In the hyperpolarizing photoreceptors of the scallop Pecten irradians the metabolic inhibitors cyani 'e and 2,4-dinitrophenol cause a rapid hyperpolarization and increase in membrane permeability to potassium ions, similar to the effect of light. Cellular metabolism appears important in maintaining the low permeability to potassium ions necessary to keep the membrane depolarized in darkness, possibly by regulating the intracellular calcium ion concentration.

Inactivation of adenylate cyclase in outer segments of retinal photoreceptor cells is proportional to the bleaching of rhodopsin. Membranes of the outer segments also contain a particulate, light-insensitive phosphodiesterase of high specific activity. In electrophysiological experiments, application of cyclic adenosine monophosphate along with a methylxanthine mimics the effects of illumination on the photoreceptor cell of the compound eye of Limulus.

Phosphorylcholine-1,2-(14)C and choline-1,2-(14)C-labeled cytidine diphosphate choline are incorporated into lecithin by whole homogenates and particulate fractions of rat retina with optimal incorporation of label by the microsomal fraction. The soluble fraction contains a factor(s) which stimulates incorporation of label with release of inorganic phosphate. Mg(++) is required for optimal incorporation of intermediates into lecithin in the presence of added diglycerides; without added diglycerides, incorporation of phosphorylcholine or cytidine diphosphate choline was moderately stimulated by preincubating the system in the absence of Mg(++) with added phosphatidic acid and by adding this mixture to fresh enzyme and the complete incubation mixture (including Mg(++)). The results show that the retina is capable of de novo synthesis of phosphatides and suggest that the rod outer segments depend on the pigment epithelium and(or) the inner rod segments for a source of phospholipids. Coenzyme A and ATP added to whole homogenate of retina did not significantly increase the incorporation of CDP-choline-1,2-(14)C into lecithin but slightly increased the radioactivity found in lysolecithin and sphingomyelin. Rats with hereditary retinitis pigmentosa have an abnormally high lipid phosphorus content of the retina, but they do not incorporate labeled CDP-choline into lecithin of retina at a higher rate than do normal animals.

Isolated pigeon and rat retinas were incubated in various media and the effect of changing ions on the P III component of the electroretinogram was studied, with particular regard to the influence of calcium on other ions.2. In pigeon, without calcium, reduction of sodium and chloride both reduce P III. This behaviour continues in the presence of calcium but only if the retina has been treated with ouabain. Under these conditions, the effect of calcium is to prevent the appearance of inverted responses.3. Under other conditions, calcium renders the pigeon P III insensitive to change in chloride. Sensitivity is rapidly restored by the use of ouabain, 2,4-DNP, or by cooling.4. Calcium does not alter the linear relationship between sodium and response amplitude.5. In pigeon, increasing potassium increases P III amplitude, though the exact relationship is altered by calcium.6. In the rat retina treated with ouabain, increasing sodium increases P III, and increasing potassium decreases P III. This result is similar to that of Sillman, Ito & Tomita (1969b). If calcium is absent, inverted P IIIs can be obtained.7. If calcium is present in the medium, reducing sodium also reduces rat P III.8. If calcium is reduced below 10(-7)M, then reducing sodium increases P III, and large responses are obtainable from solutions which contain almost no ions. These responses vanish very rapidly if the retina is cooled or treated with ouabain.

1. By applying atomized chemical solutions on to gecko and carp retinae, neuropharmacological reactions of the photoreceptors and horizontal cells were observed.2. Sodium L-glutamate and L-aspartate, glycine, ACh and GABA, had no appreciable effect on the photoreceptor activity.3. Carp horizontal cells were depolarized by both L-glutamate and L-aspartate. When maximally depolarized, the action of the endogenous transmitter from the receptor terminals was completely masked, resulting in abolition of the S-potentials.4. Responses to red light in both L- and C-type horizontal cells were more strongly affected by aspartate than responses to blue light.5. Glycine and GABA hyperpolarized the horizontal cells, and the S-potentials were diminished.6. ACh had no effect on the activity of the horizontal cells.
