Primary open-angle glaucoma (POAG) is subdivided depending on eye pressure. Patients with normal-tension glaucoma (NTG) have never had high intraocular pressure (IOP) measured while patients with ocular hypertension (OHT) have high eye pressure but no signs of glaucoma. Although IOP is considered to be a risk factor for all glaucoma patients, it is reasonable to assume that other risk factors such as inflammation play a role. We aimed to characterize the proteome and cytokine profile during hypoxia in plasma from patients with NTG (n = 10), OHT (n = 10), and controls (n = 10). Participants were exposed to hypoxia for two hours, followed by 30 min of normoxia. Samples were taken before ("baseline"), during ("hypoxia"), and after hypoxia ("recovery"). Proteomics based on liquid chromatography coupled with mass spectrometry (LC–MS) was performed. Cytokines were measured by Luminex assays. Bioinformatic analyses indicated the involvement of complement and coagulation cascades in NTG and OHT. Regulation of high-density lipoprotein 3 (HDL3) apolipoproteins suggested that changes in cholesterol metabolism are related to OHT. Hypoxia decreased the level of tumor necrosis factor-α (TNF-α) in OHT patients compared to controls. Circulating levels of interleukin-1β (IL-1β) and C-reactive protein (CRP) were decreased in NTG patients compared to controls during hypoxia. After recovery, plasma interleukin-6 (IL-6) was upregulated in patients with NTG and OHT. Current results indicate an enhanced systemic immune response in patients with NTG and OHT, which correlates with pathogenic events in glaucoma. Apolipoproteins may have anti-inflammatory effects, enabling OHT patients to withstand inflammation and development of glaucoma despite high IOP. [ABSTRACT FROM AUTHOR]

Traditional research on political parties pays little attention to the temporal focus of communication. It usually concentrates on promises, issue attention, and policy positions. This lack of scholarly attention is surprising, given that voters respond to nostalgic rhetoric and may even adjust issue positions when policy is framed in nostalgic terms. This article presents a novel dataset, PolNos, which contains six text-based measures of nostalgic rhetoric in 1,648 party manifestos across 24 European democracies from 1946 to 2018. The measures combine dictionaries, word embeddings, sentiment approaches, and supervised machine learning. Our analysis yields a consistent result: nostalgia is most prevalent in manifestos of culturally conservative parties, notably Christian democratic, nationalist, and radical right parties. However, substantial variation remains regarding regional differences and whether nostalgia concerns the economy or culture. Reductions in greenhouse gas emissions are necessary to slow global warming and decrease the likelihood of irreversible climate scenarios. There is thus an urgent need for low‐emissions fuels. This study conducted a techno‐economic analysis of two different industrial plant concepts for producing hydrogen from woody biomass. One was a large‐scale centralized 60 MWH2 option using dual fluidized bed gasification with CO2 removal. The other was a small‐scale decentralized 1 MWH2 option using fixed‐bed gasification without CO2 removal. Mass and energy balances were calculated by the process simulation software IPSEpro. Key performance indicators, including technical, economic, and environmental parameters, were derived. Overall energy efficiencies of 64.2 and 59.5% and hydrogen yields of 87 and 68 gH2 kg−1BM.db were determined for hydrogen production by dual fluidized bed and fixed‐bed gasification, respectively. The levelized costs of hydrogen amounted to 5.6 and 15.0 €2022 kg−1H2 and agreed quite well with values from the literature. Flexible and decentralized heat, electricity, and hydrogen production based on fixed‐bed gasification within a multiproduct plant were also evaluated. The results show that the multiproduct plant could be economically feasible if at least 63% of the annual operating hours were dedicated to hydrogen production, and assuming a hydrogen selling price of 17.5 €2022 kg−1H2 in Austria. In conclusion, both of the processes that were evaluated are conceivable technologies for the transition of the energy system towards renewable energy sources from a technical and economic point of view, although small‐scale hydrogen production is considerably more expensive. Introduction: Acute myeloid leukemia (AML) is a cancer of the hematopoietic system characterized by hyperproliferation of