In the normal male rat pituitary tritiated thymidine labeled mainly STH cells (somatotrophs), no labeled prolactin cell was found. Following estradiol treatment for 21 days tritiated thymidine labeled mainly prolactin cells (mammotrophs). To determine the origin of these mammotrophs tritiated thymidine was given before the estradiol treatment started, thus labeling many somatotrophs. After 21 days of estradiol, out of 42 labeled cells, 14 were mammotrophs and 13 were somatotrophs; these results suggest that there might be a true transformation of somatotrophs into mammotrophs under the influence of estradiol or that there exist two types of somatotrophs: 1) a committed somatotroph which is not transformed by estrogen treatment, and 2) an uncommitted mammosomatotroph, which under normal conditions bears the features of a somatotroph, but which transforms into a mammotroph under the influence of estradiol. In intact male rats after TRH administration for 7 and 14 days, TSH cells showed similar morphological changes to those observed after thyroidectomy. These changes were paralleled by small numerical increases in TSH cell counts. After 34 days of TRH treatment, however, most of the TSH cells had a normal appearance and the number of TSH cells also had returned to normal. TRH treatment for 7, 14 and 34 days caused morphological changes in Prolactin cells similar to those obtained after a suckling stimulus. In the three groups these changes were also paralleled by small numerical increases in Prolactin cell counts. The cell replication after TRH for 7 and 14 days, as measured by incorporation of tritiated thymidine to obtain a labeling index, was slightly but significantly increased. [ABSTRACT FROM AUTHOR]

Twelve bovine adenohypophyses were prepared for light and electron microscopy of the cell types of pars distalis. Correlation between the light and electron microscopy was effected by use of alternate thin and thick sections. Cytological changes in the experimental animals were used as criteria for the identification of six different types of secretory cells. Two types of acidophils, alpha and epsilon cells, are recognized in peripheral area of the pars distalis by light and electron microscopy. The alpha cells contain orangeophilic secretory granules of a maximum diameter of 400-450 mμ and correspond to ordinary acidophils (STH cells). The second type, epsilon cells, contains larger, fuchsinophilic granules of 600 to 900 mμ in diameter, increase in number and granulation after pregnancy and thyroidectomy, and are thought to be prolactin cells (LTH cells). Two types of amphophils, zeta and delta 1 cells, were found in the central area of the pars distalis. The zeta cells contain smaller numbers of amphophilic, cored granules (200 mμ maximum diameter) and based on the comparison with literature on other species of animals, are designated as ACTH cells. The delta 1 cells are round or oval and contain very dense, spherical granules (250-300 mμ) which are stained red or reddish purple with PAS, aldehyde thionin and PAS-methyl blue methods. They show extreme enlargement and bizarre cytoplasmic appearance after castration and are designated tentatively as LH gonadotrophs or LH cells. Two types of basophils, beta and delta 2 cells, were also identified by correlative light and electron microscopy. The beta cells are polygonal in outline, distributed exclusively in the zona tuberalis and contain large, less dense secretory granules (300-400 mμ) which are stained selectively with Gomori's aldehyde fuchsin. After thyroidectomy, they lose their secretory granules and are transformed into large, vacuolated thyroidectomy cells. They are therefore, identified as thyrotrophs or TSH cells. The delta 2 cells are round, oval or polygonal in shape and contain basophilic granules ranging from 220 to 300 mμ in diameter. They show extreme enlargement and vacuolization due to the dilation of endoplasmic reticulum, after castration, and are designated tentatively as FSH gonadotrophs or FSH cells. Prolactin cells from anterior pituitary glands of normal non-lactating female rats, and lactating animals, some of which were separated from their pups for 48 hours, were examined ultrastructurally for the presence of microfilaments. Microfilaments were found in specific intracellular locations in all cells examined. They were in association with the nuclear envelope, the Golgi complex, the endoplasmic reticulum, small vesicles of the endoplasmic reticulum, and secretory granules. Golgi methods were employed to study neurons and ependymal tanycytes in the posterior hypothalamus of the newt. The tanycytes send a few coarse, spiny or barbed processes towards the pia mater. In the periventricular grey, the neurohistological methods show common neurons, ranging from a multipolar to a plumed organization, and abundant liquor-contacting cells. These cells, possibly neurons, give rise to a process