Near-infrared (NIR) hyperspectral imaging enables rapid, non-contact imaging of hazardous materials in a non-destructive manner, allowing for analysis based on spectral reflection information. However, using traditional methods, it is challenging to identify hazardous materials with less distinct spectral reflection features. This study utilizes a self-built NIR hyperspectral imaging system and proposes a new approach. Using a convolutional neural network (CNN), This allows for the rapid completion of high-throughput spectral screening, marking suspicious spectra at spatial points. we sophisticatedly classified six hazardous material types, generating impactful warning images. The optimized CNN demonstrated superior performance (accuracy 91.08 %, recall 91.15 %, specificity 91.62 %, precision 90.17 %, and 0.924 F1 score) compared to SVM and KNN methods. Our study included multitask validation tests, revealing a sensitive detection of 10 mg/cm 2 for ammonium nitrate and trinitrotoluene, capable of identifying over 100 targets simultaneously. By simulating real-world scenarios, we successfully detected hazardous chemicals scattered on the ground and accurately identified these hazardous materials in glass and thin plastic products. Even in situations where clothing obstructed the view, we could still correctly identify hazardous chemicals and generate corresponding warning images. Our system demonstrated precise identification capabilities even amidst complex backgrounds. This method provides an accurate and rapid solution for identifying and locating hazardous chemicals, laying a strong foundation for the next steps in non-contact, long-distance quantitative determination of chemical concentrations. The CO2-assisted oxidative dehydrogenation of light alkanes offers a promising route for converting underutilized resources into valuable chemical feedstocks while addressing environmental challenges associated with CO2 emissions. CO2 plays a dual role in ODH by acting as a mild oxidant that enhances product selectivity and catalyst stability while preventing carbon deposition through RWGS and Boudouard reactions. The review has elucidated a variety of catalyst design and optimization strategies that may guide the future development of novel CO2-assisted ODH catalysts with improved alkane conversion, superior alkene selectivity, and long-term stability. It provides a comprehensive analysis of the structural characteristics, catalytic performances, and reaction mechanisms of typical catalysts. Special attention is given to the structure-performance relationship of these catalysts, emphasizing how changes in promoters, supports, and morphology affect critical properties such as redox behavior, acidity-basicity balance, dispersion of active components, and catalyst-support interactions. Finally, future research directions and perspectives for the CO2-assisted ODH of ethane and propane are proposed, with a focus on advancing catalyst design and optimization strategies. Global climate change has led to increased ultraviolet-B (UVB) radiation, which is now a major global environmental issue that poses significant threats to ecosystems. Zostera japonica, a higher plant inhabiting the intertidal zone, is particularly impacted by this increase in UVB radiation. Furthermore, its unique evolutionary history has endowed Z. japonica with distinct characteristics that differentiate its response to environmental changes from those of other marine and terrestrial plants. However, the specific response mechanisms of Z. japonica to elevated levels of UVB radiation are not fully understood. Therefore, we investigated the response mechanism of Z. japonica to varying doses of increased UVB radiation by integrating the physiological responses with transcriptomic and metabolomic analyses. The results revealed that excessive reactive oxygen species (ROS) were key substances induced by increased UVB radiation and that photosynthesis was significantly inhibited. To mitigate these adverse effects, the expression levels of genes related to antioxidants and UV-absorbing compounds, including flavonoids (PAL, 4CL, CHS, CHI, F3H) and antioxidant enzymes (DHAR, MDHAR, APX), were upregulated. The increased synthesis of these compounds serves to scavenge accumulated ROS and absorb UV radiation. Notably, flavonoids are the preferred compounds synthesized by Z. japonica, in contrast to other plant species. In addition, the weighted gene coexpression network analysis (WGCNA) method was employed to identify genes that may play important roles in the response of Z. japonica to increased UVB radiation. Notable anthropogenic heat sources such as coal-fired plants can alter the atmospheric boundary layer structure and the pollutant dispersion, thereby affecting the local environment and microclimate. Herein, in situ measurements inside a coal-fired steel plant were performed by multiple advanced lidars from 21 May to 21 June of 2021 in Yuncheng, Shanxi Province, China. Comparing with an adjacent meteorological site, we found a prominent nighttime dry heat island overhead of the factory, which was 3-10 °C hotter and 30%-60% drier than the surrounding fields. Large-eddy simulations constrained by the measured thermal contrast suggested that the heat-island-induced circulation could upward transport factory-discharged pollutants and horizontally spread them below the residual layer top, forming a mushroom-shaped cloud. The shape, size, and pollutant loading of the cloud were highly determined by thermodynamic variables such as aerodynamic wind and anthropogenic heat flux. Furthermore, these retained residual-layer pollutants can be convected downward to the ground after sunrise through the fumigation effect, causing the peaking phenomena aboveground. These peaks were statistically evidenced to be common in major urban agglomerations in China. The study provides a new insight regarding the origins of residual-layer pollutants and highlights the needs for programming representations of coal-fired heat emissions in mesoscale air-quality models.

