A comparative study of the intrinsic innervation in desert rodents (kangaroo rats) and others (albino rats) was carried out in an attempt to understand the functional anatomy of the bladder in these animals which are known to sustain severe water restraint. The bladder of the albino rat was innervated by predominantly thin nerves, more numerous beaded endings and few ganglia. That of the kangaroo rat had more numerous thick nerves (pre-ganglionic), large verve trunks, and ganglia which were extensively distributed in the wall. These findings indicate that the bladder of the albino rat depends mainly on the intrinsic innervation and facilatory micturition reflexes, while that of the kangaroo rat is intrinsically regulated, depending on a short neuron system. It was concluded that all the structural differences found might be essential for constant urine retention.

We have studied extrajunctional muscle of control and dystrophic mice by electron microscopic-cytochemistry and radiometric assay. We have found both a soluble and particulate AChE activity, which is similar proportionally in control and dystrophic muscle. The particulate AChE activity is probably due to the enzyme localized in the sarcotubular system. These sites are more numerous in muscle adjacent to the motor end-plant than in distally located extrajunctional muscle, and are increased markedly in the dystrophic mouse. Myoblasts and small muscle fibers in the dystrophic mouse also have AChE activity in the reticulum similar to fetal muscle. The soluble AChE activity identified radiometrically may represent those sites exhibiting random cytochemical end-product, such as some muscle nuclei, satellite cells, myogenic mononuclear cells in the connective tissue, and degenerating axonal boutons no longer associated with junctional folds of muscle. Enzyme activity is present in degenerating fibers, but it is randomly dispersed in the sarcoplasm rather than membrane-bound. AChE activity has not been found in debris of completely necrotic muscle. BuChE activity is higher and the number of BuChE-active sites in the sarcotubular system adjacent to the motor end-plates is greater in dystrophic muscle than in control muscle.

1 Administration of corticosterone caused a 47% reduction in the cholinesterase (ChE) activity of homogenates of the rat anococcygeus muscle. 2 ChE activity was also reduced by morphine withdrawal and this effect was abolished by the corticosteroid synthesis inhibitor, metyrapone. 3 A single dose of reserpine reduced ChE activity in normal but not in adrenalectomized rats. 4 ChE activity was increased by adrenalectomy or by metyrapone treatment. 5 The mechanism of the corticosteroid-induced reduction in ChE activity is discussed. 6 The reduction in ChE activity produced by corticosterone, morphine withdrawal, or a single dose of reserpine might explain the leftward shift of the dose-% response curve to acetylcholine produced by these procedures in the isolated anococcygeus muscle of the rat.

Acute experiments were conducted on male rats; it was revealed that electrical stimulation of the nuclei of the anterior hypothalamus led to reduction of choleresis and to a fall of the potassium content in the bile, increase of permeability of the connective tissue stroma of the liver and of cell membranes of hepatocytes. Stimulation of the posterior hypothalamic nuclei induced an increase in choleresis, of the concentration of potassium ions and cholates in the liver, and also of the cholinesterase activity in the serum and homogenates of the liver. Permeability of connective tissue structures of the liver and of the intercellular spaces of hepatocytes proved to fall. The influences of the hypothalamus on bile formation included a change in the functional activity of hepatocytes and of the production of a fluid fraction of the bile.

End-plate formation in free autologous transplants of previously denervated whole muscles in adult cats were studied with a histochemical technique for cholinesterase. Grafts from 5 to 40 weeks old have been examined. The original end-plates degenerated and could not be seen in transplants older than 9 weeks. New, immature end-plates formed at many sites in the transplants. They were seen as early as 5 weeks after transplantation. They progressively increased in number during the first 12 weeks. In the oldest transplants the end-plates were relatively mature and appeared in clusters or were scattered.