undifferentiated cells of the myeloid lineage. While most of AML therapies are focused toward tumor debulking, all-trans retinoic acid (ATRA) induces neutrophil differentiation in the AML subtype acute promyelocytic leukemia (APL). Macroautophagy has been extensively investigated in the context of various cancers and is often dysregulated in AML where it can have context-dependent pro- or anti-leukemogenic effects. On the contrary, the implications of chaperone-mediated autophagy (CMA) on the pathophysiology of diseases are still being explored and its role in AML remains elusive. Methods: We took advantage of human AML primary samples and databases to analyze CMA gene expression and activity. Furthermore, we used ATRA-sensitive (NB4) and -resistant (NB4-R1) APL cells to further dissect a potential function for CMA in ATRA-mediated neutrophil differentiation. NB4-R1 cells are unique in that they do respond to retinoic acid transcriptionally but do not mature in response to retinoid signaling alone unless maturation is triggered by adding cyclic adenosine monophosphate. Results: Here, we report that CMA-related mRNA transcripts are significantly higher expressed in immature hematopoietic cells as compared to neutrophils, contrasting the macroautophagy gene expression patterns. Accordingly, lysosomal degradation of an mCherry-KFERQ CMA reporter decreases during ATRA-induced differentiation of APL cells. On the other hand, using NB4-R1 cells we found that macroautophagy flux primed ATRA-resistant NB4-R1 cells to differentiate upon ATRA treatment but reduced the association of lysosome-associated membrane protein type 2A (LAMP-2A) and heat shock protein family A (Hsp70) member 8 (HSPA8), necessary for complete neutrophil maturation. Accordingly, depletion of HSPA8 attenuated CMA activity and facilitated APL cell differentiation. In contrast, maintaining high CMA activity by ectopic expression of LAMP-2A impeded APL differentiation. Conclusion: Overall, our findings suggest that APL neutrophil differentiation requires CMA inactivation and that this pathway predominantly depends on HSPA8 and is possibly assisted by other co-chaperones. The vespertilionid bat genus Hesperoptenus, comprising only five species and showing a distribution in the tropical regions from India to Sulawesi, was hitherto underrepresented in recent taxonomic studies. Here, modern cytogenetic techniques were applied to compare the karyotypes of two Hesperoptenus species, H. tomesi and H. blanfordi. Despite having the same chromosome number, 2n = 32, the karyotypes of H. tomesi and H. blanfordi were found to differ extensively. Instead, H. tomesi was found to share all nine derived Robertsonian fusion products with the previously published karyotype of H. doriae (2n = 26), which points to a closer relationship between both species. In line with our chromosomal data, mitochondrial DNA analyses also provided evidence for a closer relationship of H. tomesi with H. doriae and H. tickelli, for which no banded karyotype has been examined. Again, H. blanfordi was clearly separated from the remaining Hesperoptenus species. As a consequence, the subgenus Milithronycteris, comprising all species of Hesperoptenus except the type species H. doriae, is rendered paraphyletic. We therefore suggest to synonymize Milthronycteris with the subgenus Hesperoptenus. A chromosomal character, found in all three Hesperoptenus species examined but absent in other vespertilionid genera studied so far, namely a particular G-banding pattern on the homolog to Myotis chromosome MMY13, may serve as a synapomorphy for the genus Hesperoptenus. This study aimed to develop a laboratory-scale direct air capture unit for evaluating and comparing amine-based adsorbents under temperature vacuum swing adsorption conditions. The experimental campaign conducted with the direct air capture unit allowed for the determination of equilibrium loading, CO2 uptake capacity, and other main performance parameters of the investigated adsorbent Lewatit VP OC 1065®. The investigations also helped to understand the co-adsorption of CO2 and H2O on the tested material, which is crucial for improving temperature vacuum swing adsorption processes. This was achieved by obtaining pure component isotherms for CO2 and H2O and using three different co-adsorption isotherm models from the literature. It was found that the weighted average dual-site Toth model emerged as the most accurate and reliable model for simulating this co-adsorption behaviour. Its predictions closely align with the experimental data, particularly in capturing the adsorption equilibrium at various temperatures. It was also observed that this lab-scale unit offers advantages over thermogravimetric analysis when conducting adsorption experiments on the chosen amine. The final aim of this study is to provide a pathway to develop devices for testing and developing efficient and cost-effective adsorbents for direct air capture. [ABSTRACT FROM AUTHOR]