Milk is a rich source of probiotics, particularly lactic acid bacteria (LAB), which have been shown to promote gut health, support the immune system, enhance digestion, and prevent pathogen colonization. This study aimed to isolate and identify LAB strains from giant panda (Ailuropoda melanoleuca) milk, evaluate their probiotic properties, and analyze the genomic characteristics of a promising strain. Thirteen LAB strains were isolated from 12 samples of giant panda milk. Among all LAB strains, Lactiplantibacillus plantarum PM8 (PM8) demonstrated probiotic properties and safety features. It exhibited strong growth performance, high antipathogenic activity against four pathogens, and strong survival rates under simulated gastrointestinal conditions. PM8 also showed excellent adhesion capabilities to Caco-2 cells. Additionally, safety assessment revealed no hemolysin production and minimal antibiotic resistance, making it a promising candidate for probiotic applications. The genome of PM8 consists of 3,227,035 bp with a GC content of 44.60% and contains 3171 coding sequences, including 113 carbohydrate-active enzyme genes and genes related to exopolysaccharides synthesis, vitamin B biosynthesis, adhesion, antioxidant activity, and bile salt hydrolysis. Notably, it contains genes involved in nonribosomally synthesized secondary metabolite and bacteriocin production. The genomic safety analysis confirmed that PM8 lacks the capacity to transmit bacterial antimicrobial resistance and is non-pathogenic to both humans and animals. Background: The effect of statins use on the incidence of Alzheimer's disease (AD) is still under debate, and it could be modified by a series of factors., Objectives: We aimed to examine the association of statins use with the risk of cognitive impairment and AD, and assess the moderating roles of genetic susceptibility and other individual-related factors., Design: A longitudinal study was conducted from the UK Biobank where individuals completed baseline surveys (2006-2010) and were followed (mean follow-up period: 9 years)., Setting: A population-based study., Participants: A total of 371,019 dementia-free participants (mean age 56.4 years; 53.6% female)., Measurements: The effects of statins use on cognitive performance and incident AD were examined by using linear regression model and Cox proportional hazards regression model, respectively. We further evaluated the moderating roles of genetic risks and individual-related factors on both multiplicative and additive scales., Results: The findings showed statins use was associated with an increased risk of AD development [hazard ratio (HR) 1.19 (95% CI: 1.08, 1.30)] compared with no use of statins. We further found significant negative additive interactions of statins use with APOE ε4 allele. Sanguinarine constitutes the main components of Macleaya cordata, and exhibits diverse biological and pharmacological activities. This study investigated the effects of sanguinarine chloride hydrate (SGCH) on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) mice. Five groups were designed to investigate the effects of SGCH on the pathological symptoms, the mRNA expression levels of inflammatory cytokines, colonic mucosal barrier damage, microbiota composition, and SCFAs metabolism in UC mice. The administration of SGCH in DSS-induced UC mice resulted in the amelioration of pathological symptoms, as evidenced by an increase in body weight, a decrease in disease activity index score, elongation of colon length, reduction in spleen index, and improvement in colon injury. SGCH can regulate the expression of inflammatory cytokines (IL-6, TNF-α, IL-1β and IL-10) and tight junction proteins (ZO-1 and Occludin) associated with UC. SGCH exhibited a significant decrease in NF-κB P65 mRNA expression levels, accompanied by a significantly reduced protein level of NF-κB P-P65/P65. Further studies revealed SGCH effectively reversed the decrease in intestinal microbiota diversity induced by UC, thereby promoting the growth of beneficial bacteria such as Akkermansia, Alistipes, and norank_o_Clostridia_UCG-014. Correlation analysis demonstrated a positive association between butanoic acid, propanoic acid, isobutyric acid, isovaleric acid, valeric acid, hexanoic acid with Colidextribacter, while Coriobacteriaceae_UCG-002 exhibited a negative correlation with butanoic acid, acetic acid and propanoic acid. The lipid accumulation associated with diabetes causes continuous liver and kidney damage. Laminaria japonica polysaccharide (fucoidan) has been shown to regulate the disorder in lipid metabolism caused by diabetes. Herein, we established a diabetes mellitus (DM) rat model through a high-fat and high-sugar diet combined with streptozotocin. An automatic biochemical analyzer was used to detect serum lipid content, and hematoxylin and eosin, Masson, periodic acid-silver-methenamine, and Oil Red O staining were used to observe changes in the structure of the kidney and liver, including fibrosis and lipid accumulation. We confirmed that fucoidan could ameliorate renal injury, lipid metabolism, and oxidative stress in streptozotocin-induced diabetic rat models. Metabolomics analysis demonstrated that amino acid metabolism is an important process. We further demonstrated a novel role of fucoidan in regulating kidney and liver lipid metabolism through the aryl hydrocarbon receptor (AhR)-mediated CD36 signaling pathway. Similar results were found in DM rats treated with an AhR inhibitor, as well as in those treated with a combination of both an AhR