PurposeModels to study metastatic disease in rare cancers are needed to advance preclinical therapeutics and to gain insight into disease biology. Osteosarcoma is a rare cancer with a complex genomic landscape in which outcomes for patients with metastatic disease are poor. As osteosarcoma genomes are highly heterogeneous, multiple models are needed to fully elucidate key aspects of disease biology and to recapitulate clinically relevant phenotypes.Experimental designMatched patient samples, patient-derived xenografts (PDX), and PDX-derived cell lines were comprehensively evaluated using whole-genome sequencing and RNA sequencing. The in vivo metastatic phenotype of the PDX-derived cell lines was characterized in both an intravenous and an orthotopic murine model. As a proof-of-concept study, we tested the preclinical effectiveness of a cyclin-dependent kinase inhibitor on the growth of metastatic tumors in an orthotopic amputation model.ResultsPDXs and PDX-derived cell lines largely maintained the expression profiles of the patient from which they were derived despite the emergence of whole-genome duplication in a subset of cell lines. The cell lines were heterogeneous in their metastatic capacity, and heterogeneous tissue tropism was observed in both intravenous and orthotopic models. Single-agent dinaciclib was effective at dramatically reducing the metastatic burden.ConclusionsThe variation in metastasis predilection sites between osteosarcoma PDX-derived cell lines demonstrates their ability to recapitulate the spectrum of the disease observed in patients. We describe here a panel of new osteosarcoma PDX-derived cell lines that we believe will be of wide use to the osteosarcoma research community.

This paper attempts to explore the feasibility of using cholesteric liquid crystals (CLCs) for reflective displays in active matrix (AM) driving. The study focuses on reverse mode polymer-stabilized cholesteric liquid crystals (PSCLCs) to overcome the inherent bistability of CLCs and achieve faster response times for switching between bright and dark states. Key parameters such as cell gap, monomer concentration, polymerization temperature, and different monomer types and liquid crystal hosts are systematically investigated to optimize the electro-optical properties of the liquid crystals for active matrix driving. The research demonstrates that PSCLCs can operate within a 15 V driving voltage range while maintaining a good bright-dark state contrast ratio (>10) and a response time of less than 42 ms. This suggests significant potential for their application in dynamic reflective display technologies.

Abstract Pure organic phosphorescence resonance energy transfer is a research hotspot. Herein, a single-molecule phosphorescence resonance energy transfer system with a large Stokes shift of 367 nm and near-infrared emission is constructed by guest molecule alkyl-bridged methoxy-tetraphenylethylene-phenylpyridines derivative, cucurbit[n]uril (n = 7, 8) and β-cyclodextrin modified hyaluronic acid. The high binding affinity of cucurbituril to guest molecules in various stoichiometric ratios not only regulates the topological morphology of supramolecular assembly but also induces different phosphorescence emissions. Varying from the spherical nanoparticles and nanorods for binary assemblies, three-dimensional nanoplate is obtained by the ternary co-assembly of guest with cucurbit[7]uril/cucurbit[8]uril, accompanying enhanced phosphorescence at 540 nm. Uncommonly, the secondary assembly of β-cyclodextrin modified hyaluronic acid and ternary assembly activates a single intramolecular phosphorescence resonance energy transfer process derived from phenyl pyridines unit to methoxy-tetraphenylethylene function group, enabling a near-infrared delayed fluorescence at 700 nm, which ultimately applied to mitochondrial targeted imaging for cancer cells.

Abstract Comprehensive expression quantitative trait loci studies have been instrumental for understanding tissue-specific gene regulation and pinpointing functional genes for disease-associated loci in a tissue-specific manner. Compared to gene expressions, proteins more directly affect various biological processes, often dysregulated in disease, and are important drug targets. We previously performed and identified tissue-specific protein quantitative trait loci in brain, cerebrospinal fluid, and plasma. We now enhance this work by analyzing more proteins (1,300 versus 1,079) and an almost twofold increase in high quality imputed genetic variants (8.4 million versus 4.4 million) by using TOPMed reference panel. We identified 38 genomic regions associated with 43 proteins in brain, 150 regions associated with 247 proteins in cerebrospinal fluid, and 95 regions associated with 145 proteins in plasma. Compared to our previous study, this study newly identified 12 loci in brain, 30 loci in cerebrospinal fluid, and 22 loci in plasma. Our improved genomic atlas uncovers the genetic control of protein regulation across multiple tissues. These resources are accessible through the Online Neurodegenerative Trait Integrative Multi-Omics Explorer for use by the scientific community.

