- Abstract
Objective To investigate the relationships of expression levels of microRNA-143-3p (miR-143-3p) and timeless circadian regulator (TIMELESS) mRNA in epithelial ovarian cancer (EOC) tissues with the clinicopathological characteristics and prognosis of patients. Methods We selected 106 patients with EOC, and the EOC tissues and corresponding adjacent tissues were collected. Real-time fluorescence quantitative PCR was performed to detect miR-143-3p and TIMELESS mRNA expression. The binding sites of miR-143-3p and TIMELESS were predicted by online databases such as TargetScan, StarBase, and miRWalk. Pearson correlation was used to analyze the correlation between miR-143-3p and TIMELESS mRNA. We analyzed the relationships between the expression of miR-143-3p and TIMELESS mRNA and the clinicopathological characteristics of EOC patients. EOC patients were followed up for three years, and we calculated the cumulative survival rate. EOC patients were divided into high/low miR-143-3p and TIMELESS mRNA expression groups according to the mean value of their expression, and Kaplan-Meier method was used to draw survival curves for survival analysis. Multivariate Cox regression analysis was performed to analyze the effects of miR-143-3p and TIMELESS on the prognosis of EOC patients. Results Compared with the adjacent tissues, miR-143-3p was low expressed and TIMELESS mRNA was highly expressed in EOC tissues (both P<0. 05) The miR-143-3p and TIMELESS had a binding site at 3′-untranslated region 1453-1460. The miR-143-3p and TIMELESS mRNA expression in EOC tissues showed a negatively correlated (r=–0. 748,P<0. 05). The miR-143-3p and TIMELESS mRNA expression was related to International Federation of Gynecology and Obstetrics (FIGO) stage, degree of differentiation, lymph node metastasis, and distant metastasis (all P<0. 05). The 3-year overall survival rate of 106 EOC patients was 69. 81% (74/106). The high miR-143-3p expression patients had a higher 3-year overall survival rate than the low miR-143-3p expression patients, and the 3-year overall survival rate of patients with high TIMELESS mRNA expression was lower than that of patient with low TIMELESS mRNA expression patients (both P<0. 05). Independent risk factors for death in EOC patients were FIGO stage Ⅲ-Ⅳ, poor differentiation, lymph node metastasis, distant metastasis, and TIMELESS mRNA ≥ 1. 11, and the independent protective factor was miR-143-3p ≥ 0. 97(all P<0. 05). Conclusion In EOC tissues, miR-143-3p was low expressed and TIMELESS mRNA was highly expressed, and their expression changes were closely associated with FIGO stage, degree of differentiation, lymph node metastasis, distant metastasis and prognosis.
