A new species of the genus Oreolalax Myers & Leviton, 1962 is described from Sichuan Province, southwest China. Molecular phylogenetic analyses based on mitochondrial gene sequences clustered the new species as an independent clade nested with O. rugosus, O. liangbeiensis, and O. major. The new species could be distinguished from its congeners by a combination of the following characters: body size moderate (39.8–52.8 mm in male); head broad; tympanum absent; interorbital region with dark triangular pattern; 1/3 toes webbed, with broad lateral fringes, belly smooth, brown yellow or medium yellow scattered variable brown spots; skin on dorsum relatively rough with fine tiny and large warts granules; middle pectoral glands are evident in males; flanks with dark-brown warts granules; upper surface of limbs with dark bars; and iris orange above and creamy-white below. The new species inhabits subtropical alpine scrub and swamp.

Abstract Deoxyribonucleotide (DNA) is uniquely programmable and biocompatible, and exhibits unique appeal as a biomaterial as it can be precisely designed and programmed to construct arbitrary shapes. DNA hydrogels are polymer networks comprising cross-linked DNA strands. As DNA hydrogels present programmability, biocompatibility, and stimulus responsiveness, they are extensively explored in the field of biomedicine. In this study, we provide an overview of recent advancements in DNA hydrogel technology. We outline the different design philosophies and methods of DNA hydrogel preparation, discuss its special physicochemical characteristics, and highlight the various uses of DNA hydrogels in biomedical domains, such as drug delivery, biosensing, tissue engineering, and cell culture. Finally, we discuss the current difficulties facing DNA hydrogels and their potential future development. Graphical Abstract

Abstract Background A stent with characteristics of a hybrid design may have advantages in improving the patency of symptomatic iliofemoral vein obstruction. This study assessed the safety and effectiveness of the V-Mixtent Venous Stent in treating symptomatic iliofemoral outflow obstruction. Methods Eligible patients had a Clinical-Etiologic-Anatomic-Physiologic (CEAP) C classification of ≥ 3 or a Venous Clinical Severity Score (VCSS) pain score of ≥ 2. The primary safety endpoint was the rate of major adverse events within 30 days. The primary effectiveness endpoint was the 12-month primary patency rate. Secondary endpoints included changes in VCSS from baseline to 6 and 12 months, alterations in CEAP C classification, Chronic Venous Disease Quality of Life Questionnaire (CIVIQ-14) scores at 12 months, and stent durability measures. Results Between December 2020 and November 2021, 171 patients were enrolled across 15 institutions. A total of 185 endovenous stents were placed, with 91.81% of subjects receiving one stent and 8.19% receiving 2 stents. CO2 geological storage is a guaranteed technology for achieving carbon neutrality. The offshore deposit basin is far away from the human activity area, and the implementation of a CO2 geological storage is more advantageous than the land sedimentary basin. To find out the CO2 storage potential in the Weixinan sag of the Beibu Gulf Basin, based on the 8 indicators such as caprock burial depth, the maximum thickness of single layer mudstone, mud-soil ratio, fault density and reservoir porosity, formation thickness, sand-soil ratio, permeability, etc., it has established a reservoir evaluation system for the secondary structure unit of the basin. Combined with the core physical property data of lays and 3D geological models, the reservoir and caprock of the Weixinan sag are optimized, and the effective storage potential is evaluated with the US-DOE method. The results show that within 800−3000 meters below the sea floor, in the Weixinan sag, there are two sets of reservoir-cap combinations suitable for CO2 storage. Combination 1: in Neogene, the neritic mudstone of the first member of the Jiaowei Formation is the cap rock, and the neritic sandstone of the second member of the Jiaowei Formation and the Xiayang Formation is the reservoir, and the effective storage capacity is 0.749 billion tons. Combination 2: in the Paleogene, the deep lake mudstone in the second member of the Weizhou Formation is the cap rock, and the sandstone in the third member of the Weizhou Formation is the reservoir, and the effective storage capacity is 0.374 billion tons. In the Weixinan sag, two sets of reservoir-cap assemblages suitable for CO2 storage in the saline aquifers are selected, with an effective storage capacity of 1.123 billion tons. According to the evaluation of the annual CO2 emission of 70 million tons in the west of Guangdong Province, it can meet the CO2 emission reduction needs of the region for more than 16 years. Offshore geological storage is a potential path for the green and sustainable development of coastal heavy chemical clustering areas and to achieve the goal of carbon neutrality.

