The structure of ALP-D was characterized by FT-IR, monosaccharide composition and molecular weight.The optimal conditions for the green synthesis of silver nanoparticles using ALP-D were determined through single-factor experiments. The synthesized silver nanoparticles were characterized by UV-Vis spectroscopy, FT-IR, DLS, HRTEM and XRD. Finally, the in vitro antioxidant activity, antibacterial activity and anticancer activity of the nano-silver composite were evaluated.Results: Single-factor experiments identified the optimal synthesis conditions for ALP(D)-AgNPs as a reaction time of 180 min, a temperature of 100 °C, and a 10:1 volume ratio of silver nitrate to ALP-D. The free radical scavenging activity of ALP(D)-AgNPs against DPPH and ABTS was significantly enhanced compared with that of ALP-D. The minimum inhibitory concentration (MIC) values of ALP(D)-AgNPs against E coli and B subtilis were 31.25 μg/mL, while the MIC value against S aureus was 62.5 μg/mL. The minimum bactericidal concentration (MBC) values of ALP(D)-AgNPs were 125 μg/mL for E coli, B subtilis, and S aureus. The IC50 values of ALP(D)-AgNPs on the MDA-MB-231, HepG2, Caco-2, and C6 cancer cell lines were 14.72 ± 0.23, 8.19 ± 0.65, 22.73 ± 3.01, and 15.77 ± 2.91 μg/mL, respectively.Conclusion: In summary, we have identified a novel material for the green synthesis of silver nanoparticles. The results show the ALP(D)-AgNPs synthesized using the new material ALP-D exhibit excellent stability and dispersibility. The key to reducing the risk of NI is early detection and treatment in time. Nurses are close observers and primary caregivers for neonates at the bedside of the NICU, who are best positioned to capture the risk signals of NI. This study aims to develop a nurse-led prediction model for NI of preterm infants in the NICU.Patients and Methods: This study was designed as a retrospective study, preterm infants of the NICU at Renmin Hospital of Wuhan University from January 2020 to December 2023 were selected and divided into the NI group and non-NI group. Clinical data were collected and then analyzed by univariate analysis, least absolute shrinkage and selection operator (LASSO) regression analysis, and multivariate logistic regression analysis. The outcome constructed a nomogram model and its predictive efficacy was evaluated by the area under the receiver operating characteristic curve (AUC), calibration curve, and decision curve analysis (DCA). Bootstrap method was used to repeat 1,000 times for internal validation.Results: A total of 892 preterm infants were finally included and a nurse-led predictive model established, which included six variables: skin color changes, respiratory related changes, feeding deterioration, birth weight, number of arterial and venous blood draws, and days of nasogastric tube placement. The model’s AUC was 0.953, indicating good discriminatory power. The calibration plot demonstrated good calibration and the Hosmer–Lemeshow test showed high consistency. DCA indicated that the nomogram had good clinical utility. Internal validation showed the AUC of 0.952.Conclusion: This nomogram model, which is mainly based on nurses’ observations, shows good predictive ability. The craniocervical posture and facial morphology of every individual were assessed through Uceph software analysis of their cephalograms, considering 32 variables. The Independent T-test and the Mann–Whitney U-test were used to evaluate the relationships between the measures and OS. Differences in craniocervical posture and craniofacial morphology between the two groups were investigated by logistic regression.Results: Subjects with OS exhibited more severe forward head posture (FHP) (larger C0-C1, Ba-C3ia, C2ap-C4ip, craniocervical angle, CVT-OPT, and NSL-C2’) and larger facial configuration (greater mandibular body length, posterior cranial base length, and posterior facial height) than the subjects without OS. Larger C2ap-C4ip (OR 1.53; 95% CI 1.282~1.82), Craniocervical angle (OR 1.70; 95% CI 1.457~1.977) and NSL-C2’ (OR 1.18; 95% CI 1.097~1.264), were associated with greater likelihood of having OS.Conclusion: The subjects with OS were more prone to forward head posture than the control group. Objective: To evaluate the impact of dexmedetomidine (Dex) on agitation and inflammatory response during recovery from anesthesia in young children following cochlear implantation surgery., Methods: We randomly divided 80 children who underwent unilateral cochlear implantation into two equal groups. Group D received an intravenous infusion of Dex after induction of anesthesia, while those in group C received an equal volume of saline infusion. The mean arterial pressure (MAP) and heart rate (HR) of children in the two groups were recorded at four different time intervals: before induction of anesthesia (T 0 ); 30 min after intravenous infusion of Dex (T 1 ); upon admission to the post-anesthesia care unit (PACU) (T 2 ); and at the time of being transferred out of the PACU (T 3 ). At T3, we also recorded general information., Results: The MAP and HR in group D showed more consistent trends during the anesthesia recovery period when compared to those in group C. Children in group D had a significantly lower crying, requires increased O 2 administration, increased vital signs, expression and sleepless score (CRIES score), pediatric anesthesia emergence delirium (PAED) score, and incidence of agitation than in group C (P < 0.01). The rate of supplementary pain relief for the children was lower in group D than in group C (P < 0.01). At T 3 , serum levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were lower in children in group D than in group C (P < 0.01). Peritoneal metastasis (PM) is the primary site of distant metastasis in gastric cancer (GC) and is associated with an advanced disease stage and poor prognosis. Due to its high resistance to chemotherapy, disseminated peritoneal lesions are often untreatable. A primary reason for therapy resistance in cancer cells is often their defective cell death execution mechanisms. Ferroptosis, a newly identified type of regulated cell death, is strongly linked to the emergence and formation of tumors. Earlier studies have demonstrated the significant role of RNA-binding proteins in ferroptosis. Nevertheless, the fundamental process linking Staufen Double-Stranded RNA Binding Protein 1 (STAU1) to ferroptosis in the peritoneal metastasis of gastric cancer is yet to be clarified. This study shows that the RNA-binding protein STAU1 is crucial for regulating ferroptosis in gastric cancer cells. Elevated levels of STAU1 are linked to unfavorable outcomes in individuals diagnosed with gastric cancer. STAU1 was up-regulated by PLAGL2 and decreased the stability of NCOA4 mRNA by binding to the 3'-untranslated region. Decreased NCOA4 expression inhibits the accumulation of reactive iron, the occurrence of the Fenton reaction, and cellular ROS generation in the GC cells. Additionally, we showed that NCOA4 is crucial in the process of ferritinophagy triggered by the reduction of STAU1 in gastric cancer cells. Ultimately, the process safeguards GC cells from ferroptosis. These findings elucidate the function of PLAGL2/STAU1/NCOA4 in the ferroptosis of gastric cancer cells and provide theoretical backing for possible diagnostic markers and treatment targets for peritoneal metastasis in gastric cancer., Competing Interests: Declarations. Cold weather events such as cold spell and snowstorm could trigger heart attacks, but the effect of compound exposure to cold weather events remains unknown. This study aimed to estimate and compare the effect of independent and compound exposure to common cold weather events (cold spell and snowstorm) on acute myocardial infarction (AMI) mortality. Daily data on AMI deaths (74,279) and weather conditions were collected in a subtropical-temperate climate transit region of China from 2016 to 2020. We conducted a space-time-stratified case-crossover analysis to fit the association of AMI mortality with cold spell and snowstorm. We observed an increased AMI death risk associated with independent and compound exposure to cold spell or snowstorm. Furthermore, the association of compound exposure to cold spell and snowstorm was greater than the independent exposure but varied by the compound types. A greater risk was observed if the snowstorm and cold spell occurred on the same day [odds ratio (OR): 1.197 (95% confidence interval (CI): 1.153-1.243)] than the scenario that the snowstorm was followed by cold spell [OR: 1.100 (95%CI: 1.079-1.121)] or the cold spell was followed by snowstorm [OR: 1.115 (95%CI: 1.075-1.157)]. Background: Periodontitis is a periodontal inflammatory disease closely related to hypertension. However, the role of age and systemic inflammation is unclear. This study examined the effect of age and systemic inflammation on the association between severity of periodontitis and blood pressure in periodontitis patients., Methods: This cross-sectional clinical study included periodontitis patients treated at Beijing Chaoyang Hospital, Capital Medical University from January 2020 to July 2023. Patients with periodontitis were included using inclusion and exclusion criteria, and data on basic information, blood pressure, periodontal examination indicators, and biochemical examination indicators were collected. Patients were grouped by blood pressure, and