15 acetoxyethylenammonium compounds are studied as substrates for acetylcholinesterase (ACE) from bovine erythrocytes and for butyrylcholinesterase (BCE) from horse serum. Substitution of methyl groups of the ammonium grouping with other radicals and incorporation of onium nitrogen in the cycle resulted in the decrease of the hydrolysis rate under the action of BCE and ACE, the effect of BCE being more pronounced. The rate of the hydrolysis of N-acetoxyethylene-N-methylpiperidine iodide in the presence of ACE was 65 times as much as in the presence of BCE. This compound is a new specific substrate of ACE. Dipropylmethyl derivative turned not to be a good substrate for both enzymes. Dibutylmethyl and pyridinic derivatives were not attacked by ACE and BCE. Kinetic analysis of the compounds listed is performed, taking account of non-productive sorbtion. Possible role of hydrofobic regions in the orientation of substrates on the active surface of ACE and BCE is discussed.

Following a previous comparison of the peroneus longus muscle of the quail and the starling, the present paper deals with a comparative study of this muscle in two birds of the order Gallinaceae, the quail and the bantam, bearing in mind certain data found in the starling. The study deals with the types of muscle fibres, their frequency in various parts of the muscle, their lipid content and their innervation. In the quail, two types of fibres are found, with a low and high lipid content respectively, while the bantam and starling have also a third, intermediate type. These intermediate fibres have a characteristically intermediate lipid content, peripherally situated nuclei and areas with a myofibrillar structure. The proportion of the two of three types of fibres varies with the species and in the distal and proximal parts of the muscle. The innervation of the peroneus longus muscle is different in the three species. In the quail, the two types of fibres have only one arboriform motor end-plate per fibre. In the bantam, the two types of homologous fibres also have only one motor end-plate, but with fewer arborifications. The intermediate fibre type, on the other hand, is innervated by several small nerve endings for each muscle fibre. This type of multiple innervation is also found in the starling. The peroneus longus muscle is thus functionally different in the two birds of the order Gallinaceae, whereas the rapid and slow system of innervation is found in the bantam and the starling. In the quail and the bantam, there is a strong positive correlation between the diameter of the muscle fibre and the longitudinal extent of the motor end-plate. This correlation is not marked in the starling. The characteristics of the innervation revealed by the cholinesterase activity concentrated in the synaptic grooves were confirmed by a direct study of the nerve fibres, using the Bielschowsky-Gros method. In the quail only 'en plaque' endings are found, while in the bantam and the starling both 'en plaque' and 'en grappe' endings are present. A parallel is drawn between the differences in function of the peroneus longus muscle and the characteristic features of its histology and innervation in the three species.

Butanol-1 and butandiol-1,4 are shown to increase the decarbamoylation rate of N-methylcarbamoyl- and N,N-dimethylcarbamoyl-cholinesterase. It is mainly due to the formation of a ternary complex NEA which is decomposed in 2,5 times faster than corbamoyl-enzyme EA. This is an evidence for the presence of some allosteric center in cholinesterase which is capable of binding alcohols.

Cushion veins of the human nasal lining were studied in eight patients of both sexes ranging in age from 11 to 59 years. It was found that the subendothelial cushions were part of the tunica media and consisted of smooth muscle cells, collagen and elastic fibers and occasional fibrocytes. The muscle fibers of the cushion nearest to the endothelium were circular. They extended processes towards the endothelium through gaps in the endothelial basement membrane and formed appositional junctions with the endothelial cells. The rest of the cushion consisted of longitudinal muscle fibers. The sacoplasm of the muscle cells was characterized by large areas filled with vesicles of various sizes. In addition, these cells possessed cytoplasmic processes which were devoid of a basement membrane and which did not show the regular structure of sarcoplasm. The subendothelial cushion possessed a rich, intrinsic nerve supply of adrenergic and cholinergic axons. It is suggested that the cushion veins regulate the drainage of the cavernous tissue and are under nervous and humoral control. The increase in girth of the subendothelial cushion is effected by contraction of the longitudinal muscle cells and probably by uptake of extracellular fluid by means of the specialized cytoplasmic processes. The single layer of circular muscle cells situated between the endothelial lining and the longitudinal musculature, may provide protection to the endothelium against distension when the cushion expands.

