Aluminum (Al) exposure significantly interferes with the energy supply in astrocytes, which may be a potential mechanism of Al-induced neurotoxicity. This study was designed to explore the mechanisms of Al-induced energy supply impairment in rat C6 astroglioma cell line. Aluminum-maltolate (Al(mal)3) (0.1 mM, 24 h) exposure significantly decreased brain-type creatine kinase (BCK) co-localization with the endoplasmic reticulum (ER) and resulted in mitochondrial dysfunctions, accompanied by a decrease in AMPK phosphorylation. The results of molecular docking showed that Al(mal)3 increased BCK’s hydrophobicity and hindered the localization movement of BCK between subcells·H2O2 co-administration was found to exacerbate mitochondrial dysfunction, Ca2+ dyshomeostasis, and apoptosis. After treated with Al(mal)3, additional oxidative stress contributed to BCK activity inhibition but did not promote a further decrease in AMPK phosphorylation. The activation of p-AMPK by its agonist can partially restore mitochondrial function, BCK activity, and ER-localized-BCK levels in Al(mal)3-treated astrocytes. In summary, Al exposure resulted in a sustained depletion of the mitochondrial and antioxidant systems, which was associated with reduced p-AMPK activity and decreased ER-localized-BCK levels in astrocytes. This study provides a theoretical basis for exploring the mechanisms of neurotoxicity induced by Al exposure.

Cell models play a crucial role in analyzing the mechanical response of cells and quantifying cellular damage incurred during micromanipulation. While traditional models can capture the overall mechanical behavior of cells, they often lack the ability to discern among distinct cellular components. Consequently, by employing dissipative particle dynamics, this study constructed a triangular network-like representation of the cell membrane along with cross-linked cytoskeletal chains. The mechanical properties of both the membrane and cytoskeleton were then analyzed through a series of simulated mechanical tests, validated against real-world experiments. The investigation utilized particle-tracking rheology to monitor changes in the mean square displacements of membrane particles over time, facilitating the analysis of the membrane’s storage and loss moduli. Additionally, the cytoskeletal network’s storage and loss moduli were examined via a double-plate oscillatory shear experiment. The simulation results revealed that both the membrane and cytoskeleton exhibit viscoelastic behavior, as evidenced by the power-law dependency of their storage and loss moduli on frequency. Furthermore, indentation and microinjection simulations were conducted to examine the overall mechanical properties of cells. In the indentation experiments, an increase in the shear modulus of the membrane’s WLCs correlated with a higher Young’s modulus for the entire cell. Regarding the microinjection experiment, augmenting the microinjection speed resulted in reduced deformation of the cell at the point of membrane rupture and a lower percentage of high strain.

We demonstrate a fiber-guided atom interferometer in a far-off-resonant trap (FORT) of 100 μK. The differential light shift (DLS) introduced by the FORT leads to the inhomogeneous dephasing of the tightly trapped atoms inside a hollow-core fiber. The DLS-induced dephasing is greatly suppressed in π / 2 - π - π / 2 Doppler-insensitive interferometry. The spin coherence time is extended to 13.4 ms by optimizing the coupling of the trapping laser beam into a quasi-single-mode hollow-core anti-resonant fiber. The Doppler-sensitive interferometry shows a much shorter coherence time, indicating that the main limits to our fiber-guided atom interferometer are the wide axial velocity distribution and the irregular modes of the Raman laser beams inside the fiber. This work paves the way for portable and miniaturized quantum devices, which have advantages for inertial sensing at arbitrary orientations and in dynamic environments. [ABSTRACT FROM AUTHOR]