differences in basic information, blood indexes, and periodontal examination indexes between the two groups were compared by independent sample T-test. With PBOP as independent variables, systolic blood pressure (SBP) and diastolic blood pressure as dependent variables, and several indexes as covariables, the relationship between them was studied by linear regression. The interaction was used to explore the relationship between age and systemic inflammation in periodontitis and hypertension., Results: A total of 144 patients with periodontitis were enrolled. In fully adjusted analyses, the percentage of site with bleeding on probing (PBOP) was significantly associated with SBP (B = 10.08, p = 0.027) in the overall group. In patients 50 years or older, but not in those younger than 50, PBOP remained significantly associated with SBP (p = 0.018 for interaction). The strength of association between PBOP and SBP was greater in patients with higher systemic immune-inflammation index. The regression coefficients of PBOP in the systemic immune-inflammation index ≥ 400 and < 400 groups were 16.61 (95% CI, 3.32-29.90; p = 0.015) and 13.69 (95% CI, 1.15-26.23; p = 0.033), respectively (p = 0.047 for interaction). Similar results were observed in the systemic inflammation response index ≥ 9 and < 9 groups (B = 17.04 (95% CI, 2.66-31.42, p = 0.021) and 13.98 (95% CI, 1.94-26.01, p = 0.024 respectively; p = 0.028 for interaction)., Conclusions: This study indicates a significant association between the severity of periodontitis and SBP, which was modified by age and systemic inflammation., Competing Interests: Declarations. The significance of ORPD (Optimal Reactive Power Dispatch) cannot be emphasized within the operation of power systems, particularly according to the growing use of electric vehicles (EVs). Electric vehicles (EVs) have the potential to influence the power grid via their ability to augment power demand and function as distributed energy resources. The effective administration of Optimal Renewable Power Dispatch (ORPD) in conjunction with Electric Vehicle (EV) integration necessitates meticulous examination of charging schedules, battery capacity, and the desired state of charge. In the current paper, a novel optimizer known as the Novel Gooseneck Barnacle Optimization (NGBO) algorithm is introduced to address the ORPD problem within the presence of Electric Vehicles (EVs). The NGBO algorithm draws inspiration from the regular mating behavior of gooseneck barnacles involving self-fertilization and casting sperm. To evaluate its performance, the NGBO algorithm is applied to two standard exam systems, including the IEEE 118- and IEEE 57-system of bus, considering various scenarios of EV penetration. The experimental outcomes demonstrate the NGBO effectively mitigates active power loss and voltage variation in power systems, surpassing several existing metaheuristic optimization techniques by reducing power loss by up to 15% and voltage deviation by up to 10% compared to traditional methods, demonstrating the effectiveness of the method in handling EV-related uncertainties., Competing Interests: Declarations. Electrochemical nitrate reduction reaction (NO 3 RR) stands out as a promising route for sustainable ammonia synthesis, in which active hydrogen (*H) plays a crucial role in both the deoxygenation and hydrogenation steps. However, the regulation of surface *H is still overlooked, and without intervention, the competing hydrogen evolution reaction is kinetically more favored over the NO 3 RR, leaving the current system as far from satisfactory. Herein, based on reverse utilization of the Sabatier principle, a series of Fe x Ni y substitutional solid-solution alloys (SSAs) are synthesized to manipulate *H behavior for enhanced NO 3 RR. Upon precise optimization of the alloy composition, the d-band center of HER-active Ni shifts toward the Fermi level, endowing the catalyst with strong interaction to *H and greatly prolonging its lifetime, which enables abundant supply to facilitate the NO 3 RR. As expected, a maximum NH 3 yield rate of 31.46 mmol h -1 mg -1 is delivered over the optimized Fe 3 Ni 1 -SSA, which is considerably higher than most of the extensively reported works. Several in situ characterizations are combined to gain in-depth insight. Especially, in situ Fourier transform infrared spectroscopy in internal reflection mode directly observes *H enrichment on the catalyst surface, while the accompanied facilitation of the NO 3 RR process is verified by external reflection mode.

Gold nanorods (AuNRs) have shown great potential as photothermal agents for cancer therapy. However, the biosafety of AuNRs ordinarily synthesized using a cationic ligand assistance procedure has always been a subject of controversy, which limits their application in tumor therapy. In this study, we propose a novel strategy to enhance the biocompatibility of AuNRs by constructing a biological coating derived from silk fibroin (SF) on their surface. The SF coating could be easily and precisely manipulated using a layer-by-layer (LBL) assembly method. The resulting SF-coated gold nanorods (AuNRs@SF) exhibited reduced cytotoxicity and improved hemocompatibility compared to untreated AuNRs. Moreover, the treated nanorods were easily modified with a tumor-targeting peptide (AuNRs@MTSF) and efficiently loaded indocyanine green (ICG). Both in vitro and in vivo analyses demonstrated that AuNRs@MTSF could more effectively reach tumor tissue and enter MCF-7 cells. Furthermore, after loading ICG, AuNRs@MTSF exhibited superior antitumor efficacy compared to other groups by combining photodynamic therapy (PDT) with photothermal therapy (PTT) under near-infrared (NIR) irradiation without inducing any side effects. This work suggests that SF coating of gold nanorods is a potential approach for improving their biocompatibility, and that function-modified AuNRs@SF are effective nanoplatforms for targeted and multimodal tumor therapy.

Background: Our team has previously found that the stimulator of interferon genes (STING) plays a more significant anti-tumor role in host immune cells than in tumor cells. Although STING is necessary for CD8 + T cells to exert immunological activity, its effect on CD8 + T cells remains debatable. In this study, we used both in vitro and in vivo models to explore the metabolic effects of STING on CD8 + T cells., Methods: Peripheral blood lymphocytes were procured from non-small cell lung cancer (NSCLC) patients receiving anti-PD-1 therapy to investigate the correlation between STING expression levels, CD8 + T-cell subsets, and immunotherapy efficacy. STING knockout (STING-KO) mice were used for in vivo studies. RNA-seq, seahorse, flow cytometry, electron microscopy, qPCR, immunofluorescence, western blotting, and immunoprecipitation were performed to explore the underlying mechanisms of STING in regulating CD8 + T cell function., Results: We discovered that the expression level of STING in immune cells exhibited a significant correlation with immunotherapy efficacy, as well as with the proportion of central memory CD8 + T cells. Moreover, we found that the loss of the STING gene results in a reduction in the number of mitochondria and a change in the metabolic pathway selection, thereby inducing excessive glycolysis in CD8 + T cells. This excessive glycolysis generates high levels of lactate, which further inhibits IFN-γ secretion and impacts memory T cell differentiation. Correcting the glycolysis disorder partially restored function and IFN-γ secretion, rescued the central memory CD8 + T subset, and improved immunotherapy in STING-KO mice. This provides a new treatment strategy for patients with low STING expression and a poor response to immunotherapy., Conclusion: Intrinsic STING of CD8 + T cells affects their function through the HK2/Lactate/IFN-γ axis and affects memory differentiation by regulating glycolysis., Competing Interests: Declarations. Controllable and precise delivery of therapeutic agents is critical for effective tumor therapy. However, tumor targeting and the deep penetration of drugs remain among the most challenging issues in achieving controlled delivery. Herein, a novel engineered platelet microneedle patch with a microwave-responsive magnetic biometal-organic framework is proposed to facilitate the combination of the engineered platelet and microwave hyperthermia, enhancing deep drug penetration into tumors and enabling precision therapy. The prepared magnetic biometal-organic framework as nanomedicine exhibits excellent microwave thermal effects. The engineered platelets could be activated in the tumor microenvironment to release PMPs and nanomedicines combined with microwave hyperthermia for enhancing both cell uptake and deep drug penetration into tumors. The developed separable microneedle patch system allows the microneedle tip to be quickly detached from the backing layer and retained within the target tissue for repeated local cancer hyperthermia treatments. By integration of engineered platelets into the microneedle patch, the transdermal deep delivery of drugs could be effectively enhanced for local microwave thermochemotherapy of tumors. This work represents the first attempt to graft microwave-responsive inorganic nanomedicines onto platelets as cell drugs, offering a novel strategy for precise drug delivery activated by microwave thermal therapy.

