This study investigated the effect of astragalus polysaccharide (APS, an ingredient with hypoglycemic function in a traditional Chinese herbal medicine) on gut microbiota and metabolites of type 2 diabetes mellitus (T2DM) patients using a simulated fermentation model in vitro. The main components of APS were isolated, purified, and structure characterized. APS fermentation was found to increase the abundance of Lactobacillus and Bifidobacterium and decrease the Escherichia-Shigella level in the fecal microbiota of T2DM patients. Apart from increasing propionic acid, APS also caused an increase in all-trans-retinoic acid and thiamine (both have antioxidant properties), with their enrichment in the KEGG pathway associated with thiamine metabolism, etc. Notably, APS could also enhance fecal antioxidant properties. Correlation analysis confirmed a significant positive correlation of Lactobacillus with thiamine and DPPH-clearance rate, suggesting the antioxidant activity of APS was related to its ability to enrich some specific bacteria and upregulate their metabolites. 8-17 DNAzymes (8-17, 17E, Mg5, and 17EV1) are in vitro-selected catalytic DNA molecules that are capable of cleaving complementary RNAs. The conserved residues in their similar catalytic cores, together with the metal ions, were suggested to contribute to the catalytic reaction. Based on the contribution of the less conserved residues in the bulge loop residues (W12, A15, A15.0) and the internal stem, new catalytic cores of 8-17 DNAzymes were programmed. The internal stem CTC-GAG seems to be more favorable for the DNAzymes than CCG-GGC, while an extra W12.0 led to a significant loss of activity of DNAzymes, which is contrary to the positive effect of A15.0, by which a new active DNAzyme 17EM was derived. It conducts a faster reaction than 17E. It is most active in the presence of Pb2+, with the metal ion preference of Pb2+ >> Zn2+ > Mn2+ > Ca2+ ≈ Mg2+. In the Pb2+ and Zn2+-mediated reactions of 17EM and 17E, the same Na+- and pH dependence were also observed as what was observed for 17E and other 8-17 DNAzymes. This study firstly applied a Bayesian symbolic regression (BSR) to the forecasting of numerous commodities' prices (spot-based ones). Moreover, some features and an initial specification of the parameters of the BSR were analysed. The conventional approach to symbolic regression, based on genetic programming, was also used as a benchmark tool. Secondly, various other econometric methods dealing with variable uncertainty were estimated including Bayesian Model Averaging, Dynamic Model Averaging, LASSO, ridge, elastic net, and least-angle regressions, etc. Therefore, this study reports a concise and uniform comparison of an application of several popular econometric models to forecasting the prices of numerous commodities. Cotton Verticillium wilt is mainly caused by the fungus Verticillium dahliae, which threatens the production of cotton. Its pathogen can survive in the soil for several years in the form of microsclerotia, making it a destructive soil-borne disease. The accurate, sensitive, and rapid detection of V. dahliae from complex soil samples is of great significance for the early warning and management of cotton Verticillium wilt. In this study, we combined the loop-mediated isothermal amplification (LAMP) with CRISPR/Cas12a technology to develop an accurate, sensitive, and rapid detection method for V. dahliae. Initially, LAMP primers and CRISPR RNA (crRNA) were designed based on a specific DNA sequence of V. dahliae, which was validated using several closely related Verticillium spp. The lower detection limit of the LAMP-CRISPR/Cas12a combined with the fluorescent visualization detection system is approximately ~10 fg/μL genomic DNA per reaction. When combined with crude DNA-extraction methods, it is possible to detect as few as two microsclerotia per gram of soil, with the total detection process taking less than 90 min. Furthermore, to improve the method's user and field friendliness, the field detection results were visualized using lateral flow strips (LFS). The LAMP-CRISPR/Cas12a-LFS system has a lower detection limit of ~1 fg/μL genomic DNA of the V. dahliae, and when combined with the field crude DNA-extraction method, it can detect as few as six microsclerotia per gram of soil, with the total detection process taking less than 2 h. In summary, this study expands the application of LAMP-CRISPR/Cas12a nucleic acid detection in V. dahliae and will contribute to the development of field-deployable diagnostic productions. In response to the need for a space security situation assessment during orbit, the multi-satellite space environmental risk prediction and early warning system is based on the detection results of the space weather payload of the Fengyun 4A and 4B satellites, as well as the prediction results of the National Space Weather Center, for the first time. By comprehensively utilizing some open-source data, it is the first time that we have achieved a 24 h advanced prediction of the space environment high-energy proton, low-energy particle, and high-energy electron risks for the safety of the Fengyun-series high-orbit satellites, and a real-time warning of satellite single-event upset, surface charging, and deep charging risks. The automation system has preliminarily achieved an intelligent space risk assessment for the safety of multiple stationary meteorological satellites, effectively improving the application efficiency of the space environmental data and the products of Fengyun-series satellites. The business status is stable in operation, and the