Background/Aims Ubiquitination is widely involved in the progression of hepatocellular carcinoma (HCC) by regulating various cellular processes. However, systematic strategies for screening core ubiquitin-related genes, clarifying their functions and mechanisms, and ultimately developing potential therapeutics for patients with HCC are still lacking. Methods Cox and LASSO regression analyses were performed to construct a ubiquitin-related gene prediction model for HCC. Loss- and gain-of-function studies, transcriptomic and metabolomics analysis were used to explore the function and mechanism of UBE2S on HCC cell glycolysis and growth. Results Based on 1,423 ubiquitin-related genes, a four-gene signature was successfully constructed to evaluate the prognosis of patients with HCC. UBE2S was identified in this signature with the potential to predict the survival of patients with HCC. E2F2 transcriptionally upregulated UBE2S expression by directly binding to its promoter. UBE2S positively regulated glycolysis in a HIF-1α-dependent manner, thus promoting the proliferation of HCC cells. Mechanistically, UBE2S enhanced K11-linkage polyubiquitination at lysine residues 171 and 196 of VHL independent of E3 ligase, thereby indirectly stabilizing HIF-1α protein levels by mediating the degradation of VHL by the proteasome. In particular, the combination of cephalomannine, a small molecule compound that inhibits the expression of UBE2S, and PX-478, an inhibitor of HIF-1α, significantly improved the anti-tumor efficacy. Conclusions UBE2S is identified as a key biomarker in HCC among the thousands of ubiquitin-related genes and promotes glycolysis by E3 enzyme-independent ubiquitination, thus serving as a therapeutic target for the treatment of HCC.

Ionic compressors are promising applied in hydrogen refueling stations. Ionic liquid is adopted inside cylinders for sealing and cooling. Liquid slugging may occur but was rarely referred to in the previous studies, since former modeling ignored the effects of valve resistance. This paper proposes a method to involve valve resistance into gas-liquid simulation of ionic compressors. Valve flow coefficient is derived from the relationship of pressure-drop and flow rate through the valve. A multicycle simulation is conducted based on the Volume of Fluid (VOF) method. Liquid slugging and sudden pressure rise can be observed from the proposed method. Results show that liquid overflow per cycle can be separated into two parts: liquid slugging part and entrained part. Liquid slugging abates and finally disappears as the initial liquid fraction per cycle decreases. Pressure caused by liquid slugging increases beyond 1.5 times discharge pressure, demonstrating that liquid slugging may lead to deformation or even fraction of pressure-bearing parts. The results can validate the significance of involving valve resistance into the simulation model. A method is proposed to judge whether liquid slugging will occur. Predicted liquid slugging time has a good agreement with the CFD results. This study can provide a reference for numerical calculation of liquid overflow and slugging of ionic compressors.

Per- and polyfluoroalkyl substances (PFAS) have attracted significant attention due to their environmental toxicity. However, the detrimental impact of PFAS on the development of the female reproductive system remains controversial. In this study, we investigated the effects of three specific PFAS compounds perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), and perfluorodecanoic acid (PFDA) on ovarian development. Among these compounds, PFDA demonstrated the most pronounced cytotoxic effect on ovarian granulosa cells. The results showed that a 200 μM concentration of PFDA induced cell apoptosis via the intrinsic pathway by elevating reactive oxygen species (ROS) levels and activating Caspase-9 and Caspase-3. Furthermore, 200 μM PFDA triggered necroptosis, a form of regulated cell death (RCD), through the receptor-interacting serine/threonine kinase 1 (RIPK1), receptor interacting protein kinase 3 (RIPK3), and mixed-lineage kinase domain-like protein (MLKL) axis, mediated by inhibition of the canonical apoptosis proteolytic enzyme Caspase-8. In vivo experiments confirmed that mice exposed to PFDA displayed a significantly reduced ovarian index compared to the control group, accompanied by evident follicular atresia. Ovarian tissues from the PFDA-exposed group showed upregulated necroptosis markers, which were effectively mitigated by inhibiting the phosphorylation of RIPK1 at Ser166. Importantly, this study provides the first evidence that PFDA disrupts ovarian development through a novel mechanism involving the RIPK1-mediated necroptosis pathway, alongside the detection of the intrinsic apoptosis pathway. This greatly expands our insight into the effects of PFDA on cell death. This finding highlights the potential public health hazards associated with PFDA exposure and emphasizes the need for further research to fully understand its broader implications.

