Inflammaging, the chronic, progressive proinflammatory state associated with aging, has been associated with multiple negative health outcomes in humans. The pathophysiology of inflammaging is complex; however, it is often characterized by high serum concentrations of inflammatory mediators such as tumor necrosis factor alpha (TNF-α), interleukin (IL)-6, and C-reactive protein (CRP). Few studies have evaluated the effects of age on inflammatory cytokines in companion dogs, and most of these studies included dogs of a single breed. In this cross-sectional study, we measured multiple circulating inflammatory markers and hematological parameters in banked serum samples from 47 healthy companion dogs of various breeds enrolled in the Dog Aging Project. Using univariate linear models, we investigated the association of each of these markers with age, sex, body weight, and body condition score (BCS), a measure of obesity in the dog. Serum IL-6, IL-8, and TNF-α concentrations were all positively associated with age. Lymphocyte count was negatively associated with age. Platelet count had a negative association with body weight. IL-2, albumin, cholesterol, triglyceride, bilirubin, S100A12, and NMH concentrations were not associated with age, weight, BCS, or sex after adjustment for multiple comparisons. Our findings replicate previous findings in humans, including increases in IL-6 and TNF-α with age, giving more evidence to the strength of the companion dog as a model for human aging. Background: The Dog Aging Project (DAP) is a longitudinal study examining aging in US companion dogs, where missing data pose a challenge to result validity and statistical power. Early recognition and procedural adjustments are vital to address missing samples. Objectives: This research assesses missing biospecimen samples within the PRECISION cohort of the DAP, aiming to identify contributing factors and propose mitigation strategies. Methods: Ninety sample kits collected between August 2021 and October 2021 were evaluated for the missingness of individual kit components. A primary assessment reviewed the missingness of each sample type with hypothesized associated factors. A secondary assessment addressed the missingness of plasma and serum aliquots made after check‐in. Results: EDTA samples for flow cytometry, peripheral blood mononuclear cells (PBMCs), DNA and biobanking were missing at rates of 18.9%, 13.3%, 6.7% and 5.6%, respectively. Faecal samples were missing at 5.6%. Urine and plasma were each missing at 2.2%. Serum, hair and dried blood spot cards were each missing at 1.1%. No statistically significant difference in missingness was seen across regions, ease of blood collection, or cooperativeness of dog during sample collection. Higher time‐to‐receipt was associated with a higher missingness proportion. Increasing missingness proportions were seen across plasma and serum aliquots that were created in sequential order. Conclusion: The study suggests reevaluating sample collection instructions to address frequently missing sample types and prevent further loss. Investigation into collection procedures for these types is recommended to identify potential causes of sample yield loss. [ABSTRACT FROM AUTHOR]

Objective: To quantify serum amikacin concentrations in dogs undergoing wound management with topical amikacin (45 mg/mL) 3% carboxymethylcellulose hydrogel., Study Design: Prospective clinical study., Sample Population: Eleven client-owned dogs., Methods: Dogs with naturally occurring wounds, undergoing treatment with topical amikacin gel, were enrolled. A whole blood sample was collected prior to initial application of the gel. Up to a maximum dose of 30 mg/kg of gel, was applied directly on the wound and the wound was bandaged. Serial blood sampling was performed at approximately 2, 4, 8, 12, 18, 24, 32, 40, 48, 56, 64, and 72 h after application of amikacin gel. The sampling schedule was reset following each bandage change and new application of the gel. Up to 20 samples per dog were collected. The Siemens Syva EMIT Amikacin Assay was used to quantify the concentration of amikacin in each blood serum sample. The lower limit of quantification (LLOQ) of the test was 2.5 μg/mL., Results: Amikacin gel was applied a total of 31 times (dose range, 0.1-24.9 mg/kg). A total of 153 samples were analyzed. Five samples in three different dogs were above the LLOQ at approximately 2 h after gel application (range 2.75-3.82 μg/mL). All other samples were below the LLOQ., Conclusion: Routine use of amikacin gel for open wound management did not result in serum amikacin levels above 5 μg/mL., Clinical Significance: Topical amikacin gel may be a safe treatment option for wounds in dogs with resistant infections or biofilms., (© 2024 The Author(s). The article shows the case for using biosecurity measures to mitigate antimicrobial resistance (AMR) in beef and dairy production through actions of veterinarians and animal caretakers. The impact of biosecurity on the prevention of bacterial infections is discussed, with the goal being to reduce the need for antimicrobial drugs. This is expected to reduce the selection for AMR in pathogens and commensal bacterial organisms in beef and dairy animals.
