Objectives Materials and Methods Results Conclusions The purpose of this study was to determine whether indoxyl sulfate (IS) is involved in alveolar bone deterioration and to elucidate the mechanism underlying alveolar bone loss in chronic kidney disease (CKD) patients.Mice were divided into the control group, CP group (ligature‐induced periodontitis), CKD group (5/6 nephrectomy), and CKD + CP group. The concentration of IS in the gingival crevicular fluid (GCF) was determined by HPLC. The bone microarchitecture was evaluated by micro‐CT. MC3T3‐E1 cells were stimulated with IS, and changes in mitochondrial morphology and ferroptosis‐related factors were detected. RT‐PCR, western blotting, alkaline phosphatase activity assays, and alizarin red S staining were utilized to assess how IS affects osteogenic differentiation.Compared with that in the other groups, alveolar bone destruction in the CKD + CP group was more severe. IS accumulated in the GCF of mice with CKD. IS activated the aryl hydrocarbon receptor (AhR) in vitro, inhibited MC3T3‐E1 cell osteogenic differentiation, caused changes in mitochondrial morphology, and activated the SLC7A11/GPX4 signaling pathway. An AhR inhibitor attenuated the aforementioned changes induced by IS.IS activated the AhR/SLC7A11/GPX4 signaling pathway, inhibited osteogenesis in MC3T3‐E1 cells, and participated in alveolar bone resorption in CKD model mice through ferroptosis. Although the physiochemical merits of g-C3N4-based photocatalysts have garnered increasing interest in the fields of energy and environmental science, insufficient layer detachment has created a gap between fundamental research and practical applications. To unlock the intrinsic potential of g-C3N4, a bio-transformation of the ZnMn2O4 ((6)ZM) gel was employed to introduce highly-ordered modulation caused by steric hindrance during melamine pyrolysis. Phytomediated (6)ZM reorganized traditional carbon nitride into crossed C3N4 (CCN) monolayers, simultaneously engineering an auspicious Z-schematic system ((6)ZM/CCN). Phytoconverted (6)ZM retained the crystalline-amorphous configuration for facile charge transfer and provided a large surface area (288 m2 g−1) that was 2.3 times greater than that of thermally prepared g-C3N4 (TCN) monolayers. Additionally, (6)ZM exhibited a quantum confinement-promoted reduction capability and induced bulging on CCN monolayers to fully utilize photons through multilevel light scattering and reflection. Specific sequential two-step calcination of (6)ZM/CCN, furnishing affordable dual Z-schematic VO–(6)ZM/CCN, was specifically developed to introduce a third component into the structure without incurring additional operational cost or complexity. V2O5 (VO) nanoparticles were thermally anchored on (6)ZM/CCN to achieve highly efficient levofloxacin (LFC) detoxification under visible-light irradiation. After optimizing all effective synthesis parameters and experimental variables, VO–(6)ZM/CCN exhibited unsurpassed activity, achieving complete LFC photodegradation (50 mg L−1) within 120 min, which was 10.7, 8.7, and 24.7 times more kinetically efficient than the photodegradations achieved by (6)ZM, TCN, and VO, respectively. The outstanding performance of VO–(6)ZM/CCN was evident through complete mineralization of LFC, excellent decontamination of pharmaceutical wastewater within 300 min, resistance to performance deterioration during successive cycling runs, and the corresponding postcharacterization. The combination of simultaneous Z-scheme formation with photogenic (6)ZM provides a promising strategy to bridge the gap between experimental investigations and industrial applications of g-C3N4. Objectives Subjects and methods Results Conclusions To investigate the correlation between periodontitis and cerebral small vessel disease (CSVD) from the clinical and microbiological aspects.Periodontitis patients (CP group, n = 31) and CSVD patients (CSVD group, n = 30) were examined for neurological and periodontal condition. Subgingival plaque was collected and performed using 16S rRNA sequencing. Logistic regression and LASSO regression were used to analyze the periodontal parameters and subgingival microbiota related to CSVD, respectively. Inflammatory factors in gingival crevicular fluid (GCF) were also detected and compared between the two groups.Clinical attachment level (CAL), teeth number and plaque index demonstrated a significant difference between CP and CSVD group, meanwhile, CAL was independently associated with CSVD. Besides, the microbial richness and composition were distinct between two groups. Five genera related