Introduction: As the lifespan of companion animals extends, the incidence of tumor also increases. Among these tumors, lymphoma is reported as the most prevalent hematopoietic tumor with a 80-90% prevalence rate. Ongoing research spans multiple domains, aiming to uncover novel therapeutic targets, including small molecular weight inhibitors, antibody treatments, and subtype-specific selective agents. Methods: Transcriptional profiling was performed on canine lymphoma samples to identify genes and functional pathways associated with pathogenesis, treatment response, and prognosis. Additionally, genes with potential relevance to the clinical characteristics of T-cell lymphoma (TCL), which is characterized by a low treatment response and poor prognosis, were identified through a comparative analysis of different lymphoma subtypes. Results: Within the canine lymphoma group, HERC5 showed consistent upregulation, a gene similarly implicated in human acute myeloid leukemia but previously no reports exist. Additionally, noteworthy genes, including IKZF2, CCL4, SAA1, and CD40, exhibited differential expression in the TCL group compared to the B-cell lymphoma (BCL) group. Discussion: The upregulation of HERC5 may impact on canine lymphoma pathogenicity. Furthermore, the upregulation of IKZF2, CCL4, and SAA1, along with the downregulation of CD40, may contribute to adverse clinical characteristics of TCL in dogs. [ABSTRACT FROM AUTHOR]

Hypotrichosis simplex of the scalp (HSS) is a clinically rare monogenic autosomal dominant disorder associated with variants in the gene CDSN , which encodes the desmosome protein corneodesmosin. Although studies have reported that some medications can improve the symptoms of hair loss in HSS, there is still a lack of definitive and effective treatments for this disease. We report a familial case of HSS in an 8-year-old male child diagnosed with HSS caused by a mutation in CDSN , who was treated with botanical extracts in combination with minoxidil, which resulted in significant hair growth after two treatments. This is the first study describing the improvement of clinical symptoms of HSS with oral botanical extracts. This suggests that botanical extracts in combination with minoxidil may be a therapeutic approach for HSS in the clinic. [ABSTRACT FROM AUTHOR]

The surge in firearm sales from the onset of the COVID-19 pandemic have been linked to increases in firearm violence, which is of public concern given that having firearms in one's home is associated with increased risk for domestic violence and suicide. Consistent with pre-pandemic trends, individuals tended to purchase firearms for self-protection during COVID-19. Prior work indicates that protective firearm ownership is motivated not only by perceptions that the world (and one's local environment) is dangerous, but also by one's endorsement of masculinity norms found in U.S. cultures of honor (primarily southern and western states). Honor-based masculinity norms emphasis reputation defense, toughness, and an absolute intolerance of disrespect. The present research examined the relative motivating influences of various threat perceptions and masculine honor endorsement in predicting reasons for non-COVID-19 firearm ownership, firearm purchasing during COVID-19, and purchase intentions. Three separate samples (total N = 2483) of mostly White U.S. men completed online surveys during different months of the first year of the COVID-19 pandemic. Participants completed measures of their endorsement of masculine honor norms, factors associated with firearm purchasing (e.g., dangerous world beliefs, intolerance of uncertainty), and firearm purchasing behaviors. Results indicated that masculine honor endorsement was higher among (1) protective firearm owners compared to non-owners and non-protective owners, (2) firearm owners who purchased a firearm during COVID-19 compared to non-owners and non-purchasing owners, and (3) firearm owners with intentions to purchase firearms in the next year compared to those without intentions and undecided owners. Relative to other predictors (e.g., COVID-19 concerns, dangerous world beliefs), masculine honor endorsement was consistently the strongest predictor of these outcomes. Findings add to the literature by highlighting the strength of masculine honor endorsement in motivating (protective) firearm ownership. Access to firearms among individuals with mental health problems has been a source of protracted debate among policymakers, the media, and the public, writ large. At the center of this controversy are questions about the nature and consequences of gun access in the context of mental illness. The lack of substantial empirical evidence, due in part to limited access to quality data, plays a significant role in perpetuating ongoing debate. To address this problem, this