Abstract

The walls of Nocardia kirovani are composed of three main constituents: the peptidoglycan matrix, a polysaccharide polymer and a variety of free and bound lipids. The free lipids represent 17.5% (dry weight) of the walls and consist for the major part of C16-18 fatty acids and nocardic acids, and for the minor part, of nocardones, triglycerides and carotenoid pigments. The nocardic acids were identified as tri- and tetra-unsaturated, α-branched, β-hydroxylated compounds C58H110O3--C66H124O3, the nocardones as tri- and tetra-unsaturated ketones C57H106O--C63H118O, and the main carotenoid pigment as phlei-xantophylle palmitate. Esters of glycerol with C14C16C18 fatty acids and, for some of them, with odd numbered poly-unsaturated acids containing 35 to 45 carbon atoms, were also identified, Bound lipids represent about 20% (dry weight) of the walls and consist mainly of nocardic acids probably ester-linked to an arabinogalactan polymer. The peptidoglycan (about 40% dry weight) is composed of β-1,4-N-acetylglucosaminyl-N-glycolylmuramic acid disaccharide units that are substituted by diamidated L-Ala-D-αGln-(L)- A2pm-(D)-NH2 tripeptides and diamidated L-Ala-D-αGln-(L)-A2pm-(D)-NH2(L)-D_Ala tetrapeptides, where A2pm is meso-diaminopimelic acid.Crosslinking between some of the peptide units is mediated through D-Ala-(D)-A2pm linkages (peptidoglycan of ehemotype I). Abstract

Resume L'etude des lipides de cinq souches de Nocardia: N. caviae , N. erythropolis , N. farcinica , N. kirovani , N. sumatra a permis la caracterisation d'une gamme tres etendue d'acides nocardiques depuis C 34 H 68 O 3 jusqu'a C 66 H 124 O 3 . Ces acides qui possedent tous une chaine laterale saturee lineaire de C 10 H 21 a C 16 H 33 et une chaine principale acyclique comprennent des composes homologues satures mono-, di-, tri- et tetraethyleniques, le nombre de doubles liaisons augmentant avec le poids moleculaire. La presence d'acides nocardiques semble etre un critere general du genre Nocardia bien que le poids moleculaire soit assez variable suivant les especes.

Abstract

Mass spectral studies have shown that the carotenoid of Plectania coccinea is dehydro-2′-plectaniaxanthin linoleate (I). On saponification, (I) gave the free xanthin and linoleic acid, identified by TLC, gas chromatography and mass spectroscopy of the diepoxy derivative.

Abstract

Resume Structure II is established for a new peptidolipid isolated from Nocardia asteroides; this compound differs from the previously described peptidolipin NA (I) by the replacement of one l -alanine in position 6 by one l -valine. Due to previous experience with mass spectrometry of analogous compounds, the unequivocal determination of the sequence of the amino acids of this heptapeptide derivative could be very rapidly accomplished, without chemical degradation.

Resume La fraction acide des lipides d'Aspergillus flavus renferme essentiellement les acides palmitique, ol'eique, linol'eique et α hydroxyt'etracosanoique. Nous avons isol'e de la fraction insaponifiable plusieurs compos'es de nature st'erolique:l'ergost'erol, le peroxyde d'ergost'erol et un autre st'erol C28H46O3 qui est vraisemblablement le c'er'evist'erol. Ces deux derniers compos'es n'apparaissent pas lorsque les op'erations sont effectu'ees en l'absence de lumiere. Ils repr'esentent donc des art'efacts et non des m'etabolites d'A. flavus.

Abstract

By addition of 1- 14C-sodium acedate to the growth medium of Nocardia asteroides, it can be shown that the lipid content increases during the exponential phase, but does not vary during the stationary phase of the growth. Nocardic acid biosynthesis from the medium molecular weight fatty acids occurs chiefly during te stationary phase. As these compounds are localised in the cell walls, it becomes evident that the lipid envelope of the walls is still increasing when the cell growth and division have stopped.
