Robenidene has appreciable anticoccidial activity against Eimeria brunetti and E. tenella in chickens. In the case of E. tenella under continuous drug medication, this activity has been shown to be primarily coccidiostatic at the stage of the almost mature first generation schizont. The stage at which the drug acts was predicted from the mortality pattern in experiments involving restricted administration of drug and confirmed by histological examination of caeca from treated birds and by observation of parasites in tissue culture. The fine structure of sporozoites of Isospora canis closely resembled that of sporozoites of various Eimeria species, except that they had crystalloid bodies in place of refractile bodies, and 26 subpellicular microtubules instead of 24 or 25. Two crystalloid bodies, one anterior and one posterior to the nucleus, were present. Each body consisted of irregularly arranged particles, each of which had a number of dense granules embedded in a less dense matrix. The anterior crystalloid body was closely associated with rhoptries, the posterior one with amylopectin granules, and endoplasmic reticulum and mitochondria were adjacent to both. Sporozoites, trophozoites, and early schizonts of Eimeria ninakohlyakimovae grown in cultured cells were examined with the electron microscope. Intracellular sporozoites had ultrastructural characteristics similar to those of extracellular sporozoites, except that apparently functional micropores were observed. During transformation of sporozoites to trophozoites, the conoid, annuli and subpellicular tubules, as well as some portions of the inner membrane complex of the pellicle, disappeared. Trophozoites and binucleate schizonts had a reduced number of micronemes and rhoptries. In schizonts with 3 nuclei, only a few remnants of the micronemes and rhoptries were seen and the inner membrane complex had almost completely disappeared. One such schizont had one or more centrioles associated with each nucleus; in one, an eccentric spindle with a dense, coneshaped structure at each pole was observed. A moderately dense, granular crescent body, probably derived from host cell material, was seen within the parasitophorous vacuole in some specimens. [ABSTRACT FROM AUTHOR]
