Confocal Raman microscopy (CRM) is a promising in-situ visual technique that provides detailed insights into multiple lignocellulosic components and structures in plant cell walls at the micro-nano scale. In this study, we propose a novel CRM cosine similarity (CS) mapping strategy for the simultaneous in-situ visual profiling of lignin, cellulose, and hemicellulose in plant cell walls. The main stages of this strategy include: 1) a modified Otsu algorithm for extracting the regions of interest (ROI); 2) a modified subtraction method for cleaning the background signals in the ROI spectra; 3) a lignin signal subtraction method based on the pixel correction factor for eliminating the interference of strong lignin signals with weak cellulose and hemicellulose signals in the Raman full spectra of the cell walls; 4) second-order derivative spectral preprocessing for enhancing the discrimination between the characteristic peaks of cellulose and hemicellulose; 5) a CS mapping algorithm for simultaneous in-situ profiling of lignin, cellulose, and hemicellulose in plant cell walls. The effectiveness of the strategy is verified by characterizing the Brittle Culm1 (BC1) gene-mutant rice stem (IL349-BC1-KO) with known bioinformatics. Background: Digital subtraction angiography (DSA) devices are commonly used in numerous interventional procedures across various parts of the body, necessitating multiple scans per procedure, which results in significant radiation exposure for both doctors and patients. Inspired by generative artificial intelligence techniques, this study proposes GenDSA, a large-scale pretrained multi-frame generative model-based real-time and low-dose DSA imaging system., Methods: GenDSA was developed to generate 1-, 2-, and 3-frame sequences following each real frame. A large-scale dataset comprising ∼3 million DSA images from 27,117 patients across 10 hospitals was constructed to pretrain, fine-tune, and validate GenDSA. Two other datasets from 25 hospitals were used for evaluation. Objective evaluations included SSIM and PSNR. Five interventional radiologists independently assessed the quality of the generated frames using the Likert scale and visual Turing test. Scoring consistency among the radiologists was measured using the Kendall coefficient of concordance (W). The Fleiss' kappa values were used for inter-rater agreement analysis for visual Turing tests., Findings: Using only one-third of the clinical radiation dose, videos generated by GenDSA were perfectly consistent with real videos. Objective evaluations demonstrated that GenDSA's performance (PSNR = 36.83, SSIM = 0.911, generation time = 0.07 s/frame) surpassed state-of-the-art algorithms. Subjective ratings and statistical results from five doctors indicated no significant difference between real and generated videos. Metabolic enzymes perform moonlighting functions during tumor progression, including the modulation of chemoresistance. However, the underlying mechanisms of these functions remain elusive. Here, utilizing a metabolic clustered regularly interspaced short palindromic repeats (CRISPR)-Cas9 knockout library screen, we observe that the loss of glutamate-cysteine ligase modifier subunit (GCLM), a rate-limiting enzyme in glutathione biosynthesis, noticeably increases the sensitivity of colorectal cancer (CRC) cells to platinum-based chemotherapy. Mechanistically, we unveil a noncanonical mechanism through which nuclear GCLM competitively interacts with NF-kappa-B (NF-κB)-repressing factor (NKRF), to promote NF-κB activity and facilitate chemoresistance. In response to platinum drug treatment, GCLM is phosphorylated by P38 MAPK at T17, resulting in its recognition by importin a5 and subsequent nuclear translocation. Furthermore, elevated expression of nuclear GCLM and phospho-GCLM correlate with an unfavorable prognosis and poor benefit from standard chemotherapy. The plant root-knot nematode Meloidogyne spp. is an endoparasite with worldwide distribution that is detrimental to the growth of a wide range of plants. The insecticidal crystal proteins produced by Bacillus thuringiensis are widely used as a biological insecticide to control Lepidopteran, Hemiptera, and Coleopteran pests. However, there are few reports of toxicity against plant-parasitic nematodes. In this study, we report the cloning and characterization of a novel Tpp-like protein from B. thuringiensis strain GXUN31-2 that exhibits significant nematicidal activity against the plant root-knot nematode Meloidogyne enterolobii. The full-length CG_5628 gene was 951 bp and encoded a 34.4 kDa protein consisting of 316 amino acid residues. The gene was ligated into pET30a(+) and expressed in Escherichia coli BL21(DE3). Following purification of the Tpp-like protein, bioassays against the second-stage juvenile (J2) of M. enterolobii revealed lethal concentration (LC 50 ) values of 144.3 μg/ml (95 % Confidence interval: 133.5-155.7 μg/ml). Analysis of the conserved domain revealed the presence of a Ricin B lectin domain with residues located at positions 8-155. Objective: To evaluate the application and effectiveness of prenatal ultrasound in diagnosing and managing conjoined twins., Methods: A retrospective analysis was conducted on 20 cases of conjoined twins diagnosed at our hospital between January 2016 and December 2022. The types of conjoined twins, ultrasonographic characteristics, and associated anomalies were assessed., Results: The gestational age at diagnosis ranged from 10 to 35 weeks, with an average of 14.21 ± 5.69 weeks. Thirteen cases were detected in the first trimester, five in the early second trimester, one at 23 + 2 weeks, and one at 35 weeks. Thoracopagus was the most common type (11 cases, 55%), followed by omphalopagus (4 cases, 20%), cephalopagus (4 cases, 20%), and parapagus dicephalus (1 case, 5%). In the first trimester, the most common abnormalities observed included increased nuchal translucency (NT), cystic hygroma, hydrops fetalis, and generalized edema. Major birth defects identified in conjoined twins were omphalocele (3 cases), congenital heart malformations (3 cases), neural tube defects (2 cases), urachal cyst (1 case), and umbilical cyst (1 case). Pregnancy was terminated in 18 cases, one case resulted in spontaneous abortion during the second trimester, and one case was delivered by cesarean section at 37 weeks, with successful separation and recovery., Conclusion: Prenatal ultrasound is the primary diagnostic tool for conjoined twins. Obesity-related chronic kidney disease (CKD) poses a significant risk to individuals' health and wellbeing, but the pathological mechanisms and treatment strategies are currently limited. Trans-cinnamic acid (CA) is a key active monomer found in cinnamon bark and is known for its diverse pharmacological activities. However, its effect on obesity-related renal injury remains unknown. In the current study, the in vitro and in vivo experiments were combined to investigate the beneficial effect of CA on renal injury induced by HFD or PA. We found that CA significantly reduced the obesity of zebrafish body and the accumulation of fat in kidney tissues. The histopathological changes and dysfunction induced by HFD were effectively mitigated by CA administration, as evidenced by the detection of Hematoxylin-Eosin straining, NAG activity, creatinine level, and expression of functional-related genes, respectively. Additionally, the in vitro and in vivo findings demonstrated that CA dramatically reduced the oxidative stress, inflammatory, and apoptosis in HFD-induced kidney tissues or PA-treated HEK293T and HK-2 cells. Finally, the results regarding ERK, JNK, and P38 proteins phosphorylation confirmed that CA may alleviate HFD-induced renal injury by inhibiting the phosphorylation of ERK, JNK, and P38 MAPK proteins. This theory was further supported by the results of co-treatment with anisomycin (a JNK activator) or lipopolysaccharide and CA in HEK293T cells.
