Background: Individuals with subacute severe hemiplegia often undergo alternate gait training to overcome challenges in achieving walking independence. However, the ankle joint setting in a knee-ankle-foot orthosis (KAFO) depends on trunk function or paralysis stage for alternate gait training with a KAFO. The optimal degree of ankle joint freedom in a KAFO and the specific ankle joint conditions for effective rehabilitation remain unclear. Therefore, this study aimed to investigate the effects of different degrees of freedom of the ankle joint on center-of-pressure (CoP) parameters and muscle activity on the paretic side using a KAFO and to investigate the recommended setting of ankle joint angle in a KAFO depending on physical function., Methods: This study included 14 participants with subacute stroke (67.4 ± 13.3 years). The CoP parameters and muscle activity of the gastrocnemius lateralis (GCL) and soleus muscles were compared using a linear mixed model (LMM) under two ankle joint conditions in the KAFO: fixed at 0° and free ankle dorsiflexion. We confirmed the relationship between changes in CoP parameters or muscle activity under different conditions and physical functional characteristics such as the Fugl-Meyer Assessment of Lower Extremity Synergy Score (FMAs) and Trunk Impairment Scale (TIS) using LMM., Results: Anterior-posterior displacement of CoP (AP&#95;CoP) (p = 0.011) and muscle activity of the GCL (p = 0.043) increased in the free condition of ankle dorsiflexion compared with that in the fixed condition. The FMAs (p = 0.004) and TIS (p = 0.008) demonstrated a positive relationship with AP&#95;CoP. A positive relationship was also found between TIS and the percentage of medial forefoot loading time in the CoP (p < 0.001)., Conclusions: For individuals with severe subacute hemiplegia, the ankle dorsiflexion induction in the KAFO, which did not impede the forward tilt of the shank, promotes anterior movement in the CoP and muscle activity of the GCL. This study suggests that adjusting the dorsiflexion mobility of the ankle joint in the KAFO according to improvement in physical function promotes loading of the CoP to the medial forefoot., (© 2024. Objective: To investigate the immediate effects of plastic ankle-foot orthosis (AFO) on locomotor performance in patients with stroke and determine how such effects might undergo alteration when distinct plantarflexor (PF) module subtypes are considered., Design: Cross-sectional study., Setting: Two university hospitals., Participants: Fifty-two patients with stroke and 21 of those without stroke (N=73)., Interventions: Not applicable., Main Outcome Measures: Motor modules were identified through non-negative matrix factorization, and participants were classified into 3 groups: independent-normal-timing, independent-altered-timing, and merged PF modules. To assess the effects of the AFO, gait measurements reflecting locomotor performance were obtained with and without the presence of the plastic AFO for each group., Results: The independent-altered-timing group had increased paretic propulsion, greater non-paretic step length, and faster walking speed after the administration of the plastic AFO; however, these significant changes were not observed in the independent-normal-timing and merged PF module groups. Notably, patients in the independent-normal-timing and merged PF module groups exhibited longer paretic stance times., Conclusion: This study suggests that the immediate effects of plastic AFO depend on the PF module subtype. The arsenic adsorption performance of silicon (Si), iron (Fe), and magnesium (Mg) mixed hydrous oxide containing a Si: Fe: Mg metal composition ratio of 0.05:0.9:0.05 (SFM05905) was investigated. SFM05905 was synthesized by the co-precipitation method. Batch experiments on arsenic adsorption were conducted at various temperatures and concentrations. Adsorption isotherms models were represented by a linearized equations and were insensitive to temperature change. The anion selectivity of SFM05905 at single component was high for arsenite (III), arsenate (V), and phosphate (PO 4 ), indicating that PO 4 inhibits arsenic adsorption. The adsorption amount of As (III), As (V), and PO 4 were compared using a column packed with granular SFM05905, and an aqueous solution was passed by a combination of several anions that are single, binary, and ternary adsorbate systems. As (III) had the highest adsorption amount; however, As (III) and PO 4 were affected by each other under the ternary mixing condition. Although the adsorption amount of As (V) was smaller than that of As (III), it was not affected by other adsorbates in the column experiments. Finally, although the adsorption of both arsenic continued, the adsorbed PO 4 gradually desorbed.

