Prochloraz is a widely used fungicide worldwide. It is classified as an endocrine disrupting pesticide that affects the reproductive system. This study aimed to examine the impact of exposure to prochloraz of male mice on the reproductive system of their offspring male mice. Male father mice were intragastrically administered different dosages of prochloraz (group MA: 0 mg/kg/day; MB: 53.33 mg/kg/day; MD:160 mg/kg/day). Then, the testicular average weight of male offspring in the dose groups was found to be significantly lower than those in the control group (MB:0.312g, MD:0.294g, and MA:0.355 g; P < 0.05). Additionally, the testicular coefficient index in the MB and MD groups was also lower than that of the control group. Secondly,we observed that there were significantly different expressed genes clustered in groups B and D, in contrast to the control. Finally, the findings demonstrated a significant alteration in the response of male mice reproductive relative genes to prochloraz invasion. Two genes (Mt-nd6 and Slc12a4) were found to be involved in the regulation of sperm mitochondria function and six genes (Greb1, Esrrb, Catsperb, Mospd2, Sohlh1 and Specc1) were closely linked to sperm functions and estrogen response. The study revealed a significant impact of prochloraz on the reproductive system of male mice, thereby supporting further investigation into the reproductive toxicological effects of the drug. The Clearfield® technology is an useful tool for controlling weedy rice due to the effectiveness of imazamox and the cultivation of rice varieties resistant to imidazolines. However, residual imazamox in the soil probably causes phytotoxicity to subsequent non-resistant rice crops. We previously found that pyrroloquinoline quinone (PQQ), a bioactive elicitor, can enhance rice tolerance to imazamox. In this study, we explored the molecular mechanism of PQQ-mediated rice tolerance to imazamox by RNA-seq analysis, real-time quantitative PCR (RT-qPCR) assay, and enzyme activity assay. The results indicated that compared to imazamox at 66.7 mg a.i./L (IMA) alone, rice plants treated with imazamox at 66.7 mg a.i./L and PQQ at 0.66 mg a.i./L (IMA + PQQ) exhibited significantly reduced sensitivity to imazamox. Seven days post-treatment, IMA + PQQ-treated rice plants showed partial chlorosis and leaf curling, but IMA-treated rice plants had severe wilting and died. The fresh weight inhibition rate was 29.3% in the IMA + PQQ group, significantly lower than that of 56.6% in the IMA group alone. RNA-seq analysis showed differentially expressed genes were mainly involved in phenylpropanoid biosynthesis, diterpenoid biosynthesis, and MAPK signaling pathways in response to IMA + PQQ treatment. Both RNA-seq analysis and RT-qPCR assay showed that the expression of OsCATB gene in the catalase (CAT) gene family was upregulated at 12 h, the expression of OsGSTU1 gene was upregulated at 12, 24, and 48 h, while the expressions of OsABCB2, OsABCB11, and OsABCG11 genes were upregulated at 24 and 48 h. Enzyme activity assays revealed that the activity of superoxide dismutase in the IMA + PQQ group was increased by 47.45~120.31% during 12~72 h, compared to that in the IMA group. CAT activity in the IMA + PQQ group was increased by 123.72 and 59.04% at 12 and 48 h, respectively. Moreover, malondialdehyde levels indicative of oxidative damage were consistently lower in IMA + PQQ-treated plants, with a reduction of 46.29, 11.37, and 14.50% at 12, 36, and 72 h, respectively. Overall, these findings discover that PQQ has potential in reducing imazamox phytotoxicity in rice by enhancing antioxidant enzyme activities and regulating oxidative stress responses. The dried rhizomes of Coptis chinensis hold significance in Chinese medicine. Monocropping C. chinensis under the shade of a manmade scaffold, the primary planting mode, poses a threat to the ecological balance. In recent years, the practice of C. chinensis–Cunninghamia lanceolata understory cultivation has gained prevalence in southwest China. However, there is no evidence to suggest that understory cultivation enhances the viability, yield, or isoquinoline alkaloid content of C. chinensis. This study examined the physiological properties, yield indicators, and