Simple Summary: Individuals with metabolic syndrome exhibit simultaneously pro-thrombotic and a pro-inflammatory conditions which more probably can lead to cardiovascular disease progression, type 2 diabetes mellitus, and some types of cancer. The present scoping review is aimed at highlighting any relations between cancer risk, inflammation, and metabolic syndrome. A total of 20 manuscripts were identified and, among them, we identified some associations with breast cancer, colorectal cancer, esophageal adenocarcinoma, hepatocellular carcinoma (HCC), and cancer in general. Therefore, it could be deducted that cancer and its related progression may also depend on a latent chronic inflammatory condition associated with other concomitant conditions, including type 2 diabetes mellitus, metabolic syndrome, and obesity. Therefore, prevention may potentially help individuals to protect themselves from cancer. This paper reviews the published terminology, mathematical models, and the possible approaches used to characterise the risk of foodborne chemical hazards, particularly pesticides, metals, mycotoxins, acrylamide, and polycyclic aromatic hydrocarbons (PAHs). The results confirmed the wide variability of the nomenclature used, e.g., 28 different ways of referencing exposure, 13 of cancer risk, or 9 of slope factor. On the other hand, a total of 16 equations were identified to formulate all the risk characterisation parameters of interest. Therefore, the present study proposes a terminology and formulation for some risk characterisation parameters based on the guidelines of international organisations and the literature review. The mathematical model used for non-genotoxic hazards is a ratio in all cases. However, the authors used the probability of cancer or different ratios, such as the margin of exposure (MOE) for genotoxic hazards. For each effect studied per hazard, the non-genotoxic effect was mostly studied in pesticides (79.73%), the genotoxic effect was mostly studied in PAHs (71.15%), and both effects were mainly studied in metals (59.4%). The authors of the works reviewed generally opted for a deterministic approach, although most of those who assessed the risk for mycotoxins or the ratio and risk for acrylamide used the probabilistic approach. Breast cancer (BC) risks imparted by CHEK2 c.1100delC ("1100delC") germline pathogenic/likely pathogenic variant (GPV) are 20–30%, compared to CHEK2 c.470T>C ("I157T") GPV with <20%, leading to different breast screening recommendations through MRI. We compared cancer risk management (CRM) across these two GPVs. Study participants were adult females with an 1100delC or I157T GPV drawn from the Inherited Cancer Registry (ICARE) across the United States. Cancer history, clinical characteristics, and CRM were compared using chi-squared tests, t-tests, and logistic regression. Of 150 CHEK2 carriers, 40.7% had BC, with a mean age of 50. Comparing 1100delC and I157T GPVs, there were no differences in rates of (1) breast MRI among those with (65.2% versus 55.6% of 23 and 9; p = 0.612) and without (44.0% versus 44.8% of 50 and 29; p = 0.943) BC; (2) risk-reducing mastectomy among those with (50% versus 38.9% of 46 and 15; p = 0.501) and without (13.8% versus 6.5% of 58 and 31; p = 0.296) BC; and (3) risk-reducing salpingo-oophorectomy among those with (24.2% versus 22.2% of 45 and 18; p = 0.852) and without (17.5% versus 16.7% of 57 and 30; p = 0.918) BC. The results suggest over-screening with breast MRI among CHEK2 I157T GPV carriers and possible overuse of risk-reducing surgeries among CHEK2 carriers. [ABSTRACT FROM AUTHOR]

Pathogenic mutations in BRCA1 (BReast CAncer gene 1) confer high risks of both breast (up to 70%) and ovarian (up to 40%) cancers. Zinc (Zn) and copper (Cu) are essential for various physiological functions, including antioxidant reactions. Their balance, reflected in the Zn/Cu ratio, plays a crucial role in maintaining redox homeostasis, which is vital for cancer prevention. This study examines the antioxidant properties of Zn and Cu, specifically focusing on the blood Zn/Cu ratio as a potential marker for cancer risk among BRCA1 mutation carriers. The study cohort consisted of 989 initially unaffected women, followed up for 7.5 years. Blood samples were analyzed using inductively coupled plasma mass spectrometry. Although individual Zn and Cu levels did not significantly correlate with overall cancer risk, those women with a Zn/Cu ratio above 6.38 experienced a significantly lower cancer risk than women with a ratio below this cut-off point. This suggests that the Zn/Cu ratio may be a valuable biomarker for cancer prevention in this high-risk group. Given the increased cancer risk in BRCA1 mutation carriers, optimizing Zn and Cu levels through dietary and active interventions could provide a preventive strategy. [ABSTRACT FROM AUTHOR]