Tissue repair and regeneration have long been a focus of research, with growing evidence highlighting the role played by immunomodulation. The repair of tissue cells can be significantly promoted through the localized immunomodulation of tissues. Hydrogels are increasingly used in this field due to their biocompatibility, versatility, and relative ease of fabrication. This Review highlights the critical role of inflammation and immune development in the complex process of tissue repair after injury, highlighting the dynamic interactions between immune cells that ultimately influence the outcome of tissue healing. We also summarize the precise functions of immune cells and their responses in various tissue injury scenarios, which are critical for harnessing the full potential of immunomodulatory strategies to promote tissue repair. We discuss recent advances in drug-carrying hydrogels, which offer promising avenues for tissue regeneration with precise control of the immune environment for constructive tissue repair. Further advances in our understanding of the mechanisms driving inflammation and immune cell behavior during tissue injury are expected to foster the development of increasingly effective hydrogel-based immunomodulatory strategies to promote tissue repair and regeneration.

Post-translational modifications play crucial roles in regulating protein functions and stabilities. PTEN is a critical tumor suppressor involved in regulating cellular proliferation, survival, and migration processes. However, dysregulation of PTEN is common in various human cancers. PTEN stability and activation/suppression have been extensively studied in the context of tumorigenesis through inhibition of the PI3K/AKT signaling pathway. PTEN undergoes various post-translational modifications, primarily including phosphorylation, acetylation, ubiquitination, SUMOylation, neddylation, and oxidation, which finely tune its activity and stability. Generally, phosphorylation modulates PTEN activity through its lipid phosphatase function, leading to altered power of the signaling pathways. Acetylation influences PTEN protein stability and degradation rate. SUMOylation has been implicated in PTEN localization and interactions with other proteins, affecting its overall function. Neddylation, as a novel modification of PTEN, is a key regulatory mechanism in the loss of tumor suppressor function of PTEN. Although current therapeutic approaches focus primarily on inhibiting PI3 kinase, understanding the post-translational modifications of PTEN could help provide new therapeutic strategies that can restore PTEN’s role in PIP3-dependent tumors. The present review summarizes the major recent developments in the regulation of PTEN protein level and activity. We expect that these insights will contribute to better understanding of this critical tumor suppressor and its potential implications for cancer therapy in the future.

In this paper, we study the fixed-time group consensus problem for second-order multi-agent systems under the influence of actuator faults and external disturbances. First, we design a finite-time observer to quickly and accurately evaluate the effects of these disturbances and faults. Then a new non-singular terminal sliding mode surface is constructed, and a fixed-time fault-tolerant controller is designed based on the introduction of a dynamic event-triggering mechanism. The designed control protocol can effectively avoid singular phenomena and compensate and suppress the effects of these faults and disturbance information on the system to achieve fault tolerance. The Lyapunov function is applied to prove that the convergence time of the system is not influenced by the initial value setting, and the Zeno phenomenon is excluded. Finally, the performance of the proposed method is compared with previous methods through numerical simulations to prove its superiority.

This dataset presents perceived values and socioeconomic indicators collected in Siaya, a rural county in Kenya in 2022. The data was obtained from 300 household surveys and group interviews conducted in six sub-counties across eleven villages. Socioeconomic data were collected with a special focus on climate change vulnerability. Information on housing, health, water accessibility and usage, electricity accessibility and usage, extreme weather events, community service, and information accessibility were mapped across survey questions. The user-perceived value (UPV) game – a perception-based surveying approach – was used to elicit local communities’ needs and perceptions of climate change challenges. The UPV game involves asking interviewees to select which graphically depicted items would be most necessary in different situations and probing them for the reasons behind their choices (why-probing). The data was collected in two languages (Dholuo and English) and then translated into English. These surveys and interviews were conducted to better understand the needs of rural Kenyan communities and their perceptions of climate change, with the aim to identify ways to build resilience. Kenyan policymakers can use the dataset to inform county-level energy and development plans, while researchers and development practitioners can use the dataset to better design their research and programmes to reflect local needs and values.

With the increase in the proportion of photovoltaic and wind power access, the scale and form of distribution networks are becoming more and more complex. The traditional single distribution network vulnerability assessment method is difficult to use to identify the vulnerable links in the distribution network. Therefore, this paper proposes a method for identifying and evaluating vulnerable links in distribution networks based on minimum discriminant information. First, considering the influence of distributed grid connection, an improved probabilistic power flow calculation method is proposed, which improves the calculation efficiency and accuracy. Second, considering the correlation degree, transmission capacity, and voltage stability of branches in the distribution network, the identification index of vulnerable lines is defined. Based on power quality and operating state, the identification index of vulnerable nodes in a distribution network is defined. Finally, based on the indicators of vulnerable nodes and vulnerable lines, the vulnerable links in the distribution network are comprehensively evaluated based on the principle of minimum discriminant information, and the vulnerable links of the entire distribution network are evaluated according to different degrees of vulnerability. The rationality and effectiveness of the proposed method are verified via an example analysis of actual power grid data.