Halogen-substituted para-benzoquinones (p-BQs) are emerging disinfection byproducts known to induce oxidative damage both in vitro and in vivo. However, as ubiquitous oxidation byproducts, the in vivo toxicity and transport mechanism of non-halogenated p-BQs with similar structure of α, β-unsaturated ketones to halogenated p-BQs have not been thoroughly investigated. In this study, the effect of substituents on toxicity and transportation of 2-chloro-1,4-benzoquinone (CBQ) and 2-methyl-1,4-benzoquinone (MBQ) was systematically investigated. The results show that MBQ exhibits slightly lower acute toxicity to zebrafish embryos compared to CBQ. Exposure to both CBQ and MBQ at concentration of 10 μg/L and 100 μg/L significantly increased the levels of reactive oxygen species, and enhanced the activities of total superoxide dismutase, catalase, and glutathione peroxidase, while malformations were primarily observed in the 100 μg/L exposure groups. The varying developmental toxicity was associated with significant upregulation of 10 genes by CBQ compared to only 6 by MBQ. Using the high-resolution mass spectrometry and electron paramagnetic resonance spectroscopy, the hydroxylation of both CBQ and MBQ, and the production of semiquinone radicals and hydroxyl radicals in aqueous environments have been revealed. In the process of drilling and development of tight sandstone reservoirs, a large number of external fluids can invade the pore structure of the reservoir due to its strong hydrophilicity, resulting in blockage of the pore structure and a decrease in oil and gas production capacity. To reduce the aqueous phase trapping damage of tight sandstone reservoirs, the SiO 2 @KH550/FC-A nano-composite system was prepared to construct hydrophobic surfaces in core samples. First, the surface of nano-SiO 2 was modified by KH550 to prepare nanoparticles with good dispersion. FTIR, XRD, SEM, and TG were used to characterize the nanoparticles before and after modification. The fluorosurfactant FC-A was prepared in the laboratory and combined with SiO 2 @KH550 to produce the SiO 2 @KH550/FC-A composite system. The system increases the contact angle of the hydrophilic surface from approximately 20° to 130°, achieving wetting modification. Excellent stability of hydrophobicity was obtained, and the contact angle did not significantly decrease within 5 minutes. In contrast, using FC-A and SiO 2 @KH550 individually, the contact angle of the hydrophilic surface could not be increased to over 90°. The SEM results showed that after treatment with the composite system, a layer of micro-nanoscale particles was attached to the hydrophilic surface. It was proved that SiO 2 @KH550 and FC-A were adsorbed on the surface, forming a low surface free energy solid interface at the micro and nano scales, which greatly improved the surface hydrophobicity. Furthermore, after the composite system was used to treat tight sandstone cores aged at 100° for 16 hours, the amount of imbibition of the core samples treated with SiO 2 @KH550/FC-A significantly decreased from 2.6 mL of brine to only 0.5 mL after 8 hours. The core spontaneous imbibition rate was also reduced to 0.0004 g min -1 within 5 minutes, while the maximum brine water spontaneous imbibition rate was 0.27 g min -1 . The core displacement experiment further showed that the fluid in the core pores can be more easily flowed back under gas displacement after wetting modification. The water saturation of the core samples decreased to 16.3% after displacement, and the core permeability recovered to 88.4%, indicating that the SiO 2 @KH550/FC-A composite system can significantly improve the liquid phase flowback ability., Competing Interests: There are no conflicts to declare., (This journal is © The Royal Society of Chemistry.)

Pancreatic cancer (PCa) has insidious onset, high malignancy and poor prognosis. Gemcitabine (GEM) is one of the first-line chemotherapy drugs for PCa. However, GEM resistance has always been a bottleneck problem leading to recurrence and death of PCa patients. RNA-binding proteins (RBPs) are important proteins that regulate transportation, splicing, stability and translation of RNA. Abnormal expression of RBPs often lead to a series of abnormal accumulation or degradation of downstream RNA resulting in various diseases. In our study, we utilized RIP seq, RIP-qPCR, in vitro and in vivo experiments and found that pumilio2 (PUM2) was high expression in PCa, and promoted GEM resistance of PCa by regulating mRNA stability of integrin Alpha 3 (ITGA3) and other genes in focal adhesion pathway, and there was positive feedback regulation between PUM2 and transcription factor early growth response gene 1 (EGR1), that is PUM2 binding to 3'UTR region of EGR1 mRNA, and EGR1 binding to promoter region of PUM2 gene. Background: Previous neuroimaging studies have identified substantial structural and functional abnormalities in the brains of patients with rhegmatogenous retinal detachment (RRD). Nonetheless, there remains a paucity of comprehensive research on the alterations in functional connectivity (FC) within large-scale static and dynamic brain networks in these patients., Methods: This study utilized independent component analysis (ICA) to investigate alterations in large-scale brain network FC in patients with RRD. Additionally, it employed support vector machine (SVM) to classify RRD patients and healthy controls (HCs) and examined the relationship between abnormal brain regions and clinical ophthalmic parameters., Results: The RRD patients demonstrated significantly increased FC in the default mode network (DMN) and visual network (VN) compared to the HCs, whereas the FC in the auditory network (AN) and the sensorimotor network (SMN) was significantly decreased. Analysis of dynamic functional network connectivity (dFNC) revealed that the fraction of time (FT) spent in state 4 was significantly greater in RRD patients compared to HCs. SVM analysis showed that the AUC for classification using AN and FNC features reached 0.828 and 0.819, respectively. Additionally, the FC value of the right medial superior frontal gyrus (R-SFGmed) in RRD patients was positively correlated with axial length (r = 0.401, p = 0.038)., Conclusion: This study revealed that patients with RRD exhibit both damage and adaptive remodeling in their brain functional networks. Tourist risk perception is a theoretical topic of great interest, but few studies have specifically investigated vulnerable groups of people with chronic illnesses, chronic illnesses may be part of their daily lives, affecting individuals' health risk awareness and complicating travel decisions. This study is one of the first in the tourism field to explore risk perception and travel decision-making among chronically ill tourists. Adopting protection motivation theory as a guiding framework, 504 chronically ill tourists with different stages and types of illness were investigated to assess patients' perceived health risks and actual travel intentions in a quantitative method. It was found that the travel behavior and self-protective behavior of chronically ill tourists depended on the strength of risk perception, which was mainly influenced by perceived threat, response cost and self-efficacy. Further studies confirmed the role of risk perception as a mediator between PMT factors and decision-making behavior and the moderating effect of Plog psychological type. The study reveals the logic that shapes the decision-making behaviors of chronically ill tourists' travel and expands accessible tourism from creating enjoyable experiences to facilitating positive travel, and the findings can provide theoretical guidance for the design of destination experiences and encourage the exploration of travel behaviors of people with chronic illnesses., Competing Interests: Declarations. Challenges remain in the design and manufacture of acoustic devices with excellent broadband sound absorption performance. Herein, an efficient acoustic absorber with hierarchical pore structure and additional acoustic-electrical energy conversion mechanism is reported in which combined zirconia porous ceramics and P(VDF-TrFE) piezoelectric aerogels. Reticular cross-scale pore structure enables sound waves to propagate and dissipate effectively. The vibrations generated by piezoelectric aerogels under acoustic excitation further consume sound waves through converting acoustic energy into electrical energy based on the local piezoelectric and triboelectric effect, which improves the medium and low-frequency sound absorption capability. The average sound absorption coefficient of the prepared acoustic composites reaches 0.87 while the noise reduction coefficient is 0.54. Furthermore, the composites also possess low density, high compressive strength, good hydrophobicity and thermal insulation properties for applications. Urban drainage system (UDS) plays a key role in city urbanization, where defective pipes can lead to seepage. Previous studies have identified the locations of defects in UDS using inverse optimization models. However, the unique optimal solution neglects uncertainty analysis, which may lead to misdiagnosis. In addition, the multi-point defect diagnosis has heavy computational burden due to high dimensional parameters space. To address these issues, this paper proposes a hybrid method that leverages the genetic algorithm (GA) to identify probable space, and then utilizes the