Background/purpose: Oral submucous fibrosis (OSF) is a precancerous lesion in the oral cavity, commonly results from the Areca nut chewing habit. Arecoline, the main component of Areca nut, is known to stimulate the activation of myofibroblasts, which can lead to abnormal collagen I deposition. Meanwhile, Resveratrol is a non-flavonoid phenolic substance that can be naturally obtained from various berries and foods. Given that resveratrol has significant anti-fibrosis traits in other organs, but little is known about its effect on OSF, this study aimed to investigate the therapeutic impact of resveratrol on OSF and its underlying mechanism. Materials and methods: The cytotoxicity of resveratrol was tested using normal buccal mucosal fibroblasts (BMFs). Myofibroblast phenotypes such as collagen contractile, enhanced migration, and wound healing capacities in dose-dependently resveratrol-treated fBMFs were examined. Results: Current results showed that arecoline induced cell migration and contractile activity in BMFs as well as upregulated the expressions of α-SMA, type I collagen, and ZEB1 markers. Resveratrol intervention, on the other hand, was shown to inhibit arecoline-induced myofibroblast activation and reduce myofibroblast hallmarks and EMT markers. Additionally, resveratrol was also demonstrated to restore the downregulated miR-200a in the arecoline-stimulated cells. Conclusion: In a nutshell, these findings implicate that resveratrol may have an inhibitory influence on arecoline-induced fibrosis via the regulation of miR-200a. Hence, resveratrol may be used as a therapeutic strategy for OSF intervention.

Metal particle and air gap defects on the surface of insulators can induce localized electric field distortions and exacerbate charge accumulation, subsequently triggering surface flashover. In this study, we first enhanced traditional capacitive probes to improve stability and effectively suppress charge leakage. The modified probes achieved a charge leakage time constant exceeding 5600 s, with a leakage amount of only 4.9% over a single data acquisition period (210 s). Based on this improvement, we developed a system for measuring surface charges on insulators, investigating the characteristics and influencing factors of surface charge accumulation under metal micro-particles and air gap defects. The results demonstrate that metal micro-particles adhering to the insulator surface accumulate charges of opposite polarity at different locations under applied voltage. These charges tend to diffuse over time and with increased voltage. The presence of air gap defect accelerates charge accumulation in its vicinity, with charge polarity and location being closely related. When the air gap is near the high-voltage electrode, the insulator surface predominantly accumulates positive charges. Conversely, when the air gap is near the ground electrode, negative charges accumulate at both the high-voltage and ground electrodes, with positive charge accumulating in the central region. Both the amplitude and duration of the applied voltage affect the charge accumulation on the air gap surface. Interestingly, with prolonged voltage application, the surface positive charge density in the air gap decreases. Different defect models exhibit charge inversion phenomena occur as the voltage amplitude increases.

Periodontitis, characterized by inflammation and loss of periodontal tissue, is a significant health complication for individuals with diabetes mellitus (DM). Buildup of advanced glycation end-products (AGEs) in DM poses an increased risk of periodontitis via inflammaging. Ganoderma immunomodulatory protein (GMI) shows promise in suppressing inflammaging by mitigating oxidative stress and inflammation via Nrf2 modulation. However, its specific protective effects are not fully understood. Thus, this study aimed to investigate GMI’s anti-inflammaging properties and its underlying mechanism in diabetic-associated periodontitis (DP). We first simulated DP by culturing human gingival fibroblasts (HGFs) with AGEs and lipopolysaccharides from P. gingivalis (LPS). We then evaluated the impact of GMI on cell proliferation, migration and wound healing. Additionally, we assessed GMI’s effects on the components of inflammaging such as reactive oxygen species (ROS) formation, cellular senescence expression, IL-6 and IL-8 secretions, and NF-κB phosphorylation. Next, we explored whether GMI’s anti-inflammaging effects are mediated through the Nrf2 pathway by evaluating Nrf2 and HO-1, followed by the assessment of IL-6 and IL-8 post-Nrf2 knockdown. Our findings revealed that GMI treatment suppressed ROS production, cell senescence, IL-6 and IL-8 and NF-κB phosphorylation. Furthermore, GMI upregulated Nrf2/HO-1 expression and its protective effects were reversed when Nrf2 was knocked down. In conclusion, GMI exerts its anti-inflammaging effect via the modulation of the Nrf2/NF-κB signaling axis in DP in vitro, highlighting its potential as an effective adjunct treatment for diabetes-related periodontitis.

