In a randomized double-blind trial hydroxyurea was compared to placebo in conjunction with radiotherapy of head and neck cancer. Chemotherapy was given orally twice a week (80 mg∕kg) concurrently with irradiation. Comparison of the 2 groups for primary tumor regression, cervical node response, and early survival fails to show statistically significant differences. Combined treatment may be useful in massive node metastases. Increased local radiation reactions were frequent in the hydroxyurea group and severe with interstitial implants. The drug was generally well tolerated. Complications of treatment and an apparently higher incidence of distant metastases are discussed.
