With the exponential growth in data density and user ends of wireless networks, fronthaul is tasked with supporting aggregate bandwidths exceeding thousands of gigahertz while accommodating high-order modulation formats. However, it must address the bandwidth and noise limitations imposed by optical links and devices in a cost-efficient manner. Here we demonstrate a high-fidelity fronthaul system enabled by self-homodyne digital-analog radio-over-fiber superchannels, using a broadband electro-optic comb and uncoupled multicore fiber. This self-homodyne superchannel architecture not only offers capacity boosting but also supports carrier-recovery-free reception. Our approach achieves a record-breaking 15,000 GHz aggregated wireless bandwidth, corresponding to a 0.879 Pb/s common public radio interface (CPRI) equivalent data rate. Higher-order formats up to 1,048,576 quadrature-amplitude-modulated (QAM) are showcased at a 100 Tb/s class data rate. Furthermore, we employ a packaged on-chip electro-optic comb as the sole optical source to reduce the cost, supporting a data rate of 100.5 Tb/s with the 1024-QAM format. These demonstrations propel fronthaul into the era of Pb/s-level capacity and exhibit the promising potential of integrated-photonics implementation, pushing the boundaries to new heights in terms of capacity, fidelity, and cost. Here the authors propose a self-homodyne fronthaul architecture, utilizing DA-RoF super channels and multicore fiber, paving the way for the Pb/s era in fronthaul transmission, enabling ultra-highspeed Internet access. G‐protein‐coupled receptors (GPCRs) transmit downstream signals predominantly via G‐protein pathways. However, the conformational basis of selective coupling of primary G‐protein remains elusive. Histamine receptors H2R and H3R couple with Gs‐ or Gi‐proteins respectively. Here, three cryo‐EM structures of H2R‐Gs and H3R‐Gi complexes are presented at a global resolution of 2.6‐2.7 Å. These structures reveal the unique binding pose for endogenous histamine in H3R, wherein the amino group interacts with E2065.46 of H3R instead of the conserved D1143.32 of other aminergic receptors. Furthermore, comparative analysis of the H2R‐Gs and H3R‐Gi complexes reveals that the structural geometry of TM5/TM6 determines the primary G‐protein selectivity in histamine receptors. Machine learning (ML)‐based structuromic profiling and functional analysis of class A GPCR–G‐protein complexes illustrate that TM5 length, TM5 tilt, and TM6 outward movement are key determinants of the Gs and Gi/o selectivity among the whole Class A family. Herpes simplex virus type 1 (HSV-1) plays an important role in the field of gene therapy and viral vaccines, especially as an oncolytic virus. However, the mass production of HSV-1 viral vectors remains a challenge in the industry. In this study, a microcarrier-mediated serum-reduced medium culture was used to improve the bioprocess of HSV-1 production and increase HSV-1 yields. The composition of the culture media, which included a basal medium, serum concentration, and glutamine additive, was optimized. The process was successfully conducted in a 1 L bioreactor, and virus production was threefold greater than that of conventional processes with a 10% serum medium. The bead-to-bead transfer process was also developed to further increase scalability. In spinner flasks, the detachment rate increased from 49.4 to 80.6% when combined agitation was performed during digestion; the overall recovery proportion increased from 37.9 to 71.1% after the operational steps were optimized. Specifically, microcarrier loss was reduced during aspiration and transfer, and microcarriers and detached cells were separated with filters. Comparable cell growth was achieved with the baseline process using 2D culture as the inoculum by exchanging the subculture medium. To increase virus production after bead-to-bead transfer, critical parameters, including shear stress during digestion, TrypLE and EDTA concentrations in the subculture, and the CCI, were identified from 47 parameters via correlation analysis and principal component analysis. The optimized bead-to-bead transfer process achieved an average of 90.4% overall recovery and comparable virus production compared to that of the baseline process. This study is the first to report the optimization of HSV-1 production in Vero cells cultured on microcarriers in serum-reduced medium after bead-to-bead transfer. The SPRY domain-containing SOCS box proteins SPSB1, SPSB2, and SPSB4 utilize their SPRY/B30.2 domain to interact with a short region in the N-terminus of inducible nitric oxide synthase (iNOS), and recruit an E3 ubiquitin ligase complex to polyubiquitinate iNOS, resulting in the proteasomal degradation of iNOS. Inhibitors that can disrupt the endogenous SPSB-iNOS interactions could be used to augment cellular NO production, and may have antimicrobial and anticancer activities. We previously reported the rational design of a cyclic peptide inhibitor, cR8, cyclo(RGDINNNV), which bound to SPSB2 with moderate affinity. We, therefore, sought to develop SPSB inhibitors with higher affinity. Here, we show that cyclic peptides cR7, cyclo(RGDINNN), and cR9, cyclo(RGDINNNVE), have ~6.5-fold and ~2-fold, respectively, higher SPSB2-bindng affinities than cR8. We determined high-resolution crystal structures of the SPSB2-cR7 and SPSB2-cR9 complexes, which enabled a good understanding of the structure–activity relationships for these cyclic peptide inhibitors. Moreover, we show that these cyclic peptides displace full-length