Bisphenol A (BPA) is a widespread endocrine disruptor that mimics estrogen. The accumulation of BPA within the human body has been shown to be detrimental to ovarian function. However, few studies have focused on the specific mechanisms by which it causes harm to granulosa cells (GCs), pivotal ovarian cells that are responsible for the growth and function of oocytes. In vitro research was conducted using human GC lines (KGN cells). The cells were exposed to various concentrations of BPA (0.1, 1, 10, or 100 µM) for either 24 or 48 hours. Here, our findings indicate that 100 μM BPA inhibits KGN cell proliferation and promotes cell autophagy through inhibiting the PI3K/Akt/mTOR pathway. Interestingly, these effects could be partly reversed by an NTRK2 activator (LM22b-10). NTRK2 is the receptor for BDNF. Moreover, via the use of bioinformatics tools, miR-204 was predicted to target BDNF. Additionally, our findings confirmed that miR-204 has the ability to directly target BDNF through a luciferase assay. Downregulation of miR-204 abrogated the BPA exposure-mediated effects on proliferation and autophagy. Furthermore, the inhibition of miR-204 significantly reversed the downregulation of PI3K/Akt/mTOR pathway-related molecules. Similarly, we validated miR-204 as a novel miRNA involved in BPA-mediated damage to GC proliferation and autophagy, and our data provide the first in vitro evidence that increasing miR-204 expression and inhibiting the BDNF/NTRK2-mediated PI3K/Akt/mTOR signaling pathway are involved in the BPA-induced toxic effects in KGN cells.

Emerging evidence shows that disrupted gut microbiota-bile acid (BA) axis is critically involved in the development of neurodegenerative diseases. However, the alterations in spatial distribution of BAs among different brain regions that command important functions during aging and their exact roles in aging-related neurodegenerative diseases are poorly understood. Here, we analyzed the BA profiles in cerebral cortex, hippocampus, and hypothalamus of young and natural aging mice of both sexes. The results showed that aging altered brain BA profiles sex- and region- dependently, in which TβMCA was consistently elevated in aging mice of both sexes, particularly in the hippocampus and hypothalamus. Furthermore, we found that aging accumulated-TβMCA stimulated microglia inflammation in vitro and shortened the lifespan of C. elegans, as well as behavioral impairment and neuroinflammation in mice. In addition, metagenomic analysis suggested that the accumulation of brain TβMCA during aging was partially attributed to reduction in BSH-carrying bacteria. Finally, rejuvenation of gut microbiota by co-housing aged mice with young mice restored brain BA homeostasis and improved neurological dysfunctions in natural aging mice. In conclusion, our current study highlighted the potential of improving aging-related neuro-impairment by targeting gut microbiota-brain BA axis.

Abstract In chloroplasts, insertion of proteins with multiple transmembrane domains (TMDs) into thylakoid membranes usually occurs in a co-translational manner. Here, we have characterized a thylakoid protein designated FPB1 (Facilitator of PsbB biogenesis1) which together with a previously reported factor PAM68 (Photosynthesis Affected Mutant68) is involved in assisting the biogenesis of CP47, a subunit of the Photosystem II (PSII) core. Analysis by ribosome profiling reveals increased ribosome stalling when the last TMD segment of CP47 emerges from the ribosomal tunnel in fpb1 and pam68. FPB1 interacts with PAM68 and both proteins coimmunoprecipitate with SecY/E and Alb3 as well as with some ribosomal components. Thus, our data indicate that, in coordination with the SecY/E translocon and the Alb3 integrase, FPB1 synergistically cooperates with PAM68 to facilitate the co-translational integration of the last two CP47 TMDs and the large loop between them into thylakoids and the PSII core complex.

