This paper presents the uncertainty quantification and sensitivity analysis of the standard k-ɛ turbulence model applied to the numerical prediction of the non-cavitating and cavitating flow around a 2D NACA66MOD hydrofoil. The turbulence model parameters are treated as epistemic uncertain variables, and the forward propagation of uncertainty is evaluated using the non-intrusive polynomial chaos approach. The required simulations are performed using a commercial CFD solver. The Sobol indices are used to rank the relative contribution of closure coefficients to the total uncertainty in the output quantities. For the considered case, the ranking of the model parameters is not influenced by the presence of the sheet cavity flow, while it varies with respect to the considered output quantity.

Several neutrino detectors, KamLAND, Daya Bay, Double Chooz, RENO, and the forthcoming large-scale JUNO, rely on liquid scintillator to detect reactor antineutrino interactions. In this context, inverse beta decay represents the golden channel for antineutrino detection, providing a pair of correlated events, thus a strong experimental signature to distinguish the signal from a variety of backgrounds. However, given the low cross-section of antineutrino interactions, the development of a powerful event selection algorithm becomes imperative to achieve effective discrimination between signal and backgrounds. In this study, we introduce a machine learning (ML) model to achieve this goal: a fully connected neural network as a powerful signal-background discriminator for a large liquid scintillator detector. We demonstrate, using the JUNO detector as an example, that, despite the already high efficiency of a cut-based approach, the presented ML model can further improve the overall event selection efficiency. Moreover, it allows for the retention of signal events at the detector edges that would otherwise be rejected because of the overwhelming amount of background events in that region. We also present the first interpretable analysis of the ML approach for event selection in reactor neutrino experiments. This method provides insights into the decision-making process of the model and offers valuable information for improving and updating traditional event selection approaches.

Amniotic fluid is a complex and dynamic biological matrix that surrounds the fetus during the pregnancy. From this fluid, is possible to isolate various cell types with particular interest directed towards stem cells (AF-SCs). These cells are highly appealing due to their numerous potential applications in the field of regenerative medicine for tissues and organs as well as for treating conditions such as traumatic or ischemic injuries to the nervous system, myocardial infarction, or cancer. AF-SCs, when subcultured in the presence of basic Fibroblast Growth Factor (bFGF), have been shown to survive and migrate when transplanted into the striatum of the rat brain, exhibiting behavior characteristics of neuronal/glial progenitor cells. In this work, we performed an electrophysiological characterization to ascertain the propensity of AF-SCs to differentiate into glial and neuronal cells by bFGF. By using patch clamp technique we characterized a fibroblast-like morphology that display a barium-sensitive inward-rectifying potassium current (Kir) and calcium-activated potassium currents (KCa). The electrophysiological and calcium dynamics of histamine, a marker of undifferentiated neural progenitors, was further studied. Histamine promoted intracellular calcium increase by Fura-2 recording and calcium-activated potassium current activation with a similar temporal profile in AF-SC. The data presented in this paper ultimately confirm the expression in AF-SCs of the Kir and KCa currents, also showing regulation by endogenous stimuli such as histamine for the latter.

Alcohol is responsible for an ever-increasing number of deaths worldwide, and many road accidents are caused by irresponsible drinking and driving. The use of biomarkers that can support a diagnosis of alcohol abuse is a very important tool that can improve the prevention of many alcohol-related diseases and serious traffic accidents. The main aim of our study was the full validation of a rapid and simple method by ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) to detect ethyl glucuronide in hair (hEtG). The method was successfully applied to n = 171 real hair samples collected from drivers convicted of driving while impaired by alcohol or drugs. A comparison of hEtG and serum Carbohydrate-Deficient Transferrin percentages (% CDT) was also performed to carefully evaluate the data in relation to the specific detection windows of the two different biomarkers. Most of the drivers with hEtG > 30 pg/mg were males in their thirties. None of the hEtG-positives had a serum % CDT above the cutoff (≥2%). Although some researchers suggest caution until solid data are available on the possible effects of interindividual variability that may influence EtG incorporation and metabolism, hEtG is a very useful biomarker of long-term alcohol exposure that shows greater reliability than traditional blood markers. Circadian clocks are biological oscillators that evolved to coordinate rhythms in behaviour and physiology around the 24-hour day. In mammalian tissues, circadian rhythms and metabolism are highly intertwined. The clock machinery controls rhythmic levels of circulating hormones and metabolites, as well as rate-limiting enzymes catalysing biosynthesis or degradation of macromolecules in metabolic tissues, such control being exerted both at the transcriptional and post-transcriptional level. During infections, major metabolic adaptation occurs in mammalian hosts, at the level of both the single immune cell and the whole organism. Under these circumstances, the rhythmic metabolic needs of the host intersect with those of two other players: the pathogen and the microbiota. These three components cooperate or compete to meet their own metabolic demands across the 24 hours. Here, we review findings describing the circadian regulation of the host response to infection, the circadian metabolic adaptations occurring during host–microbiota–pathogen interactions and how such regulation can influence the immune response of the host and, ultimately, its own survival. The purpose of this study was to evaluate the efficacy of the construction of mental images as a cognitive strategy to improve comprehension of expository texts in elementary school children. Since these texts require greater cognitive effort, we investigated whether the creation of mental images facilitates the reading comprehension process. A quantitative approach with a quasi-experimental design and a hypothetico-deductive method was used. Fifty-four elementary school students participated (mean age = 9.24, SD = 0.43; 31 boys and 23 girls), distributed in two groups: an experimental group, which applied the strategy during eight 90-minute sessions, and a control group, which received conventional instructions. The results indicated that the experimental group showed a significant improvement in the comprehension of expository texts, with 63% of the students reaching a satisfactory level of achievement, while the control group did not present relevant advances. In conclusion, the strategy of constructing mental images proved to be effective in the development of reading comprehension skills, improving the literal, inferential, and critical levels. The practical implications indicate that implementing this strategy as a key pedagogical tool could positively impact reading comprehension in primary education. [ABSTRACT FROM AUTHOR]