adaptive Metropolis (AM) to provide an estimation of the posterior probability distribution (PPD). Firstly, a multi-population GA is employed for the maximum exploration within the model space. Then, AM algorithm is used to explore the final PPD of each pipe defect parameter. The metrics accuracy (ACC), Matthews correlation coefficient (MCC) and mean absolute error (MAE) are used to evaluate the diagnosis performance. A synthetic UDS case with randomized multi-point seepage scenarios is used to validate the method. Results indicate that the proposed hybrid method is effective in diagnosing multi-point defect, with 0.91, 0.78 of the hybrid method and 0.87, 0.69 of the DiffeRential Evolution Adaptive Metropolis method for the ACC and MCC, respectively. Meanwhile, the diagnosis speed has increased by 32 %. The result PPD passes the 90 % confidence interval validation. Ambient low temperature is a risk factor for myocardial infarction (MI) worldwide, with limited evidence about the effect of low temperature at different periods of a day. We aimed to characterize the association of MI mortality with daytime and nighttime low temperatures in rural areas. Daily daily temperature data and MI deaths from 2016 to 2020 were obtained for the rural areas of Anhui Province. We first applied the distributed lag nonlinear model to examine the association of MI death risk with daily maximum (daytime) and minimum (nighttime) temperature for each rural area. Then, we used the random-effects meta-analysis to pool the rural-specific effect estimates and subgroups stratified by sex, age, and geographical region. Daytime low temperature and nighttime low temperature were both associated with an increased risk of MI death. On the same day of exposure to daytime low temperature and nighttime low temperature, the risk of MI death respectively increased by 12.6% [95% confidence interval (CI): 0.7% ~ 25.8%] and 3.2% (95%CI: 0% ~ 6.6%). Subgroup results showed an increased MI death risk associated with daytime and nighttime low temperatures in the elderly (≥ 65 years) and males. Besides, there was an obvious geographical variation in the lag effects of daytime and nighttime low temperatures on MI mortality. This study provides evidence of different effect of daytime and nighttime low temperature on MI mortality in China. PAK4 has been widely reported to function in somatic cells. However, its role and the underlying mechanisms in meiotic oocytes are largely unknown. Here, we show that PAK4 deficiency significantly disrupts maturational progression and meiotic apparatus in mouse oocytes. Furthermore, based on the kinase substrate binding preference and systematic functional screening, our mechanistic investigation demonstrated that PAK4 promotes cytoskeletal organization and oocyte maturation through phosphorylating serine 597 on DDX17. Of note, we identified a marked reduction of PAK4 protein in oocytes from diabetic mice. Importantly, ectopic expression of hyperphosphorylation-mimicking DDX17 mutant (DDX17-S597D) partly prevented the meiotic defects in these diabetic oocytes, indicating that the decreased phosphorylation of DDX17 due to PAK4 insufficiency is responsible for the impaired oocyte quality. Toll-like receptors (TLR) are phylogenetically conserved mediators of innate immunity that are essential for establishing adaptive immune responses against invading pathogens. TLR7 is an endosomal receptor expressed predominantly in myeloid and B cells. Activation of TLR7 induces Type I interferon and proinflammatory responses; therefore, targeting TLR7 is a promising strategy for antitumor therapy. Although the use of bacterial components to trigger innate immune responses in cancer patients started a century ago, the effectiveness of systemic TLR agonists has been rather underwhelming in clinical trials, partly due to nonspecific immune activation leading to safety and tolerability issues. Antibody-drug conjugates (ADCs) constitute a proven therapeutic modality amenable to systemic administration with limited toxicity concerns via a targeted delivery platform. We generated TLR7 agonist-antibody conjugates that recognize tumor antigens expressed on the surface of tumor cells. Generated ADCs demonstrated robust activity in in vitro tumor antigen-presenting cell (APC) coculture systems as indicated by dose-dependent upregulation of PD-L1 and CD86 on macrophages. TLR7 agonist-ADC provided superior tumor growth control compared to intravenously (IV) administrated free TLR7 agonist. Treatment with TLR7 agonist-ADC led to prolonged activation of myeloid cells in the tumor microenvironment (TME) with minimum immune activation in the periphery. Systemic and tissue exposure studies demonstrated tumor-specific free drug release by targeted ADC treatment. In summary, the TLR7 agonist-ADC can potentially activate immune cells in the TME to generate tumor antigen-specific T-cell responses, making it an attractive approach for precision cancer therapy.

Copper, one of the essential nutrients for the human body, acts as an electron relay in multiple pathways due to its redox properties. Both deficiencies and excesses of copper lead to cellular fragility. Therefore, it can manifest pro- and anti-cancer properties in tumors. Therefore, it is crucial to clarify the copper activity within the cell. We have thoughtfully summarized the metabolic activities of copper from a macro and micro perspective. Cuproptosis, as well as other forms of cell death, is directly or indirectly interfered with by Cu 2+ , causing cancer cell death. Meanwhile, we did pan-cancer analysis of cuproptosis-related genes to further clarify the roles of these genes. In addition, copper has been found to be involved in multiple pathways within the metastasis of cancer cells. Given the complexity of copper's role, we are compelled to ask: is copper a friend or a foe? Up to now, copper has been used in various clinical applications, including protocols for measurement of copper concentration and bioimaging of radioactive 64 Cu. But therapeutically it is still a continuation of the old medicine, and new possibilities need to be explored, such as the use of nanomaterials. Some studies have also shown that copper has considerable interventional power in metabolic cancers, which provides the great applications potential of copper therapy in specific cancer types. This paper reviews the dual roles played by cuproptosis in cancer from the new perspectives of oxidative stress, cell death, and tumor metastasis, and points out the value of its application in specific cancer types, summarizes the value of its testing and imaging from the perspective of clinical application as well as the current feasible options for the new use of the old drugs, and emphasizes the prospects for the application of nano-copper., (© 2025 The Author(s). Integrins, a family of transmembrane receptor proteins, are well known to play important roles in cancer development and metastasis. However, a comprehensive understanding of these roles has not been achieved due to the complex relationships between specific integrins, cancer types, and the stages of cancer progression. Publicly accessible repositories from the Genotype-Tissue Expression (GTEx) and The Cancer Genome Atlas (TCGA) projects provide rich datasets for exploring these relationships using machine learning (ML). In this study, integrin RNA-Seq expression data of ~ 8 healthy tissues in GTEx and corresponding tumors in TCGA were selected. Integrin expression was used to train ML models to distinguish between different healthy tissues, solid tumors, as well as normal and tumor samples from the same tissue type. These ML models can classify samples by tissue origin or disease status with high accuracy, and the integrins essential to these classifiers were identified. In some cases, the expression of only one or two integrins was needed to classify tissue type, tumor type or disease status with accuracy > 0.9. For example, expression of ITGA7 alone can distinguish healthy and cancerous breast tissue. Additionally, integrin co-expression networks in healthy and cancerous breast tissues were compared and were found to change significantly from healthy to cancer, indicating changes in functional involvement of integrins due to cancer. Integrin expression in metastatic tumors were further examined using data from the AURORA project for Metastatic Breast Cancer (MBC), and several integrins such as ITGAD, ITGA4, ITGAL, and ITGA11 were found to have significantly lower expression in metastases than in primary tumors., Competing Interests: Declarations. The integration of single-cell RNA sequencing (scRNA-seq) and spatial transcriptomics (ST) data is crucial for understanding gene expression in spatial context. Existing methods for such integration have limited performance, with structural similarity often below 60\%, We attribute this limitation to the failure to consider causal relationships between genes. We present CausalGeD, which combines diffusion and autoregressive processes to leverage these relationships. By generalizing the Causal Attention Transformer from image generation to gene expression data, our model captures regulatory mechanisms without predefined relationships. Across 10 tissue datasets, CausalGeD outperformed state-of-the-art baselines by 5- 32\% in key metrics, including Pearson's correlation and structural similarity, advancing both technical and biological insights.

