Abstract

In a paper read before the Royal Society of Edinburgh in April 1843, Dr Maclagan described the general properties of the alkaloid, whose presence had been indicated in the bark of the bebeeru or greenheart tree, by Dr Rodie of Demerara, and described the mode of preparation of its sulphate for medicinal use. The fact that bebeerine appeared to possess marked antiperiodic properties, rendered its careful chemical study desirable, and accordingly the alkaloid, purified as far as possible, was subjected to analysis by Drs Maclagan and Tilley. It resulted from this research that bebeerine is an uncrystallisable base, very soluble in alcohol, less so in ether, and very sparingly so in water. It forms with acids salts which are all uncrystallisable. With perchloride of gold, mercury, copper, and platinum, it gives precipitates which are soluble to a certain extent in water and alcohol, but which are deposited in a non-crystalline form when the solution cools. To this base the author assigned the formula C35H20N2O6(C = 6). Von Planta subsequently attempts to purify further the alkaloid, and assigned to it the formula C36H21O6N(C = 6) or C18H21O3N(C = 12.)

Bebeerine is a vegetable alkaloid, discovered by Dr Rodie of Demerara, in the bark of the Bebeeru tree,Nectandra Rodiei, Schomburgk. The properties of the alkaloid and its application in the form of sulphate, in the treatment of disease, were formerly described by Dr Maclagan in a paper read before the Society, and published in their Transactions, vol. xv., part 3.

Abstract

This chapter describes the alkaloids containing an isoquinoline or reduced isoquinoline ring-system. Structure determination and synthesis is discussed for the derivatives of isoquinoline and tetrahydroisoquinoline, which comprise of the simple tetrahydroisoquinolines, 1-Phenyltetrahydroisoquinolines, such as the cryptostylines, cherylline, and papaverin, one of the principal alkaloids of opium. Also synthetic methods including biosynthesis are shown for the cularines, the dibenzopyrrocoline alkaloids, pavinanes and isopavinanes, the bisbenzylisoquinolines, aporphinoids, the protoberberines, the protopine alkaloids, the phthalideisoquinoline alkaloids, the rhoeadines and papaverrubines, the benzo[c]phenanthridines, the spirobenzylisoquinolines, the Ipecacuanha alkaloids, colchicine, and 1-phenethylisoquinolines. It has become apparent, as the result of a large number of studies on the biosynthesis of the more highly condensed alkaloids of isoquinoline (protoberberins, protopines, aporphines, and so on) that norlaudanosoline may be regarded as the common precursor of all of them. The bisbenzylisoquinoline alkaloids, which are widely distributed in plants of the Menispermaceae family growing in South America and the Far East, and in Daphnandra, Berbis, Magnolia and Nectandra families, is one of the largest sub-groups of the isoquinoline alkaloids. The n.m.r, spectra of benzo[c]phenanthridines have attracted some attention, and are now of considerable importance in structural studies. The spectra of noravicine, nornitidine and norsanguinarine in trifluoracetic acid solution are highly characteristic.

Abstract

When the wood of the bebeeru tree is subjected to a process similar to that recommended in the British Pharmacopœia for the separation of bebeerine from the bark, a mixture of several alkaloids is obtained.In this memoir the authors specially describe the properties of one of these alkaloids, to which they have assigned the name Nectandria and the formula C20H23NO4. (Bebeerine, according to V. Planta, has the formula C18H21NO3). Nectandria is obtained by treating the mixed alkaloids obtained from the wood with chloroform, in which it is abundantly soluble.
