Volumetric subcellular imaging has long been essential for studying structures and dynamics in cells and tissues. However, due to limited imaging speed and depth of field, it has been challenging to perform live-cell imaging and single-particle tracking. Here we report a 2.5D fluorescence microscopy combined with highly inclined illumination beams, which significantly reduce not only the image acquisition time but also the out-of-focus background by ∼2-fold compared to epi-illumination. Instead of sequential z-scanning, our method projects a certain depth of volumetric information onto a 2D plane in a single shot using multi-layered glass for incoherent wavefront splitting, enabling high photon detection efficiency. We apply our method to multi-color immunofluorescence imaging and volumetric super-resolution imaging, covering ∼3–4 µm thickness of samples without z-scanning. Additionally, we demonstrate that our approach can substantially extend the observation time of single-particle tracking in living cells.

Background Multi - modal neuroimaging examinations play a crucial role in the diagnosis and treatment of functional neurosurgery. However, there is currently a lack of effective management for these complex data in clinical practice. This study attempts to establish a feasible multimodal neuroimaging data information management system and evaluate its application effects. Methods By standardizing clinical diagnosis and treatment processes, analyzing the nodes where imaging data were generated, and streamlining data flow routes, establishing storage naming conventions, setting up storage servers, and training specialized personnel, we designed and applied a multi - modal neuroimaging data information management system. The primary evaluation indicators were the archiving rates of 5 types of data: structural sequences, other preoperative images, postoperative electrode CT, electrode reconstruction, and postoperative CT/MRI. The secondary evaluation indicators included the total man-hours consumed for data archiving and the average man-hours consumed per case. Results Without multi-modal neuroimaging data information management (control group, n = 64), the total manpower consumption was 192 man-hours, with an average of 3 man-hours per case. With multi-modal neuroimaging data information management (data management group, n = 50), the total manpower consumption was 84 man-hours, with an average of 1.68 man-hours per case. The data management group had higher archiving rates compared to the control group: structural sequences [100% (50/50) vs. 32.81% (21/64); χ2 = 11.383, P = 0.001], other preoperative images [96% (48/50) vs. 26.56% (17/64); χ2 = 13.839, P = 0.000], postoperative electrode CT [96% (48/50) vs. 32.81% (21/64); χ2 = 10.409, P = 0.001], electrode reconstruction [96% (48/50) vs. 32.81% (21/64); χ2 = 10.409, P = 0.001], postoperative CT/MRI [96% (48/50) vs. 15.63% (10/64); χ2 = 22.169, P = 0.000]. Conclusions Designing a multi-modal neuroimaging data information management system that aligns with clinical practice and reasonably setting data collection and archiving nodes can effectively improve data archiving rates, save manpower resources, ensure the complete storage of clinical data, and ensure the smooth operation of clinical tasks, and enhance clinical diagnosis and treatment levels.

Research has shown that an effective peer critique process improves the quality of student assignments in various disciplines. Despite its benefits, peer critique is not without challenges. The major concern is the quality of the feedback. Students are not willing to take critical stances in reviewing their peers' work and they tend to give more positive comments on their peers' work. Students often complain that most feedback is not useful. Due to poor feedback, students do not feel they have learned from their experiences. This paper reports a longitudinal study investigating peer critique conducted in the online learning environment. The purpose of this study is to observe how students conduct peer critique in an online environment, where interventions are infused and applied to promote group dynamics. Student online critiques were archived. Content analysis was applied in coding student critiques. Student-written reflections were collected to obtain student perceptions about online peer critique.

In order to investigate the effect of low-temperature air cooling on the postmortem energy metabolism and meat quality of yellow-feathered broilers during mixed cooling (first water cooling and then air cooling), 85-day-old broilers were slaughtered and divided into three groups: treatment (water cooling at 0–4 ℃ followed by air cooling at −8, −18, −25 or −31 ℃), control (traditional water cooling at 0–4 ℃) and traditional mixed cooling (water cooling at 0–4 ℃ followed by air cooling at 0–4 ℃) groups. Finally, all carcasses were cooled to an internal temperature of 4 ℃, and the optimal air cooling temperature was determined by measuring cooling rate, pH, energy metabolism and quality indexes. The results showed that the cooling time of air cooling at −25 and −31 ℃ was 2 537 and 2 272 s, respectively, which was shortened by 27.78% and 35.33% compared with the traditional water cooling and by 51.87% and 56.90% compared with the traditional mixed cooling, respectively. Air cooling at −25 and −31 ℃ delayed the decrease in pH, reduced the drip loss and cooking loss of broiler breast muscle, significantly increased the shear force and the content of immobilized water compared with the control group (P < 0.05). Compared with the other groups, the activities of muscle hexokinases, phosphate-3-phosphate dehydrogenase and lactate dehydrogenase in the air cooling treatments at −25 and −31 ℃ were lower, and the consumptions of adenosine triphosphate (ATP) and glycogen, the production of lactic acid and adenosine monophosphate (AMP) and AMP/ATP ratio were the smallest (P < 0.05). The above findings showed that air cooling at −25 and −31 ℃ significantly inhibited the rate of anaerobic glycolysis during the early postmortem period in chicken meat, delayed the rate of pH decline, and effectively improved muscle water retention. From the perspective of industrial energy saving, −25 ℃ is determined as the best temperature for air cooling treatment.

With the growing popularity of eSport games, eSport-related issues have gradually gained attention and discussion in academic research. However, the positive benefits (values) brought by playing eSport have not received too much attention in current research. Therefore, after reviewing related research in the past, this study proposed that eSport has the 5 Cs educational Value of cultivating the cooperative attitude, communication skills, critical thinking, self-confidence and continuous improvement attitude based on the three-domain model (TDM) of cognitive, affective and psychomotor, and developed an eSport educational values scale. In this study, a conceptual sampling method was adopted and players with eSport experience were invited to fill out the questionnaire. A total of 316 participants filled out the questionnaire, 51 invalid samples were deleted, the number of effective participants was 265, and the effective recovery rate was 83.9%. Then SPSS 23.0 and AMOS 20.0 were used to analyze the reliability and validity of the scale, and the verification results show the scale developed by this study has good reliability and validity. In addition, in this study, it was also found that the participants had a positive view (M > 3.9) on the 5 Cs educational value of the MOBA type eSport, which shows that eSport is not only a casual game, moderate playing this game can also bring educational significance to players.
