This study aims to investigate the feasibility of including silt, a by-product of limestone aggregate production, as a filler in geopolymer cement. Two separate phases were planned: The first phase aimed to determine the optimum calcination conditions of the waste silt obtained from Società Azionaria Prodotti Asfaltico Bituminosi Affini (S.A.P.A.B.A. s.r.l.). A Design of Experiment (DOE) was produced, and raw silt was calcined accordingly. Geopolymer cement mixtures were made with sodium or potassium alkali solutions and were tested for compressive strength and leaching. Higher calcination temperatures showed better compressive strength, regardless of liquid type. By considering the compressive strength, leaching, and X-ray diffraction (XRD) analysis, the optimum calcination temperature and time was selected as 750 °C for 2 h. The second phase focused on determining the optimum amount of silt (%) that could be used in a geopolymer cement mixture. The results suggested that the addition of about 55% of silt (total solid weight) as filler can improve the compressive strength of geopolymers made with Na or K liquid activators. Based on the leaching test, the cumulative concentrations of the released trace elements from the geopolymer specimens into the leachant were lower than the thresholds for European standards.

Every year, up to 3 billion tons of non-renewable natural aggregates are demanded by the construction sector and approximately 623 million tons of waste (mining and quarrying) was produced in 2018. Global efforts have been made to reduce the number of virgin aggregates used for construction and infrastructure sectors. According to the revised waste framework directive in Europe, recycling at least 70% of construction and demolition waste materials by 2020 was obligatory for all member states. Nonetheless, quarries must work at full capacity to keep up with the demands, which has made quarry/mining waste management an important aspect during the past decades. Amongst the various recycling methods, quarry waste can be included in cement mortar mixtures. Thus, the current research focuses on producing cement mortars by partially substituting natural sand with the waste silt obtained from the limestone aggregate production in S.A.P.A.B.A. s.r.l. (Italy). A Design of Experiments (DOE) method is proposed to define the optimum mix design, aiming to include waste silt in cement mortar mixtures without affecting the final performance. Three cement mortar beams were produced and tested for each of the 49 randomized mixtures defined by the DOE method. The obtained results validate the design approach and suggest the possibility of substituting up to 20% of natural sand with waste silt in cement mortar mixtures.

Carbon footprint reduction of paving materials could be explored through recycling mining by-products into different applications, which will preserve natural resources and decrease environmental issues. One possible approach is to reuse quarry dust and mining ore waste as precursors in geopolymer applications. geopolymers are mineral polymers rich in aluminosilicates with an amorphous to a semi-crystalline three-dimensional structure. The current review aims to summarize the studies conducted during the past decade on geopolymers containing quarry dust and mine tailings. The first section discusses various precursors used for geopolymer cement production such as metakaolin, ground granulated blast furnace slag (GGBFS), fly ash, and quarry/mining ore wastes including silt, tungsten, vanadium, copper, gold, zinc, marble, iron, basalt, and lithium. Different calcination treatments and curing conditions have been summarized. In some cases, the precursors are required to be calcined to increase their reactivity. Both ambient temperature and elevated temperature curing conditions have been summarized. Less attention has been paid to room temperature curing, which is necessary for field and industrial implementations. Engineering properties such as compressive strength, density, durability and acid resistance, water absorption and abrasion of geopolymers containing mining waste were reviewed. One of the main barriers preventing the widespread use of waste powders, in addition to economic aspects, in geopolymers could be due to their unstable chemical structure. This was shown through extensive leachate of Na + or K + cations in geopolymer structures. The review of over 100 articles indicated the need for further research on different aspects of quarry waste geopolymer productions before its full industrial implementation.

Kambo is a substance obtained from the skin secretions of a frog, Phyllomedusa bicolor, popular in the Amazon region, which is administered via the transdermal route. We report a case of 42-year-old man found dead in his house. Near the corpse, a plastic box labeled as "Kambo sticks" was found. The man was a chronic consumer of Kambo and no previous pathology or genetic disease emerged in clinical history from the declaration of his general practitioner. Autopsy investigations and toxicological analysis were performed. The histopathological examination showed left ventricular hypertrophy. Toxicological screening was negative for ethanol and other drugs. Phyllocaerulein, phyllokinin, and deltorphin A were isolated from the Kambo sticks but, only deltorphin A was detected in blood sample. We describe the first forensic case of death associated with Kambo administration. HBV vaccine needs 3 injections over 6 months to induce immunity. Thus, the use of adjuvants capable of inducing earlier immune protection would be highly desirable. Most adjuvants may act by inducing cytokines, and among them, type I interferons (IFNs), deserve a special attention in view of the potent immunomostimulatory activity observed in mouse models and on dendritic cell functions. The aim of the present trial was to evaluate the effects of IFN-alpha administered as an adjuvant of HBV vaccine in healthy unvaccinated individuals. No significant enhancing effect on the antibody response was observed, in spite of an early and transient upregulation of costimulatory molecule expression on peripheral blood mononuclear cells, which may be suggestive of an IFN-mediated activation of antigen presenting cells. We conclude that, under the conditions used in this trial, natural IFN-alpha does not act as an adjuvant of the HBV vaccine in healthy unvaccinated individuals.