Abstract Wearable strain sensors have attracted research interest owing to their potential within digital healthcare, offering smarter tracking, efficient diagnostics, and lower costs. Unlike rigid sensors, fiber‐based ones compete with their flexibility, durability, adaptability to body structures as well as eco‐friendliness to environment. Here, the sustainable fiber‐based wearable strain sensors for digital health are reviewed, and material, fabrication, and practical healthcare aspects are explored. Typical strain sensors predicated on various sensing modalities, be it resistive, capacitive, piezoelectric, or triboelectric, are explained and analyzed according to their strengths and weaknesses toward fabrication and applications. The applications in digital healthcare spanning from body area sensing networks, intelligent health management, and medical rehabilitation to multifunctional healthcare systems are also evaluated. Moreover, to create a more complete digital health network, wired and wireless methods of data collection and examples of machine learning are elaborated in detail. Finally, the prevailing challenges and prospective insights into the advancement of novel fibers, enhancement of sensing precision and wearability, and the establishment of seamlessly integrated systems are critically summarized and offered. This endeavor not only encapsulates the present landscape but also lays the foundation for future breakthroughs in fiber‐based wearable strain sensor technology within the domain of digital health.

Abstract Background Nucleoside analogues are currently applied as a first-line treatment for chronic hepatitis B (CHB) patients. However, the long-term effects of this type of treatment on kidney and bone tissue need to be further investigated. Methods We conducted a search of entecavir (ETV), tenofovir disoproxil fumarate (TDF), and tenofovir alafenamide fumarate (TAF) for treatment of CHB patients through October 29, 2023. Side effects of the three drugs were compared. Standardized mean difference (SMD), 95% confidence interval (95%CI), and surface under the cumulative ranking curve (SUCRA) were reported for each outcome. Further subgroup analysis was conducted according to duration of administration. Results ETV and TAF exhibited less effect on estimated glomerular filtration rate (eGFR) than TDF (SMD = -3.60 (95%CI: -1.94 ~ -5.26) and SMD = -4.27 (95%CI: -2.62 ~ -5.93)). ETV also exhibited less effect on creatinine rise than TAF and TDF (SMD = -0.55 (95%CI: -0.09 ~ -1.01) and SMD = -0.61 (95%CI: -0.15 ~ -1.06)). Moreover, the effect of TAF on bone mineral density (BMD) was less than that of TDF (SMD = -0.02 (95%CI: -0.01 ~ -0.02)). The probabilities of the three drugs changing relevant indicators exhibited similar patterns: eGFR (TDF (100.0%) > ETV (41.2%) > TAF (8.8%)), creatinine (TDF (94.7%) > TAF (54.7%) > ETV (0.6%)), BMD (TDF (79.7%) > ETV (50.6%) > TAF (19.6%)), and blood phosphorus (TDF (90.6%) > TAF (49.8%) > ETV (9.7%)). After 6 and 24 months of treatment, no statistically significant difference in renal function or bone tissue was observed between ETV and TDF. However, greater adverse effects on renal function were observed for TDF than ETV at 60 months compared to 12 months. TDF also exhibited greater adverse effects on bone tissue than ETV at 36 months than at 12 months. Conclusions Long-term administration of TDF has resulted in stronger adverse effects than TAF and ETV in regard to both renal function and bone tissue in CHB patients. The effect of TAF on creatinine increase was greater than ETV. The difference in side effects between ETV and TDF was independent of treatment duration.

Abstract Nonalcoholic fatty liver disease (NAFLD) is usually characterized with disrupted bile acid (BA) homeostasis. However, the exact role of certain BA in NAFLD is poorly understood. Here we show levels of serum hyodeoxycholic acid (HDCA) decrease in both NAFLD patients and mice, as well as in liver and intestinal contents of NAFLD mice compared to their healthy counterparts. Serum HDCA is also inversely correlated with NAFLD severity. Dietary HDCA supplementation ameliorates diet-induced NAFLD in male wild type mice by activating fatty acid oxidation in hepatic peroxisome proliferator-activated receptor α (PPARα)-dependent way because the anti-NAFLD effect of HDCA is abolished in hepatocyte-specific Pparα knockout mice. Mechanistically, HDCA facilitates nuclear localization of PPARα by directly interacting with RAN protein. This interaction disrupts the formation of RAN/CRM1/PPARα nucleus-cytoplasm shuttling heterotrimer. Our results demonstrate the therapeutic potential of HDCA for NAFLD and provide new insights of BAs on regulating fatty acid metabolism.

