Abstract

A radioimmunoassay procedure for cortisol is described that requires only 5 µl of serum or plasma. Native cortisol-binding proteins are denatured by heating 5 µl of sample in 0.2 ml of glutamate buffer in a boiling water bath for 15 min. After a 2-h incubation of heat-treated serum or plasma with [H3]cortisol and cortisol antibody, "free" and "bound" cortisol are separated by use of hemoglobin-coated charcoal. The method is easy and sensitive, and results compare favorably with those of a well-accepted acid-fluorescence method. In addition, we show that serum and heparinized plasma yield the same results by this method, and therefore either can be assayed by this procedure.

Abstract

Exposure of Biomphalaria glabrata infected with Schistosoma mansoni to 14 C-glucose results in a greater uptake of original total snail label by the parasitized digestive gland-gonad, site of the developing daughter sporocysts and cercariae, than by the digestive gland-gonad of control animals. As a consequence of this greater uptake by the infected digestive gland-gonad, the albumen gland and remainder of the carcass of parasitized snails receive less label than do those areas in normal snails. Emergence of cercariae from the snail and daughter sporocyst mass account for a diversion of 12.6% of original total label from the infected snail itself. This diversion of label from the snail to the parasite may explain carbohydrate depletion in parasitized snails.

Abstract

Three types of reaction are of particular value in the synthesis of fluoro sugars, namely, nucleophilic displacements with fluoride salts, epoxide cleavage reactions, and glycal addition reactions. These reactions have been developed to the point where effective syntheses of a wide range of fluoro sugars can be planned on a rational basis. Nucleophilic displacement usually involves the treatment of sulphonates with fluoride salts. Reagents of the type tetrabutylammonium fluoride-dipolar aprotic solvent are particularly effective for the displacement of secondary sulphonates. An understanding of the steric and polar factors which can adversely influence the displacement of carbohydrate secondary sulphonates and of competing reaction pathways usually allows the evaluation of this route in designing effective syntheses of fluoro sugars. Selection of ring size (furanose, pyranose, septanose derivatives) is an important parameter in synthesis design. The cleavage of carbohydrate epoxides to give Trans-fluorohydrins can be effected with reagents such as HF or KHF2. The reactions usually occur stereo-specifically and predictably if the epoxide ring is part of a rigid molecular system such as 1, 6-anhydrohexopyranose. The reagent CF3OF, which is an effective source of electrophilic fluorine, readily adds to O-acetylated glycals to give 2-deoxy-2-fluoro derivatives. 2-Deoxy-2-fluoro-D-glucose can be converted into 3,4,6-tri-O-acetyl-2-fluoro-D-glucal and thence by treatment with CF3OF, into ‘2,2-difluoro-D-glucose.’

Numerous attempts have been madeto demonstrate the specific cause of the extremely prevalent catarrhal affections of the upper respiratory tract, popularly known as "common colds." In reviewing the literature, however, one is struck by the multiplicity of organisms to which an etiologic relation has been ascribed. B. influenzae, Strep, hemolyticus, Strep, viridans, the pneumococcus, M. catarrhalis, B. septus, Friedlanders bacillus and Tunicliff's B. rhinitis have each held the stage. Some investigators believe that micro-organisms bear no relation whatever to the causation of colds; but that many factors?wet and cold, drafts, irritating vapors, overheated rooms, worry, fatigue, sexual excesses, dietetic errors, alcohol, and what not?enter into the etiology. Still another group takes the stand that a number of conditions?notably chilling, sudden changes in temperature, and fatigue? act as predisposing factors by lowering resistance, and thus pave the way for infection by micro-organisms commonly believed to be present normally in the nasal cavities. In short, present knowledge of the etiology of the common cold is in a most chaotic state. As remarked by Stitt,1 "Of all the diseases common in man this condition has been surrounded by greater etiological and epidemiological obscurity than any other."

Abstract

For the convenience of this article I shall use the term " modern science " in a restricted sense, so as to exclude from consideration its most recent developments. Thus by " modern physics" I mean what is now sometimes called " the classical physics ". I do this not because I wish to imply that what I say of it is not true also of the most recent developments of science, but becaiqse I do not wish to raise the question here whether it is or not. I approach my subject by way of a consideration of modern philosophy, and I apply the term " modern " to philosophy with a similar restriction, meaning by " modern philosophy " the philosophy which arose at the end of the Middle Ages and developed alqng the two main lines of Empiricism and Rationalism from Hobbes to Hume and from Descartes to Leibniz. About this philosophy as a whole I shall make two assumptions which I think will not be disputed. The first is that it was devoted (in so far as it was concerned with a theory of nature) mainly to establishing the possibility or justifying the presuppositions of the modern science of nature. It is necessary to use these two alternative descriptions because the relation of philosophy to science varied according to the degree of development which the latter had achieved at the time. By the later centuries of the modern era the sciences of nature had become so firmly established that they formed a datum from which philosophical speculation could start. This does not of course mean that the philosopher -dogmatically accepted the truth of any scientific hypothesis.

Detailed analyses have been made of the 1H and 19F n.m.r. spectra of the α- and β-anomers of 2,4,6-tri-O-acetyl-3-deoxy-3-fluoro-D-glucopyranosyl fluoride and of 2,4-di-O-acetyl-3-deoxy-3-fluoro-β-D-xylopyranosyl fluoride. The 4JF,F, couplings demonstrate a stereospecific dependence with 4Ja,a = + 10.4, 4Ja,e = + 1.0, and 4Je,e = −3.0 Hz.

The 1H and 19F n.m.r. spectra of the α- and β-anomers of 3,4,6-tri-O-acetyl-2-deoxy-2-fluoro-D-glucopyranosyl fluoride and of the corresponding D-manno derivatives have been fully analyzed. The signs of most of the 19F–1H and of all the l9F–19F couplings have been determined by a heteronuclear double-resonance method. The vicinal 19F–19F couplings are absolutely negative in sign, with Ja,a = – 20.0 Hz, and Ja,e and Je,e = – 13.5 to – 18.8 Hz. Sizeable long-range couplings (4J) between an axial fluorine and an axial proton are absolutely positive in sign.

The mass spectra of these derivatives were rationalised in terms of the position of the fluorine substituent on the carbon chain.

Abstract

A radioimmunoassay procedure for thyroxine (T4) is described, for which only 25 µl of serum is required. The antibody, commercially available, is produced in rabbits in response to injections of a T4—albumin conjugate. Native T4—binding proteins are denatured by heating 25 µl of serum in 0.2 ml of a glutamate buffer in a boiling water bath. After a 1-h incubation of heat-treated serum with I125-T4 and T4 antibody, "free" and "bound" T4 are separated by use of hemoglobin-coated charcoal. The method has excellent sensitivity, is rapid, and possesses significant advantages over a standard competitive protein binding method with which it is compared.