inhibitor and fucoidan. Importantly, we observed a higher expression of AhR/CD36 in the kidneys and liver of rats with DM, and the level of AhR/CD36 correlated with lipid accumulation and kidney function, suggesting that AhR/CD36 signaling could be a promising therapeutic target for fucoidan in treating lipid metabolism in DM. PRACTICAL APPLICATION: As the main component of Laminaria japonica, fucoidan has excellent antioxidant properties and protective effects against liver and kidney damage in diabetes mellitus. The constrained enzymatic activity and aggregation challenges encountered of nanozymes pose an obstacle to their practical utility, necessitating a strategy to alleviate this problem and improve the enzymatic catalytic efficiency. Herein, MOF-confined ultrafine ZnIrO x clusters (ZnIrO x /ZnIrMOFs) nanozyme have been synthesized by controlling the in-situ growth of ZnIrO x via a one-pot method. Notably, due to the domain-limiting effect of MOF, the ZnIrO x clusters with small size grew uniformly in the structure of ZnIrMOFs nanosheets, while the presence of Zn atoms improved the crystallinity of the MOF phase and the dispersion of ZnIrO x clusters. Moreover, the obtained ZnIrO x /ZnIrMOFs exhibited excellent oxidase- and peroxidase-like activities and better substrate affinity, which can directly and effectively catalyze chromogenic reaction to produce corresponding color and signal changes. Adopting this strategy, we established a visual, sensitive, and selective colorimetric method for glucose detection with a linear range and detection limit of 2.66-319 μM and 1.9 μM, respectively. The Vpx protein encoded by HIV-2/simian immunodeficiency virus (SIV) can antagonize the restriction of the host intrinsic restriction factor, SAMHD1, in nondividing cells by promoting its polyubiquitination and subsequent degradation, thereby facilitating viral replication and immune evasion. However, the role of deubiquitinating enzymes (DUBs) in the dynamics of virus and host remains poorly understood. Here, we demonstrate that DUB USP37 significantly reverses the Vpx-mediated degradation of SAMHD1 in various HIV-2/SIV subtypes by interacting with SAMHD1 and removing its ubiquitin chains. Notably, USP37 deubiquitinates SAMHD1 by directly recognizing SAMHD1 rather than by targeting the E3 ubiquitin ligase. The deubiquitinase activity of USP37 and its ubiquitin interacting motifs are essential for the deubiquitination of SAMHD1, whereas the phosphorylation state of USP37 does not influence its activity. Additionally, USP37 enhances the suppression of the retrotransposition of LINE-1 elements by SAMHD1 via stabilizing SAMHD1. Our findings provide important evidence that enhancing the deubiquitinating activity of some DUBs results in the stability of the host restriction factor and might be a viable strategy against HIV/SIV infections.IMPORTANCESAMHD1 is a multifunctional protein, including restricting virus replication, maintaining genomic integrity through DNA repair, modulating the immune response by influencing the production of type I interferons and other cytokines, and affecting cancer cell proliferation and sensitivity to chemotherapy. However, HIV-2/simian immunodeficiency virus (SIV)-encoded Vpx and the host E3 ligase TRIM21 can induce the degradation of SAMHD1 via the ubiquitin-proteasome pathway. Therefore, it is necessary to find the strategy to stabilize SAMHD1. Our study demonstrates that the deubiquitinase USP37 reverses Vpx- and TRIM21-mediated degradation of SAMHD1, thereby inhibiting SIV replication and LINE-1 activity by stabilizing SAMHD1. Accurately and swiftly segmenting breast tumors is significant for cancer diagnosis and treatment. Ultrasound imaging stands as one of the widely employed methods in clinical practice. However, due to challenges such as low contrast, blurred boundaries, and prevalent shadows in ultrasound images, tumor segmentation remains a daunting task. In this study, we propose BCT-Net, a network amalgamating CNN and transformer components for breast tumor segmentation. BCT-Net integrates a dual-level attention mechanism to capture more features and redefines the skip connection module. We introduce the utilization of a classification task as an auxiliary task to impart additional semantic information to the segmentation network, employing supervised contrastive learning. A hybrid objective loss function is proposed, which combines pixel-wise cross-entropy, binary cross-entropy, and supervised contrastive learning loss. Experimental results demonstrate that BCT-Net achieves high precision, with Pre and DSC indices of 86.12% and 88.70%, respectively. Two-dimensional (2D) materials with spontaneous polarization can exhibit large second-order nonlinear optical (NLO) effects. Here, we present a series of stable distorted MX 2 Y monolayers by using first-principles calculations and lattice vibration analysis. The structural distortion leads to a lower polar symmetry, giving rise to intrinsic ferroelectricity with a Curie point up to room temperature. We show that the bulk photovoltaic effect (BPVE) in polar MX 2 Y monolayers has an enhanced shift current, an order of magnitude larger than that in the undistorted nonpolar counterparts. Meanwhile, the second harmonic generation (SHG) susceptibility reaches up to the order of 10 6 pm 2 /V, superior to that of 2D conventional materials such as MoS 2 , h-BN, and GeS. Our study advances the research in 2D ferroelectric materials and would stimulate more efforts in developing optoelectronic devices based on NLO effects.