Conventional dual-signal electrochemiluminescence (ECL) sensors feature high sensitivity and reliability, but the involvement of coreactants inevitably results in a complex configuration and shows reproducibility risk. Here, we propose an exogenous coreactant-free dual-signal platform, comprising luminol (anodic luminophore), CdSe quantum dots (cathodic luminophore), and Co 3 O 4 /Ti 3 C 2 electrocatalyst (coreaction promoter). At different redox potentials, Co 3 O 4 /Ti 3 C 2 induces water oxidation and oxygen reduction to produce • OH and O 2 •- radicals, which subsequently drive cathodic and anodic ECL emission, respectively. The dual-signal ECL pathways are confirmed by reactive oxygen species (ROS) scavenging, and the effect of dissolved oxygen is excluded. By integrating with molecular imprinting technology, this dual-signal ECL sensor is employed for the quantitative detection of trenbolone. It exhibits high sensitivity, broad linear range (1.0 × 10 1 to 1.0 × 10 8 fg mL -1 ), low detection limit, and detection capability of real samples. This work presents a new endogenous ROS-driven dual-signal ECL sensor that excludes the use of exogenous coreactants and offers new insights into ROS generation in a coreactant-free sensor design.

The deep brain stimulation (DBS) in the subthalamic nucleus (STN) has attracted more attention for primary Meige syndrome due to easier target location and lower power consumption. However, potential and reliable preoperative predictors of longitudinal outcomes of STN-DBS to guide therapeutic decisions remain largely unexplored. Herein, we used preoperative structural MRI and Burke-Fahn-Marsden Dystonia Rating Scale (BFMDRS) from 55 patients with primary Meige syndrome who finished STN-DBS after 5 years. They were further classified into response ( n = 23) and super-response ( n = 32) based on the improvement rates of BFMDRS. Voxel-based morphology, partial correlation analyses, receiver operating characteristic (ROC) analyses and support vector machine were performed. We identified that improved rates of BFMDRS were 63, 71.97, 76.64, 79.51, 81.02, 81.36, 81.16, 80.80 and 80.93% at 1, 3, 6, 12, 18, 24, 36, 48 and 60 months after STN-DBS, respectively, and remained steady across 1-5 years. Further voxel-based morphology analyses revealed significantly lower grey-matter volume in the right hippocampus, left putamen, right supramarginal gyrus and left superior frontal gyrus in response when compared with super-response. The grey-matter volumes in the left putamen, right supramarginal gyrus and left superior frontal gyrus were not only positively correlated with improvement rates of BFMDRS after STN-DBS for 5 years in the primary Meige syndrome, but also presented a reliable classification ability in distinguishing response and super-response (area under curve = 0.855). Photoelectrochemical (PEC) immunosensors are highly promising tools for monitoring biochemical molecules. Constructing high-performance heterojunctions is a general method to improve the sensitivity of PEC immunosensors. The internal electric field (IEF) formed at the heterojunction interface plays a crucial role in coordinating the separation of photogenerated carriers. Although IEF in heterojunctions shows extraordinary talents in PEC immunosensors, it is powerless to suppress the recombination of photogenerated carriers in the bulk phase. Thus, modulating the IEF of heterojunctions more than just at interfaces remains a challenge. Herein, a "signal-off" PEC immunosensor for CYFRA21-1 detection was constructed. The immunosensor was based on sulfurized bismuth oxyhydroxide compound Bi 2 S 3 /Bi 2 O 2 (OH) 2 nanorods as the platforms, N-doped hollow homogenous Mo 2 C nanospheres with Ru NPs (Mo 2 C:N@Ru) as the quenching label. The Bi 2 O 2 (OH) 2 were composed of [Bi 2 O 2 ] 2+ and OH - alternating layers, which provided an IEF throughout the bulk phase. Meanwhile, the heterogeneous interface further promoted the separation of photogenerated carriers and output strong photocurrent signals by energy level matching of Bi 2 O 2 (OH) 2 with Bi 2 S 3 . Subsequently, Mo 2 C:N@Ru was quantitatively introduced in the presence of CYFRA21-1 through specific recognition between antigens and antibodies. The main structure of Mo 2 C:N@Ru was a homojunction formed by cubic and hexagonal phases Mo 2 C, which accelerated the separation of photogenerated carriers. Moreover, doped N element and Ru NPs further enhanced the competition between Mo 2 C:N@Ru and the Bi 2 S 3 /Bi 2 O 2 (OH) 2 platforms for ascorbic acid and then achieved a quenching effect. The limit of detection for CYFRA21-1 was 17 fg/mL, and linearity range was 50 fg/mL -100 ng/mL. This PEC immunosensor achieves highly sensitive detection of CYFRA21-1 and has good scalability, which provides new ideas for the development analysis and detection platforms of tumor biomarkers. Objective: To investigate the expression pattern of pan-TRK protein in colorectal cancers with NTRK gene fusion and mismatch repair deficient (dMMR) and to analyze its molecular pathological characteristics. Methods: A total of 117 dMMR colorectal cancers diagnosed in the Department of Pathology of Henan Provincial People's Hospital, Zhengzhou, China from 2020 to 2023 were collected. Immunohistochemistry (IHC), fluorescence in situ hybridization (FISH) and DNA/RNA-based next-generation sequencing (NGS) were used to detect pan-TRK protein expression and fusion partner genes in tumors, and to further explore the correlation between pan-TRK staining patterns and partner genes. Results: IHC and FISH were performed successfully in formalin-fixed paraffin-embedded tissues from 117 dMMR colorectal cancer patients. There were 15 (15/117, 12.8%) cases with positive pan-TRK, including 6 cases with strong staining in tumor cell membrane and cytoplasm, 2 cases with weakly granular staining in tumor cytoplasm, 2 cases with moderate dot-like staining in near 5% tumor cell nuclei, 1 case with moderately to strongly granular staining in the cytoplasm and membrane of tumor cells, 1 case with moderately to weakly granular staining in about 60% of the tumor cells, 1 case with strongly staining in about 1% of the tumor cells, 1 case with moderately to strongly staining in about 3% of the tumor cells and 1 case with diffuse, moderate para-nuclear dot-like and weakly perinuclear granular staining. NTRK1 gene disruption was detected in 6 cases (6/117, 5.1%) and consistent with diffusely strong expression of pan-TRK. Based on DNA/RNA NGS, it was further confirmed that the 6 cases with NTRK1 gene disruption all carried TPM3-NTRK1 fusion gene, and all had high microsatellite instability and high tumor mutation burden. No KRAS, NRAS, BRAF V600E or TP53 gene mutations were detected. Four patients carried frame shift mutations in RNF43. Other molecular changes included 3 cases with ROS1 gene mutation, 2 cases with BRAC, ALK, and EGFR gene mutations, 2 cases with ATM gene mutation, and 2 cases with KIT gene mutation. These were missense/frame shift mutations that were associated with no clinical significance. The nine pan-TRK-positive cases without NTRK gene fusion detected with DNA-based NGS were further confirmed with RNA-based NGS, and no NTRK gene fusion was found. The sensitivity and specificity of NTRK gene fusion detected using IHC were 100.0% and 92.5%, respectively. The sensitivity and specificity of diffusely strong membranous/cytoplasmic staining were both 100.0%. Conclusions: Pan-TRK protein has various expression patterns in dMMR colorectal cancer. Its diffusely strong expression is highly suggestive of NTRK1 gene fusion. TPM3-NTRK1 gene fusion is a common form of NTRK gene fusion in dMMR colorectal cancer.