The purification of the pregnancy zone protein by means of immunoadsorbents is described. The pregnancy zone protein antibody was isolated from an absorbed rabbit antiserum and coupled with CNBr-activated sepharose. The pregnancy zone protein was isolated from pregnancy serum by the specific antibody cross-linked with sepharose. Contaminating serum proteins were eliminated by "inverse" immunoadsorption using antibodies against these proteins coupled with sepharose. An immunoelectrophoretically pure pregnancy zone protein was obtained. By means of a combination of immunoprecipitation and enzyme reaction in agar gel could be excluded that the pregnancy zone protein possesses activities of the following 11 enzymes: ceruloplasmin, leucine amino peptidase, alkaline phosphatase, carboxylic esterase, lactate dehydrogenase, malate dehydrogenase, glycerophosphate dehydrogenase, glucose-6-phosphat-dehydrogenase, cholinesterase, acetyl cholinesterase and oxytocinase.

In two new cases of centronuclear myopathy, histochemical findings included failure to differentiate fiber type with oxidative enzymes, the presence of core glycogenosis, and core acid phosphatase activity. Fluorescence microscopy demonstrated autofluorescent lipochrome in fiber centers and nonspecific fiber injury in some immunofluorescent preparations. Electron microscopical findings included the observation of unusually small myofibrils arrayed between central nuclei.

Rainbow trout (Salmo gairdneri) were exposed to IMOL S-140, a synthetic high-temperature lubricating oil composed of tri-aryl phosphates and, although no sign of acute toxicity was evident, the fish slowly developed symptoms of chronic posioning. Thus, floating food pellets were refused, the activities of glutamic oxalacetic transaminase and lactate dehydrogenase in serum were greatly elevated, and internal fatty tissues became discoloured. Neither serum nor brain cholinesterase activities were inhibited.

Following 25 mug/day synthetic alpha-MSH administration, the liver regeneration of partially hepatectomized rats proved to be increased. The hormone treatment resulted in an enhanced alanine incorporation of the liver proteins, but this effect was uncertain on partially hepatectomized rats. Due to the hormone treatment the low liver protein content of the operated rats became normal. The pseudocholinesterase activity of the liver homogenate of alpha-MSH treated rats was also elevated. On the basis of these experiments authors are supposing some protein synthesis increasing effect of synthetic alpha-MSH.

The effects of acute (10 mg/kg) and chronic 10 mg/kg for 30 days) administration of delta-9-tetrahydrocannabinol (delta9-THC) have been studied histochemically in the rat adrenal medulla, which include total catecholamines, noradrenaline, histometric measurements of adrenal medullary areas, calcium content of the medullary cells along with adenosine triphosphatase (ATPase), acetyl cholinesterase (AChE) and butyryl cholinesterase (BChE) activities. Acute delta9-THC treatment reduced the total catecholamine content (including noradrenaline) of the gland, was accompanied by increased ATP-ase, AChE, BChE activities and increased calcium distribution in the gland. Chronic delta9-THC treatment caused significant hypertrophy of the chromaffin tissue, with decreased total catecholamine content, although noradrenaline containing areas exhibited no notable change. The calcium content and ATPase activity were increased along with a concomitant increase in AChE and BChE activities. Although the changes in adrenal medullary enzyme activities following both acute and chronic delta9-THC treatment are qualitatively similar, marked quantitative increase is noted in the chronically treated groups. The results indicate an increased total catecholamine releasing activity of the adrenal medulla following acute delta9-THC treatment, while chronic delta9-THC administration produces a preferential release of adrenaline.