Dexamethasone sodium phosphate (DSP) and betamethasone sodium phosphate (BSP) imprinted hydrogels embedded with two-dimensional photonic crystals (2DPC) were developed as hormones-sensitive photonic hydrogel sensors with highly sensitive, selective, anti-interference and reproducible recognition capability. The DSP/BSP molecularly imprinted photonic hydrogels (denoted as DSP-MIPH and BSP-MIPH) can specifically recognize DSP/BSP by rebinding the DSP/BET molecules to nanocavities in the hydrogel network. This recognition is enabled by the similar shape, size, and binding sites of the nanocavities to the target molecules. The rebinding of hormones molecules causes the hydrogel to swell, resulting in a particle spacing increase of the embedded 2DPC of the hydrogel. The particle spacing increase can be used as sensing signal and can be acquired by simply measuring the Debye diffraction diameters of the photonic hydrogel sensor before and after exposure with a laser pointer and a ruler. The particle spacing increments of the DSP-MIPH and BSP-MIPH sensors linearly change when DSP and BSP concentrations changes within the ranges 0.025-1 μM, 10-100 μM, and 100-500 μM, and the limits of detection (LoD) are 21.8 nM for DSP and 12.6 nM for BSP, respectively. These photonic hydrogel sensors were successfully applied to the determination of DSP/BSP contents in the real pharmaceutical injections, providing an ideal strategy for the development of portable hormones sensors., Competing Interests: Declarations. Chikungunya virus (CHIKV) is an arbovirus that can lead to chronic arthritis and significantly diminish the quality of life of patients. Given the expanding global prevalence of CHIKV and the absence of specific antiviral therapies, there is an urgent need to explore effective treatment options. This study aimed to evaluate the antiviral effects of shikimic acid (SA) against CHIKV through a combination of network pharmacology, molecular docking, and in vitro assays. Network pharmacology analysis identified 26 potential targets through which SA could inhibit CHIKV, including key pathogenic targets such as TNF, IL-6, and MAPK3. This hypothesis was further supported by molecular docking. The molecular docking analysis revealed that SA could interact with multiple CHIKV-related targets, including EGF, with vina scores generally lower than -6, indicating a high propensity for stable complex formation. The results also suggested that SA could potentially disrupt the IL-17 signaling pathway by engaging with various targets to form complexes. In vitro experiments confirmed that SA significantly enhanced the viability of 293T and BHK-21 cells infected with CHIKV by ~25% and reduced viral load by over 20% at concentrations ranging from 1,000 to 31.25 μM. Additionally, SA was found to markedly downregulate the expression of CHIKV-related attachment factors ACTG1, TSPAN9, and TIM-1 in 293T cells infected with CHIKV. Furthermore, RT-qPCR analysis demonstrated that SA effectively decreased the expression of NFKB1, PTGS2, RELA, and EGF related to the IL-17 signaling pathway. Background: PD-1 inhibitors have shown promising efficacy in enhancing OS and AEs as second-line therapies for patients with advanced esophageal squamous cell carcinoma (ESCC). However, there remains no clear consensus on which PD-1 inhibitor provides the best balance between efficacy and safety. To address this key issue in the second-line treatment of ESCC, we conducted a network meta-analysis (NMA) with a focus on OS benefits, particularly in patients with different levels of PD-L1 expression., Methods: A systematic search of relevant literature was conducted in Web of Science, Embase, PubMed, and Cochrane Library, covering publications from the inception of these database to June 2024. The evaluated endpoints included OS, progression-free survival (PFS), objective response rate (ORR), AEs, and Grade ≥ 3 adverse events (Grade ≥ 3 AEs). A systematic review and Bayesian network meta-analysis were performed to assess the efficacy and safety of various immunotherapy regimens in patients with advanced ESCC. To ensure transparency, novelty, and reliability, this study was prospectively registered in the systematic review registry (CRD42024540581)., Results: Five randomized controlled trials (RCTs), encompassing 2,078 patients and six treatment regimens, were included in this study. Among advanced ESCC patients not selected based on PD-L1 expression, Sintilimab demonstrated the greatest OS benefit (HR = 0.70, 95% CI: 0.50-0.98). Camrelizumab showed the most favorable improvement in PFS compared to chemotherapy (HR = 0.64, 95% CI: 0.47-0.87) and also achieved the best ORR benefit (OR = 3.72, 95% CI: 1.98-6.99). In terms of safety, Nivolumab (OR = 0.10, 95% CI: 0.05-0.19) and Tislelizumab (OR = 0.18, 95% CI: 0.10-0.33) exhibited significant safety advantages over chemotherapy concerning AEs. Moreover, Nivolumab (OR = 0.13, 95% CI: 0.08-0.20) was associated with a markedly lower risk of Grade ≥ 3 AEs compared to chemotherapy. Subgroup analysis based on PD-L1 expression revealed that Tislelizumab (HR = 0.53, 95% CI: 0.37-0.76) offered the greatest OS benefit for patients with PD-L1 ≥ 10%, while Camrelizumab (HR = 0.71, 95% CI: 0.57-0.89) was the most likely regimen to provide an OS advantage for patients with PD-L1 < 10%., Conclusion: Compared to chemotherapy, PD-1 inhibitors may provide improved survival outcomes for patients with advanced ESCC. Among