Skeletal muscle satellite cells (SMSCs), a type of myogenic stem cell, play a pivotal role in postnatal muscle regeneration and repair in animals. Circular RNAs (circRNAs) are a distinct class of non-coding RNA molecules capable of regulating muscle development by modulating gene expression, acting as microRNAs, or serving as protein decoys. In this study, we identified circ_14820, an exonic transcript derived from adenosine triphosphatase family protein 2 (ATAD2), through initial RNA-Seq analysis. Importantly, overexpression of circ_14820 markedly enhanced the proliferation of goat SMSCs while concomitantly suppressing their differentiation. Moreover, circ_14820 exhibited predominant localization in the cytoplasm of SMSCs. Subsequent small RNA and mRNA sequencing of circ_14820-overexpressing SMSCs systematically elucidated the molecular regulatory mechanisms associated with circ_14820. Our preliminary findings suggest that the circ_14820-miR-206-CCND2 regulatory axis may govern the development of goat SMSCs. These discoveries contribute to a deeper understanding of circRNA-mediated mechanisms in regulating skeletal muscle development, thereby advancing our knowledge of muscle biology.

Green roof systems are regarded as a viable solution for mitigating urban environmental challenges and offering a multitude of environmental benefits. Currently, green roofs are increasingly being utilized for the management of rainwater runoff and wastewater. The integration of decentralized rainwater and sewage on-site treatment technology with urban green buildings is being gradually promoted. Green roofs can also be considered as a form of decentralized rainwater and sewage on-site technology, which holds great potential for widespread adoption in the future. Several studies have suggested that green roofs may serve as a potential source of pollutants; however, there are also studies that clearly demonstrate the efficient removal of nutrients and organic pollutants by green roofs. This article critically examines the existing literature on water treatment aspects associated with green roofs and elucidates their classification and operational mechanisms. Through an analysis of previous research cases, it becomes evident that both substrate and vegetation play a significant role in influencing the treatment performance of green roofs. By designing and configuring appropriate substrate and vegetation, green roofs can play a pivotal role in the purification of water quality. Finally, a brief outlook is presented for the future research directions of green roofs, with the anticipation that green roofs will feature more innovative and environmentally friendly designs, as well as expanded prospects for application.

Background and aims: Thromboangiitis obliterans (TAO), also known as Buerger’s disease, is a rare vasculitis. Observational epidemiology studies have suggested a relationship between the gut microbiota and TAO. However, due to confounding factors and reverse causality, the causal relationship remains unclear. Based on the assumption of their association, this study sought specific gut microbiota causally linked to TAO. Methods: The case–control study was conducted at the Xiangya Hospital of Central South University from November 2022 to January 2023 including twelve TAO patients and nine healthy controls. We conducted a Mendelian randomization (MR) study using summary statistics from a genome-wide association study (GWAS) of gut microbiota and TAO. Considering the scale and accessibility of the data, the MiBioGen consortium served as the exposure, whereas the FinnGen consortium GWAS study served as the outcome. Finally, we compared the results of the MR with those of the case–control studies. Results: The inverse variance weighted (IVW) (OR = 0.119, 95% CI: 0.021–0.688, p = 0.017) and maximum likelihood (ML) (OR = 0.121, 95% CI: 0.020–0.742, p = 0.022) estimates suggest that Ruminiclostridium 5 has a suggestive protective effect on TAO while the IVW (OR = 5.383, 95% CI: 1.128–25.693, p = 0.035) and ML (OR = 5.658, 95% CI: 1.142–28.021, p = 0.034) estimates suggest that Eubacterium (xylanophilum group) has a suggestive risk effect on TAO, and the ML (OR = 0.055, 95% CI: 0.004–0.755, p = 0.030) estimates suggest that Lachnospira has a suggestive protective effect on TAO. No significant heterogeneity of instrumental variables or horizontal pleiotropy was found. The results of the case–control study showed that the TAO had a lower relative abundance of Ruminiclostridium 5 (p = 0.015) and Lachnospira (p = 0.048), and a higher relative abundance of Eubacterium (xylanophilum group) (p = 0.029) than the healthy controls. These results were consistent with the MR analysis. Conclusions: Our study demonstrates that Ruminiclostridium 5, Lachnospira, and Eubacterium (xylanophilum group) are causally related to TAO, suggesting their potential significance for the prevention and treatment of TAO.