BackgroundThe blood urea nitrogen-to-creatinine ratio (BUNCR) has been proposed as a potential biomarker for critical illness-induced catabolism. However, its specific relevance and significance in the context of non-traumatic intracranial hemorrhage (NTIH) remains unclear. As such, the primary objective of this study was to determine the role of BUNCR in the prognosis of patients with NTIH.Materials and methodsAll data were sourced from the Medical Information Mart for Intensive Care-IV 2.0 (MIMIC-IV) database. Study outcomes included 30-day and 1-year mortality rates. Univariate and multivariate logistic regression analyses were used to calculate adjusted odds ratio with corresponding 95% confidence interval, and generalized additive model were used to identify both linear and non-linear relationships between BUNCR and mortality rates. A two-piecewise regression model was performed to calculate the saturation effect. Subgroup analyses were performed to evaluate outcome stability in various groups.ResultsA retrospective study of 3,069 patients with NTIH revealed a U-shaped relationship between BUNCR levels and 30-day/1-year mortality. The two-piecewise regression model showed that the inflection points for 30-day and 1-year mortality were 10.455 and 16.25, respectively. On the left side of the inflection point, the 30-day and 1-year mortality rate decreased by 17.7% (OR = 0.823, 95%CI: 0.705–0.960; p = 0.013) and 5.3% (OR = 0.947, 95%CI: 0.899–0.999; p = 0.046), respectively, per 1 unit increment of BUNCR. A coumarin-modified tris[4-(4-pyridyl)phenyl]amine compound was encapsulated in the cavity of cucurbit[8]uril with a 2 : 3 stoichiometric ratio, which not only effectively enhances the guest fluorescence emission with a hypsochromic shift of about 51 nm by macrocycle confinement, but also allows cascade assembly with β-cyclodextrin-grafted hyaluronic acid (HACD) to construct a ternary supramolecular network. Furthermore, doping of sulfo-cyanine 5 into the β-cyclodextrin cavity of HACD through host–guest interaction achieved a high efficiency Förster resonance energy transfer system from the supramolecular network to dyes, which was successfully applied to near-infrared mitochondria-targeted imaging in HeLa cells. Total leaf area per plant is an important measure of the photosynthetic capacity of an individual plant that together with plant density drives the canopy leaf area index, that is, the total leaf area per unit ground area. Because the total number of leaves per plant (or per shoot) varies among conspecifics and among mixed species communities, this variation can affect the total leaf area per plant and per canopy but has been little studied. Previous studies have shown a strong linear relationship between the total leaf area per plant (or per shoot) (AT) and the total number of leaves per plant (or per shoot) (NT) on a log–log scale for several growth forms. However, little is known whether such a scaling relationship also holds true for bamboos, which are a group of Poaceae plants with great ecological and economic importance in tropical, subtropical, and warm temperate regions. To test whether the scaling relationship holds true in bamboos, two dwarf bamboo species (Shibataea chinensis Nakai and Sasaella kongosanensis 'Aureostriatus') with a limited but large number of leaves per culm were examined. For the two species, the leaves from 480 and 500 culms, respectively, were sampled and AT was calculated by summing the areas of individual leaves per culm. Linear regression and correlation analyses reconfirmed that there was a significant log–log linear relationship between AT and NT for each species. For S. chinensis, the exponent of the AT versus NT scaling relationship was greater than unity, whereas that of S. kongosanensis 'Aureostriatus' was smaller than unity. The coefficient of variation in individual leaf area increased with increasing NT for each species. The data reconfirm that there is a strong positive power‐law relationship between AT and NT for each of the two species, which may reflect adaptations of plants in response to intra‐ and inter‐specific competition for light. This study aims to solve the problem of optimal performance parameter selection of the virtual synchronous generator (VSG) control strategy using an improved artificial jellyfish search (IMJS) algorithm. VSG technology helps the inverter to improve the anti-disturbance ability. The VSG control effect depends on the setting of its performance parameter values. The population initialization strategy and dynamic adaptive factor are used to improve the optimization ability of the artificial jellyfish search algorithm. The IMJS algorithm is used to optimize the VSG, and a control strategy for the VSG based on the IMJS (IMJS-VSG) is proposed to improve the control capability. The results show that the active power achieves smooth changes without oscillations under the IMJS-VSG control strategy when the system is running in grid-connected operation. The system voltage drop under the IMJS-VSG strategy is less than 40% of the VSG strategy when the system is in an islanded operation state. [ABSTRACT FROM AUTHOR]