1. Female albino rats were treated with a total of 28 mg of chlormadinone acetate (CMA) for 28 days. In the adrenal cortex, the ovary, the vagina, and the uterus the activities of 3-beta-ol-steroiddehydrogenase, of dl-beta-OH-butric acid dehydrogenase, of alcaline and acid phosphatases, of DPN-diaphorase, of ATP-ase, and of non-specific esterases do not differ from untreated controls. 2. In the external muscle layer of the myometrium strong cholinesterase (ChE) activity was induced by C.M.A. A corresponding high ChE activity is normally found in immature rats or in estrus. 3. Furthermore, by treatment with CMA, ChE activity was induced in the tubular glands of the endometrium. This activity is found in the small parts of glomerate glandular terminals only but not in the rest of the glandular epithelium, nor in the epithelium of the cavum. It could be demonstrated that a corresponding ChE activity normally appears in the second third of pregnancy. The ChE activity induced by CMA was considerably higher and more widespread than during normal pregnancy. 4. It is concluded that in the endometrial glands a development similar to pregnancy is initiated by CMA. But development stops at the stage of ChE activity, thus leading to accumulation of ChE active cells.

Patterns of mortality and inhibition of brain and plasma ChE in birds treated with ChE inhibitors were studied in an attempt to determine the validity of using ChE activity as a monitoring and diagnostic technique. Analysis of brain ChE activity proved to be reliable for diagnosing and monitoring effects of selected ChE inhibitors in birds. Brain ChE inhibition exceeding 20 percent indicated exposure, and inhibition greater than 50 percent was sufficient for diagnosing cause of death. Individuals that died from dietary exposure to parathion-1 or carbofuran had brain ChE activities below 55 percent of normal; although individuals could survive with brain ChE activity lower than 50 percent. Problems associated with collection, storage, and analysis of tissues for ChE activity are discussed.

(1) Crude synaptosomal fractions (P2) derived from guinea-pig cerebral cortex were incubated in the presence of 50 mM KCl in a Krebs-glucose medium. Torpedo marmorata electric organs were stimulated electrically in vivo at 5 pulses/sec for 30 min by electrodes placed on the electric lobe. Synaptic vesicles were isolated from each source and the phospholipid compositions analysed and compared with vesicles from unstimulated controls. (2) Lysophosphatidylcholine was the only lysophosphoglyceride demonstrable in the synaptic vesicles from either source and its low levels did not increase as a result of chemical or electircal stimulation. In each case there was a close similarity of the phospholipid distributions in the vesicles taken from control and stimulated samples. (3) Control experiments indicated extensive decreases in the acetylcholine content of the vesicles from the stimulated electric organ and smaller decreases in the acetylcholine content of the synaptic vesicles from stimulated crude synaptosomal fractions. These fractions were found to respire linearly in the presence of 10 mM glucose and the vesicle fractions were shown to have low levels of contaiminating membranes as judged by marker enzyme analyses. (4) Crude synaptosomal fractions from guinea-pig cerebral cortex were incubated in a Krebs-glucose medium with labelled fatty acids and [3H]glucose in the presence or absence of 50 mM KCl. Subsynaptosomal fractionation was carried out and specific radioactivities of phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine and phosphatidylinositol were determined in fractions D (synaptic vesicles), E (microsomes) and H (disrupted synaptosomes). The release of neurotransmitter did not significantly enhance the labelling of phospholipids in any of the fractions studied as compared with phospholipids from unstimulated fractions. This was found after two incubation times and using [14C]oleate, [14C]arachidonate, [3H]palmitate and [3H]glucose.

In blood serum of patients with lymphogranulomatose, as compared with healthy persons, a decrease in activity of one of the arylesterase fractions was observed. In lymphogranulomatous process content of this fraction and content of cholinesterase were decreased in liver tissue more distinctly than in the blood. At the same time the arylesterase activity was increased; the enzyme was found only in trace amount in normal liver tissue. The impairments in content of lactate dehydrogenase isozymes were less distinct. In lymphogranulomatose the activity of alkaline phosphatase was increased, especially in cases accompanied by impairment of liver tissue.