that reaches the cerebro-spinal fluid, and terminates in a spindle-shaped swelling, with a thin thread at its tip. In other cells, the intraventricular endings are bulbous or finger-like. The occurrence of: (1) branches of the liquor-contacting process, running parallel to the infundibular surface; (2) infundibular processes which end at the base or between the ependymal cell bodies; and (3) axons coursing in the same position, all indicates that the subependymal layer is a site for complex intercellular relationships. The significance of liquor-contacting cells and tanycytes is discussed, in view of the possibility that they may represent part of a system for hypothalamic regulation in response to changes in the CSF. A histochemical and ultrastructural investigation of the early development of the adenohypophysis was carried out on a human material. Special attention was paid to an accumulation of hyaluronic acid and chondroitin-4-and/or-6-sulfate in the mesenchyma; its role in morphogenesis of the Rathke's pouch is discussed. The role of the vessels as a 'critical factor' in the budding of the parenchyma and in the differentiation of secretory cells is discussed. Canalicular extensions from the original lumen of the pouch into the core of parenchymal buds, which migrate into the mesenchyma, is a new observation. The participation of canaliculi in formation of follicular structures of pars distalis is described and discussed. The primitive cell type lining the pouch is also found in the wall of canaliculi and follicular structures. The cell type is described and its role as the real progenitor cell of the adenohypophysis is discussed. Three types of colloid are noticed in pars distalis: 1. In the follicular structures, 2. in the slits or clefts caused by partial occlusion of the lumen of the pouch, and 3. mesenchymal extravasal colloid presumably representing material squeezed out from the aforementioned clefts or slits. It is concluded that the colloid contains material secreted from different types of granulated cells as well as material from the surface coat of the luminal cells. [ABSTRACT FROM AUTHOR]

Ectopic differentiation of retinal structures in contact with the pituitary, after insertion of a methylene blue stained celloidin lamina at the level of the median eminence was observed in the adult crested newt ( Triturus cristatus carnifex Laur.). Retinal components ranging from pigmented cells to early retinal vesicles and fully developed neural retina and visual cells are described. Indications pointing to the ependymal cells which line the infundibular sac sequestered between the intermediate lobe and the pars distalis, as the source of the neural retina and visual cells are presented. The specificity of Alcian blue staining has been investigated on a material comprising the adenohypophysis from 26 human foetuses. Two distinct types of alcianophilic cells were observed. The first cell type appears about 28 mm CRL forming small groups at the epithelial-mesenchymal junction and beneath the anlage of the fibrous capsule. Besides its alcianophilic property the cell shows a pronounced maltase-resistant PAS-positivity. The alcianophilia is due to carboxylgroups - probably of proteins as no sialic acid could be detected. By the performic acid - Alcian blue method the cell seems to show a high content of SH and S-S groups. The second type of cell appears about 70 mm CRL and is mostly located singly. It shows a pronounced alcianophilia - probably due to sulphate groups and to some extent to carboxyl groups. The role of the alcianophilic properties of the cells in typing definite cells of the foetal adenohypophysis was discussed. - Finally the pitfalls caused by different fixatives and by the use of the general polychrome staining methods for the typing of pituitary parenchymal cells were discussed. The development of TSH producing pituitary tumours in mice that were induced by severe thyroid inhibition was analysed during 47 weeks. This was done on the basis of the gland weights, counts and measurements of the cells with the aid of the electron microscope, calculation of changes in the absolute number of cells per hypophysis, autoradiography afterH-thymidine labeling, and morphological observations at the ultrastructural level. The results indicated that there was a short initial phase during which the thyroidectomy cells had a high mitotic activity, followed by a phase that lasted for several months during which the mitotic activity had a much lower level, so that the growth proceeded only slowly. After 38 weeks, sharply demarcated nodules of very fast proliferating thyroidectomy cells were found in most hypophyses. These nodules must have originated very early in the process of tumour formation. The morphology as well as the size of the thyroidectomy cells remained constant during the period of the experiment, except for those in the nodules that were