36. Effects of Local Immunization with Streptococcus mutanson Induction of Salivary Immunoglobulin A Antibody and Experimental Dental Caries in Rats

Abstract

The effect of local immunization with Streptococcus mutanson dental caries in conventional and gnotobiotic rats was studied. Injection of these animals with S. mutansstrain 6715 incorporated into complete Freund adjuvant consistently resulted in the presence of antibody in saliva directed to this organism. This antibody was primarily of the immunoglobulin A class as demonstrated by specific antiglobulin augmentation and gel filtration of antibody activity. Serum antibody was also present. Five experiments have been completed in conventional rats and two in gnotobiotic animals. The immunized group of animals always had lower mean caries scores than comparably sham-immunized or nonimmunized control groups. The numbers of lesions were also always lower in the immunized animals, suggesting a possible interference with the formation of new lesions in immunized animals. The reductions in dental caries and lesions were greater on smooth surfaces than on occlusal surfaces. which might be explained as interference with adherence phenomena demonstrated by S. mutans.It is proposed that salivary immunoglobulin A antibody may be viewed as an ecological determinant in the oral cavity by affecting oral microorganisms and possibly their by-products.

THE USE OF PLAY THERAPY WITH MENTALLY SUBNORMAL CHILDREN IS EXPLORED, AS IT ENABLES SUCH CHILDREN TO REALIZE THEIR POTENTIALS AND TO INTERACT MORE ADEQUATELY WITH THEIR ENVIRONMENT. A CONSIDERATION OF GENERAL AND THEORETICAL PROBLEMS AND A CONCEPTION OF THE DEVELOPMENT OF MAN'S ESSENTIAL QUALITIES ARE PRESENTED. A GENERALIZED THEORY OF PLAY THERAPY IS ESTABLISHED AND RELATED TO THE LEARNING PROCESS. THEORETICAL IMPLICATIONS OF PLAY THERAPY APPROPRIATE FOR SUBNORMAL CHILDREN ARE DISCUSSED. TECHNIQUES AND PROCEDURES OF PLAY THERAPY ARE EXAMINED. 39. Ultracentrifugal and Immunological Comparisons of Succinylated Human Thyroglobulin and 27 S Iodoprotein

Abstract

Human thyroglobulin (Tg) and thyroid 27 S iodoprotein were dissociated with succinic anhydride (SA), to examine similarities in subunit organization and in immunological reactivity of these subunits. In the analytical ultracentrifuge, S-9, S-11, and 5-16 components arose from 19 S Tg dissociation. Four species, including S-7, S-12, S-15, and S-19 subunits, were formed upon dissociation of 27 S protein. More than 80% of the Tg was transformed into the S-9 component at higher SA concentrations. At similar SA levels, all four of the 27 S protein subunits remained. Immunological reactivity of the intact and succinylated iodoproteins was tested with rabbit anti-Tg sera and human autoimmune thyroiditis sera. Products of both Tg and 27 S protein dissociation behaved similarly in immunodiffusion (ID) and in inhibition of tanned cell hemagglutination (TCHI). All succinylated products, even after purification, were active as inhibitors in TCHI against both types of sera. Also, every dissociated sample precipitated with the heteroimmune sera. However, moderately dissociated Tg or 27 S iodoprotein lost the ability to precipitate in gel diffusion with the autoimmune serum.

A polystyrene replica technique, similar to that employed in electron microscopy, has been used to prepare extremely thin films of identical surface structure. These thin transparent films can be transferred to any desirable surface. Replicas made from gloss standards are shown to be reproducible in surface structure and spatial reflectance characteristics.

41. Homologous Transplantation of Canine Lungs: Technique with Contralateral Pulmonary Artery Ligation

Abstract

Homologous transplantation of vital organs, except under very specific conditions, still cannot be successfully carried out. Rejection of the donor tissue is presumed to be due to the development by the host of antibodies to the foreign antigen. Once this problem is solved, homologous transplantation of organs may become a clinical reality.The development of a suitable technique for the homografting of lungs is thought to be worthwhile for two reasons: First, when organ transplantation does become a clinical reality, a suitable technique will have been established; second, a transplanted, and therefore, completely denervated lung makes an excellent preparation for physiologic study. The transplant can be compared grossly, histologically, and functionally with the contralateral intact lung. The functions of the lung, including ventilation and respiration, can be studied differentially. The lung can be studied radiographically. Abstract

EFFICIENT and effortless swallowing at the upper end of the esophagus requires, among other events, coordinated and powerful contraction of the pharyngeal constrictors coupled with relaxation of the cricopharyngeus muscle, or upper esophageal sphincter, at the precise moment of arrival of the bolus from above. This highly coordinated sequence of events in normal persons has been demonstrated by a number of investigators using pressure recording,1,2 routine radiographic,3,4 and cineradiographic techniques.5-8The peristaltic wave begun by the pharyngeal constrictors is propagated into the esophagus proper but first must traverse the zone of increased pressure at the level of the cricopharyngeal sphincter. Code and his associates have shown in normal swallowing that there is a precisely timed and pronounced relaxation of the sphincter beginning immediately prior to pharyngeal contraction, "ensuring that the doorway to the esophagus is open as the swallowed material is forced toward it" (Fig 1).