About 30 Sendai Virus (SV) preparations, examined for their capacity to induce natural human interferon alpha from fresh human leukocytes (Le-IFN-alpha) of healthy donors, were characterized for hemagglutinating (HA) and hemolytic (HemA) activities and for SDS-PAGE proteic pattern. The SV preparations were produced by a single passage or by serial propagations through eggs in different conditions of multiplicity of infection (m.o.i.). The produced SV subpopulations showed variable IFN-inducing capacity, the values of which are distributed over a 6-fold range resembling a Gaussian distribution. No detectable difference was observed comparing the SV preparation obtained by serial propagations with those obtained by a single passage. The variability of the measured HA activity and HemA activity and as well as the SDS-PAGE proteic pattern of the SV preparations did not correlate with the induced amount of IFN per cell. In the same experimental conditions to produce Le-IFN-alpha, u.v.-treated SV preparations were unable to induce interferon depending on the u.v.-treatment. So it can be concluded that the distinct nHu-IFN-alpha-inducing capacity of SV subpopulation could be mainly associated with divergent compositions of the viral RNAs rather than with a different contents of viral proteins, among those detectable in SDS-PAGE and those responsible for HA activity and for HemA activity.

Benzene is a ubiquitous environmental pollutant primarily metabolized by a cytochrome P-450 (CYP-450) isoenzyme, CYP-450 IIE1. A consistent induction of CYP450 IIE1 has been observed in both rat and human affected by diabetes mellitus. The aim of this study was to evaluate whether streptozotocin (STZ)-induced diabetes determines modifications in the metabolic pathways of benzene in rat. Benzene (100 mg/kg per day, dissolved in corn oil) was administered i.p. once a day for 5 days. Urine samples were collected every day in STZ-treated and normoglycaemic animals, treated and untreated with benzene (n = 10). Urinary levels of trans,trans-muconic acid and of phenol, catechol and hydroquinone (free and conjugated with sulphuryl and glucuronic group) were measured by high-performance liquid chromatography (HPLC). In normoglycaemic rats during the 5 days of treatment with benzene we observed a progressive and significant decrement in the urinary excretion of phenol, phenyl sulphate and glucuronide, catechol, catechol glucuronide, hydroquinone, hydroquinone glucuronide and t,t-muconic acid (P < 0. 05). In the diabetic animals, conversely, the same metabolites showed progressively increasing urinary levels (P < 0.05). Catechol sulphate and hydroquinone sulphate levels were below the instrument's detection limit. In the comparison between diabetic and normoglycaemic benzene treated rats, the inter-group difference was significant (P < 0.05) from day 3 of treatment for t,t-muconic acid, and from day 1 for free and conjugated phenol, free and glucuronide catechol and free hydroquinone. In the normoglycaemic rat exposed to benzene the decreasing trend observed in urinary excretion of free and conjugated metabolites may be due to their capability to reduce cytochromial activity. Conversely, in the diabetic rat, urinary levels of benzene metabolites tended to increase progressively, probably due to the consistent induction of CYP-450 IIE1 observed in diabetes, which would overwhelm the inhibition of this isoenzyme caused by phenolic metabolites. Furthermore, the metabolic switch towards detoxification metabolites observed after administration of high doses of benzene is not allowed in the diabetic because of reduced glutathione-S-transferase activity. As a consequence, higher levels of hydroquinone, phenol and catechol, considered the actual metabolites responsible for benzene toxicity, will accumulate in the diabetic rat. Extrapolating these data to human, we may thus suggest that occupational exposure to benzene of a diabetic subject poses a higher risk level, as his metabolism tends to produce and accumulate higher levels of reactive benzene catabolites.

Type I IFNs constitute a family of proteins exhibiting high homology in primary, secondary, and tertiary structures. They interact with the same receptor and transmit signals to cellular nucleus through a similar mechanism, eliciting roughly homogeneous biological activity. Nevertheless, the members of that family, IFN alpha species, IFN beta and IFN omega, due to local differences in the structure sometime show distinct properties. From the reported data it results that even minute changes or differences in the primary sequences could be responsible for a significant variety of biological actions, thus inducing to the hypothesis that Type I IFNs, rather than to be the result of a redundant replication during the evolution play definite roles in the defense of living organisms to foreign agents.

Recent reports suggest that several viruses, besides human immunodeficiency virus, induce apoptosis in infected cells. We report here that Sendai virus or Herpes simplex virus type 1 (HSV-1), two potent inducers of interferon-alpha, caused cell death in a consistent number of human peripheral blood mononuclear cells. A careful analysis of infected cells by different techniques, such as optical and electron microscopy, DNA agarose gel electrophoresis, and cytofluorimetric analysis of DNA content, showed that cell death was of apoptotic type. Sendai virus was more apoptogenic than HSV-1, and it was further studied to understand the mechanism(s) by which it induced apoptosis. Physical (uv and heat) and chemical (beta-propiolactone) inactivation reduced or abolished the apoptogenic power of Sendai virus. The use of a novel technique, which allows the study of mitochondrial membrane potential (MMP) in intact cells by flow cytometry, showed that a decrease of MMP is concomitant with the appearance of the hypodiploid peak. These results suggest that Sendai virus and HSV-1 can be added to the list of viruses causing apoptosis, which appears to be a general mechanism occurring during viral infection.