A key objective of tissue engineering (TE) is to produce in vitro funcional grafts that can replace damaged tissues or organs in patients. TE uses bioreactors, which are controlled environments, allowing the application of physical and biochemical cues to relevant cells growing in biomaterials. For soft musculoskeletal (MSK) tissues such as tendons, ligaments and cartilage, it is now well established that applied mechanical stresses can be incorporated into those bioreactor systems to support tissue growth and maturation via activation of mechanotransduction pathways. However, mechanical stresses applied in the laboratory are often oversimplified compared to those found physiologically and may be a factor in the slow progression of engineered MSK grafts towards the clinic. In recent years, an increasing number of studies have focused on the application of complex loading conditions, applying stresses of different types and direction on tissue constructs, in order to better mimic the cellular environment experienced in vivo. Such studies have highlighted the need to improve upon traditional rigid bioreactors, which are often limited to uniaxial loading, to apply physiologically relevant multiaxial stresses and elucidate their influence on tissue maturation. To address this need, soft bioreactors have emerged. They employ one or more soft components, such as flexible soft chambers that can twist and bend with actuation, soft compliant actuators that can bend with the construct, and soft sensors which record measurements in situ. This review examines types of traditional rigid bioreactors and their shortcomings, and highlights recent advances of soft bioreactors in MSK TE. Challenges and future applications of such systems are discussed, drawing attention to the exciting prospect of these platforms and their ability to aid development of functional soft tissue engineered grafts.

Electricity is the lifeblood of modern society. However, the predominant source of electricity generation still relies on non-renewable fossil fuels, whose combustion releases greenhouse gases contributing to global warming. The increasing demand for energy and escalating environmental concerns necessitate proactive measures to develop innovative green energy technologies capable of both cooling the Earth and generating electricity. Here, we look forward to an interdisciplinary power system integrating solar absorbers, radiative coolers, and thermoelectric generators. This system can simultaneously harvest thermal energy from the sun and from cold space, thereby transforming the challenges posed by global warming into opportunities for the production of clean electricity. We underscore recent advancements in this field and address key challenges while also exploring forward-looking opportunities in the foreseeable future. The proposed integrated energy technology achieves uninterrupted power supply through the unrestricted capture of thermal energy, offering a robust alternative pathway for next-generation sustainable energy technologies.

PurposeThe effectiveness of probiotics or synbiotics in adults with metabolic syndromes (MetS) remains controversial, this meta-analysis will further analyze the effects of probiotics or synbiotics on cardiovascular factors in adults with MetS.MethodsWe searched Web of Science, PubMed, Embase, Cochrane Library and other databases for randomized controlled trials (RCTs) on the effects of probiotics or synbiotics on MetS in adults up to July 2023, and used RevMan 5.4.0 software for statistical analysis.ResultsThis analysis included eleven RCTs (n = 608 participants), and the results showed that compared with the control group, supplementation with probiotics or synbiotics reduced body mass index (weighted mean difference, WMD = -0.83, 95% CI = [-1.21, -0.44], P

Summary: Here, we present a protocol for the complete stereoselective synthesis of a molecular 51 knot. Enantiopure chiral ligands serve as the starting point, while Zn(OTf)2 acts as the template, facilitating the quantitative formation of pentameric circular helicates with 100% d.e. A subsequent sequence of ring-closing metathesis and demetalation steps transforms the structure into a fully organic 51 knot. Abstract Multiple strain sensors are required to identify individual forces/stresses on human joints and recognize how they work together in order to determine the motion’s direction and trajectory. However, current sensors cannot detect and differentiate the individual forces/stresses and their contributions to the motion from the sensors’ electrical signals. To address this critical issue, we propose a concept of unimodal tension, bend, shear, and twist strain sensors with piezoelectric poly L-lactic acid films. We then construct an integrated unimodal sensor (i-US) using the unimodal sensors and prove that the i-US can detect and differentiate individual strain modes, such as tensioning, bending, shearing, and twisting in complex motion. To demonstrate the potential impact of unimodal sensors, we design a sleeve and a glove with the i-US that can capture wrist motions and finger movements. Therefore, we expect unimodal strain sensors to provide a turning point in developing motion recognition and control systems.