patients not selected based on PD-L1 expression, Sintilimab is most likely to deliver the best survival benefit. Background: The co-occurrence of smoking behaviors and major depressive disorder (MDD) has been widely documented in populations. However, the underlying mechanism of this association remains unclear., Methods: Genome-wide association studies of smoking behaviors and MDD, combined with multi-omics datasets, were usedto characterise genetic correlations, identify shared loci and genes, and explore underlying biological mechanisms. Mendelian randomization (MR) analyses were conducted to infer causal relationships between smoking behaviors and MDD. Druggability analyses were performed to identify potential drugs with both antidepressant and smoking cessation effects., Results: Extensive overall genetic correlations were found between smoking behaviors and MDD. Furthermore, eighteen local regions showed significant genetic correlations, which could be partly explained by gene co-expression patterns. We identified 24 shared loci and 120 genes, which were enriched in limbic system, GABAergic and dopaminergic neurons, as well as in synaptic pathways. Through integrating with tissue specific information, seven key genes (ANKK1, NEGR1, USP4, TCTA, SORCS5, SPPL3, and USP28) were pinpointed. Notably, druggability analyses supported ANKK1 as a potential drug target for the treatment of MDD and tobacco dependence. MR analyses suggested a bidirectional causal relationship between smoking initiation and MDD. Hypoxia is a common feature of glioblastoma (GBM). Circular RNAs (circRNAs) are identified as regulators in cancers. However, the role of circRNAs in GBM remains elusive. Here, circPLOD2a and circPLOD2b, spliced from the same parental gene PLOD2, are identified as hypoxia-responsive circRNAs. Overexpression of circPLOD2a and b enhance while knockdown inhibit GBM cell aggressiveness. The protein partners and downstream molecules were investigated by RNA-pulldown, mass spectrometry and RNA-seq. Mechanistically, HIF1α induces the expression of circPLOD2a and b, which competitively bind to HuR, causing a degradation of XIRP1 in vitro and in vivo. Clinical data demonstrate circPLOD2a and b are highly expressed in GBM negatively correlated with XIRP1, whose lower expression associates with higher glioma grade and worse prognosis. In conclusion, hypoxia-induced circPLOD2a and b are oncogenic regulators of tumour aggressiveness through attenuating the interaction between HuR and XIRP1 in glioblastoma cells and may be potential therapeutic targets for this disease., Competing Interests: Competing interests: The authors declare no competing interests. As organic solar cells (OSCs) achieve notable advancements, a significant consensus has been highlighted that the device performance is intricately linked to the active layer morphology. With conjugated molecules being widely employed, intermolecular interactions exert substantial influence over the aggregation state and morphology formation, resulting in distinct molecular packing motifs, also known as polymorphism. This phenomenon is closely associated with processing conditions and exerts a profound impact on functional properties. Consequently, understanding the mechanisms underlying polymorphism formation and establishing a definitive correlation between polymorphism and photophysical behavior is crucial for driving high-performance OSCs. In this review, a comprehensive synthesis of recent developments is provided and emphasizing its pivotal role in the field of OSC polymorphism. The thermodynamic and kinetic principles governing polymorphism formation are examined. Then, representative polymorphisms are classified in OSC materials, segmenting them into homopolymers, copolymers, and IDTT- and BTP-based small molecules. Additionally, prevalent strategies are evaluated for manipulating polymorphism. This review culminates with an analysis of the critical effects of polymorphism on OSCs, including charge carrier characteristics, photovoltaic efficiency, and long-term stability. This paper introduces RWA-BFT, a reputation-weighted asynchronous Byzantine Fault Tolerance (BFT) consensus algorithm designed to address the scalability and performance challenges of blockchain systems in large-scale IoT scenarios. Traditional centralized IoT architectures often face issues such as single points of failure and insufficient reliability, while blockchain, with its decentralized and tamper-resistant properties, offers a promising solution. However, existing blockchain consensus mechanisms struggle to meet the high throughput, low latency, and scalability demands of IoT applications. To address these limitations, RWA-BFT adopts a two-layer blockchain architecture; the first layer leverages reputation-based filtering to reduce computational complexity by excluding low-reputation nodes, while the second layer employs an asynchronous consensus mechanism to ensure efficient and secure communication among high-reputation nodes, even under network delays. This dual-layer design significantly improves performance, achieving higher throughput, lower latency, and enhanced scalability, while maintaining strong fault tolerance even in the presence of a substantial proportion of malicious nodes. Experimental results demonstrate that RWA-BFT outperforms HB-BFT and PBFT algorithms, making it a scalable and secure blockchain solution for decentralized IoT applications.