BackgroundRupture of abdominal aortic aneurysm (rAAA) is a fatal event in the elderly. Elevated blood pressure and weakening of vessel wall strength are major risk factors for this devastating event. This present study examined whether the expression profile of mechanosensitive genes correlates with the phenotype and outcome, thus, serving as a biomarker for AAA development.MethodsIn this study, we identified mechanosensitive genes involved in AAA development using general bioinformatics methods and machine learning with six human datasets publicly available from the GEO database. Differentially expressed mechanosensitive genes (DEMGs) in AAAs were identified by differential expression analysis. Molecular biological functions of genes were explored using functional clustering, Protein-protein interaction (PPI), and weighted gene co-expression network analysis (WGCNA). According to the datasets (GSE98278, GSE205071 and GSE165470), the changes of diameter and aortic wall strength of AAA induced by DEMGs were verified by consensus clustering analysis, machine learning models, and statistical analysis. In addition, a model for identifying AAA subtypes was built using machine learning methods.Results38 DEMGs clustered in pathways regulating 'Smooth muscle cell biology' and 'Cell or Tissue connectivity'. By analyzing the GSE205071 and GSE165470 datasets, DEMGs were found to respond to differences in aneurysm diameter and vessel wall strength. Thus, in the merged datasets, we formally created subgroups of AAAs and found differences in immune characteristics between the subgroups. Finally, a model that accurately predicts the AAA subtype that is more likely to rupture was successfully developed.ConclusionWe identified 38 DEMGs that may be involved in AAA. This gene cluster is involved in regulating the maximum vessel diameter, degree of immunoinflammatory infiltration, and strength of the local vessel wall in AAA. The prognostic model we developed can accurately identify the AAA subtypes that tend to rupture.

Fenugreek (Trigonella foenum-graecum L.) is a traditional medicinal plant for treating human diseases that is widely cultivated in many countries. However, the component and related metabolic pathways are still unclear. To understand the changes in expression of the component and related genes during seed development, this study employed metabolomic and transcriptomic analyses and integrative analysis to explore the metabolites and pathways involved in the growth of fenugreek. The antifungal activity of the fenugreek seeds was also analyzed. A total of 9499 metabolites were identified in the positive ion mode, and 8043 metabolites were identified in the negative ion mode. Among them, the main components were fatty acyls, prenol lipids, steroids, steroid derivatives, flavonoids, and isoflavonoids. Among these enriched pathways, the top 20 pathways were “flavone and flavonol biosynthesis”, “isoflavonoid biosynthesis”, and “flavonoid biosynthesis”. 3,7-Di-O-methylquercetin, flavonoids, pseudobaptigenin, isoflavonoids, methylecgonine, alkaloids, and derivatives were the most significantly upregulated metabolites. There were 38,137 differentially expressed genes (DEGs) identified via transcriptomic analysis. According to the KEGG pathway enrichment analysis, 147 DEGs were significantly enriched in “flavonoid biosynthesis”. Ten DEGs of the six key enzymes were found to be involved in three pathways related to flavonoid and alkaloid synthesis in fenugreek. The antifungal activity test revealed the inhibitory effect of the ethanol extract of fenugreek seeds on Alternaria tenuissima (Kunze)Wiltshire and Magnaporthe oryzae. These findings further prove that the use of botanical pesticides in fenugreek fruit has research value.

In this article, an observer-based adaptive non-singular fast-reaching terminal sliding mode control strategy is proposed to tackle the problem of actuator faults and uncertain disturbance in aerial robot systems. Firstly, a model of an aerial robot system is established through dynamic analysis. Next, an adaptive observer, combined with a fast adaptive fault estimation (FAFE) algorithm, is proposed to estimate system states and actuator failure and compensate for faults in a precise and prompt manner. In addition, a non-singular fast terminal sliding surface is defined, taking into account the fast convergence of the tracking errors in order to provide appropriate trajectory tracking results. Since the upper bounds of the disturbances caused by the manipulator of the system in practice are unknown, the control approach may benefit from the addition of an adaptive control strategy that can suppress the influence of uncertain disturbances. The Lyapunov stability theory demonstrates that tracking errors are able to converge stably and quickly. In the end, the contrast experiment is conducted to exhibit the effectiveness of the proposed control strategy. The results demonstrate quicker convergence and improved estimating accuracy.

