Background: Gastric cancer (GC) is a prevalent malignancy with limited therapeutic options for advanced stages. This study aimed to identify novel therapeutic targets for GC by profiling HSP90 client kinases. Methods: We used mass spectrometry-based activity-based protein profiling (ABPP) with a desthiobiotin-ATP probe, combined with sensitivity analysis of HSP90 inhibitors, to profile kinases in a panel of GC cell lines. We identified kinases regulated by HSP90 in inhibitor-sensitive cells and investigated the impact of MASTL knockdown on GC cell behavior. Global proteomic analysis following MASTL knockdown was performed, and bioinformatics tools were used to analyze the resulting data. Results: Four kinases—MASTL, STK11, CHEK1, and MET—were identified as HSP90-regulated in HSP90 inhibitor-sensitive cells. Among these, microtubule-associated serine/threonine kinase-like (MASTL) was upregulated in GC and associated with poor prognosis. MASTL knockdown decreased migration, invasion, and proliferation of GC cells. Global proteomic profiling following MASTL knockdown revealed NEDD4-1 as a potential downstream mediator of MASTL in GC progression. NEDD4-1 was also upregulated in GC and associated with poor prognosis. Similar to MASTL inhibition, NEDD4-1 knockdown suppressed migration, invasion, and proliferation of GC cells. Conclusions: Our multi-proteomic analyses suggest that targeting MASTL could be a promising therapy for advanced gastric cancer, potentially through the reduction of tumor-promoting proteins including NEDD4-1. The dominant narrative in much of the world is that public safety is provided by policing, evidenced by supportive rhetoric from institutional forces including politicians, media, and large budget allocations in all levels of government. Alongside a long history of police violence, especially against Black, Brown, poor, and other marginalized people, many social movements reject the idea that policing provides safety and seek other methods for community wellness. The present study utilizes critical narrative analysis (CNA) to describe how marginalized residents of a small city in Iowa construct their understanding of personal and community safety. Their stories and the dialectic exchange during interviews illustrated several counternarratives and moments of conscientization for participants and researchers where safety was deconstructed and understood outside the power of recycled institutional narratives. Participants rejected popular notions of safety such as police, and instead embraced safety through robust relationships, community resources, and forms of self‐knowledge such as mental health. We analyzed their interviews as efforts to be humanly recognized within violent white supremacist structures, and their stories help to radicalize popular messages about safety. Sargassum horneri (S. horneri), a brown seaweed excessively proliferating along Asian coastlines, are damaging marine ecosystems. Thus, this study aimed to enhance nutritional value of S. horneri through lactic acid bacteria fermentation to increase S. horneri utilization as a functional food supplement, and consequently resolve coastal S. horneri accumulation. S. horneri supplemented fermentation was most effective with Lactiplantibacillus pentosus SH803, thus this product (F-SHWE) was used for further in vitro studies. F-SHWE normalized expressions of oxidative stress related genes NF-κB, p53, BAX, cytochrome C, caspase 9, and caspase 3, while non-fermented S. horneri (SHWE) did not, in a H2O2-induced HT-29 cell model. Moreover, in an LPS-induced HT-29 cell model, F-SHWE repaired expressions of inflammation marker genes ZO1, IL1β, IFNγ more effectively than SHWE. For further functional assessment, F-SHWE was also treated in 3T3-L1 adipocytes. As a result, F-SHWE decreased lipid accumulation, along with gene expression of adipogenesis markers PPARγ, C/EBPα, C/EBPβ, aP2, and Lpl; lipogenesis markers Lep, Akt, SREBP1, Acc, Fas; inflammation markers IFN-γ and NF-κB. Notably, gene expression of C/EBPβ, IFN-γ and NF-κB were suppressed only by F-SHWE, suggesting the enhancing effect of fermentation on obesity-related properties. Compositional analysis attributed the protective effects of F-SHWE to acetate, an organic acid significantly higher in F-SHWE than SHWE. This retrospective study reports the isolation and characterization of Severe Acute Respiratory Syndrome coronavirus 2 (SARS-CoV-2) from a household cat in South Korea. The cat, which was presented with respiratory symptoms, was identified during a retrospective analysis of samples collected between April 2021 and March 2022. Genomic sequencing revealed that the isolated virus belonged to the Omicron variant (BA.1), coinciding with its global emergence in early 2022. This case study provides evidence for the potential of direct human-to-cat transmission of the Omicron variant in South Korea during its period of widespread circulation. Our findings underscore the importance of continuous monitoring of SARS-CoV-2 in both human and animal populations to track viral evolution and potential spillover events. [ABSTRACT FROM AUTHOR]