Nerve guidance conduits (NGCs) have attracted much attention due to their great necessity and applicability in clinical use for the peripheral nerve repair. Great efforts in recent years have been devoted to the development of high-performance NGCs using various materials and strategies. The present review provides a comprehensive overview of progress in the material innovation, structural design, advanced engineering technologies and multi functionalization of state-of-the-art nerve guidance conduits NGCs. Abundant advanced engineering technologies including extrusion-based system, laser-based system, and novel textile forming techniques in terms of weaving, knitting, braiding, and electrospinning techniques were also analyzed in detail. Findings arising from this review indicate that the structural mimetic NGCs combined with natural and synthetic materials using advanced manufacturing technologies can make full use of their complementary advantages, acquiring better biomechanical properties, chemical stability and biocompatibility. Finally, the existing challenges and future opportunities of NGCs were put forward aiming for further research and applications of NGCs.

Accumulating evidence indicates gut microbiota contributes to aging-related disorders. However, the exact mechanism underlying gut dysbiosis-related pathophysiological changes during aging remains largely unclear. In the current study, we first performed gut microbiota remodeling on old mice by fecal microbiota transplantation (FMT) from young mice, and then characterized the bacteria signature that was specifically altered by FMT. Our results revealed that FMT significantly improved natural aging-related systemic disorders, particularly exerted hepatoprotective effects, and improved glucose sensitivity, hepatosplenomegaly, inflammaging, antioxidative capacity and intestinal barrier. Moreover, FMT particularly increased the abundance of fecal A.muciniphila, which was almost nondetectable in old mice. Interestingly, A.muciniphila supplementation also exerted similar benefits with FMT on old mice. Notably, targeted metabolomics on short chain fatty acids (SCFAs) revealed that only acetic acid was consistently reversed by FMT. Then, acetic acid intervention exerted beneficial actions on both Caenorhabditis elegans and natural aging mice. In conclusion, our current study demonstrated that gut microbiota remodeling improved natural aging-related disorders through A.muciniphila and its derived acetic acid, suggesting that interventions with potent stimulative capacity on A. muciniphila growth and production of acetic acid was alternative and effective way to maintain healthy aging. Abstract Wearable strain sensors are arousing increasing research interests in recent years on account of their potentials in motion detection, personal and public healthcare, future entertainment, man–machine interaction, artificial intelligence, and so forth. Much research has focused on fiber-based sensors due to the appealing performance of fibers, including processing flexibility, wearing comfortability, outstanding lifetime and serviceability, low-cost and large-scale capacity. Herein, we review the latest advances in functionalization and device fabrication of fiber materials toward applications in fiber-based wearable strain sensors. We describe the approaches for preparing conductive fibers such as spinning, surface modification, and structural transformation. We also introduce the fabrication and sensing mechanisms of state-of-the-art sensors and analyze their merits and demerits. The applications toward motion detection, healthcare, man–machine interaction, future entertainment, and multifunctional sensing are summarized with typical examples. We finally critically analyze tough challenges and future remarks of fiber-based strain sensors, aiming to implement them in real applications.

Cooperation evolution emerges in diversities of social networks by way of individuals’ interactions and influence propagations. The influence on cooperation evolution owing to the multiplex nature of social networks and interactions is still an open question. In this work, we focus on the impacts of the individuals’ heterogeneous properties and the networks’ multilayer nature on cooperation evolution, and propose a heterogeneous diffusion model to describe and analyze the cooperation dynamics processes in diverse layers of multiplex networks with weighted values. We investigated how the interlayer and intralayer interactions with these heterogeneities influence individuals’ behaviors and strategies, and discussed how some important parameters impact the cooperation densities and evolution processes in the interactions of different strategy games. The experimental results show that the tighter interlayer interaction can obviously improve the cooperation densities in the strategy games SG, but has little impact on the cooperation evolution in the strategy games PD. The effect of individuals’ trust on cooperation diffusion is smaller in the games PD than in the games SG. The heterogeneity of degree distribution between individuals from interlayer networks also affects cooperation processes. This work explores and reveals the mechanism and regular pattern of cooperation diffusion considering the heterogeneities of individuals in multiplex networks with weighted values.

Construction vehicles have high requirements for load carrying capacity， with suspension stiffness large， vibration damping performance poor， and working environment harsh， severe vibration in the three directions of vertical， pitch and roll， which seriously affects the physical and mental health of the driver and the vehicle operational stability. To solve this problem， a 2-SPS+SR multi-dimensional vibration-damping suspension for construction vehicle seats is proposed. Firstly， the degree of freedom analysis of two translations and one rotation of the seat suspension is carried out by using the single-open chain theory. Then， the position equation of the seat suspension is established to analyze the position of the seat suspension， and the positive solution is given for the suspension structure. The first and second order motion coefficients of the suspension are obtained by taking the first and second derivatives of the position equation. Finally， the simulation analysis is performed in Adams/view and programming simulation of mathematical model is performed in Matlab to verify the correctness of the established mathematical and theoretical analysis. The results of analysis provide important significance for the study of the control simulation of the suspension and the research on the ride comfort of the construction vehicle on the three-dimensional road in the later period.