Lysine lactylation is a newly discovered protein post-translational modification that plays regulatory roles in cell metabolism, growth, reprogramming, and tumor progression. It utilizes lactate as the modification precursor, which is an end product of glycolysis while functioning as a signaling molecule in cells. Unlike previous reviews focused primarily on eukaryotes, this review aims to provide a comprehensive summary of recent knowledge about lysine lactylation in prokaryotes and eukaryotes. The current identification and enrichment strategies for lysine lactylation are introduced, and the known readers, writers, and erasers of this modification are summarized. In addition, the physiological and pathological implications of lysine lactylation are reviewed for different organisms, especially in prokaryotic cells. Pancreatic cancer patients urgently need new treatments, and we explored the efficacy and safety of combination therapy with AL2846 and gemcitabine in pancreatic cancer patients. This was a single-arm, single-center, open-label phase I/IIa study (NCT06278493). The dose-escalation phase was designed to evaluate the maximum tolerated dose (MTD) of AL2846 combined with gemcitabine. One or two dose levels were chosen for the dose-expansion phase. Treatment continued until disease progression, intolerable toxicity, patient withdrawal, or at the investigators' discretion. The primary study endpoint is to evaluate the safety and MTD of AL2846 combined with gemcitabine. The secondary endpoints included objective response rate (ORR), progression-free survival (PFS), overall survival (OS), and disease control rate (DCR). Between August 2018 and July 2021, 33 pancreatic cancer patients were enrolled in the study. A total of 15 patients were enrolled in the dose-escalation phase, and the MTD was not determined. Eventually 90 mg and 120 mg of AL2846 were chosen for the dose-expansion phase, in which 11 patients (90 mg) and 7 patients (120 mg) were administered. Treatment-related adverse events (TRAEs) of any grade were reported in 30 (90.91%) patients, and those of grade ≥ 3 were reported in 16 (48.48%) patients. The most frequently reported grade ≥ 3 TRAEs were thrombocytopenia (18.18%), neutropenia (12.12%), elevated γ-glutamyltransferase (6.06%), proteinuria (6.06%), and gastrointestinal hemorrhage (6.06%).The ORR was 6.06%, and the DCR was 72.73%. The median PFS was 3.71 months (95% CI: 3.38-4.11), and the median OS was 5.59 months (95% CI: 4.11-8.71). Gemcitabine and Al2846 combination therapy exhibited tolerable safety, but there was no improvement in efficacy over standard treatment. Epinephrine (Ep) is an important neurotransmitter, which plays an important role in the nervous system and glycogen metabolism of living organisms. Hence, a novel NCQDs/FeCoFe-PBA composite with FeCoFe-Prussian blue analogues (PBA) as the core and nitrogen-doped carbon quantum dots (NCQDs) as the shell was constructed by a one-pot hydrothermal method, and it was used for the efficient detection of Ep. As a good electroactive material, NCQDs in the composite not only improved the weak conductivity of FeCoFe-PBA, but also limited the self-aggregation of FeCoFe-PBA, and formed a uniform shell on FeCoFe-PBA. The heterogeneous structure formed between the core and shell layer resulting in NCQDs/FeCoFe-PBA nanocomposites with more active sites, electron transport channels and a larger effective surface area. Further, under optimal conditions, the electrochemical method was used to evaluate the NCQDs/FeCoFe-PBA sensor, and the results revealed that the sensor had exceptional sensing performance for Ep, with an excellent linear range from 0.01 to 306.7 μM and a low detection limit of 0.002 μM. Background: Camellia oleifera Abel ( C. oleifera ) is widely cultivated and serves as an important source of edible oil. Yet, during oil production, pruned branches generate significant waste and contribute to environmental pollution., Objectives: In this work, we obtain a natural polysaccharide from the branches of C. oleifera and optimize its extraction using Box-Behnken design (BBD), which is a statistical method commonly used in response surface methodology. Additionally, we study its properties, such as monosaccharide composition, structural features, antioxidant, and anti-inflammatory abilities., Results: BBD was employed to optimize polysaccharide extraction (solid-liquid ratio = 1:40; 90 °C; 130 min) for a higher yield. After purification, the major monosaccharides of branches of C. oleifera's polysaccharide (CBP) were disclosed as glucose and galactose. Subsequent structural features of CBP were measured. The antioxidant and anti-inflammatory abilities were measured. The highly scavenging rates of the 2,2-diphenyl-1-picrylhydrazyl and hydroxyl radicals, with the chelating capacity of Fe 2+ , indicate potent antioxidant activity of CBP., Conclusions: In general, CBP demonstrated significant anti-inflammatory activity with down-regulating the expression of IL-6 and IL-1β in the LPS-induced macrophage RAW264.7 model. This bioactive polysaccharide adds value to waste branches by offering a novel approach to waste recycling and the development of C. oleifera .