A composite material of tungsten carbide and mesoporous carbon was synthesized by the sol-gel polycondensation of resorcinol and formaldehyde, using cetyltrimethylammonium bromide as a surfactant and Ludox HS-40 as a porogen, and served as a support for Pd-based electrodes. Phosphorus-modified Pd particles were deposited onto the support using an NH3-mediated polyol reduction method facilitated by sodium hypophosphite. Remarkably small Pd nanoparticles with a diameter of ca. 4 nm were formed by the phosphorus modification. Owing to the high dispersion of Pd and its strong interaction with tungsten carbide, the Pd nanoparticles embedded in the tungsten carbide/mesoporous carbon composite exhibited a hydrogen oxidation activity approximately twice as high as that of the commercial Pt/C catalyst under the anode reaction conditions of proton exchange membrane fuel cells. Beyond the existing in-solution plasma (iSP) process used for nanoparticle synthesis, polymer film synthesis has recently been demonstrated via iSP, which occurs at a specific cycle in a substrate with electrical potential. Herein, we propose an iSP reactor designed to tune the characteristics of polypyrrole (PPy) films by applying varied DC biases to the substrate during the solution plasma process. Applying DC bias to the substrate notably enhances the iSP during the negative cycle, thereby generating ionic precursors from pyrrole monomers for PPy film formation. These films demonstrated morphological variances as a function of the applied VDC. At a low voltage (VDC = 0.5 kV), a uniform film was formed via a layer-by-layer growth mechanism, and as the voltage was increased, the film exhibited a structured morphology. Specifically, at VDC = 0.5 kV, the PPy film was successfully coated onto a finely patterned electrode. The proposed iSP process, under specific DC biasing conditions, enables polymer coating without necessitating additional chemicals, catalysts, or masks. South Korea has experienced outbreaks of foot-and-mouth disease (FMD) of serotypes O and A, leading to nationwide vaccination with a bivalent vaccine. Since the FMD virus (FMDV) Asia1 group-V genotype occurred in North Korea in 2007, an Asia1/MOG/05 vaccine strain belonging to the Asia1 group-V genotype was developed using a genetic recombination method (Asia1/MOG/05-R). This study aimed to evaluate the antigen productivity and viral inactivation kinetics of Asia1/MOG/05-R to assess its commercial viability. The antigen yield of Asia1/MOG/05-R produced in flasks and bioreactors was approximately 4.0 μg/mL. Binary ethylenimine (BEI) inactivation kinetics of Asia1/MOG/05-R showed that 2 mM and 1.0 mM BEI treatment at 26 °C and 37 °C, respectively, resulted in a virus titer <10−7 TCID50/mL within 24 h, meeting the inactivation kinetics criteria. During incubation at 26 °C and 37 °C, 10% antigen loss occurred, but not due to BEI treatment. When pigs were inoculated twice with the Asia1/MOG/05-R antigen, the virus neutralization titer increased to approximately 1:1000; therefore, it can sufficiently protect against Asia1/MOG/05-R and Asia1 Shamir viruses. In this study, we focus on the estimation of the regression function in the single-index model based on B-splines using penalization techniques. We adopt a spherical coordinates reparameterization of an index vector to deal with an identification problem of the single-index model. To provide a spatially adaptive method, two types of penalties are applied to the estimation of the index vector and the regression function. A special penalty called the localized penalty is introduced to handle the sparsity of the index vector using the spherical coordinates, and the total variation penalty is considered to deal with the smoothing function. Using a coordinate descent algorithm with a grid search of the two tuning parameters, the