iNOS from SPSB2, SPSB1, and SPSB4, and that their inhibitory potencies correlate well with their SPSB2-binding affinities. Background: Mediation analysis is a powerful tool to identify factors mediating the causal pathway of exposure to health outcomes. Mediation analysis has been extended to study a large number of potential mediators in high-dimensional data settings. The presence of confounding in observational studies is inevitable. Hence, it's an essential part of high-dimensional mediation analysis (HDMA) to adjust for the potential confounders. Although the propensity score (PS) related method such as propensity score regression adjustment (PSR) and inverse probability weighting (IPW) has been proposed to tackle this problem, the characteristics with extreme propensity score distribution of the PS-based method would result in the biased estimation. Methods: In this article, we integrated the overlapping weighting (OW) technique into HDMA workflow and proposed a concise and powerful high-dimensional mediation analysis procedure consisting of OW confounding adjustment, sure independence screening (SIS), de-biased Lasso penalization, and joint-significance testing underlying the mixture null distribution. We compared the proposed method with the existing method consisting of PS-based confounding adjustment, SIS, minimax concave penalty (MCP) variable selection, and classical joint-significance testing. Results: Simulation studies demonstrate the proposed procedure has the best performance in mediator selection and estimation. The proposed procedure yielded the highest true positive rate, acceptable false discovery proportion level, and lower mean square error. In the empirical study based on the GSE117859 dataset in the Gene Expression Omnibus database using the proposed method, we found that smoking history may lead to the estimated natural killer (NK) cell level reduction through the mediation effect of some methylation markers, mainly including methylation sites cg13917614 in CNP gene and cg16893868 in LILRA2 gene. Conclusions: The proposed method has higher power, sufficient false discovery rate control, and precise mediation effect estimation. Guided waves-based structural health monitoring (SHM) methods have potential for practical applications, since they are sensitive to small damages and are able to realize large area monitoring. Among these methods, the Reconstruction Algorithm for Probabilistic Inspection (RAPID), using a Piezoelectric transducer (PZT) sensor array, is one of the most widely used imaging algorithms to perform active damage monitoring and localization. However, since the sensing paths are distributed inside the sensor array with the non-uniform density, the RAPID algorithm can only localize damage when it is occurring inside of the array. If the damage occurs outside of the array or both inside and outside of the array, that is, multi-type damage, the performance of RAPID algorithm would not be satisfactory. In this paper, a scattering coefficient-based RAPID algorithm with damage indexes separation and imaging fusion is proposed. The amplitude of damage scattered signal at the corresponding time of fight is adopted as the weight in the probability distribution function, and damage indexes are then classified into two categories in the RAPID algorithm for the inside and outside damage localization respectively. Finally, an experiment on the complex composite plate, with the center large hole and surrounding bolt holes, is carried out to verify this proposed method. Secondary organic aerosols (SOAs) can exist in liquid, semi-solid, or amorphous solid states. Chemical transport models (CTMs), however, usually assume that SOA particles are homogeneous and well-mixed liquids, with rapid establishment of gas–particle equilibrium for simulations of SOA formation and partitioning. Missing the information of SOA phase state and viscosity in CTMs impedes accurate representation of SOA formation and evolution, affecting the predictions of aerosol effects on air quality and climate. We have previously developed a parameterization to estimate the glass transition temperature (Tg) of an organic compound based on volatility and to predict viscosity of SOA. In this study, we apply this method to predict the phase state of SOA particles over China in summer of 2018 using the Weather Research and Forecasting model coupled with Chemistry (WRF-Chem). The simulated Tg of dry SOA (Tg,org) agrees well with the value estimated from ambient volatility measurements at an urban site in Beijing. For the spatial distributions of Tg,org , simulations show that at the surface the values of Tg,org range from ∼287 to 305 K, with higher values in northwestern China, where SOA particles have larger mass fractions of low-volatility compounds. Considering water uptake by SOA particles, the SOA viscosity shows a prominent geospatial gradient in which highly viscous or solid SOA particles are mainly predicted in northwestern China. The lowest and highest SOA viscosity values both occur over the Qinghai–Tibet Plateau, where the solid phase state is predicted over dry and high-altitude areas and the liquid phase state is predicted mainly in the south of the plateau with high relative humidity during the summer monsoon season. Sensitivity simulations show that, including the formation of extremely low-volatility organic compounds, the percent time that a SOA particle is in the liquid phase state decreases by up to 12 % in southeastern China during the simulated period. With an assumption that the organic and