To explore the possible novel microRNA (miRNA) regulatory pathways in Zhengmai 1860, a newly cultivated drought-tolerant wheat (Triticum aestivum L.) cultivar, miRNA transcriptome sequencing of the flag leaves of Zhengmai 1860, drought-sensitive variety Zhoumai 18, and drought-resistant variety Bainong 207 was performed during the grain filling stage. We also observed changes in the chloroplast ultrastructure, phytohormone levels, and antioxidant- and photosynthesis-related physiological indicators in three wheat varieties. The results showed that the flag leaves of the drought-tolerant variety Zhengmai 1860 had higher chlorophyll contents and net photosynthetic rates than those of Zhoumai 18 under drought stress during the grain filling stage; in addition, the chloroplast structure was more complete. However, there was no significant difference between Zhengmai 1860 and Bainong 207. MiRNA transcriptome analysis revealed that the differential expression of the miRNAs and mRNAs exhibited variable specificity. The KEGG pathway enrichment results indicated that most of the genes were enriched in the MAPK signaling pathway, plant hormone signal transduction, photosynthetic antennae protein, and amino acid and carbohydrate metabolism. In the drought-tolerant cultivar Zhengmai 1860, tae-miR408 was targeted to regulate the allene oxide synthase (AOS) gene, inhibit its expression, reduce the AOS content, and decrease the synthesis of jasmonic acid (JA) and abscisic acid (ABA). The results of this study suggest that Zhengmai 1860 could improve the photosynthetic performance of flag leaves by inhibiting the expression of genes involved in the JA pathway through miRNAs under drought conditions. Moreover, multiple miRNAs may target chlorophyll, antioxidant enzymes, phytohormone signal transduction, and other related pathways; thus, it is possible to provide a more theoretical basis for wheat molecular breeding. Background: Nitrogen use efficiency (NUE) is closely related to crop yield and nitrogen fertilizer application rate. Although NUE is susceptible to environments, quantitative trait nucleotides (QTNs) for NUE in wheat germplasm populations have been rarely reported in genome-wide associated study. Results: In this study, 244 wheat accessions were phenotyped by three NUE-related traits in three environments and genotyped by 203,224 SNPs. All the phenotypes for each trait were used to associate with all the genotypes of these SNP markers for identifying QTNs and QTN-by-environment interactions via 3VmrMLM. Among 279 QTNs and one QTN-by-environment interaction for low nitrogen tolerance, 33 were stably identified, especially, one large QTN (r2 > 10%), qPHR3A.2, was newly identified for plant height ratio in one environment and multi-environment joint analysis. Among 52 genes around qPHR3A.2, four genes (TraesCS3A01G101900, TraesCS3A01G102200, TraesCS3A01G104100, and TraesCS3A01G105400) were found to be differentially expressed in low-nitrogen-tolerant wheat genotypes, while TaCLH2 (TraesCS3A01G101900) was putatively involved in porphyrin metabolism in KEGG enrichment analyses. • The effect of size on molecular dynamics simulation results are considered. • Tensile and shear properties of amorphous silicon nitride with different densities calculated using molecular dynamics simulations. • The empirical equation between mechanical properties (Young's modulus, shear modulus, tensile strength, and shear strength) and density of amorphous silicon nitride is established. Silicon nitride fiber is among the most popular candidates for high-temperature wave-transparent ceramic matrix composites (CMC) in aerospace engineering, and the measurement of its mechanical properties has always been the focus of researchers. In this paper, a simple and effective molecular dynamics method is used to generate molecular dynamics models of amorphous silicon nitride (a -Si 3 N 4) materials with different densities, and it is verified that the developed models reflect the microstructure of the fiber. Based on analyzing the effects of size on the results, tensile and shear simulations are carried out to obtain the mechanical properties of the models with different densities. According to the research data obtained, the empirical equation between mechanical properties (Young's modulus, shear modulus, tensile strength, and shear strength) and density of fiber is established, and validated by the published experimental results. The empirical equation can not only predict the mechanical properties of fibers directly from the density but also provide a new way to measure the mechanical properties of fibers nondestructively. Oxygen plays a crucial role in the synthesis of boron nitride (BN) and the preparation of BN ceramics. The effects of oxygen content on the microstructure and properties of BN fibers treated at 1600 °C have not been studied yet. In this work, polymer-derived BN fibers are cured at 300 °C under oxygen and pyrolyzed at 600 °C, 700 °C and 800 °C under ammonia as the pre-heated BN fibers. Then, these pre-heated BN fibers are treated at 1600 °C under nitrogen as the final BN fibers. The compositions and microstructures of the pre-heated and final BN fibers are investigated. As the pre-heated temperature increases, the oxygen contents of the pre-heated BN fibers decrease to 24 wt%, 17 wt% and 5 wt%, respectively. The structures of the pre-heated BN fibers transform from amorphous BN to turbostratic BN. The oxygen in the pre-heated BN fiber volatilizes as boron oxide during treatment at 1600 °C, promoting the formation and growth of h-BN in the final BN fiber, with the maximum grain size reaching 74 × 20 nm (La × Lc). As the grain size of the final BN fiber increases, the Young's modulus, density and oxidation resistance level of the BN fiber improve, and the dielectric properties decrease. The final BN fiber that underwent pre-heating at 700 °C has the best comprehensive properties, with a tensile strength of 1.11 GPa and a Young's modulus of 265 GPa. This study researches the relationship between composition, structure and performance, and provides a scientific basis for the preparation process of BN fibers. • The initial BN fibers with gradient oxygen content are obtained by controlling the nitridation temperature after air curing. • The higher the oxygen content in the initial BN fibers, the better the crystallinity of the BN fibers treating at 1600 °C. • The tensile strength of BN fiber with a grain size of 34 × 11 nm (La × Lc) is 1.11 GPa, and the Young's modulus is 265 GPa.