During rapid urbanization, environmental issues significantly affect urban residents health. Urban parks and green spaces play a crucial role in urban planning and layout, significantly impacting residents quality of life and livability. This study constructs a comprehensive landscape evaluation model, "AHP-TOPSIS-POE" from the perspective of behavioral psychological perception. It uses four urban parks in Jinan City (Qianfoshan Park, Baotu Spring Park, Daming Lake Park, and Quancheng Park) as case study samples. This method validates its feasibility by converting subjective perceptions into objective data. Objective: The global incidence of severe fever with thrombocytopenia syndrome (SFTS) has markedly increased over the past decade. There is an urgent need to establish a reliable scoring system for predicting mortality of SFTS patients., Methods: In this ambispective study, 714 SFTS patients were recruited from 11 sites in China. Among these, 544 patients hospitalized for SFTS from May 2012 to June 2022 were included retrospectively in the training cohort, and 170 were prospectively enrolled between April 2021 and November 2023 in the validation cohort. Logistic regression analysis was performed to identify risk factors for 30-day mortality. A nomogram model (SFTS-logistic model) and a simplified scoring system (SFTS-Wuhan model) were established for predicting mortality. The performance of these models in terms of calibration, discrimination, and clinical utility was evaluated and validated., Results: The 30-day mortality rate was 12.89% (92/714). The mean age was 65 years old (IQR 57-71), and 322 (45.10%) patients were male. The SFTS-logistic model and SFTS-Wuhan model were developed based on seven independent risk factors, including age (AOR=1.062, 95%CI (1.019-1.106)), temperature at admission (AOR=1.599, 95%CI (1.095-2.336)), white blood cell count (AOR=0.799, 95%CI (0.653-0.978)), platelet count (AOR=0.977, 95%CI (0.959-0.996)), aspartate aminotransferase (AOR=1.001, 95%CI (1.000-1.003)), creatinine (AOR=1.006, 95%CI (1.001-1.011)), and vasopressors use (AOR=6.270, 95%CI (1.397-28.146)). Dietary uptake is the main pathway of exposure to polycyclic aromatic hydrocarbons (PAHs). However, there is no data regarding the pollution and health risks posed by PAHs in Lilium davidii var. unicolor. We measured the concentrations of 16 PAHs in lily bulbs from Lanzhou; analyzed the bioaccumulation, sources, and pollution pathways of PAHs; assessed the influence of baking on PAH pollution in the bulb; and assessed the cancer risks associated with PAH exposure via lily consumption. The total PAH concentrations in raw bulbs were 30.39-206.55 μg kg-1. The bioconcentration factors of total PAHs ranged widely from 0.92 to 5.71, with a median value of 2.25. Pearson correlation analysis revealed that the octanol-water partition coefficients and water solubility values played important roles in the bioaccumulation of naphthalene, fluorene, phenanthrene, pyrene, and fluoranthene in the raw bulb by influencing PAH availability in soil. Correlation analysis and principal component analysis with multivariate linear regression indicated that biomass and wood burning, coal combustion, diesel combustion, and petroleum leakage were the major sources of PAHs in the raw bulbs. The paired t-test showed that the PAH concentrations in the baked bulbs were higher than those in the raw bulbs. PAH compositions in lily bulb changed during the baking process. Baked bulbs exhibited a higher cancer risk than raw bulbs. Local adults had low carcinogenic risks from consuming lily bulbs. Photosynthesis has attracted much attention due to its extremely high energy transfer efficiency in the primary light capture stage. Numerical simulations based on energy transfer theory are helpful in exploring the physical mechanisms behind the dynamics of energy transfer. This article starts from the process of photosynthesis and explains the specific contents of two classical energy transfer theories, namely coherent and incoherent theories. Based on these theories, we discuss the numerical simulation methodology of perturbation approximation on the Fenna-Matthews-Olson (FMO) complex model. Finally, we compare the non-perturbation results with the perturbation results and further explore some energy transfer mechanisms. This work has the potential to stimulate ideas on attaining carbon neutrality through a more efficient photosynthesis process.

Cerebral ischemia/reperfusion (I/R) injury is an important pathophysiological condition of ischemic stroke that involves a variety of physiological and pathological cell death pathways, including autophagy, apoptosis, necroptosis, and phagoptosis, among which autophagy is the most studied. We have reviewed studies published in the past 5 years regarding the association between autophagy and cerebral I/R injury. To the best of our knowledge, this is the first review article summarizing potential candidates targeting autophagic pathways in the treatment of I/R injury post ischemic stroke. Background: Recent advancements in artificial intelligence (AI) like ChatGPT have expanded possibilities for patient education, yet its impact on perioperative anxiety in total knee arthroplasty (TKA) patients remains unexplored., Methods: In this single-blind, randomized controlled pilot study from April to July 2023, 60 patients were randomly allocated using sealed envelopes to either ChatGPT-assisted or traditional surgeon-led informed consent groups. In the ChatGPT group, physicians used ChatGPT 4.0 to provide standardized, comprehensive responses to patient queries during the consent process, while maintaining their role in interpreting and contextualizing the information. Outcomes were measured using Hospital Anxiety and Depression Scales (HADS), Perioperative Apprehension Scale-7 (PAS-7), Visual Analogue Scales for Anxiety and Pain (VAS-A, VAS-P), Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC), and satisfaction questionnaires., Results: Of 55 patients completing the study, the ChatGPT group showed significantly lower anxiety scores after informed consent (HADS-A: 10.48 ± 3.84 vs 12.75 ± 4.12, P = .04, Power = .67; PAS-7: 12.44 ± 3.70 vs 14.64 ± 2.11, P = .01, Power = .85; VAS-A: 5.40 ± 1.89 vs 6.71 ± 2.27, P = .02, Power = .75) and on the fifth postoperative day (HADS-A: 8.33 ± 3.20 vs 10.71 ± 3.83, P = .01, Power = .79; VAS-A: 3.41 ± 1.58 vs 4.64 ± 1.70, P = .008, Power = .85). The ChatGPT group also reported higher satisfaction with preoperative education (4.22 ± 0.51 vs 3.43 ± 0.84, P<.001, Power = .99) and overall hospitalization experience (4.11 ± 0.65 vs 3.46 ± 0.69, P = .001, Power = .97). No significant differences were found in depression scores, knee function, or pain levels., Conclusions: ChatGPT-assisted informed consent effectively reduced perioperative anxiety and improved patient satisfaction in TKA patients. Objectives: This study aimed to describe the pulmonary toxicity of cyclin-dependent kinase 4/6 inhibitors (CDK 4/6 inhibitors) (palbociclib, ribociclib, and abemaciclib) in patients being treated for breast cancer using the Food and Drug Administration Adverse Event Reporting System (FAERS) database., Research Design and Methods: Disproportionality analysis was performed to assess pulmonary toxicity associated with CDK 4/6 inhibitors. Clinical characteristics, onset time, sensitivity analysis, subgroup analyses, drug combinations, comorbidities, and co-reported events were performed., Results: Out of 83,505 CDK 4/6 inhibitor-related adverse events (AEs) documented in the FAERS database during the study period, 437 cases of pneumonitis, 555 cases of pulmonary edema, and 181 cases of pulmonary thrombosis related to CDK 4/6 inhibitors were analyzed. Pneumonitis and pulmonary thrombosis had the strongest signal strength in abemaciclib; pulmonary edema had the strongest signal strength in ribociclib. The median latency for pneumonitis, pulmonary edema, and pulmonary thrombosis was 66-173.5 days, 27-131 days, and 68-279 days, respectively. Pulmonary toxicity is statistically significant disproportionality in females as well as in patients over 60 years old., Conclusion: Abemaciclib was most strongly associated with pneumonitis and pulmonary thrombosis. Ribociclib was most strongly associated with pulmonary edema. The correlation with pulmonary toxicity was, in descending order, abemaciclib, ribociclib, and palbociclib.