inorganic compounds are internally mixed in one phase, we show that the water absorbed by inorganic species can significantly lower the simulated viscosity over southeastern China. This indicates that constraining the uncertainties in simulated SOA volatility distributions and the mixing state of the organic and inorganic compounds would improve prediction of viscosity in multicomponent particles in southeastern China. We also calculate the characteristic mixing timescale of organic molecules in 200 m SOA particles to evaluate kinetic limitations in SOA partitioning. Calculations show that during the simulated period the percent time of the mixing timescale longer than 1 h is >70 % at the surface and at 500 hPa in most areas of northern China, indicating that kinetic partitioning considering the bulk diffusion in viscous particles may be required for more accurate prediction of SOA mass concentrations and size distributions over these areas. To quantify the volatility of organic aerosols (OA), a comprehensive campaign was conducted in the Chinese megacity. Volatility distributions of OA and particle‐phase organic nitrate (pON) were estimated based on five methods: (a) empirical method and (b) kinetic model based on the measurement of a thermodenuder (TD) coupled with an aerosol mass spectrometer; (c) Formula‐based SIMPOL model‐driven method; (d) Element‐based estimations using molecular formula measurements of OA; and (e) gas/particle partitioning. Our results demonstrate that the ambient OA volatility distribution shows good agreement between the two heating methods and the formula‐based method when assuming ambient OA was mainly composed of organic nitrate (pON), organic sulfate and acid groups using the SIMPOL model. However, the element‐based method tends to overestimate the volatility of OA compared to the above three methods, suggesting large uncertainties in the parameterizations or in the representativeness of the molecular measurements that need further refinement. The volatility of ambient OA is generally lower than that of the laboratory‐derived secondary OA, emphasizing the impact of aging. A large fraction at the higher and lower volatility ranges (approximately log C* ≤ −9 and ≥2 μg m−3) was found for pON, implying the importance of both extremely low volatile and semi‐volatile species. Overall, this study evaluates different methods for volatility estimation and gives new insight into the volatility of OA and pON in urban areas. Plain Language Summary: Volatility, which controls the gas/particle partitioning of organic compounds, is one of most the important physiochemistry properties of organic aerosols (OA). Multiple methods have been used to estimate OA volatility, whereas the accuracy of each method is still unclear. The purpose of this study is to verify the volatility of ambient OA and its key component (i.e., particulate organic nitrate, pON) in urban areas by comparing the estimated results using different methods. With the help of state‐of‐art mass spectrometers, the estimation methods, including two heating methods, one formula‐based method, one element‐based method, and one gas/particle partitioning method, were achieved. In general, we found the heating methods show good agreement with the formula‐based method with a reasonable assumption of functional groups contained in OA and pON. In general, we observed a large faction of extremely low volatile compounds exist in OA (30%–40%), and both extremely low volatile and semi‐volatile species exist in pON, suggesting the complex evolution of atmospheric particles. The results provide a better understanding of the volatility of OA and pON in urban areas and benefit accurate volatility estimation that facilitates the model simulations of OA. Oral squamous cell carcinoma (OSCC) is a prevalent malignancy of the head and neck with rising global incidence. Despite advances in treatment modalities, OSCC prognosis remains diverse due to the complex molecular and cellular heterogeneity within tumours, as well as the heterogeneity in tumour microenvironment (TME). In this study, we utilized single‐cell RNA sequencing (scRNA‐seq) analysis to explore distinct subpopulations of tumour cells in OSCC tissues and their interaction with components in TME. We identified four major tumour cell subpopulations (C0, C1, C2 and C3) with unique molecular characteristics and functional features. Pathway enrichment analysis revealed that C0 primarily expressed genes involved in extracellular matrix interactions and C1 showed higher proliferation levels, suggesting that the two cell subpopulations exhibited tumour aggressiveness. Conversely, C2 and C3 displayed features associated with keratinization and cornified envelope formation. Accordingly, C0 and C1 subpopulations were associated with shorter overall and disease‐free survival times, while C2 and C3 were weakly correlated with longer survival. Genomic analysis showed that C1 demonstrated a positive correlation with tumour mutation burden. Furthermore, C0 exhibited resistant to cisplatin treatment, while C1 showed more sensitive to cisplatin treatment, indicating that C0 might exhibit more aggressive compared to C1. Additionally, C0 had a higher level of communication with fibroblasts and endothelial cells in TME via integrin‐MAPK signalling, suggesting that the function of C0 was maintained by that pathway. In summary, this study provided critical insights into the molecular and cellular heterogeneity of OSCC, with potential implications for prognosis prediction and personalized therapeutic approaches.