morphologically very different and that were considerably smaller. All other hormone-producing cells were virtually not affected during most of the experimental period. A comparison was made between the results presented here and a mathematical model of the development of TSH producing pituitary tumours that was proposed by Clifton. In some of the basophilic trophoblastic cells of the basal plate and islands of the mature human placenta there are glycoprotein inclusions, many of which are within vacuoles. As established by the histochemical results these inclusions are composed of a neutral mucopolysaccharide and a basophilic protein with SH groups. They gradually increase in number from the 4th month of pregnancy and are not influenced by labor. From the first to the fourth month of gestation small vacuoles, initially infra-nuclear but later supranuclear, may be seen in the syncitial cells of the villi; these likewise contain glycoprotein granules. The same inclusions are to be found in some trophoblastic cells of the chorionic plate and of the chorion laeve, and in an occasional epithelial cell of the amnion. On the basis of the histochemical behavior of these inclusions, their similarity with the δ-granules of the hypophysis, and the obvious high activity of the trophoblastic cells which bear them, it is postulated that these inclusions contain gonadotropin. Their occurrence during pregnancy parallels the biochemical concentration and immunofluorescent histological demonstration of gonadotropin in the placenta. Thus, at the end of pregnancy the basal trophoblastic cells of the human placenta appear to form at least two different hormones: relaxin, probably represented morphologically by the acidophilic protein inclusions (described elsewhere), and gonadotropin most likely represented by the basophilic glycoprotein inclusions. The results of these experiments clearly indicate that the PAS-red and PAS-purple gonadotrophs of the rat anterior hypophysis are functionally as well as tinctorially distinct cell types. The PAS-red cells located peripherally in control animals ('peripheral gonadotrophs' of Purves and Griesbach) produce LH. Following castration they begin to appear in greater numbers in the central areas and by 45 days following castration they are predominant in both central and peripheral portions of the glands. At this time after castration the gonadotrophic content of the pituitary gland is predominantly luteinizing in character. The PAS-purple cells which are found in the central portions of control glands ('central gonadotrophs' of Purves and Griesbach) produce FSH. They appear peripherally following castration and are the predominant type of gonadotrophic cell in the glands of short term (10-day) castrates. The gonadotrophic content of such glands is chiefly FSH as shown by bioassay. By 45 days after castration the LH producing 'peripheral gonadotrophs' and the FSH producing 'central gonadotrophs' have lost their characteristic distribution patterns. Obviously, then, these two gonadotrophic types cannot be accurately followed on the basis of a restricted regional location in the pituitary but must be differentiated on the basis of specific cytological features and staining characteristics. A modified PAS-methyl blue staining method is described which, in conjunction with aldehyde fuchsin, enables the tinctorial differentiation of two types of gonadotrophs in the pituitary gland of the rat. That these two cell types are both gonadotrophic in function is indicated by their development into distinct PAS-red and PAS-purple castration cells following gonadectomy. In addition to these two kinds of castration cells, thyroidectomy cells which are PAS-negative, aldehyde fuchsin-negative, and have an affinity for orange G are seen in the pituitary glands of animals that have been both gonadectomized and thyroidectomized. Thus, in such glands the three types of modified basophiles are tinctorially different. Differences in the distribution of the PAS-red and PAS-purple gonadotrophs are characteristic of the glands of normal animals. Following gonadectomy, however, extensive shifts in the distribution of these cells occur so that the normal pattern is soon lost. [ABSTRACT FROM AUTHOR]

Thyroidectomy cells of the rat pituitary gland were studied by the peroxidase-antibody labeling procedure and by electron microscopy. Secretory granules accumulated in these cells in response to a short-term treatment with thyroxine, and the cells were then reactive to the peroxidase-antibody labeling procedure. An intravenous injection of synthetic thyrotrophin releasing hormone (TRH) to thyroxine-treated, thyroidectomized rats provoked an acute and active extrusion of secretory granules from the thyroidectomy cells. The secretory granules in these cells were mostly 'haloed' after primary fixation in osmium tetroxide. It is concluded that TRH causes thyroidectomy cells to release their secretory granules, and presumably TSH, by the usual process of exocytosis or granule extrusion. [ABSTRACT FROM AUTHOR]