This study aims to analyze and describe the various forms of collaborative work that teachers and university students in the province of Cusco accomplish together using the descriptive methodology. All professionals must know their area of work as well as lead in all work environments. The participation was anonymous and voluntary. The participating students are from the first cycle to the third cycle of the different professional careers between males and females from the rural and urban areas of the province of Cusco, Peru. The findings indicate that university teachers play an effective and significant pedagogical role in designing the collaborative work model that assigns group work in their respective classrooms with the various careers they supervise. Additionally, the findings show that the teacher fulfils the social role of motivating students by performing as a tutor in the classroom. Therefore, it has a positive and significant effect among university students. This has implied that the formation of working groups in university students generates student leaders and demonstrates broad knowledge in communication and information technologies which will generate many expectations in their professional training. Brain abscesses are invasive infections of the central nervous system with a high level of treatment complexity especially in pediatric patients. Here, we describe a 3-month-old infant with multiple brain abscesses caused by methicillin-susceptible Staphylococcus aureus (MSSA). The patient was initially treated with empirical antibiotics (ceftriaxone, metronidazole, vancomycin). Upon MSSA identification, therapy was optimized by switching vancomycin to linezolid to improve tissue penetration. Therapeutic drug monitoring (TDM) was performed to check linezolid levels in the plasma and pus of the abscess, confirming drug penetration into brain tissue. A two-stage surgical drainage approach, consisting of repeated pus aspiration through an intracystic catheter, was then performed to achieve a significant reduction in abscess size. After nine weeks of antibiotic therapy, the patient was discharged in good clinical condition. This case highlights the role of linezolid for the treatment of complicated CNS infections and the importance of a multidisciplinary approach, combining TDM-based antibiotic therapy with timely and eventually repeated surgery, in order to effectively treat brain abscesses. Macroautophagy (often-named autophagy), a catabolic process involving autophagy-related (Atg) genes, prevents the accumulation of harmful cytoplasmic components and mobilizes energy reserves in long-lived and self-renewing cells. Autophagy deficiency affects antigen presentation in conventional dendritic cells (DCs) without impacting their survival. However, previous studies did not address epidermal Langerhans cells (LCs). Here, we demonstrate that deletion of either Atg5 or Atg7 in LCs leads to their gradual depletion. ATG5-deficient LCs showed metabolic dysregulation and accumulated neutral lipids. Despite increased mitochondrial respiratory capacity, they were unable to process lipids, eventually leading them to ferroptosis. Finally, metabolically impaired LCs upregulated proinflammatory transcripts and showed decreased expression of neuronal interaction receptors. Altogether, autophagy represents a critical regulator of lipid storage and metabolism in LCs, allowing their maintenance in the epidermis. [ABSTRACT FROM AUTHOR]

Mouse models are vital tools for discerning the relative contributions of host and microbial genetics to disease, often requiring the transplantation of microbiota between different mouse strains. Transfer methods include antibiotic treatment of recipients and colonization using either co-housing with donors or the transplantation of fecal or cecal donor material. However, the efficiency and dynamics of these methods in reconstituting recipients with donor microbes is not well understood. We thus directly compared co-housing, fecal transplantation, and cecal transplantation methods. Donor mice from Taconic Biosciences, possessing distinct microbial communities, served as the microbial source for recipient mice from Jackson Laboratories, which were treated with antibiotics to disrupt their native microbiota. We monitored microbial populations longitudinally over the course of antibiotics treatment and reconstitution using 16S rRNA gene sequencing, quantitative PCR, and shotgun sequencing of viral-like particles. As expected, antibiotic treatment rapidly depleted microbial biomass and diversity, with slow and incomplete natural recovery of the microbiota in non-transplanted control mice. While all transfer methods reconstituted recipient mice with donor microbiota, co-housing achieved this more rapidly for both bacterial and viral communities.