Purpose: Depression is associated with low-grade systemic inflammation and impaired intestinal function, both of which may reduce dietary iron absorption. Low iron status has been associated with depression in adults and adolescents. In Swiss adolescents, we determined the associations between paediatric major depressive disorder (pMDD), inflammation, intestinal permeability and iron status. Methods: This is a matched case–control study in 95 adolescents with diagnosed pMDD and 95 healthy controls aged 13–17 years. We assessed depression severity using the Children's Depression Rating Scale-Revised. We measured iron status (serum ferritin (SF) and soluble transferrin receptor (sTfR)), inflammation (C-reactive protein (CRP) and alpha-1-acid-glycoprotein (AGP)), and intestinal permeability (intestinal fatty acid binding protein (I-FABP)). We assessed history of ID diagnosis and treatment with a self-reported questionnaire. Results: SF concentrations did not differ between adolescents with pMDD (median (IQR) SF: 31.2 (20.2, 57.0) μg/L) and controls (32.5 (22.6, 48.3) μg/L, p = 0.4). sTfR was lower among cases than controls (4.50 (4.00, 5.50) mg/L vs 5.20 (4.75, 6.10) mg/L, p < 0.001). CRP, AGP and I-FABP were higher among cases than controls (CRP: 0.16 (0.03, 0.43) mg/L vs 0.04 (0.02, 0.30) mg/L, p = 0.003; AGP: 0.57 (0.44, 0.70) g/L vs 0.52 (0.41, 0.67) g/L, p = 0.024); I-FABP: 307 (17, 515) pg/mL vs 232 (163, 357) pg/mL, p = 0.047). Of cases, 44% reported having a history of ID diagnosis compared to 26% among controls (p = 0.020). Finally, 28% of cases had iron treatment at/close to study inclusion compared to 14% among controls. Conclusion: Cases had significantly higher systemic inflammation and intestinal permeability than controls but did not have lower iron status. Simple Summary: Patients with metastasized, castration-resistant prostate cancer can be treated locally with radioactive radium-223, which usually comprises six cycles. We wanted to know whether this treatment affects immune function. We performed cell culture experiments with white blood cells from the patients and added components of microorganisms. We tested the ability of the cells to proliferate and produce two cytokines (interferon-gamma and interleukin-10) that are important for the balance of the immune system. Our data in 21 patients indicate that the immune cells show impairment in their defense against microorganisms after treatment. As determined by bone scintigraphy, a reduction in tumor burden was observed in 67% of patients. Interestingly, even before treatment, the number of cells producing interleukin-10, an anti-inflammatory cytokine, was an indicator of tumor burden at the end of treatment. Thus, a blood test can help assess whether the treatment is likely to be successful. Previous data indicate that one cycle of treatment with radium-223 (223Ra) did not significantly impair lymphocyte function in patients with metastasized, castration-resistant prostate cancer. The aim of the current study was to assess in 21 patients whether six cycles of this therapy had an effect on lymphocyte proliferation and interferon-γ and interleukin (IL)-10 ELISpot results. Lymphocyte proliferation after stimulation with microbial antigens and the production of interferon-γ continuously decreased after six cycles of radionuclide therapy, reaching statistical significance (p < 0.05) at months 1, 2, 4, and/or 6 after therapy. One month after the last cycle of therapy, 67% of patients showed a decrease in tumor burden. The tumor burden correlated negatively with IL-10 secretion at baseline, e.g., after stimulation with tetanus antigen (p < 0.0001, r = −0.82). As determined by receiver operating characteristic (ROC) curve analysis, tetanus-specific IL-10 spots at baseline had the highest predictive value (p = 0.005) for tumor burden at month 6, with an area under the curve (AUC) of 0.90 (sensitivity 100%, specificity 78%). In conclusion, we observed an additive effect of treatment with 223Ra on immune function and found that IL-10 secretion at baseline predicted tumor burden at month 6 after treatment.