Carbon dioxide removal (CDR) is the essential ‘net’ in net zero. However, a thriving CDR industry will not come into being without government intervention. As governments start to devise CDR support policies, this paper solicits the views of market participants in two of the most prominent CDR methods: bioenergy with carbon capture and storage (BECCS) and direct air carbon capture and storage (DACCS). We survey 47 BECCS and DACCS project developers and financiers active in Europe, conducting in-depth interviews with 27 of them to identify their key challenges and preferred policy interventions to address them. We find that participants prefer compliance markets, such as links to emissions trading systems, to generate demand but seek government support to cushion early market risks. They acknowledge the need for stringent monitoring and regulation to ensure environmental integrity. Bearing industry expectations in mind, policymakers face five key challenges in developing CDR: reaching scale, striking a balance with emissions cuts, safeguarding integrity, ensuring fairness and accelerating the speed of deployment.

Phycocyanin 620 (PC620) is the outermost light-harvesting complex in phycobilisome of cyanobacteria, engaged in light collection and energy transfer to the core antenna, allophycocyanin. Recently, long-lived exciton–vibrational coherences have been observed in allophycocyanin, accounting for the coherent energy transfer [Zhu et al., Nat. Commun. 15, 3171 (2024)]. PC620 has a nearly identical spatial location of three α84–β84 phycocyanobilin pigment pairs to those in allophycocyanin, inferring an existence of possible coherent energy transfer pathways. However, whether PC620 undergoes coherent or incoherent energy transfer remains debated. Furthermore, accurate determination of energy transfer rates in PC620 is still necessary owing to the spectral overlap and broadening in conventional time-resolved spectroscopic measurements. In this work, the energy transfer process within PC620 was directly resolved by polarization-controlled two dimensional electronic spectroscopy (2DES) and global analysis. The results show that the energy transfer from α84 to the adjacent β84 has a lifetime constant of 400 fs, from β155 to β84 of 6–8 ps, and from β155 to α84 of 66 ps, fully conforming to the Förster resonance energy transfer mechanism. The circular dichroism spectrum also reveals that the α84–β84 pigment pair does not form excitonic dimer, and the observed oscillatory signals are confirmed to be vibrational coherence, excluding the exciton–vibrational coupling. Nodal line slope analysis of 2DES further reveals that all the vibrational modes participate in the energy dissipation of the excited states. Our results consolidate that the ultrafast energy transfer process in PC620 is incoherent, where the twisted conformation of α84 is suggested as the main cause for preventing the formation of α84–β84 excitonic dimer in contrast to allophycocyanin. Background: Lung cancer (LC) is currently the number one malignancy death rate disease in China, and its disease burden is serious. The study aimed to analyze trends of LC and its risk factor attributable disease in China from 1990 to 2019 and predict the next 41 years. Methods: The average annual percentage change (AAPC) was used to analyze the trend of LC and its risk factor attributable incidence, deaths, and disability‐adjusted life years (DALYs) rate in China from 1990 to 2019, collected in the Global Burden of Disease 2019. Cochran–Armitage trends examine trends in lung cancer disease burden by sex, age, and attributable risk factor groups in China from 1990 to 2019. In addition, based on data on death and DALYs rate due to LC and its risk factors between 1990 and 2019, an autoregressive integrated moving average (ARIMA) model was developed to predict the change in the trend of burden of disease due to LC and its risk factors over the next 41 years, and the model was evaluated using the model parameters root mean square error, mean absolute error, and mean absolute percentage error. Results: From 1990 to 2019, the incidence, mortality and DALYs of LC were all increased. Among the eight risk factors associated with lung cancer, the DALYs rate and mortality rate of lung cancer risk factors for Chinese residents increased from 1990 to 2019, except for household air pollution from solid fuels and diet low in fruit, which showed a decrease; among them, the DALYs rate and mortality rate due to ambient particulate matter pollution showed the greatest increase with AAPC values of 2.880 and 3.310, respectively, while DALYs and mortality rates due to household air pollution from solid fuels showed the largest decreases, with AAPC values of −4.755 and −4.348, respectively. The results of the ARIMA model predictions show that both the mortality rate and the rate of DALYs for lung cancer are increasing yearly, and it is predicted that the rate of DALYs for lung cancer by 2060 will reach 740.095/100 000 and the mortality rate will reach 35.151/100 000. It is expected that by 2060, the top four risk factors for lung cancer in China will be, in order of DALYs rate and mortality rate, smoking, ambient