study uses data from the National Comorbidity Survey Replication to evaluate the relative importance of several clinical, cultural, and criminological factors in explaining gun access and carrying among adults with and without mental illnesses. Multivariate analyses reveal that, whereas past year disorder (of any type or severity) and other clinical characteristics were unrelated to firearm access, several cultural factors such as childhood rurality (e.g., ORrural vs. urban: 3.59; 95% CI: 2.52, 5.12) and the criminological experience of early intimate partner violence (e.g., ORvictim only vs. noexp: 1.84; 95% CI: 1.50, 2.26) were. None were predictive of carrying. Further, none of the relationships observed were conditioned on any of the clinical characteristics. These results indicate that people with mental illnesses likely own and carry guns for the same reasons and in the same contexts as others. Additional updated and quality data is needed to further explore these issues; however, these finding suggest that suicide and violence prevention efforts targeting people with mental illnesses need to be sensitive to the cultural and personal significance of guns. Background/Objectives: Dementia is not a single disease but an umbrella term that encompasses a range of symptoms, such as memory loss and cognitive impairments, which are severe enough to disrupt daily life. One of the most common forms of dementia is Alzheimer's Disease (AD), a complex neurodegenerative condition influenced by both genetic and environmental factors. Recent research has highlighted diet as a potential modifiable risk factor for AD. Decades of research have explored the role of dietary patterns, including the Mediterranean Diet (MD) and its components, in neuroprotection and cognitive health. Systematic review examines studies investigating the impact of the Mediterranean Diet, Mediterranean-like diets, the Nordic Diet (ND), dietary intake patterns, and specific components such as extra virgin olive oil and rapeseed oil on cognitive function, disease onset, and progression in AD and dementia. Methods: A comprehensive search of PubMed, the Directory of Open Access Journals, and the Social Science Research Network was conducted independently by two reviewers using predefined search terms. The search period included studies from 2006 to 2024. Eligible studies meeting the inclusion criteria were systematically reviewed, yielding 88 studies: 85 focused on the MD and its relationship to AD and dementia, while only 3 investigated the ND. Results: The findings suggest that adherence to the Mediterranean and Nordic diets is generally associated with improved cognitive function and delayed cognitive decline and that adherence to both these diets can improve cognitive function. Some studies identified that higher legume consumption decreased dementia incidence, while fruits and vegetables, carbohydrates, and eggs lowered dementia prevalence. Most studies demonstrated that high MD or ND adherence was associated with better cognitive function and a lower risk of poor cognition in comparison to individuals with lower MD or ND adherence. However, some studies reported no significant benefits of the MD on cognitive outcomes, while two studies indicated that higher red meat consumption was linked to better cognitive function. Conclusion: Despite promising trends, the evidence remains varying across studies, underscoring the need for further research to establish definitive associations between diet and cognitive function. These findings highlight the essential role of dietary interventions in the prevention and management of dementia and AD, therefore offering critical insights into the underlying mechanisms by which the diet may impact brain health. Deep learning significantly advances object detection. Post processes, a critical component of this process, select valid bounding boxes to represent the true targets during inference and assign boxes and labels to these objects during training to optimize the loss function. However, post processes constitute a substantial portion of the total processing time for a single image. This inefficiency primarily arises from the extensive Intersection over Union (IoU) calculations required between numerous redundant bounding boxes in post processing algorithms. To reduce these redundant IoU calculations, we introduce a classification prioritization strategy during both training and inference post processes. Additionally, post processes involve sorting operations that contribute to their inefficiency. To minimize unnecessary comparisons in Top-K sorting, we have improved the bitonic sorter by developing a hybrid bitonic algorithm. These improvements have effectively accelerated the post processing. Given the similarities between the training and inference post processes, we unify four