Stereospecifically labeled mevalonic acid was incorporated into the carotenoids of Rhodotorula. The randomized results are discussed in relation to mechanisms proposed for the conversion of isopentenyl pyrophosphate to dimethylallyl pyrophosphate and the prenol transferase enzyme system to Rhondotorula rubra.

The effects of diphenylamine, dimethyl sulfoxide, streptomycin, AMO-1618, and beta-ionone on the carotene composition of a wild-type and three mutant strains of Phycomyces blakesleeanus have been examined. Diphenylamine increased the phytoene and phytofluene concentrations of all strains while reducing the levels of the color carotenes. Dimethyl sulfoxide reduced the concentration of both cyclic and acyclic carotenes, whereas AMO-1618 increased the levels of all carotenes in all the strains. The wild type and mutants responded differently to the presence of streptomycin and beta-ionone. The possible mode of action of the above agents on carotenoid biosynthesis is discussed.

It was determined that lycopene could be cyclized directly by Rhodotorula glutinis. It was also shown the the temperature effect (i.e., increased beta-carotene synthesis in response to lower incubation temperatures) in R. glutinis was controlled by changes in enzyme concentration.

The biosynthesis of 4-keto and 4,4'-diketo carotenoids in Brevibacterium sp. KY-4313 was studied. Echinenone and canthaxanthin were isolated from the cultures grown on a medium containing several n-alkanes. When glutathione was added to the bacterial cultures, the formation of canthaxanthin was inhibited while beta-carotene and its hydroxy derivatives accumulated. It is suggested that these 4-hydroxy compounds, isocryptoxanthin, isozeaxanthin, and 4-hydroxy-4'-keto-beta-carotene, are intermediates in the biosynthesis of canthaxanthin. In the presence of 2-(4-chlorophenylthio)-triethylamine hydrochloride or nicotine, lycopene and neurosporene accumulated. The beta-carotene level decreased slightly but beta-zeacarotene remained unchanged. beta-carotene and its derivatives were resynthesized upon removal of the inhibitors. It was concluded that cyclization can take place at either the neurosporene or lycopene level in Brevibacterium sp. KY-4313.

Fruits of the lutescent tomato genetic line were exposed to gamma-radiation at different stages of maturity to determine the effect of ionizing radiation on carotenoid synthesis in the ripening fruit. Irradiation generally resulted in the inhibition of carotenogenesis. The effect was more pronounced at the higher dosage and in less mature fruit. Lycopene synthesis was inhibited more extensively than beta-carotene synthesis. The total carotenoid content was also generally lower in irradiated fruits. It was proposed that the beta-carotene in the tomato fruit is formed by a pathway not involving lycopene.

1. The incorporation of [2-(14)C]acetate, [2-(14)C]mevalonic acid and [2-(14)C,2-(3)H(2)]-mevalonic acid into torulene and torularhodin by Rhodotorula rubra and Rhodotorula glutinis was studied. 2. A recovery of 14.3% of the label was obtained on decarboxylation of the torularhodin biosynthesized from [2-(14)C]mevalonic acid. 3. An analysis of the (3)H/(14)C ratio in torularhodin gave a value of 9.44:8. 4. These results, obtained by different experimental techniques, show that the reactions in the conversion of the dimethyl group of isopentenyl pyrophosphate into the 16',17'-position of torularhodin must be free from randomization. A mechanism for the isomerization of isopentenyl pyrophosphate to dimethylallyl pyrophosphate is suggested.

Bacteriol. 88:1688-1694. 1964.-The biosynthesis of carotenoids was followed in Rhodotorula glutinis and in a new strain, 62-506. The treatment of the growing cultures by methylheptenone, or ionone, vapors permitted observations of the intermediates in the biosynthetic pathway. On the basis of concentration changes and accumulation in blocked pathways, the sequence of carotenoid formation is postulated as phytoene, phytofluene, zeta-carotene, neurosporene, beta-zeacarotene, gamma-carotene, torulin, a C(40) aldehyde, and torularhodin. Torulin and torularhodin were established as the main carotenoids of 62-506.

1. beta-Ionone vapour has been shown to cause an increase in the more saturated carotenes and a decrease in the less saturated carotenes of Rhodotorula glutinis. Lycopersene (dihydrophytoene) has been proposed as a precursor to phytoene. Attempts were made to isolate lycopersene from beta-ionone-treated cultures of R. glutinis. 2. Large samples of beta-ionone-treated cultures were examined for the presence of lycopersene. Spots were detected on silicic acid plates that could not be differentiated from synthetic lycopersene on the basis of column and thin-layer chromatographic separations and staining techniques. The lycopersene-like substance could be obtained from non-treated pigmented yeast as well as baker's yeast. 3. An extraction of bacterial-grade yeast extract also yielded a lycopersene-like substance. The extracts of R. glutinis cells cultured on media not containing yeast extract did not contain the lycopersene-like compound. 4. No significant carbon was incorporated into the lycopersene zone from (14)C-labelled mevalonate, acetate and glucose by R. glutinis and baker's yeast. 5. These results indicate that compounds may exist with chromatographic properties similar to lycopersene, but that lycopersene could not be detected in either a pigmented or a non-pigmented yeast.