particulate matter pollution, high fasting plasma glucose (HFPG), and secondhand smoke, with HFPG showing the greatest increase. Conclusions: The LC burden increased from 1990 to 2019 in China, the LC burden that could be attributed to HFPG will continue to increase in the next 40 years, and will be the third most factor by 2060. Effective tiller number (ETN) directly affects panicle number and is closely related to wheat grain yield. Mining quantitative trait loci (QTLs) associated with wheat tiller number and analyzing the correlation between tiller number and other important agronomic traits can provide the theoretical basis for molecular breeding. In this study, we first proposed and established a novel genetic analysis framework of "multiple environmental assessments-depth analysis of individual traits-comprehensive evaluation of various traits-friendly marker development-verification in different backgrounds". Using this approach, low-tillering QTLs were identified and validated base on an F6 recombinant inbred line population (MC population) derived from the low-tillering plant msf and Chuannong 16 (CN16), phenotypic data of effective tillers from multiple environments, and a 16K chip-based constructed genetic linkage map. QTL mapping results showed that there were four QTLs controlling tillering on chromosomes 1A, 5A, and 6D, respectively. Qltn.sau-MC-1A was a stable and major low-tillering QTL explaining 13.39%--60.40% of the phenotypic variation rate, and its positive allele was from msf. Phenotypic analysis showed that ETN of the lines carrying the positive allele of Qltn.sau-MC-1A was significantly less than those with the negative alleles. Correlation analysis showed that ETN had a significantly positive correlation with plant height (PH), and a significantly negative correlation with thousand kernel weight (TKW), kernel number per spike (KNPS), kernel weight per spike (KWPS), and flag leaf width (FLW), but no significantly correlation with flag leaf length (FLL) and anthesis date (AD). Genetic analysis showed that positive allele of Qltn.sau-MC-1A had a significant effect on increasing KNPS, KWPS, and TKW, but delaying AD. Validation results in different backgrounds suggested that the ETN of lines carrying positive alleles from Qltn.sau-MC-1A could be significantly decreased. Collectively, we established a new genetic mapping approach and further used it to identify and validate a major QTL controlling low-tiller number, Qltn.sau-MC-1A, which laid a foundation for further fine mapping and understanding the mechanism of tiller formation. Immunosenescence contributes to systematic aging and plays a role in the pathogenesis of Alzheimer's disease (AD). Therefore, the objective of this study was to investigate the potential of immune rejuvenation as a therapeutic strategy for AD. To achieve this, the immune systems of aged APP/PS1 mice were rejuvenated through young bone marrow transplantation (BMT). Single-cell RNA sequencing revealed that young BMT restored the expression of aging-and AD-related genes in multiple cell types within blood immune cells. The level of circulating senescence-associated secretory phenotype proteins was decreased following young BMT. Notably, young BMT resulted in a significant reduction in cerebral Aß plaque burden, neuronal degeneration, neuroinflammation, and improvement of behavioral deficits in aged APP/PS1 mice. The ameliorated cerebral amyloidosis was associated with an enhanced Aß clearance of peripheral monocytes. Geoherb usually represents high-quality medicinal herbs with better clinical therapeutic effects, and elucidating the geoherbalism is essential for the quality improvement of traditional Chinese Medicine. However, few researches were conducted to clarify the geoherbalism based on a large scale of transcriptomics. In the present study, we compared the transcriptomes of Rheum palmatum complex derived from top-geoherb and non-geoherb areas to show the geoherbalism properties of rhubarb. A total of 412.32 Gb clean reads were obtained with unigene numbers of 100,615 after assembly. Based on the obtained transcriptome datasets, key enzyme-encoding genes involved in the anthraquinones biosynthesis were also obtained. We also found that 21 anthraquinone-related unigenes were differentially expressed between two different groups, and some of these DEGs were correlated to the content accumulation of five free anthraquinones, indicating that the gene expression profiles may promote the geoherbalism formation of rhubarb. In addition, the selective pressure analyses indicated that most paired orthologous genes between these two groups were subject to negative selection, and only a low proportion of orthologs under positive selection were detected. Functional annotation analyses indicated that these positive-selected genes related to the functions such as gene expression, substance transport, stress response and metabolism, indicating that discrepant environment also enhanced the formation of geoherbalism. Our study not only provided insights for the genetic mechanism of geoherbalism of rhubarb, but also laid more genetic cues for the future rhubarb germplasms improvement and utilization.