4-day cyclic adult female Wistar rats were injected subcutaneously with testosterone propionate on diestrus 1 at 16:00 and on diestrus 2 at 10:00 respectively. Non-injected females served as controls. Autopsy was performed on diestrus 2 at 23:00, and on proestrus at 14:00 and 17:00 respectively. The blue Alcian-PAS staining was used to evidence FSH (β) and LH (γ) pituitary cells. In control animals and in diestrus 2 injected females only a small number of FSH cells could be detected on diestrus 2 at 23:00. This number increased markedly on proestrus at 14:00 and decreased on proestrus at 17:00. A similar evolution was observed in diestrus 1 testosterone injected females, but the number of FSH cells appeared higher at any stage of autopsy in these females than in diestrus 2 injected females and in control rats. In control females, numerous LH cells were observed on diestrus 2 at 23:00. The number of these cells was diminished on proestrus at 14:00 and still more at 17:00. On the contrary few LH cells were detected in testosterone injected females on the evening of diestrus 2. An increase of these cells occurred on proestrus at 14:00, followed at 17:00 by only a weak diminution as established by comparison with control animals. An inhibition of FSH release and a suppression of the proestrus surge of LH were therefore supposed to cause, on one hand, the slowing up of follicular growth observed in diestrus 1 injected females and, on the other hand, the blockage of ovulation noted in both diestrus 1 and diestrus 2 treated animals. In the crested newt, the ultrastructural organization of the pars nervosa is analogous to that already known in non-mammal tetrapods. An orderly array of ependymal cells makes up the inner limiting layer while less abundant pituicytes are irregularly distributed within this organ. 'Light' and 'dark' pituicytes can be distinguished on the basis of the relative density of the cytoplasmic matrix and the distribution of the cell organelles. Both the ependymal cells and pituicytes are rich in dense bodies and possess extensive processes which ramify among the nerve fibers, often reaching the pericapillary space which they can line for long distances. The main components of the pars nervosa are nerve fibers and nerve terminals (type A), containing electron dense granules 1200-2000 Å in diameter together with clear vesicles averaging 250-400 Å. These fibers are likely to correspond to the aldehyde fuchsin positive neurosecretory fibers revealed by light microscopy. Differences in the granule size within the fibers and terminals lead to further recognition of two subgroups (A and A). Other fibers and terminals (type B) containing clear vesicles and granular vesicles 600 to 1000 Å in diameter, possibly of aminergic type, are also encountered. These fibers are rare and can be seen only in the portion of the pars nervosa near the pars intermedia of the adenohypophysis. Lastly, fibers and terminals containing only clear vesicles ranging from 250 to 400 Å (type C) are occasionally found. Nerve endings are often formed by type A fibers on the perivascular space and on the perivascular processes of the ependymal cells and pituicytes. In agreement with recent findings available in the literature, the occurrence of synaptoid contacts between these terminals and both pituicytes and ependymal cells may confirm the active role of these cells in transport and release of neurosecretion. The neurohypophysis of the South American lungfish Lepidosiren paradoxa has been studied with light and electron microscopy, including the Falck-Hillarp technique for catecholamines. The pars nervosa hypophyseos is a well-marked, dorsally located subdivision of the pituitary gland composed of lobes or follicles, each one constituted of a central core of ependymal cells, a subependymal hilar region made up of nerve fibers and a peripheric palisade zone of nerve endings which contact capillary vessels. Four types of neurosecretory axons can be distinguished under the electron microscope. Type I, the most common, contains spherical elementary granules of high electron density, 1500-1800 Å in diameter. The scarce type II axons contain irregularly-shaped elementary granules. Type III contains only small clear vesicles, 400-600 Å in diameter. Type IV, mostly present in regions of the gland contacting the pars intermedia, contain large granulated vesicles, 900-1000 Å in diameter. The Falck-Hillarp technique revealed axons with a positive reaction for catecholamines at sites corresponding approximately to the location of type IV of the electron microscope. Ependymal cells are of large size, linking the cerebrospinal fluid, the nerve endings and the blood vessels. A conspicuous membrane-bound, spherical dense material, 1400-2000 Å in diameter, is observed in both