We have devised innovative anti-loosening bolts with a double thread-mechanism (denoted DTB-IIC) composed of coaxial single and multiple coarse threads. In this study, we first developed a high-speed and high-precision grinding system for dedicated thread rolling dies of the DTB-IIC. Compared to conventional electro-discharge machining dies, ground dies significantly reduced processing time and costs, and achieved more than 10 times higher durability in thread rolling tests. Comparative Junker vibration loosening tests based on an ISO standard were conducted on several types of DTB-IICs. The amount of backlash δ between the inner multiple-thread nut and DTB-IIC bolt has a great effect on the initial drop of the bolt axial preload, and the rolled DTB-IIC with a relatively small δ secured a residual axial load rate of ≥85%, which satisfies the rating-1 of good self-locking behavior. A 3D FEM model was employed to simulate the initial loosening process in the Junker test, and the analytical results agreed well with the experimental ones by adjusting δ appropriately. Both experimental and analytical results indicate that the high loosening resistance of DTB-IIC is due to the synergistic effect of the interference mechanism of two types of nuts and the jammed locking state., Competing Interests: The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (© 2024 The Authors. The HHP inactivation behaviors of Niigata sake yeast Saccharomyces cerevisiae strain S9arg and its aerobic respiratory-deficient mutant strains were investigated after cultivating them in a YPD media containing 2% to 15% glucose, as well as in moromi mash, in a laboratory-scale sake brewing process. The piezotolerance of strain S9arg, shown after cultivation in a YPD medium containing 2% glucose, decreased to become piezosensitive with increasing glucose concentrations in YPD media. In contrast, the piezosensitivity of a mutant strain UV1, shown after cultivation in the YPD medium containing 2% glucose, decreased to become piezotolerant with increasing glucose concentrations in the YPD medium. The intracellular ATP concentrations were analyzed for an S. cerevisiae strain with intact aerobic respiratory ability, as well as for strain UV1. The higher concentration of ATP after cultivation suggested a higher energy status and may be closely related to higher piezotolerance for the yeast strains. The decreased piezotolerance of strain S9arg observed after a laboratory-scale sake brewing test may be due to a lower energy status resulting from a high glucose concentration in moromi mash during the early period of brewing, as well as a lower aeration efficiency during the brewing process, compared with cultivation in a YPD medium containing 2% glucose.

Nuclear factor of activated T cells 5 (NFAT5; also called TonEBP/OREBP) is a transcription factor that is activated by hypertonicity and induces osmoprotective genes to protect cells against hypertonic conditions. In the kidney, renal tubular NFAT5 is known to be involved in the urine concentration mechanism. Previous studies have suggested that NFAT5 modulates the immune system and exerts various effects on organ damage, depending on organ and disease states. Pathophysiological roles of NFAT5 in renal tubular cells, however, still remain obscure. We conducted comprehensive analysis by performing transcription start site (TSS) sequencing on the kidney of inducible and renal tubular cell-specific NFAT5 knockout (KO) mice. Mice were subjected to unilateral ureteral obstruction to examine the relevance of renal tubular NFAT5 in renal fibrosis. TSS sequencing analysis identified 722 downregulated TSSs and 1,360 upregulated TSSs, which were differentially regulated ≤-1.0 and ≥1.0 in log 2 fold, respectively. Those TSSs were annotated to 532 downregulated genes and 944 upregulated genes, respectively. Motif analysis showed that sequences that possibly bind to NFAT5 were enriched in TSSs of downregulated genes. Gene Ontology analysis with the upregulated genes suggested disorder of innate and adaptive immune systems in the kidney. Unilateral ureteral obstruction significantly exacerbated renal fibrosis in the renal medulla in KO mice compared with wild-type mice, accompanied by enhanced activation of immune responses. In conclusion, NFAT5 in renal tubules could have pathophysiological roles in renal fibrosis through modulating innate and adaptive immune systems in the kidney. NEW & NOTEWORTHY TSS-Seq analysis of the kidney from renal tubular cell-specific NFAT5 KO mice uncovered novel genes that are possibly regulated by NFAT5 in the kidney under physiological conditions. The study further implied disorders of innate and adaptive immune systems in NFAT5 KO mice, thereby exacerbating renal fibrosis at pathological states. Our results may implicate the involvement of renal tubular NFAT5 in the progression of renal fibrosis. Further studies would be worthwhile for the development of novel therapy to treat chronic kidney disease.