Hepatocellular carcinoma (HCC) is one of the most lethal malignant diseases worldwide. Despite advances in the diagnosis and treatment of HCC, its overall prognosis remains poor. Recent studies have shown that long noncoding RNAs (lncRNAs) play crucial roles in various pathophysiological processes, including liver cancer. In the current study, we report that lncRNA SLC2A1‐AS1 is frequently downregulated in HCC samples, as shown by quantitative real‐time polymerase chain reaction analysis. SLC2A1‐AS1 deletion is significantly associated with recurrence‐free survival in HCC. By performing glucose uptake, lactate production and ATP detection assays, we found that SLC2A1‐AS1‐mediated glucose transporter 1 (GLUT1) downregulation significantly suppressed glycolysis of HCC. In vitro Cell Counting Kit‐8, colony formation, transwell assays as well as in vivo tumorigenesis and metastasis assays showed that SLC2A1‐AS1 overexpression significantly suppressed proliferation and metastasis in HCC through the transcriptional inhibition of GLUT1. Results from fluorescence in situ hybridization, ChIP and luciferase reporter assays demonstrated that SLC2A1‐AS1 exerts its regulatory role on GLUT1 by competitively binding to transketolase and signal transducer and activator of transcription 3 (STAT3) and inhibits the transactivation of Forkhead box M1 (FOXM1) via STAT3, thus resulting in inactivation of the FOXM1/GLUT1 axis in HCC cells. Our findings will be helpful for understanding the function and mechanism of lncRNA in HCC. These data also highlight the crucial role of SLC2A1‐AS1 in HCC aerobic glycolysis and progression and pave the way for further research regarding the potential of SLC2A1‐AS1 as a valuable predictive biomarker for HCC recurrence.

Src homology 2 domain-containing protein tyrosine phosphatase 1 (SHP1), a non-receptor member of the protein tyrosine phosphatase (PTP) family, negatively regulates several signaling pathways that are responsible for pathological cell processes in cancers. In this study, we report a series of 3-amino-4,4-dimethyl lithocholic acid derivatives as SHP1 activators. The most potent compounds, 5az-ba, showed low micromolar activating effects (EC50: 1.54–2.10 μM) for SHP1, with 7.63–8.79-fold maximum activation and significant selectivity over the closest homologue Src homology 2 domain-containing protein tyrosine phosphatase 2 (SHP2) (>32-fold). 5az-ba showed potent anti-tumor effects with IC50 values of 1.65–5.51 μM against leukemia and lung cancer cells. A new allosteric mechanism of SHP1 activation, whereby small molecules bind to a central allosteric pocket and stabilize the active conformation of SHP1, was proposed. The activation mechanism was consistent with the structure–activity relationship (SAR) data. This study demonstrates that 3-amino-4,4-dimethyl lithocholic acid derivatives can be selective SHP1 activators with potent cellular efficacy.

The acute wound healing process requires a quick and short-stage vascular endothelial growth factor (VEGF) supply, especially, in the early-stage of injury. To date, research has rarely met the demand for controlled VEGF quick release. Herein, a novel wheel spinning technique was introduced to fabricate alginate (Alg)/silk fibroin (SF) composite fiber for loading VEGF and achieving controllable release. In turn, it was demonstrated that the parameter of wheel spinning and Alg/SF material morphological combination significantly influence VEGF release behavior and fiber degradation speed, which could be controlled by adjusting spinning solution concentration and modifying SF morphology. The loading concentration and SF structural features interacted with each other to affect the VEGF release behavior, indicating that growth factors such as VEGF could be released in a controllable fashion. It could therefore be developed as a novel approach to control the wound healing process.