resulting error between the predicted results of various risk indices and the measured data was less than one level. The warning accuracy was better than 90%. This article uses the system for the first time, to use Fengyun satellite data to, accurately and in a timely manner, predict and warn us about the low-energy particles and surface charging high-risk levels of the Fengyun 4A and 4B satellites during the typical space weather event on 21 April 2023, in response to the impact of complex spatial environmental factors on the safety of Fengyun-series high-orbit satellites. The construction and operation of a multi-satellite space environmental risk prediction and early warning system can provide a reference for the safety work of subsequent satellite ground system operations. Efficiency in stock markets is essential for economic stability and growth. This study investigates the efficiency and herding behavior of the stock markets from the top economies of the world (known as G20 countries). We classify stock market indices using MSCI classification for the developed and emerging markets to provide a comparative examination using the latest data and by employing the robust multifractal detrended fluctuation (MFDFA) method. In addition to the full sample, the analysis uses sub-sample periods to reveal the hidden features and efficiencies of the G20 markets during the Russia–Ukraine War and COVID-19 for the first time. The findings show the availability of varied multifractality among all G20 stock markets during the overall and crisis periods, exhibit long-range correlations, and may support the fractal market hypothesis. In addition, Italy remains the least efficient, while Germany remains the most efficient stock market. The sub-sample results further reveal unevenness in the local fluctuations and resultant higher inefficiency considering the sheer magnitude and impact of crises on the G20 stock markets. However, the efficiency of developed stock markets performed better as compared to emerging markets. The study of G20 stock markets is useful and provides several implications for a wider audience. Objective: To investigate the expression patterns and clinical significance of specific lymphocyte subsets in the peripheral blood of patients with severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) infection. Methods: Between December 2022 and February 2023, a cohort of 165 patients from the First Affiliated Hospital of Guangzhou University of Chinese Medicine were analyzed. The participants represented various stages of coronavirus infection severity: mild, moderate, severe, and critical. Additionally, 40 healthy individuals constituted the control group. The FC 500MPL flow cytometer and associated reagents for flow cytometry. Results: Compared with the healthy control group, activated B lymphocytes witnessed a pronounced increase (p <.05). A significant decrease was observed in the levels of Breg, Cytotoxic T cells or Suppressor T‐cell (Tc/s), late‐activated T, late‐activated Th, and late‐activated Tc/s lymphocytes (p <.05). Th, initial Th, initial Tc/s, total Treg, natural Treg, induced Treg, early activated T, and early activated Th lymphocyte levels showed no significant difference (p >.05). As the disease progressed, there was an uptick in midterm activated T lymphocytes (p <.05), while Breg, T, Tc/s, senescent Tc/s, and total senescent T levels dwindled (p <.05). Noteworthy patterns emerged across different groups for B1, T‐lymphocytes, Tc/s, B2, CD8+ Treg cells, and other subsets, highlighting variance in immune responses relative to disease severity. When juxtaposed, no significant difference was found in the expression levels of lymphocyte subsets between patients who died and those deemed critically ill (p >.05). Conclusion: Subsets of Treg and B‐cells could act as yardsticks for the trajectory of SARS‐CoV‐2 infection and might have potential in forecasting patient trajectories. A comprehensive evaluation of lymphocyte subsets, especially in real‐time, holds the key to discerning the clinical severity in those with COVID‐19. This becomes instrumental in monitoring treatment outcomes, tracking disease evolution, and formulating prognostications. Future index prices are viewed as a critical issue for any trader and investor. In the literature, various models have been developed for forecasting index prices. For example, the geometric Brownian motion (GBM) model is one of the most popular tools. This work examined four types of GBM models in terms of the presence of memory and the kind of volatility estimations. These models include the classical GBM model with memoryless and constant volatility assumptions, the SVGBM model with memoryless and stochastic volatility assumptions, the GFBM model with memory and constant volatility assumptions, and the SVGFBM model with memory and stochastic volatility assumptions. In this study, these models were utilized in an empirical study to forecast the future index price of the energy sector in the Saudi Stock Exchange Market. The assessment was led by utilizing two error standards, the mean square error (MSE) and mean absolute percentage error (MAPE). The results show that the SVGFBM model demonstrates the highest accuracy, resulting in the lowest MSE and MAPE, while the GBM model was the least accurate of all the models under study. These results affirm the benefits of combining memory and stochastic volatility assumptions into the GBM model, which is also supported by the findings of numerous earlier studies. Furthermore, the findings of this study show that GFBM models are more accurate than GBM