. The animals were divided into three groups: group C (passive control), group XBG (treated with Bio-Oss), and group P-XBG (treated with puerarin + Bio-Oss combination). All animals were euthanized at 28 days postoperative. Stereologic analyses were performed. New bone area and connective tissue volumes were measured. Results: Mean new bone volumes were 1.57 +/- 0.13mm3, 1.24 +/- 0.10 mm3, and 0.31 +/- 0.09 mm3 in groups P-XBG, XBG and C respectively. Material and Method: Twenty-four rats were divided into three groups: Group C (passive control), Group G (active control) and Group G+PLM. A 5-mm diameter critical-size defect was created in the calvarium of each animal. In Group C, the defects were left empty. In Group G defects were filled with only autogenous graft and an absorbable collagen sponge treated with 300 mg sterile saline solution was applied on grafted area. In Group G+PLM defects were filled with autogenous graft, and 300 mg palmatine treated absorbable collagen sponge was applied on grafted area. All animals were euthanized at 28 days postoperative. Stereologic analyses were performed. New bone volume and connective tissue volumes were measured. Results: Stereologic analysis showed that Group G and G+PLM significantly had more new bone at four weeks compared with group C. Connective tissue volumes were also significantly higher in autografted groups. New bone and connective tissue volumes' difference were not statistically significant between group G and G+PLM groups. Discussion: Locally administered 500 mg palmatine doesn't enhance bone regeneration in critical size calvarial rat defects filled with autologous graft.