Gastric cancer is one of the most common malignant tumours in the world. As one of the crucial hallmarks of cancer reprogramming of metabolism and the relevant researches have a promising application in the diagnosis treatment and prognostic prediction of malignant tumours. This study aims to identify a group of metabolism-related genes to construct a prediction model for the prognosis of gastric cancer.A large cohort of gastric cancer cases (1121 cases) from public database was included in our analysis and classified patients into training and testing cohorts at a ratio of 7: 3. Abstract It has been proposed that the circadian rhythm generally plays important roles in tumor suppression, but there is also evidence that disruption of the canonical circadian pathway has anticancer effects. In this study, we systematically analyzed the aberrances of circadian clock genes across cancers based on data from The Cancer Genome Atlas (TCGA). These data showed that the frequencies of mutations and copy number alterations in core clock genes (PER1/2/3, CLOCK, CRY1/2, and ARNTL) were low, but that the expression levels of core clock genes were downregulated by the higher levels of DNA methylation in most tumors. The circadian clock index (CCI) was established through a principal component analysis, and this measure well represents the overall expression of the core clock genes. In fact, the CCI was significantly lower in hepatocellular carcinoma with HBV infection than in other cancers. Furthermore, pathways such as the MAPK, JAK‐STAT, and immune‐related signaling pathways were enriched in tumors with high CCI values. Interestingly, the CCI was generally positively related to the immunophenoscores and immunophenotypes of tumors. Additionally, the expression levels of core clock genes and the CCI were also generally positively related to survival across cancers. Taken together, the results of this study provide a comprehensive analysis of circadian clock aberrances in cancer, and the results should aid further investigations of the molecular mechanisms of cancer and the development of therapeutic strategies.

Summary: Ferroptosis is a type of cell death related to cancer; however, the characteristics of ferroptosis in cancers are still uncertain. Based on the data in The Cancer Genome Atlas, we found that most ferroptosis regulator genes (FRGs) were differentially expressed in tumors, somatic copy number alterations (SCNA) and DNA methylation contributed to their aberrant expression. We established the ferroptosis potential index (FPI) to reveal the functional roles of ferroptosis and noticed that the FPI was higher in tumors than in normal tissues in most cancers and was associated with subtypes and clinical features. The FPI was negatively correlated with several metabolic pathways but positively associated with several important metastasis-related pathways and immune-related pathways. High FPI predicted poor prognosis in several tumors, whereas FPI and FRGs impacted drug sensitivity. Our study presents a systematic analysis of ferroptosis and its regulatory genes and highlights the potential of ferroptosis-based cancer therapy.

Photothermal conversion is an environmentally friendly process that harvests energy from the sun and has been attracting growing research interest in recent years. However, nanostructured strategies to improve light capture performance deserve further development, and the application of solar heating effects for clean energy needs to be explored. Herein, a multiscale nanomaterial was prepared by in situ polymerizing the polyaniline (PANI) nanoparticles into porous anodic aluminum oxide (AAO) membrane. As a result, the as-prepared PANI-AAO shows broadband solar absorption and provides a platform for efficient photothermal conversion. What is more, we introduced a typical thermoelectricity generator (TEG) with excellent output performance and combined it with PANI-AAO to prepare a solar thermoelectric generator (s-TEG). The s-TEG harvests solar energy and converts it into electricity, showing an outstanding power generation capability in outdoor conditions. Thus, the nanostructured broadband solar absorber and the integrated solar thermoelectric generator offer a promising candidate for a sustainable and green energy source in the future.

Vibrio parahaemolyticus causes aquatic vibriosis. Its biofilm protects it from antibiotics; therefore, a new different method is needed to control V. parahaemolyticus for food safety. Phage therapy represents an alternative strategy to control biofilms. In this study, the lytic Vibrio phage vB_VpaP_FE11 (FE11) was isolated from the sewers of Guangzhou Huangsha Aquatic Market. Electron microscopy analysis revealed that FE11 has a typical podovirus morphology. Its optimal stability temperature and pH range were found to be 20–50 °C and 5–10 °C, respectively. It was completely inactivated following ultraviolet irradiation for 20 min. Its latent period is 10 min and burst size is 37 plaque forming units/cell. Its double-stranded DNA genome is 43,397 bp long, with a G + C content of 49.24% and 50 predicted protein-coding genes. As a lytic phage, FE11 not only prevented the formation of biofilms but also could destroy the formed biofilms effectively. Overall, phage vB_VpaP_FE11 is a potential biological control agent against V. parahaemolyticus and the biofilm it produces.