Messenger RNA (mRNA) therapeutics have garnered considerable attention due to their remarkable efficacy in the treatment of various diseases. The COVID-19 mRNA vaccine and RSV mRNA vaccine have been approved on the market. Due to the inherent nuclease-instability and negative charge of mRNA, delivery systems are developed to protect the mRNA from degradation and facilitate its crossing cell membrane to express functional proteins or peptides in the cytoplasm. However, the deficiency in transfection efficiency and targeted biological distribution are still the major challenges for the mRNA delivery systems. In this review, we first described the physiological barriers in the process of mRNA delivery and then discussed the design approach and recent advances in mRNA delivery systems with an emphasis on their tissue/cell-targeted abilities. Finally, we pointed out the existing challenges and future directions with deep insights into the design of efficient mRNA delivery systems. We believe that a high-precision targeted delivery system can greatly improve the therapeutic effects and bio-safety of mRNA therapeutics and accelerate their clinical transformations. Background and Purpose: To date, no previous studies have used multishell diffusion MRI to identify striatal microstructural damage in vivo in amyotrophic lateral sclerosis (ALS) patients. Thus, in the present study, we aimed to comprehensively explore connectivity-based selective striatal subregion microstructural damage in sporadic ALS patients and its associations with motor disability, cognitive deficits, and serum biomarkers., Methods: In this retrospective study, 79 ALS patients and 53 healthy controls (HCs) who underwent clinical assessment, serum neurofilament light (NfL) measurement, genetic testing, and multishell diffusion MRI scanning were included. Using a probabilistic tractography approach, the striatum was segmented into six subregions based on their corticostriatal connectivity. Three neurite orientation dispersion and density imaging (NODDI) parameters, the neurite density index (NDI), orientation dispersion index (ODI), and isotropic volume fraction (ISO), of the connectivity-based striatal subregions were measured., Results: Compared with HCs, ALS patients had a significantly lower NDI in the bilateral motor and right frontal subregions, a significantly lower ODI in the right motor and frontal subregions, and a significantly higher ISO in the bilateral motor and frontal subregions of the striatum after familywise error (p < 0.05). Moreover, striatal subregion microstructural damage was significantly correlated with motor disabilities, cognitive deficits, and serum NfL levels in ALS patients (p = 0.020-0.002)., Conclusions: Our study provides clear evidence demonstrating that connectivity-based selective striatal subregion microstructural damage is a definite feature of sporadic ALS patients and suggesting that striatal damage may play an important role in motor disability and cognitive deficits in ALS patients., (© 2024 The Author(s). Genomic and evolutionary analysis of epidemic porcine hepatitis E virus (HEV) in the Tibetan Plateau was performed. Faecal samples were collected from 216 Tibetan pigs and 78 Tibetan Yorkshire (Large White) and 53 tissue samples from Yorkshire from the Linzhi City slaughterhouse. Total RNA was extracted from faeces and fragments of HEV open reading frame 2 (ORF2) detected by reverse transcription and nested polymerase chain reaction (RT-nPCR) and cloned. Twenty-three samples (23/347; 6.63%) were positive for the virus, including 6.94% (15/216) Tibetan pig and 6.11% (8/131) Yorkshire samples. No tissue samples tested positive for the virus. Cloned sequences were uploaded to GenBank (accession numbers: OR392679-OR392685, OR355817-OR355824 and OR909495-OR909502) and a phylogenetic tree constructed. The entire viral genome was amplified using primers for the 5-month-old Tibetan pig sequence which confirmed that the strain belonged to HEV type 4, subtype d (GenBank accession number: OQ981960) and showed 93.30% homology with Sichuan Tibetan pig sequence, MK410044. Bayesian tree analysis showed that the earliest divergence was in 1999 and evidence of homologous recombination was found. Genomic and evolutionary analysis of HEV in the Tibetan Plateau is presented. The importance of continuous surveillance and genomic analysis of HEV is highlighted, especially in regions like the Tibetan Plateau where new strains may emerge. Introduction: Patent foramen ovale (PFO) patients may experience states of hypoxia and hypoperfusion, which may increase the burden of enlarged perivascular spaces (EPVS). However, to our knowledge, no data are available regarding EPVS in PFO patients. This study sought to investigate if patients with PFO exhibit a heightened burden of EPVS and to identify the mediating factors between PFO and EPVS., Methods: A total of 108 consecutive PFO patients (PFO group) and 110 healthy controls (HC group) from January 2022 to February 2024 were enrolled. The differences in centrum semiovale EPVS (CSO-EPVS) and basal ganglia EPVS (BG-EPVS) scores between PFO and HC groups were compared. The correlations among PFO diameters, laboratory indexes, and EPVS burdens were analyzed. The relationships among them were obtained using mediation analysis., Results: Mean age of PFO and HC group was 47.68 ± 14.47 and 48.14 ± 12.84 years. The CSO-EPVS and BG-EPVS scores were higher in PFO group than HC group (P < 0.001). The CSO-EPVS and BG-EPVS scores for PFO group were concentrated in the ranges 1-3 and 1-2 points, while for HC group were concentrated in the range 0-1 points. A positive correlation among PFO diameters and CSO-EPVS score (r = 0.62, P < 0.001), BG-EPVS score (r = 0.63, P < 0.001), and homocysteine (HCY)(r = 0.21, P = 0.03) was observed. Mediation analysis indicated that higher HCY significantly mediated the relationship between PFO diameter and BG-EPVS burden in PFO patients (P < 0.05)., Conclusion: These findings revealed the presence of glymphatic dysfunction in patients with PFO. Objectives: Noise risk control or management based on noise level has been documented, but noise risk management based on a combination of noise level and noise's temporal structure is rarely reported. This study aimed to develop a framework for industrial noise risk management based on noise kurtosis (reflecting noise's temporal structure) and its adjustment for the noise level., Design: A total of 2805 Chinese manufacturing workers were investigated using a cross-sectional survey. The noise exposure data of each subject included L EX,8h , cumulative noise exposure (CNE), kurtosis, and kurtosis-adjusted L EX,8h (L EX,8h -K). Noise-induced permanent threshold shifts were estimated at 3, 4, and 6 kHz frequencies (NIPTS 346 ) and 1, 2, 3, and 4 kHz frequencies (NIPTS 1234 ). The prevalence of high-frequency noise-induced hearing loss