isoquinoline alkaloid content to investigate variations in C. chinensis in response to understory cultivation. Transcriptome analysis was conducted to elucidate potential mechanisms driving these alterations. The results indicate that understory cultivation significantly enhances the viability, yield, and levels of epiberberine, palmatine, and berberine in C. chinensis while reducing coptisine content. Transcriptomic analyses identified 2062 upregulated and 1853 downregulated genes in the understory cultivation system. Pathways such as "phenylpropanoid biosynthesis," "zeatin biosynthesis," "photosynthesis," "tyrosine metabolism," "isoquinoline alkaloid biosynthesis," and "starch and sucrose metabolism" exhibited significant enrichment of differentially expressed genes (DEGs). DEGs involved in these pathways were thoroughly analyzed. INV, BGL-2, BGL-4, SPS-2, AMY-3, Psb B, Psb R, Psb S, Psa D, Psa E, Psa H, Psa O, Pet C, Pet H, deta, and b exhibited significant positive correlations with plant fresh weight, aboveground fresh weight, and underground fresh weight. 6-OMT-2 and COMT1-3 displayed significant positive correlations with coptisine content, but negative correlations with epiberberine, palmatine, and berberine content. ZOG-1, ZOG-3, TAT, PPO, POD-13 and UGT 73C5-1 showed noteworthy positive correlations with berberine content. Conversely, MIFH, POD-4, POD-5, and POD-8 displayed significant positive correlations with epiberberine, palmatine, and berberine content. POD-5, and POD-7 were significantly negatively correlated with coptisine content. Furthermore, gene expression levels determined by qRT–PCR aligned with the transcriptomic sequencing results, confirming the reliability of the transcriptomic findings. Osteoarthritis (OA) is a severe chronic inflammatory disease. As the main active component of Astragalus mongholicus Bunge, a classic traditional ethnic herb, calycosin exhibits anti-inflammatory action and its mechanism of exact targets for OA have yet to be determined. In this study, we established an anterior cruciate ligament transection (ACLT) mouse model. Mice were randomized to sham, OA, and calycosin groups. Cartilage synthesis markers type II collagen (Col-2) and SRY-Box Transcription Factor 9 (Sox-9) increased significantly after calycosin gavage. While cartilage matrix degradation index cyclooxygenase-2 (COX-2), phosphor-epidermal growth factor receptor (p-EGFR), and matrix metalloproteinase-9 (MMP9) expression were decreased. With the help of network pharmacology and molecular docking, these results were confirmed in chondrocyte ADTC5 cells. Our results indicated that the calycosin treatment significantly improved cartilage damage, this was probably attributed to reversing the imbalance between chondrocyte synthesis and catabolism. Urinary extracellular vesicles (uEVs) are rich in valuable biomolecule information which are increasingly recognized as potential biomarkers for various diseases. uEV long RNAs are among the critical cargos capable of providing unique transcriptome information of the source cells. However, consensus regarding ideal reference genes for relative long RNAs quantification in uEVs is not available as of date. Here we explored stable reference genes through profiling the long RNA expression by RNA‐seq following unsupervised analysis and validation studies. Candidate reference genes were identified using four algorithms: NormFinder, GeNorm, BestKeeper and the Delta Ct method, followed by validation. RNA profile showed uEVs contained abundant long RNAs information and the core transcriptome was related to cellular structures, especially ribosome which functions mainly as translation, protein and RNA binding molecules. Analysis of RNA‐seq data identified RPL18A, RPL11, RPL27, RACK1, RPSA, RPL41, H1‐2, RPL4, GAPDH, RPS27A as candidate reference genes. RT‐qPCR validation revealed that RPL41, RPSA and RPL18A were reliable reference genes for long RNA quantification in uEVs from patients with diabetes mellitus (DM), diabetic nephropathy (DN), IgA nephropathy (IgAN) and prostate cancer (PCA). Interestingly, RPL41 also outperformed traditional reference genes in renal tissues of DN and IgAN, as well as in plasma EVs of several types of cancers.