As a widespread post-translational modification of proteins, calpain-mediated cleavage regulates a broad range of cellular processes, including proliferation, differentiation, cytoskeletal reorganization, and apoptosis. The identification of proteins that undergo calpain cleavage in a site-specific manner is the necessary foundation for understanding the exact molecular mechanisms and regulatory roles of calpain-mediated cleavage. In contrast with time-consuming and labor-intensive experimental methods, computational approaches for detecting calpain cleavage sites have attracted wide attention due to their efficiency and convenience. In this study, we established a novel computational tool named DeepCalpain (http://deepcalpain.cancerbio.info/) for predicting the potential calpain cleavage sites by adopting deep neural network and the particle swarm optimization algorithm. Through critical evaluation and comparison, DeepCalpain exhibited superior performance against other existing tools. Meanwhile, we found that protein interactions could enrich the calpain-substrate regulatory relationship. Since calpain-mediated cleavage was critical for cancer development and progression, we comprehensively analyzed the calpain cleavage associated mutations across 11 cancers with the help of DeepCalpain, which demonstrated that the calpain-mediated cleavage events were affected by mutations and heavily implicated in the regulation of cancer cells. These prediction and analysis results might provide helpful information to reveal the regulatory mechanism of calpain cleavage in biological pathways and different cancer types, which might open new avenues for the diagnosis and treatment of cancers.

The interface affects the transmission behavior of electrons and phonons, which in turn determines the performance of thermoelectric materials. In this paper, metals (Cu, Ag, Au)/Al2O3/Bi2Te3 heterostructures have been fabricated from bottom to up to optimize the thermoelectric power factor. The introducing metals can be alloyed with Bi2Te3 or form interstitials or dopants to adjust the carrier concentration and mobility. In addition, the metal-semiconductor interface as well as the metal-insulator-semiconductor interface constructed by the introduced metal and Al2O3 would further participate in the regulation of the carrier transport process. By adjusting the metal and oxide layer, it is possible to realize the simultaneous optimization of electric conductivity and Seebeck coefficient. This work will enable the optimal and novel design of heterostructures for thermoelectric materials with further improved performance.

How to operate an unmanned aerial vehicle (UAV) safely and efficiently in an interactive environment is challenging. A large amount of research has been devoted to improve the intelligence of a UAV while performing a mission, where finding an optimal maneuver decision-making policy of the UAV has become one of the key issues when we attempt to enable the UAV autonomy. In this paper, we propose a maneuver decision-making algorithm based on deep reinforcement learning, which generates efficient maneuvers for a UAV agent to execute the airdrop mission autonomously in an interactive environment. Particularly, the training set of the learning algorithm by the Prioritized Experience Replay is constructed, that can accelerate the convergence speed of decision network training in the algorithm. It is shown that a desirable and effective maneuver decision-making policy can be found by extensive experimental results.

As an abundant post-translational modification, reversible phosphorylation is critical for the dynamic regulation of various biological processes. prkC, a critical serine/threonine-protein kinase in bacteria, plays important roles in regulation of signaling transduction. Identification of prkC-specific phosphorylation sites is fundamental for understanding the molecular mechanism of phosphorylation-mediated signaling. However, experimental identification of substrates for prkC is time-consuming and labor-intensive, and computational methods for kinase-specific phosphorylation prediction in bacteria have yet to be developed. In this study, we manually curated the experimentally identified substrates and phosphorylation sites of prkC from the published literature. The analyses of the sequence preferences showed that the substrate recognition pattern for prkC might be miscellaneous, and a complex strategy should be employed to predict potential prkC-specific phosphorylation sites. To develop the predictor, the amino acid location feature extraction method and the support vector machine algorithm were employed, and the methods achieved promising performance. Through 10-fold cross validation, the predictor reached a sensitivity of 91.67% at the specificity of 95.12%. Then, we developed freely accessible software, which is provided at http://free.cancerbio.info/prkc/. Based on the predictor, hundreds of potential prkC-specific phosphorylation sites were annotated based on the known bacterial phosphorylation sites. prkC-PSP was the first predictor for prkC-specific phosphorylation sites, and its prediction performance was promising. We anticipated that these analyses and the predictor could be helpful for further studies of prkC-mediated phosphorylation.