models, regardless of the type of volatility. Furthermore, under the same type of memory, the models with a stochastic volatility assumption are more accurate than the corresponding models with a constant volatility assumption. In general, all models considered in this work showed a high accuracy, with MAPE ≤ 10%. This indicates that these models can be applied in real financial environments. Based on the results of this empirical study, the future of the energy sector in Saudi Arabia is forecast to be predictable and stable, and we urge financial investors and stockholders to trade and invest in this sector. Ferroptosis is a necrotic form of iron‐dependent regulatory cell death. Estrogen withdrawal can interfere with iron metabolism, which is responsible for the pathogenesis of postmenopausal osteoporosis (PMOP). Here, it is demonstrated that estrogen withdrawal induces iron accumulation in the skeleton and the ferroptosis of osteocytes, leading to reduced bone mineral density. Furthermore, the facilitatory effect of ferroptosis of osteocytes is verified in the occurrence and development of postmenopausal osteoporosis is associated with over activated osteoclastogenesis using a direct osteocyte/osteoclast coculture system and glutathione peroxidase 4 (GPX4) knockout ovariectomized mice. In addition, the nuclear factor erythroid derived 2‐related factor‐2 (Nrf2) signaling pathway is confirmed to be a crucial factor in the ferroptosis of osteocytic cells. Nrf2 regulates the expression of nuclear factor kappa‐B ligand (RANKL) by regulating the DNA methylation level of the RANKL promoter mediated by DNA methyltransferase 3a (Dnmt3a), which is as an important mechanism in osteocytic ferroptosis‐mediated osteoclastogenesis. In the field of electric aircraft, the axial force of the conical motor is used to offset propeller thrust and reduce bearing wear. For the problem of accurate calculation of the no‐load axial force of the conical motor, combined with the characteristics that the rotor magnetic bridge of the hybrid conical inner‐mounted permanent magnet synchronous motor (HC‐IPMSM) is easy to be saturated and difficult to be calculated analytically, an equivalent analytical calculation method is proposed. According to the principle of constant air‐gap magnetic flux, the HC‐IPMSM is equivalent to many cylindrical surface‐mounted permanent magnet synchronous motors (CY‐SPMSM), and an analytical model is gained. Combining with the scalar magnetic potential Laplace equation and Poisson equation, the air‐gap flux density distribution on the rotor surface is calculated. And finally, the no‐load axial force of the motor is obtained by using the cone angle and the Maxwell stress method. We examine how ageing without children is linked to older adults' social participation and psychological distress in selected East and Southeast Asian countries (South Korea, China, Thailand, Vietnam, and Myanmar). Recognizing the nuances of such ageing experiences, we distinguish between actual childlessness (older persons without children) and defacto childlessness (older persons whose children all lived far away). We harmonize and analyze recent nationally representative survey data from the five abovementioned countries, utilizing both descriptive and multivariate approaches. First, we investigate the prevalence and dimensions of actual and defacto childlessness. Second, we examine the extent to which actual and defacto childlessness are associated with social participation and psychological distress in old age. The analyses focus on gender and wealth differences. We find that actual childlessness is associated with lower levels of social participation in Southeast Asian countries and greater levels of psychological distress in Korea, Thailand, and Myanmar. The effects of defacto childlessness are, however, mixed. We discuss the social and psychological implications of childlessness for older adults across varying societal contexts in the five Asian countries. [ABSTRACT FROM AUTHOR]

The objective of this study was to systematically analyze and identify genetic characteristics associated with immune infiltration in UC.Patients and Methods: Gene expression data from three independent datasets obtained from the Gene Expression Omnibus (GEO) were utilized. By employing the ssGSEA and CIBERSORT algorithms, we estimated the extent of immune cell infiltration in UC samples. Subsequently, Weighted Correlation Network Analysis (WGCNA) was performed to identify gene modules exhibiting significant associations with immune infiltration, and further identification of hub genes associated with immune infiltration was accomplished using least absolute shrinkage and selection operator (LASSO) regression analysis. The relationship between the identified hub genes and clinical information was subsequently investigated.Results: Our findings revealed significant activation of both innate and adaptive immune cells in UC. Notably, the expression levels of CD44, IL1B, LYN, and ITGA5 displayed strong correlations with immune cell infiltration within the mucosa of UC patients. Immunohistochemical analysis confirmed the significant upregulation of CD44, LYN, and ITGA5 in UC samples, and their expression levels were found to be significantly associated with common inflammatory markers, including the systemic immune inflammation indices, C-reactive protein, and erythrocyte sedimentation rate.Conclusion: CD44, LYN, and ITGA5 are involved in the immune infiltration pathogenesis of UC and may be potential therapeutic targets for UC.Keywords: Ulcerative colitis, Immune infiltration, WGCNA, CIBERSORT, LASSO