Pepper ( Capsicum spp.) is an important global vegetable and spice, with fruit color being a key determinant of its commercial quality. However, the regulatory mechanisms underlying pepper fruit color are still not fully understood. This study focuses on the MADS-RIPENING INHIBITOR (MADS-RIN), a MADS-box transcription factor that regulates various aspects of fruit ripening, including pigmentation. We identified CaRIN , a homolog of tomato's SlRIN , whose expression is closely associated with fruit ripening in pepper. Silencing CaRIN through virus-induced gene silencing (VIGS) resulted in increased chlorophyll and chlorophyll a content, reduced carotenoid accumulation, and uneven fruit coloration. Integrative analysis of the RNA-seq and DAP-seq data identified 77 target genes regulated by CaRIN, which was involved in processes such as chlorophyll metabolism and plant hormone signaling. Yeast one-hybrid (Y1H) and dual-luciferase (LUC) assays demonstrated that CaRIN directly bound to the promoter of CaLhcb-P4 , repressing its expression. Downregulation of CaLhcb-P4 in pepper fruits via VIGS accelerated chlorophyll degradation. Additionally, CaRIN indirectly regulated multiple genes associated with chlorophyll and carotenoid metabolism, sugar transport, and cell wall degradation. These findings provide novel insights into the regulatory mechanisms of chlorophyll degradation during pepper fruit ripening, offering a foundation for further research and potential genetic improvement strategies.

Purpose: Hemodialysis (HD) patients commonly experience enhanced oxidative stress and inflammation, potentially increasing the risk of malnutrition. Selenium levels are typically lower in HD individuals. Selenium deficiency has been identified as a risk factor for oxidative stress and inflammatory responses. Therefore, we intended to perform a meta-analysis to assess the impact of selenium supplementation on HD patients., Methods: PubMed, Embase, Cochrane Library, and Web of Science were searched for randomized controlled trials (RCTs) on the effect of selenium supplementation on HD patients until September 31, 2024. The data analysis was conducted using Stata15.0 software., Results: Seven RCTs, with 2080 participants, were finally included, with 1029 in the selenium group and 1051 in the control group. The meta-analysis unraveled no statistically significant impact of selenium supplementation on body mass index (SMD = 0.15, 95% CI (- 0.20, 0.49), triglyceride [(SMD = - 0.02, 95% CI (- 0.44, 0.41)], total cholesterol [(SMD = - 0.18, 95%CI (- 0.41, 0.05)], low-density lipoprotein [(SMD = - 0.37, 95% CI (- 1.04, 0.31)], high-density lipoprotein cholesterol [(SMD = 0.15, 95% CI (- 0.54, 0.84)], C-reactive protein [(SMD = - 0.10, 95% CI (- 0.46, 0.27)], ferritin [(SMD = - 0.02, 95% CI (- 0.29, 0.26)], Hx-C reactive protein [(SMD = 0.00, 95% CI (- 1.02, 1.03)], and hemoglobin [(SMD = 0.01, 95% CI (- 0.57,0.59)]. The misuse of antibiotics has led to frequent detection of antibiotics in the environment and triggered a series of environmental pollution problems. As the upper reaches of the Yangtze and Yellow Rivers, Sichuan Province has a large population, and the safety of its drinking water is of great importance to the local and even downstream areas. In this study, ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was used for targeted screening 41 antibiotics from 4 categories including tetracyclines, sulfonamides, macrolides, and quinolones in 35 water sources in Sichuan Province, China. Results showed that 30 antibiotics existed in the water samples. The total concentration of antibiotics (∑ 30 antibiotics) was 0.03-33.09 ng·L -1 , with sulfonamide antibiotics exhibiting the highest concentration. In terms of different types of water sources, the concentration of ∑ 30 antibiotics showed the trend of in the river type (8.15 ± 10.18 ng·L -1 ) > the lake and reservoir type (2.62 ± 1.60 ng·L -1 ) > the groundwater type (0.33 ± 0.31 ng·L -1 ). Veterinary antibiotics predominated in the groundwater and river type sources, while veterinary-agricultural antibiotics dominated in the lake and reservoir type sources. The order of ∑ 30 antibiotics concentration of in water sources from different economic zones showed that in the South Sichuan Economic Zone > Chengdu Plain Economic Zone > Northeast Sichuan Economic Zone > Panxi Economic Zone > Northwest Sichuan Ecological Demonstration Zone. Cluster analysis showed that antibiotics in water sources mainly originated from human medical treatment, breeding and agricultural use. The risk evaluation indicated that there were no high-risk points in the study area, and most of the points were at the no-risk to low-risk levels. Background: Invasive fungal infections (IFI) represent a significant contributor to mortality among sepsis patients in the Intensive Care Unit (ICU). Early diagnosis of IFI is challenging, and currently, there are no predictive tools for identifying sepsis patients who may develop IFI. Our study aims to develop a predictive scoring system to assess the risk of IFI in patients with sepsis admitted to the ICU., Methods: A retrospective collection of data from a total of 549 patients was conducted. Data-driven, clinically knowledge-driven, and decision tree models were used to identify predictive variables for risk of IFI in ICU patients with sepsis. Demographic data, vital signs, laboratory values, comorbidities, medication use, and clinical outcomes were all collected. The optimal model was selected based on model performance and clinical utility to establish a risk score., Results: Among adult patients with sepsis admitted to the ICU, 127 patients (23.1%) developed IFI. The final data-driven model included four predictive factors, the clinically knowledge-driven model included three predictive factors, and the decision tree model included two. Based on the good performance and clinical utility of the clinically knowledge-driven model, it was chosen as the optimal risk scoring model (C-statistics: 0.79 (95% confidence interval (CI): 0.75-0.83); Hosmer-Lemeshow (H-L) test P = 0.884). The ICU sepsis patient invasive fungal infection risk (AMI) score, created based on the clinically knowledge-driven model, includes mechanical ventilation, application of immunosuppressants, and the types of antibiotics used. The C-statistics for this risk score was 0.79 (95% CI:0.75-0.84) with good calibration (H-L test P = 0.992 and see calibration curve: Fig. 2). Understanding the cellular and genetic mechanisms driving human-specific features of cortical development remains a challenge. We generated a cell-type resolved atlas of transcriptome and chromatin accessibility in the developing macaque and mouse prefrontal cortex (PFC). Comparing with published human data, our findings demonstrate that although the cortex cellular composition is overall conserved across species, progenitor cells show significant evolutionary divergence in cellular properties. Specifically, human neural progenitors exhibit extensive transcriptional rewiring in growth factor and extracellular matrix (ECM) pathways. Expression of the human-specific progenitor marker ITGA2 in the fetal mouse cortex increases the progenitor proliferation and the proportion of upper-layer neurons. These transcriptional divergences are primarily driven by altered activity in the distal regulatory elements. The chromatin regions with human-gained accessibility are enriched with human-specific sequence changes and polymorphisms linked to intelligence and neuropsychiatric disorders. Aims: The aim of this study was to develop the inpatient dietary behaviour score (IDBS) and validate its predictive performance in assessing the risk of hypoglycemia in patients with type 2 diabetes mellitus (T2DM)., Methods: The study included patients with T2DM admitted to the Endocrinology Department of the First Affiliated Hospital of Chongqing Medical University between 1 March 2021 and 1 March 2023. Patients were categorised into hypoglycemia and non-hypoglycemia groups using a paired approach, with hypoglycemia defined as any point-of-care glucose test results ≤ 3.9 mmol/L. Clinical data were collected, and the IDBS was calculated using a self-designed scale with 11 variables. Subsequently, measures for predicting hypoglycemia risk in hospitalised T2DM patients were formulated., Results: A total of 1478 patients were included in the survey, with 739 patients in each of the hypoglycemic and non-hypoglycemic groups after pairing. Baseline differences were observed in education, marital status, job status, DM duration, glycosylated haemoglobin A1c (HbA1c), body mass index (BMI), hypoglycemia history, diabetic peripheral artery disease (DM-PAD), diabetic foot (DF), diabetic kidney disease (DKD), and diabetic retinopathy (DR) between the hypoglycemic and non-hypoglycemic groups. Significant correlations were found between the updated and baseline changes in the IDBS. The predictive efficacy was highest in DKD patients (OR = 1.82, 95% CI: 1.69-1.97, P = 6.75E-53). In multivariate analyses, only BMI, DM duration, diabetic foot and marriage response were included in the regression model. The area under the curve (AUC) and mean squared error (MSE) of the IDBS were 0.77 and 0.00039, respectively. A linear relationship existed between the IDBS and OR values, indicating that the risk of hypoglycemia increased with higher scores., Conclusions: IDBS demonstrated a robust predictive effect on hypoglycemia. The variable data were easily obtainable, and the evaluation and calculation methods were straightforward. In cyanobacteria and most Archaeplastida, Arg regulates its formation via allosteric inhibition of the controlling enzyme, N-acetyl-L-glutamate kinase (NAGK) that requires PII protein to properly sense the feedback inhibitor. Although PII expression has been shown to be reduced in Dunaliella salina compared to other green algae, the potential impact of this protein on DsNAGK activity remains unclear. We here performed coupled enzyme assay and surface plasmon resonance analysis and show that DsNAGK is activated by NAG and inhibited by Arg but is not controlled by DsPII. Moreover, DsPII has likely lost its function as an effective glutamine sensor. Replacement of the C-terminus from DsPII with the C-terminus from Chlamydomonas PII restored sensitivity to glutamine in a recombinant DsPII protein, demonstrating the importance of C-terminal residues close to the Q-loop for PII functions. Background: The challenge of early and rapid diagnosis of periprosthetic joint infection (PJI) remains important. This study aimed to assess the efficacy of dual-energy computed tomography (DECT) iodine maps for diagnosing PJI in total hip arthroplasty., Methods: We prospectively enrolled 68 patients who had postoperative joint pain after hip arthroplasty. All patients underwent preoperative DECT iodine imaging to quantify iodine concentration (IC) in periprosthetic tissues during arterial and venous phases. The diagnostic efficacy of DECT iodine maps was evaluated by constructing receiver operating characteristic curves according to the Musculoskeletal Infection Society criteria., Results: Compared with erythrocyte sedimentation rate (area under the curve [AUC] = 0.837), polymorphonuclear cell percentage (AUC = 0.703), and C-reactive protein (AUC = 0.837), periprosthetic tissue venous-phase IC (AUC = 0.970) and arterial-phase IC (AUC = 0.964) exhibited outstanding discriminative capability between PJI and aseptic failure. Introduction: The internet has become a common space for adolescents to connect with others and find information. However, it remains unclear whether online encounters can serve as new sources of social support for Chinese adolescents. The present study aimed to identify distinct patterns in Chinese adolescents' social support from both offline and online sources and to examine how these patterns relate to individual and family factors and adolescents' mental health conditions., Methods: Data were obtained from a cross-sectional survey of 1031 Chinese adolescents (47.1% female, mean age = 13.67) in Foshan, Guangdong province. Latent profile analysis was performed with Mplus. Associations between the identified classes and auxiliary variables were examined with the "R3STEP" procedure and the DCON command in Mplus., Results: We identified four groups. Three groups showed consistent patterns in terms of receiving social support from offline and online sources, namely, the high support group (24.8%), moderate support group (43.8%), and low support group (15.3%), respectively. The fourth group received a moderate level of offline but low level of online support (16%), named the moderate offline-low online support group. The groups differed in individual and family characteristics. Higher support was associated with higher life satisfaction, lower depression, and social anxiety, but also higher problematic internet use. Overall, the moderate offline-low online support group showed better mental health than the other groups., Conclusions: Our study extends social support research by revealing heterogeneity in adolescents' social support patterns. With the development of optical anti-counterfeiting and the increasing demand for high-level information encryption, multimodal luminescence (MML) materials attract much attention. However, the discovery of these multifunctional materials is very accidental, and the versatile host suitable for developing such materials remains unclear. Here, a grossite-type fast ionic conductor CaGa 4 O 7 , characterized by layered and tunnel structure with excellent defect tolerance, is found to meet the needs of various luminescent processes. Almost all luminescent modes, including down/up-conversion luminescence (DCL/UCL), long persistent luminescence (LPL), mechanoluminescence (ML), and X-ray excited optical luminescence (XEOL), are realized in this single host. Full-spectrum (from violet to near-infrared) photoluminescence and ML as well as multicolor XEOL are achieved by simply changing the doped luminescent center. A series of anti-counterfeiting devices, including the quasi-dynamic display of famous paintings, digital information encryption, and multi-color handwritten signatures, are designed to show the encryption of information in temporal and spatial dimensions. It is of great significance to monitor bisphenol A (BPA) in the environment because of its potential environmental and health risks. However, the detection of trace or ultratrace BPA in complicated environmental samples is challenging due to the relatively low affinity and poor selectivity of existing adsorbents used in sample pretreatment. Herein, we report a high-affinity, low environment-dependent and strong interference-resistant abiotic affinity ligand, a N-methacryloyl-l-lysine-NH 2 (MLys)-based hydrogel copolymer (HP 17) screened from a small focused polymer library engineered by incorporating various combinations and ratios of candidate functional monomers. The selection of these monomers was guided by molecular mechanism between BPA and the ligand-binding pocket of its estrogen receptors. The BPA-HP17 binding is mainly a synergistic effect of π-cation and hydrophobic interactions. The screened HP 17 has high adsorption capacity (349.4 mg/g) for BPA under wide pH (3.0-10.0) and ionic strength (0-150 mM) range. To improve its practicability, a hydrogel copolymer/Fe 3 O 4 composite microspheres (Fe 3 O 4 @HP 17) was synthesized and applied for magnetic solid phase extraction-high-performance liquid chromatography (MSPE-HPLC) analysis of BPA in tap water, lake water and industrial effluents. Synapse has been considered a critical neuronal structure in the procession of Alzheimer's disease (AD), attacked by two pathological molecule aggregates (amyloid-β and phosphorylated tau) in the brain, disturbing synaptic homeostasis before disease manifestation and subsequently causing synaptic degeneration. Recently, evidence has emerged indicating that soluble oligomeric amyloid-β (AβO) and tau exert direct toxicity on synapses, causing synaptic damage. Synaptic degeneration is closely linked to cognitive decline in AD, even in the asymptomatic stages of AD. Therefore, the identification of novel, specific, and sensitive biomarkers involved in synaptic degeneration holds significant promise for early diagnosis of AD, reducing synaptic degeneration and loss, and controlling the progression of AD. Currently, a range of biomarkers in cerebrospinal fluid (CSF), such as synaptosome-associated protein 25 (SNAP-25), synaptotagmin-1, growth-associated protein-43 (GAP-43), and neurogranin (Ng), along with functional brain imaging techniques, can detect variations in synaptic density, offering high sensitivity and specificity for AD diagnosis. However, these methods face challenges, including invasiveness, high cost, and limited accessibility. In contrast, biomarkers found in blood or urine provide a minimally invasive, cost-effective, and more accessible alternative to traditional diagnostic methods. The anti-tumor efficacy of current pharmacotherapy is severely hampered due to the adaptive evolution of tumors, urgently needing effective therapeutic strategies capable of breaking such adaptability. Metabolic reprogramming, as an adaptive survival mechanism, is closely related to therapy resistance of tumors. Colorectal cancer (CRC) cells exhibit a high energy dependency that is sustained by an adaptive metabolic conversion between glucose and glutamine, helping tumor cells to withstand nutrient-deficient microenvironments and various treatments. We discover that transition metal vanadium (V) effectively inhibits glucose metabolism in CRC and synergizes with glutaminase inhibitors (BPTES) to disrupt CRC's energy dependency. Thus, a dual energy metabolism suppression nanosystem (VSi-BP@HA) is engineered by loading BPTES into V-doped hollow mesoporous silica nanoparticles. This nanosystem effectively dampens CRC energy metabolism, eradicating 33% of tumors in mice. Strikingly, the cell biological and preclinical model datasets provide compelling evidence showing that VSi-BP@HA not only reverses CRC cells chemo-resistance but also drastically potentiates anti-PD1 immunotherapy. Therefore, this nanosystem provides not only a promising approach to suppress CRC, but also a potential adjunct tool for enhancing chemotherapy and immunotherapy., (© 2024 The Author(s).