Summary: Self‐incompatibility plays a vital role in angiosperms, by preventing inbreeding depression and maintaining genetic diversity within populations. Following polyploidization, many angiosperm species transition from self‐incompatibility to self‐compatibility.Here, we investigated the S‐locus in Brassicaceae and identified distinct origins for the sRNA loci, SMI and SMI2 (SCR Methylation Inducer 1 and 2), within the S‐locus. The SMI loci were found to be widespread in Cruciferae, whereas the SMI2 loci were exclusive to Brassica species.Additionally, we discovered four major S‐haplotypes (BnS‐1, BnS‐6, BnS‐7, and BnS‐1300) in rapeseed. Overexpression of BnSMI‐1 in self‐incompatible Brassica napus ('S‐70S1300S6') resulted in a significant increase in DNA methylation in the promoter regions of BnSCR‐6 and BnSCR‐1300, leading to self‐compatibility. Conversely, by overexpressing a point mutation of BnSmi‐1 in the 'S‐70S1300S6' line, we observed lower levels of DNA methylation in BnSCR‐6 and BnSCR‐1300 promoters. Furthermore, the overexpression of BnSMI2‐1300 in the 'SI‐326S7S6' line inhibited the expression of BnSCR‐7 through transcriptional repression of the Smi2 sRNA from the BnS‐1300 haplotype.Our study demonstrates that the self‐compatibility of rapeseed is determined by S‐locus sRNA‐mediated silencing of SCR after polyploidization, which helps to further breed self‐incompatible or self‐compatible rapeseed lines, thereby facilitating the utilization of heterosis. Brassica napus L. is an important oil crop. Salt stress is one of the major environmental conditions affecting the growth and development of B. napus, which may lead to yield reduction, quality deterioration, and even the death of B. napus. In this study, the B. napus semi-winter cultivar ZS11 was used as the experimental material to perform transcriptome sequencing on the leaf and root tissues with salt stress treatment (0, 0.25, 0.5, 1, 3, 6, 12, and 24 h). The measured 90 RNA-seq data provided the high-resolution time dynamic transcriptional expression spectrum of rapeseed tissues responding to salt stress was obtained. Correlation analysis showed that the samples exhibited significant clustering differences in early response and late response before and after 1 h salt stress treatment. Using DESeq2 for differential gene analysis, we identified 20,462 and 29,334 differential genes for the response of root and leaf tissues, respectively, indicating that the overall response of leaf tissue in rapeseed was more severe than the root. Furthermore, WGCNA was used to construct gene co-expression networks for the salt stress response in root and leaf tissues, respectively, and tan and yellow modules were significantly related to the early response to salt stress, and green and red modules were significantly related to the late response to salt stress. GO enrichment analysis was then performed, and 41 and 26 core transcription factors responding to salt stress at the early and late stages, respectively, were selected from these networks. Functional annotation showed that the known Arabidopsis homologous genes involved in salt stress responses at different stages existed in all four modules, and core genes, such as BnWRKY46 and BnWRKY57, had abundant SNPs variation and haplotypes in 505 salt stress-treated rape population variation data, suggesting that these core transcription factors might be key candidate genes for rape salt stress response. Rapeseed (Brassica napus) is an important oilseed crop worldwide. Plant vascular tissues are responsible for material transport and provide mechanical support. The lateral roots (LRs) absorb sufficient water and nutrients. The genetic basis of vascular tissues and LRs development in rapeseed remains unknown. This study characterized an EMS-mutagenized rapeseed mutant, T16, which showed dwarf stature, reduced LRs, and leaf wilting. Scanning electron microscopy observations showed that the internode-cell shortened. Observations of the tissue sections revealed defects in the development of vascular bundles in the stems and petioles. Genetic analysis revealed that the phenotypes of T16 were controlled by a single semi-dominant nuclear gene. Map-based cloning and genetic complementarity confirmed that BnaA03.IAA13 is the functional gene, a G-to-A mutation in second exon changed the glycine at the 79th position to glutamic acid, disrupting the conserved degron motif VGWPP. Transcriptome analysis in roots and stems showed that auxin and cytokinin signaling pathways were disordered in T16. Evolutionary analysis showed that AUXIN/INDOLE-3-ACETIC ACID was conserved during plant evolution. The heterozygote of T16 significantly reduced the plant height while maintaining other agronomic traits. Our findings provide novel insights into the regulatory mechanisms of vascular tissues and LRs development, and provide a new germplasm resource for rapeseed breeding., (© The Author(s) 2024. We demonstrate the suppression of inhomogeneous dephasing of cold 87 Rb atoms optically trapped inside a hollow-core fiber. The differential light shift (DLS) for the clock transition caused by the trapping beam is reduced by one order of magnitude through the use of a weak compensation laser beam that is spatially mode-matched to the trapping beam. The coherence of the DLS-compensated system is characterized by microwave Ramsey interferometry, which shows Ramsey fringes with a contrast of over 0.6 at a separation time of 10 ms. The dephasing time, measured by Ramsey spectroscopy at different separation times, reaches tens of milliseconds after DLS cancellation, limited by the residual DLS caused by mode mismatching between the two laser beams. This work paves the way for compact and portable fiber-guided atom interferometers.