the apical and vascular processes of these cells. The ependymal processes which traverse the hilar and palisade regions contain structures resembling degenerated neurosecretory axons. These results are discussed in relation with the currently available information on the comparative anatomy of the pars nervosa. The possible functional significance of ependymal cells and of each type of axon are also discussed. The cytological changes in the pars distalis of pituitary of male and female green frog, Rana esculenta during the reproductive cycle have been studied by examining the form, cytoplasmic granulation and numerical proportions of the different cell types. Some sex differences were recorded, particularly in relative total number of cells in a median sagittal section of the pars distalis. No extensive cytological changes occur during the hibernation period, but there are striking alterations at the time of spring awakening, during spawning months and during replenishment of the spent gonads i.e. during late summer and autumn months. Important changes are to be observed in the acidophils type one (A1) and basophils type two (B2) and three (B3). Changes in the A1 cells during breeding season have been speculated as being in support of the spawning activity of the animal. The B2 cells exhibit most profound changes which correlate precisely with the morphological changes in gonads, and these cells are considered as the FSH-gonadotrops. The functional significance of the B3 cells is discussed and it is suggested that they might be the source of an ICSH- or LH-like hormone. [ABSTRACT FROM AUTHOR]

Adenohypophyses from more than two hundred white-crowned sparrows of both sexes and different ages, and from different periods of their annual reproductive cycle, have been used for this investigation. In addition to examination of these normal birds, we have also studied the adenohypophyses of 23 castrates and 24 controls held in different photoperiodic conditions. Cytologically the pars distalis of the adenohypophysis of the white-crowned sparrow is typically avian with distinct cephalic and caudal lobes, each with characteristic cell-types. Four basic cell-types, the acidophils, basophils, amphophils, and chromophobes, have been identified in the pars distalis by means of Matsuo tetrachrome and Matsuo modified PAS-methyl blue staining methods. Three types of acidophils, orange, red, and small, are confined to the caudal lobe of the pars distalis. Their possible functions are discussed. Light basophils (PAS-light red cells) and deep basophils (PAS-deep red cells) are equally distributed in both lobes. It is suggested that basophils may be involved in gonadotropic function since their appearance correlates well with the annual gonadal cycle and photoperiodic stimulation of gonadal growth and with the results of castration. The amphophils or PAS-purple cells (aldehyde-fuchsin positive) are found only in the cephalic lobe. Their probable function is discussed. Two types of chromophobes, specific and ordinary chromophobes, have been observed. The specific chromophobes are found only in the cephalic lobe and are similar to the 'Kernhaufen' described by Romeis (1940). The ordinary chromophobes are similar to those of the pars distalis of other avian species and of mammals. The castration cells are found in both lobes of the photosensitive castrates under natural photoperiodic conditions as well as in those subjected artificially to photostimulation (20-hour daily photoperiods). Similar cells have also been observed in the pars tuberalis of the castrated photostimulated birds. An electron microscope study of excised thyroid fragments from a case of De Quervain's subacute thyroiditis is reported. Deposit of material along the basement membrane making the latter apparently thickened, cytoplasmic alterations and stratification of thyroid cells seem to be induced by the thyroiditis process itself. The characteristic giant cells are syncytia resulting from fusion of inflammatory cells. The pathology is specific of a granuloma and is strikingly different from that of the other thyroidites. A case of nonbeta-islet-cell adenoma of the pancreas with profuse watery diarrhea, hypokalemia, acidosis, achlorhydria and negative gastrin assay of the tumor, is presented. All these symptoms and signs disappeared after the removal of the tumor. This case brings the total number of cases with proven WDHA syndrome to 10. Some unusual features, such as flushing, hypercalcemia and normalization of hypercalcemia after operation, are presented and discussed. After excision of the pancreatic tumor, marked acid hypersecretion together with wide fluctuations in gastric secretory pattern, were observed, and are emphasized. It is the second time this finding is reported. Data concerning kidney disease are briefly commented on, in face of the known deleterious effect of potassium depletion upon the kidney.