prevalence (HFNIHL%) and noise-induced hearing impairment (NIHI%) were determined. Risk 346 or Risk 1234 was predicted using the ISO 1999 or NIOSH 1998 model. A noise risk management framework based on kurtosis and its adjustment was developed., Results: Kurtosis could identify the noise type; Kurtosis combining noise levels could identify the homogeneous noise exposure group (HNEG) among workers. Noise kurtosis was a risk factor of HFNIHL or NIHI with an adjusted odds ratio of 1.57 or 1.52 ( p < 0.01). At a similar CNE level, the NIPTS 346 , HFNIHL%, NIPTS 1234 , or NIHI% increased with increasing kurtosis. A nonlinear regression equation (expressed by logistic function) could rebuild a reliable dose-effect relationship between L EX,8h -K and NIPTS 346 at the 70 to 95 dB(A) noise level range. After the kurtosis adjustment, the median L EX,8h was increased by 5.45 dB(A); the predicted Risk 346 and Risk 1234 were increased by 11.2 and 9.5%, respectively; NIPTS 346 -K of complex noise at exposure level <80, 80 to 85, and 85 to 90 dB(A), determined from the nonlinear regression equation, was almost the same as the Gaussian noise. Risk management measures could be recommended based on the exposure risk rating or the kurtosis-adjusted action levels (e.g., the lower and upper action levels were 80 and 85 dB(A), respectively)., Conclusions: The kurtosis and its adjustment for noise levels can be used to develop an occupational health risk management framework for industrial noise. A hydroponic experiment was conducted to investigate the variations in membrane permeabilities, chlorophyll contents, antioxidase activities, the ascorbic acid (AsA)-glutathione (GSH) cycle, and the glyoxalase system in the leaves of Pontederia cordata with 0 ∼ 15.0 mg L -1 lead ion (Pb 2+ ) exposure. The concentrations of Pb 2+ accumulated in the plant roots, stems, and leaves were also evaluated. After 7 days of exposure, the plants maintained normal growth, and there was a significant increase in ascorbate peroxidase and dehydroascorbate reductase activities. With 5.0 mg L -1 Pb 2+ exposure for 28 days, nearly 66.36% of Pb 2+ accumulated in the roots, while excess Pb 2+ immobilized in the leaves was not observed. Exposure to 10.0 and 15.0 mg L -1 Pb 2+ for 28 days significantly increased Pb 2+ contents in the leaves. This led to decrease in chlorophyll a, b , and carotenoid contents, and to increase in the methylglyoxal content in the leaves. With 10 and 15 mg L -1 Pb 2+ exposure, NPT and PCs contents in leaves increased. however, the glyoxalase system did not function well in the plant tolerant to Pb 2+ at higher concentrations. The AsA-GSH cycle did not cooperate with the glyoxalase system in the plant defense against Pb 2+ exposure in the present investigation.

A sustainable biosorbent, silver nanoparticles-decorated coffee-ground waste (CWAg), was synthesized through a simple in-situ reduction method. CWAg is extensively characterized via SEM-EDX, PZC, FTIR, XRD, HR-TEM, and XPS analyses. The biosorbent was tested to remove chromium (Cr(VI)) and methylene blue (MB) from wastewater, and its antibacterial properties was evaluated. CWAg demonstrated efficient sorption efficiencies, achieving ⁓149.9 mg/g at 323 K and pH: 2.0 for Cr(VI) and ⁓113.4 mg/g at 293 K and pH: 6.0 for MB, at a biosorbent dose of 1 g/L. The equilibration times were 90 and 120 min for Cr(VI) and MB, with half-sorption times (t HST ) of 9.2 and 13.9 min, respectively. The sorption mechanisms were successfully fitted with Sips isotherm and Pseudo-second-order models. Simultaneous sorption of MB and Cr(VI) at pH 2.0 exhibited antagonism, while a synergistic effect was observed at pH 6.0. Desorption and regeneration were achieved using NaOH and HCl solutions for Cr(VI) and MB, respectively. The efficiencies were just reduced by 9-13%. Finally, CWAg exhibited remarkable effectiveness for removing Cr(VI) from complex tannery effluent. Moreover, CWAg demonstrated positive antibacterial effects against both Gram-positive and Gram-negative bacteria, surpassing raw coffee-ground waste, underscoring the significant role of silver nanoparticles in enhancing bacterial inhibition., Competing Interests: Declarations. Background and Objective: Automatic generation of medical reports reduces both the burden on radiologists and the possibility of errors due to the inexperience of radiologists. The model that utilizes attention mechanism and contrastive learning can generate medical reports by capturing both general and specific semantics. However, existing contrastive learning methods ignore the specificity of medical data, that is, a patient may suffer from multiple diseases at the same time. This means that the lack of fine-grained relationships for contrastive learning will lead to the problem of insufficient specificity., Methods: To address the above problem, a label correlated contrastive learning method is proposed to encourage the model to generate higher-quality reports. Firstly, the refined similarity description matrix of the contrastive relationship between the reports is obtained by calculating the similarities between the multi-label classification of the reports. Secondly, the representations of image features and the embeddings containing semantic information from the decoder are projected into a hidden space. Thirdly, label correlated contrastive learning is performed with the hidden representations of the image, the embeddings of the text, and the similarity matrix. Through contrastive learning, the "hard" negative samples that share more labels with the target sample are being assigned more weights. Finally, label correlated contrastive learning and attention mechanism are combined to generate reports., Results: Comprehensive experiments are conducted on widely used datasets, IU X-ray and MIMIC-CXR. Specifically, on IU X-ray dataset, our method achieves METEOR and ROUGE-L scores of 0.198 and 0.392, respectively. On MIMIC-CXR dataset, our method achieves precision, recall, and F-1 scores of 0.384, 0.376, and 0.304, respectively.