entire solution paths of the index coefficients and the regression functions for tuning parameters can be obtained efficiently. To generate a stable and effective air–liquid discharge in an open atmosphere, we investigated the effect of the dielectric barrier on the discharge between the pin electrode and liquid surface in an atmospheric-pressure plasma reactor. The atmospheric-pressure plasma reactor used in this study was based on a pin–plate discharge structure, and a metal wire was used as a pin-type power electrode. A plate-type ground electrode was placed above and below the vessel to compare the pin–liquid discharge and pin–liquid barrier discharge (PLBD). The results indicated that the PLBD configuration utilizing the bottom of the vessel as a dielectric barrier outperformed the pin–liquid setup in terms of the discharge stability and that the concentration of reactive species was different in the two plasma modes. PLBD can be used as a digestion technique for determining the phosphorus concentration in natural water sources. The method for decomposing phosphorus compounds by employing PLBD exhibited excellent decomposition performance, similar to the performance of thermochemical digestion—an established conventional method for phosphorus detection in water. The PLBD structure can replace the conventional chemical-agent-based digestion method for determining the total dissolved phosphorus concentration using the ascorbic acid reduction method. Lipid biosynthesis is recently studied its functions in a range of cellular physiology including differentiation and regeneration. However, it still remains to be elucidated in its precise function. To reveal this, we evaluated the roles of lysophosphatidic acid (LPA) signaling in alveolar bone formation using the LPA type 2 receptor (LPAR2) antagonist AMG‐35 (Amgen Compound 35) using tooth loss without periodontal disease model which would be caused by trauma and usually requires a dental implant to restore masticatory function. In this study, in vitro cell culture experiments in osteoblasts and periodontal ligament fibroblasts revealed cell type‐specific responses, with AMG‐35 modulating osteogenic differentiation in osteoblasts in vitro. To confirm the in vivo results, we employed a mouse model of tooth loss without periodontal disease. Five to 10 days after tooth extraction, AMG‐35 facilitated bone formation in the tooth root socket as measured by immunohistochemistry for differentiation markers KI67, Osteocalcin, Periostin, RUNX2, transforming growth factor beta 1 (TGF‐β1) and SMAD2/3. The increased expression and the localization of these proteins suggest that AMG‐35 elicits osteoblast differentiation through TGF‐β1 and SMAD2/3 signaling. These results indicate that LPAR2/TGF‐β1/SMAD2/3 represents a new signaling pathway in alveolar bone formation and that local application of AMG‐35 in traumatic tooth loss can be used to facilitate bone regeneration and healing for further clinical treatment. We present the multiwavelength flaring activity of the blazar AO 0235 + 164 during its recent active period from 2013 to 2019. From a discrete correlation function analysis, we find a significant (⁠|$\gt 95~{{\ \rm per\ cent}}$|⁠) correlation between radio and gamma-ray light curves with flares at longer wavelengths following flares at shorter wavelengths. We identify a new jet component in 43 GHz Very Long Baseline Array data that was ejected from the radio core on MJD |$57246^{+26}_{-30}$| (2015 August 12), during the peak of the 2015 radio flare. From the analysis of the jet component, we derived a Doppler factor of δvar = 28.5 ± 8.4, a bulk Lorentz factor of |$\Gamma =16.8^{+3.6}_{-3.1}$|⁠ , and an intrinsic viewing angle of |$\theta _{\rm v}=1.42^{+1.07}_{-0.52}\textrm {~degrees}$|⁠. Investigation of the quasi-simultaneous radio data revealed a partially absorbed spectrum with the turnover frequency varying in the range of 10−70 GHz and the peak flux density varying in the range of 0.7−4 Jy. We find the synchrotron self-absorption magnetic field strength to be |$B_{\rm SSA}=15.3^{+12.6}_{-14.0}\textrm {~mG}$| at the peak of the 2015 radio flare, which is comparable to the equipartition magnetic field strength of |$B_{\rm EQ}=43.6^{+10.6}_{-10.4}\textrm {~mG}$| calculated for the same epoch. Additional analysis of the radio emission region in the relativistic jet of AO 0235 + 164 suggests that it did not significantly deviate from equipartition during its recent flaring activity.