typical post processing algorithms and design a hardware accelerator based on this framework. Our accelerator achieves at least 7.55 times the speed in inference post processing compared to that of recent accelerators. When compared to the RTX 2080 Ti system, our proposed accelerator offers at least 21.93 times the speed for the training post process and 19.89 times for the inference post process, thereby significantly enhancing the efficiency of loss function minimization. The training effect can be enhanced when trainees/learners interact with real-world environments and construct personal knowledge from those direct experiences. Leveraging such experiential learning strategies for occupational training has been widely discussed due to its effectiveness. The construction industry has also been focusing on experiential safety training to address the limitations in conventional classroom-based training, such as passive learning and limited interaction with actual physical hazards. Recently, government organizations and construction companies have started to operate safety training facilities, where trainees can physically experience the negative consequences of unsafe behaviors (without actual injuries). Although the effect of experiential safety training at those facilities has been anecdotally noted, no study has empirically investigated its effectiveness in enhancing trainees' risk perception toward unsafe behaviors. To this end, this study examined the effectiveness of experiential safety training in enhancing construction managers' risk perception toward workers' unsafe behaviors and their intention to stop workers from working in dangerous situations. The results, based on answers to survey questions showing scene images of unsafe behavior related to the risk of a fall, show that construction managers who participated in experiential safety training perceived a higher risk regarding workers' unsafe behaviors in less obviously risky situations, and exhibited a stronger intention to immediately stop workers from working in subtly unsafe conditions. This study contributes empirical evidence about the effectiveness of experiential safety training at safety training centers, thereby promoting the wide adoption of experiential safety training and advancing safety engineering and management strategies in the construction industry. Objective: Spinopelvic sagittal balance ensures efficient posture and minimizes energy expenditure by aligning the spine, pelvis, and lower extremities. Deviations can cause clinical issues like back pain and functional limitations. Key radiographic parameters, including pelvic tilt (PT), pelvic incidence (PI), sacral slope (SS), and lumbar lordosis (LL), are essential for evaluating spinal pathologies and planning surgeries. Accurate PI measurement is challenging in certain conditions, necessitating alternative parameters. This study aimed to introduce a new, easily measurable parameter and examine its reliability and correlation with established sagittal parameters., Methods: This study analyzed 107 asymptomatic adult volunteers (57 males and 50 females), with an average age of 36.75 years. Whole-spine radiographs in the standing position were taken using EOS technology. The established spinopelvic sagittal parameters and a novel parameter, sacrum1-pubic angle (S1PA), were measured. The correlation coefficient of each parameter, the regression equation of PI using S1PA, and the regression equation of PTα using PTβ were obtained. The intraclass correlation coefficients (ICCs) was calculated to evaluate the measurement reliability., Results: Morphologic (S1PA, PI) and positional parameters (PTα, PTβ, PTγ, SS, LL) showed no significant gender differences (p > 0.05). S1PA had strong correlations with PI (r = -0.883, p < 0.001) and other parameters. PTα demonstrated a strong correlation with PTβ (r = -0.929, p < 0.001). PI could be predicted according to the regression equation: PI = 71.672 - 4.537 × S1PA (R² = 0.779, p < 0.001). The PTα could be predicted using the following equation: PTα = 67.245 - 0.865 × PTβ (R² = 0.864, p < 0.001). Reliability analysis showed high intra- and inter-rater agreement in all the spinopelvic parameters., Conclusion: The S1PA is a dependable parameter for evaluating the morphology and orientation of the pelvis. PI could be precisely predicted using the S1PA. Context: Gun violence negatively impacts not only victims but also their families and surrounding communities. Resources and counseling services may be available to support families affected by gun violence, but the families and their clinicians may not know about these resources or how to access them., Objectives: The objective of this study was to investigate the impact of a clinician-directed educational program on patient reports of their discussions with their physician