To increase the efficiency of human and financial resource allocations, the El Dorado County, California, Office of Education initiated a planning, programing, budgeting system (PPBS). This document is a progress report on the effectiveness of the system after one year of operation. It contains (1) a statement on goals, (2) charts of the program structure, and (3) descriptions, budgets, and financial plans for 69 instructional and administrative programs. The molecular weight of purified human erythropoietin was determined by SDS-polyacrylamide gel electrophoresis. Five crystallized proteins (BSA, aldolase, trypsin, hemoglobin, and lysozyme) were used for the calibration curve. All the proteins were labeled with 131I prior to electrophoresis in order to determine their mobility. The molecular weight of human erythropoietin is 23.000 according to the data obtained.

The present paper describes a scheme of immunization for the production of anti-erythropoietin (anti-ESF), which has been shown to be very effective. The capacity for the neutralization of erythropoietic activity was 12, 35, 125, and 480 units of erythropoietin (Standard B) per milliliter of antiserum for each one of the immunized rabbits. The absorption of the antiserum with urinary proteins did not modify the titers of the antisera. The velocity of the complex ESF-anti-ESF formation has been studied. We have also investigated its dissociation by means of pH and temperature. The precipitation capacity of the antiserum with the higher titer is proved using the Ouchterlony double-immunodiffusion method.

Partition chromatography on paper makes possible the separation of rubber hydrocarbon from latex. This method offers a means which is both simple and rapid for determining the presence of rubber in plant latexes and for the subsequent estimation of its percentage in a given latex. The method also gives a fraction which is composed of the nonrubber components. In addition to this, the method makes possible a comparison of the partition coefficients of rubber hydrocarbon isolated from latexes of different plants. Experiments are now in progress, the purpose of which is the application of the chromatographic method to the rapid determination of plant rubbers. By its simplicity, this method unquestionably has certain advantages in the identification of rubber in any particular flora and in studies of this hydrocarbon in its various phases of the biological cycle of the plant.

1. 1. Erythropoietin, purified from urine extract by several purification steps, with a spec. act. of 8000 units/mg of protein was tested for heterogeneity and partially characterized. 2. 2. The purified material, subjected to gel filtration on Sephadex G-100, electrophoresis on polyacrylamide gel and double immunodiffusion in agar gel was essentially homogeneous. 3. 3. The absorption spectrum shows a maximum at 279 nm. The E1%1cm at this wavelength is 9.26. 4. 4. The composition in total hexoses, hexosamines, sialic acid, proteins and amino acid is reported. 5. 5. Erythropoietin is an acid glycoprotein with a 30% carbohydrate content according to the presented results.

1. 1. Human erythropoietin has been separated into several components by means of polyacrylamide gel electrophoresis. 2. 2. Chromatography on DEAE-cellulose using a linear gradient of NaCl for elution also permits separation of the activity into several peaks. 3. 3. Fractions separated by chromatography on DEAE-cellulose present different mobilities in polyacrylamide gel electrophoresis, but similar elution volumes when they are filtered through a Sephadex G-100 column. Immunological differences are not observed when these fractions are neutralized with the antibody antierythropoietin 4. 4. Erythropoietin pre-incubated with sodium dodecylsulphate gives only one symmetric peak when it is eluted with a linear gradient of sodium chloride from DAEE cellulose. 5. 5. The activity profile on sodium dodecylsulphate polyacrylamide gel electrophoresis gives only a narrow peak, but gel electrophoresis in urea does not modify the pattern of activity along the gel. 6. 6. The experimental data obtained suggest that the heterogeneity of erythropoietin could be due to differences of charge. No substantial differences in molecular weight, or aggregation of sub-units may be inferred.

The electrophoretic behavior of erythropoietin in polyacrylamide is studied. The biological activity is distributed in a wide area located between the pre- and the postalbumin region, separated into several components. Reelectrophoresis of the components with different mobility shows that there is no interchange from one to another. Isoelectric point values of major active components determined by electrofocusing, are in pH range 3.0–3.9 and minor components in pH 4.1, 4.5, and 6.0. The possibility that erythropoietin could be related to a family of protein with differences in charge is discussed.