BRCA1 mutations predispose women to breast and ovarian cancer. The anticancer effect of zinc is typically linked to its antioxidant abilities and protecting cells against oxidative stress. Zinc regulates key processes in cancer development, including DNA repair, gene expression, and apoptosis. We took a blood sample from 989 female BRCA1 mutation carriers who were initially unaffected by cancer and followed them for a mean of 7.5 years thereafter. There were 172 incident cases of cancer, including 121 cases of breast cancer, 29 cases of ovarian cancers, and 22 cancers at other sites. A zinc level in the lowest tertile was associated with a modestly higher risk of ovarian cancer compared to women with zinc levels in the upper two tertiles (HR = 1.65; 95% CI 0.80 to 3.44; p = 0.18), but this was not significant. Among those women with zinc levels in the lowest tertile, the 10-year cumulative risk of ovarian cancer was 6.1%. Among those in the top two tertiles of zinc level, the ten-year cumulative risk of ovarian cancer was 4.7%. There was no significant association between zinc level and breast cancer risk. The region surrounding the Elbrus volcano in the Great Caucasus is a popular tourist destination, home to native people and an area of the richest ore mineral resources. Based on the chemical composition of surface water, mineral, and groundwater in the Elbrus area, as well as statistical and thermodynamic calculations, waters with high concentrations of Mn, Cu, Zn, Mo, Ba, W, Pb, and As were identified, possible mineral phases that may lead to these metal concentrations in water were estimated, and risks to human health were assessed. Two main sources of surface water pollution have been identified: (1) water from tailings that are enriched due to oxidation of sulfide minerals is considered a source of As (181 µg/L) and Mo (2070 µg/L) pollution in the area of the Tyrnyauz W-Mo deposit; and (2) CO2-rich waters of deep horizons discharge on the earth surface with high concentrations of As (828 µg/L) and W (504 µg/L). It has been shown that compared to all toxic elements studied, only Mo and As have adverse non-carcinogenic effects on adult health. The estimated carcinogenic risk of As in the mine area was 1.7 × 10−3 and exceeded the generally accepted range of 10−4 to 10−6. In order to minimize the health risks, regular monitoring of water quality is required when mining activities on the Tyrnyauz W-Mo deposit are renewed. [ABSTRACT FROM AUTHOR]

Urban vegetable farming in wetlands and riverbanks are common features of Ugandan cities. However, urbanization has led to various anthropogenic activities that can lead to the pollution of water resources, enrichment of pollutants in soils and, consequently, pollutant bioaccumulation in edible tissues of plants cultivated on such soils. In this study, we report on the levels of six trace metals (TMTs) in 75 samples of leafy vegetables (Brassica oleracea L., Spinacia oleracea L., Amaranthus hybridus L., Cucurbita pepo L. and Solanum nigrum L.) and soils (n = 75) grown on the banks of River Rwizi, the second longest river in Uganda only after the Nile River. The concentrations of TMTs (Mn, Zn, Cd, Pb, Cr and Cu) in edible vegetable tissues and soils were quantified using flame atomic absorption spectrometry. The mean concentrations (in mg kg−1) of the TMTs in the soil samples were 205–373.84 (Mn), 12.72–65.04 (Zn), 0.26–0.42 (Cd), 3.36–16.80 (Pb), 5.96–25.06 (Cr) and 2.83–35.27 (Cu). In vegetable samples, the concentrations ranged from 43.25 to 110.00 (Mn), 1.08 to 1.83 (Cd), 41.06 to 71.20 (Zn), 4.31 to 6.16 (Pb), 0.65 to 0.81 (Cr) and 5.70 to 14.35 (Cu). With the exception of Mn and Cr, the rest of the TMTs were bioaccumulated in the edible vegetable tissues (bioconcentration factors = 1.03 to 10.71). Considering chronic daily intake through ingestion, dermal contact and inhalation of the TMTs in soils from the banks of River Rwizi, there are no potential non-cancer and carcinogenic health effects that could be experienced in both adults and children. Consumption of leafy vegetables could pose both non-cancer health risks (from ingestion of Zn, Pb, Cr, Mn and Cd) and cancer health risks (due to intake of Cd) in both children and adults.