regarding gun violence, prevention, and available resources for support and treatment., Methods: This quasi-experimental, cross-sectional, survey-based, quality-improvement study included pre-, mid-, and posttraining surveys administered to patients and clinicians participating in an educational program at two urban healthcare centers in Philadelphia. The educational program included office enhancements (handouts and posters) and lunchtime presentations for clinicians regarding gun violence prevalence, prevention strategies, local support resources, and impacts on mental health for patients and their families. The anonymous patient survey was offered to all patients seen at two urban healthcare centers in Philadelphia during three nonconsecutive weeks over 3 months., Results: Among 542 patients seen over the 3 weeks of survey collection, 428 completed the survey (response rate of 79 %). Sixty-four percent acknowledged being impacted by gun violence including the death of a loved one, witnessing a shooting, or being shot themselves. Over the course of the educational program, patients reported significant increases in (1) awareness of materials related to gun violence in the waiting areas, by 17.2 %, (2) discussions of gun violence with their clinician, by 12.1 %, and (3) discussions of methods to prevent gun injury, by 9.7 %. At the end of the study, 19.3 % of patients reported having discussions with their clinician about gun violence, and 14.3 % discussed strategies to prevent gun injury. Participating clinicians reported high levels of satisfaction and increased confidence when talking to patients about gun violence at the end of the program., Conclusions: Providing clinician-directed education and printed materials increased the frequency with which clinicians discussed gun violence, prevention, and available resources with their patients. Increases were modest, with opportunities for improvement. Background: Emergency department (ED) visits may serve as opportunities for firearm injury prevention and intervention efforts. Our objective was to determine ED utilization by youth before and after firearm injury., Methods: We performed a retrospective cohort study of ED encounters by youth (0-18 years old) with firearm injury from eight states using the 2019 State ED and Inpatient Databases. Our primary outcome was an ED encounter (1) 90 days before or (2) 90 days after index injury. We used generalized estimating equations, accounting for hospital clustering, to determine associations between ED utilization and ED type (pediatric vs. general), youth age, sex, race and ethnicity, urbanicity, and insurance status., Results: We identified 1035 ED encounters for firearm injury (median [IQR] age 17 (15-18) years, 85.3% male, 63.3% non-Hispanic Black, 68.6% publicly insured, 90.5% living in a metropolitan area, 52.8% general ED). In the 90 days before an index injury, 12.8% of youth had an ED encounter; of these, 68.2% occurred in general EDs, and 18.2% were for trauma. In the 90 days after an index injury, 22.1% of youth had an ED encounter; of these, 50.0% occurred in general EDs, and 22.6% were for trauma. We found no significant association between ED type and ED utilization patterns. Few youths changed ED type across longitudinal encounters., Conclusions: Youth have high rates of ED utilization before and after firearm injury. Half of firearm-injured youth receive their emergency care exclusively in general EDs. Quantifying the mechanical response of the biological milieu (such as the cell's interior) and complex fluids (such as biomolecular condensates) would enable a better understanding of cellular differentiation and aging and accelerate drug discovery. Here we present time-shared optical tweezer microrheology to determine the frequency- and age-dependent viscoelastic properties of biological materials. Our approach involves splitting a single laser beam into two near-instantaneous time-shared optical traps to carry out simultaneous force and displacement measurements and quantify the mechanical properties ranging from millipascals to kilopascals across five decades of frequency. To create a practical and robust nanorheometer, we leverage both numerical and analytical models to analyse typical deviations from the ideal behaviour and offer solutions to account for these discrepancies. We demonstrate the versatility of the technique by measuring the liquid-solid phase transitions of MEC-2 stomatin and CPEB4 biomolecular condensates, and quantify the complex viscoelastic properties of intracellular compartments of zebrafish progenitor cells. In Caenorhabditis elegans, we uncover how mutations in the nuclear envelope proteins LMN-1 lamin A, EMR-1 emerin and LEM-2 LEMD2, which cause premature aging disorders in humans, soften the cytosol of intestinal cells during organismal age.