to periodontal pathogens (Treponema, Prevotella, Streptococcus, Fusobacterium, Porphyromonas) were screened out by LASSO regression, suggesting a potential association with CSVD. Finally, the levels of inflammatory factors in GCF were statistically higher in CSVD group than those in CP group.Cerebral small vessel disease patients demonstrated worse periodontal condition, meanwhile the interaction between microbiota dysbiosis and host factors (inflammation) leading to a better understanding of the association between periodontitis and CSVD. Background Currently, research on the factors influencing the health status of older adults with multimorbidity in China is scattered, and it is difficult to give a comprehensive consideration from a holistic perspective, and the contribution of factors leading to health disparities is not explored, resulting in the ineffectiveness of current health management often programs in older adults with multimorbidity. Objective By introducing a health bifactor model, this study aims to understand the endogenous and exogenous influencing factors and their contributions to the health of older adults with multimorbidity and provide a practical basis for developing precise health management plans for older adults with multimorbidity. Methods In this study, using the China Health and Retirement Longitudinal Survey (CHARLS) 2018 data and introducing the two-factor model of health developed from the Grossman health production function (including both endogenous and exogenous aspects of health determinants). First, the Wilcoxon rank sum test was used to analyze whether there were differences in the health status of older adults with multimorbidity by gender. Secondly, the OLS regression model was used to analyze the mechanism of the two-factor model of health on the health of older adults with multimorbidity. Finally, the Shapley value method was further used to decompose the contribution of health endogenous factors to their health. Results The study found that factors such as still drinking alcohol, having no disease control methods, being satisfied with medical services, having a high level of education, having a pension, not using health services, attending free health checks, caring for grandchildren and being satisfied with their children's relationship were more likely to improve the health of older people with multiple chronic conditions. The results of the Shapley decomposition showed that socioeconomic status was the most important factor in the full sample, while family health support, health-related behaviors, and health-related behaviors were the most important factors. Coping strategies was the next most important, and social health resources was the least influential. In the gender subgroup analysis, socioeconomic status remained the most important factor; for older men with multiple chronic illnesses, health-related behaviors were the next most important factor; for older women with multiple chronic illnesses, coping strategies were the next most important. Conclusion The health status of older men with multimorbidity is better than that of women, socioeconomic status is the most important factor affecting the health of older adults with multimorbidity, and the remaining four dimensions have different contributions to the health differences between men and women. It is recommended that precise health management should be implemented according to the size of the contribution of each dimension of different health endogenous factors to improve the health status of older adults with multimorbidity with maximum benefit.

Voltage-gated sodium (Na V ) channels mediate a plethora of electrical activities. Na V channels govern cellular excitability in response to depolarizing stimuli. Inactivation is an intrinsic property of Na V channels that regulates cellular excitability by controlling the channel availability. The fast inactivation, mediated by the Ile-Phe-Met (IFM) motif and the N-terminal helix (N-helix), has been well-characterized. However, the molecular mechanism underlying Na V channel slow inactivation remains elusive. Here, we demonstrate that the removal of the N-helix of Na V Eh (Na V Eh ΔN ) results in a slow-inactivated channel, and present cryo-EM structure of Na V Eh ΔN in a potential slow-inactivated state. The structure features a closed activation gate and a dilated selectivity filter (SF), indicating that the upper SF and the inner gate could serve as a gate for slow inactivation. In comparison to the Na V Eh structure, Na V Eh ΔN undergoes marked conformational shifts on the intracellular side.
